  • Trans-resveratrol, quercetin, and epicatechin are natural polyphenolic substances with a wide range of biological activities, including anti-oxidant and anti-cancer properties. The compositions and levels of these chemicals were determined in grape (Vitis vinifera) juice and wine of different geographic origins. Significant differences in Hunter color values, pH, total acidity, and ethanol and free sugar contents, were found depending on both the nature and country of processing. Overall, trans-resveratrol and quercetin levels in wines were higher than in juices, indicating that wine is a rