Investigation about enzymatic properties of the gene encoding catechol 1,2-dioxygenase from Phenol-degrading, Rhodococcus sp. EL-GT

  • 본 연구는 방향족 화합물질 중 페놀폐수에 대한 생물학적 처리를 위해 본 실험실에서 분리한 페놀분해능이 우수한 Rhodococcus sp. EL-GT를 이용하여 catechol 분해 catechol 1,2-dioxygenase 분해활성을 측정하였고, 이것이 ortho-pathway임을 확인할 수 있었다. 또한 다른 연구에서 보고된 Rhodococrus rhodochrous NCIMB 13259 균주의 catechol 1,2 dioxygenase를 기초로한 primer를 이용하여 PCR을 수행하였으며 이 분해 유전자의 cloning실험을 수행 중이다. 이들 실험을 통하여 Rhodoroccus sp. EL-GT의 페놀분해 균의 유전적 구조 및 특성을 검토하고 이를 이용하여 방향족 화합물의 분해능이 보다 우수한 균주의 개발을 시도하고자 한다.

  • Biological efficacy assays, including antimicrobial, tyrosinase inhibition and anticancer activities of Chlorella hydrolysate, were carried out. In tyrosinase inhibition assay of Chlorella hydrolysate, $IC_{50}$(inhibitory concentration) was measured as 12%. Chlorella hydrolysate showed high anti-lung cancer activity of 88.2% at a concentration of 0.15%.

  • 중수숭급기 충진재의 부식원인인 염분 유입 및 열분해로 인한 산화물 형성 과정 조사를 위해 zeolite 의 열분해 및 염분 유입을 조사하였다. 그리고 원자력 발소의 dryer 모사 장치를 만들어 열분해 특성 조사 실험을 하였다. zeolite 열분해 특성 조사 결과 zeolite에 고온의 증기를 통과시키면 가수분해가 일어나 구조가 바뀌게 되며, $500\~700^{\circ}C$에서 완전히 열분해 된다. 그리고 열분해가 일어난 zeolite에 염분이 유입되면, zeolite의 $H^+$와 염분의 $Na^+$ 가 자리바꿈을 하게 되고, 수중에 $H^+$$Cl^-$ 가 존재하여 HCl이 형성될 것으로 조사되었고, 실험 결과 pH가 4.5, 4.53, 4.72, 4.64, 4.86, 5.03, 4.61로 낮게 측정되었다.

Investigation of Physicochemical Properties of Bio-oils Produced from Pitch Pine (Pinus rigida) at Various Temperatures (열분해 온도에 따른 리기다소나무 바이오오일의 물리·화학적 특성 평가)

  • In this study, fast pyrolysis of pitch pine (Pinus rigida) was performed in a fluidized bed reactor under the temperature ranges between 400 and $550^{\circ}C$ at the residence time of 1.9 sec. Essential pyrolytic products (bio-oil, biochar, and gas) were produced and their yield was clearly influenced by temperature. The maximum yield of bio-oil was observed to 64.9 wt% (wet basis) at the temperature of $500^{\circ}C$. As pyrolysis temperature increased, the yield of biochar decreased from 36.8 to 11.1 wt%, while gas amount continuously increased from 16.1 to 33.0 wt%. Water content as well as heating value of bio-oils were obviously sensitive to the pyrolysis temperature. The water contents in the bio-oil clearly decreased from 26.1 ($400^{\circ}C$) to 11.9 wt% ($550^{\circ}C$), with increasing the fast pyrolysis temperature, while their higher heating values were increased from 16.6 MJ/kg to 19.3 MJ/kg. According to GC/MS analysis, 22 degradation compounds were identified from the bio-oils and 10 compounds were derived from carbohydrate, 12 compounds were derived from lignin.

Physico-chemical stability of pesticide formulations under different storage conditions (저장조건별 농약제품의 물리.화학적 안정성)

  • To confirm physico-chemical stability of formulated products, this study was conducted under different storage conditions; room temperature and accelerated temperature of $54^{\circ}C$. The tested pesticide formulations were dichlorvos 50% EC, acephate 50% WP, hymexazol 4% DP, thiram 80% WP and isoprothiolane 12% GR. The selected formulations were stored for 10 weeks and 5 years under the given temperature in maximum and contents of active ingredients were also analyzed by GLC or HPLC after each time of storage. The degradation rates of 5 active ingredients under the two conditions showed a similar trend except acephate. Acephate was rapidly decomposed at $54^{\circ}C$ but slowly decomposed at room temperature, and the degradation rate under the accelerated condition was 2.4 to 5-fold higher than that under the room temperature. Consequently, the stability test on active ingredients in pesticide formulations was able to recommend to be carried out under the accelerated condition except acephate. And the physical properties of all formulations tested under two storage conditions were good.

  • In this study, we evaluated optimal conditions for ethanol production of the spruce hydrolysate (SH) obtained from diethyl oxalate pretreatment. Fermentable sugar concentration in SH was 29.04 g/${\ell}$ except arabinose. Monosaccharides obtained from the oligomer degradation were mainly mannose (39.26 g/${\ell}$) and galactose (12.83 g/${\ell}$). Concentration of 5-HMF and furfural which are inhibitors on ethanol fermentation were 0.09 g/${\ell}$ and 0.04 g/${\ell}$ respectively. Concentration of acetic acid and total phenolic compounds in SH were 1.4 g/${\ell}$ and 2.83 g/${\ell}$. Ethanol production using hydrolysate was 11.7 g/${\ell}$ at optimal pH 6.0 after 48 h. Specific ethanol production was 0.15 (g/(${\ell}^*h$)) at pH 5.0 and 5.5. while that was 0.24 (g/(${\ell}^*h$)) at pH 6.0. Specific ethanol production has difference depend on initial pH for fermentation. Ethanol production was 14.3 g/${\ell}$ after 48 h when xylanase 20 IU was added in SH for degradation of oligomer during fermentation. It implied that ethanol production increased by 22.2% compare with control (without xylanase).

Correlation between Glycemic Index and in vitro Starch Hydrolysis of Cereals (곡류의 혈당지수와 전분 가수분해율과의 상관관계)

  • To see the correlation between the rate of in vitro starch hydrolysis and the glycemic index, an in vitro digestion was carried out by incubating the cereal samples for 2 hours with ${\alpha}-amylase$ in dialysis tubing. Also the levels of blood glucose were measured over 2 hours after feeding healthy volunteers with 50 g carbohydrate portions. Hydrolysis area, hydrolysis index (HI) and the dialysate content of carbohydrate throughout the digestion time for barley was significantly below those for other cereals (p<0.05), and unpolished glutinous rice was significantly above (p<0.05). The GI-glucose of barley $(57%{\pm}7)$ to glucose as standard was significantly (p<0.05) lower than those of other cereals whereas the GI-glucose of glutinous rice $(110%{\pm}8)$ was significantly higher (p<0.05) than other cereals. The GI-rice values to rice as standard were $122%{\pm}4$ for glutinous sorghum, $116%{\pm}13$ for job's tear, $115%{\pm}13$ for glutinous millet, $106%{\pm}6$ for unpolished glutinous rice, $102%{\pm}7$ for glutinous rice, $100%{\pm}0$ for rice, $90%{\pm}12$ for unpolished rice, $85%{\pm}6$ for foxtail millet, $79%{\pm}5$ for buckwheat and $63%{\pm}6$ for barley. The GI-rice was significantly correlated to hydrolysis area and HI (r=0.75, p<0.01). It suggests that the in vitro starch hydrolysis offers good potential to predict the in vivo glycemic response of starch foods.

Non-isothermal TGA Analysis on Thermal Degradation Kinetics of Modified-NR Rubber Composites (비등온 TGA에 의한 개질NR고무복합재료지 열분해 Kinetics에 관한 해석)

  • Thermal degradation behavior of CR (chloroprene) -modified NR (natural rubber) compounds, having different sulfur/accelerator compositions, was studied by non-isothermal TGA method. Data were analyzed using both Kissinger and Flynn-Wall-Ozawa analysis to assess the activation energies. Activation energy obtained from Kissinger analysis was $147.0{\pm}2.0$ kJ/mol for all samples, showing little effect of sulfur/accelerator composition changes in the samples. On the other hand, activation energy from Flynn-Wall-Ozawa analysis exhibited much variations with conversion, showing average value of $211.6{\pm}19.0$ kJ/mol. From the results, it was considered that whole thermal degradation processes of the samples were composed of complex multiple step processes, of which reaction mechanisms were different from each other.

PAHs Degrading Bacterium Separation and Identification for Biological Treatment (PAHs의 생물학적 처리를 위한 분해 미생물 분리 동정)

  • Pseudomonas sp. KM1 was separated from soil contaminated by petroleum and identified. The isolated strain is Gram-positive, rod-shaped and immotile. In batch culture, the optimum cultivation temperature and pH was $35^{\circ}C$ and 7, respectively. Biodegradation of PAHs experiment with soil slurry system was performed using Pseudomonas sp. KM1. Pseudomonas sp. KM1 could degrade 7 PAHs including naphthalene, acenaphthylene, acenaphthene, fluorene, phenanthrene, pyrene, and fluoranthene. These mixed PAHs was easily degraded within one day except fluoranthene, which was degraded much slowly, taking several days by this isolated bacteria. Pseudomonas sp. KM1 is good candidate for bioremediation of PAHs contaminated soils. Biodegradation rates of naphthalene, phenanthrene and pyrene in soils were different at each soil, and the rates were decreased as sorption capacity increased.

  • The development of protein utilization with keratin(cattle hair and human hair) have heen investigated. The include a) conditioning of hydrolysis of keratin, b) isolation of crude protein, c) pepsin HCl digestibility of isolated protein, d) the amino acid composition of isolated protein and amino acid concentrate. The results are obtained as follow ; 1. Human hair and cattle hair were softened and hydrolysed with hot dilute alkali(especially 1% NaOH) 2. Crude protein were isolated from hydrolysed keratin by means of different solubility and isoelectric point. 3. The protein isolated was of good quality on the point of color, protein content and pepsin digestibility. 4. The amino acid concentrate obtained by acids processing was of good quality on the point of color, taste and amino acid composition.
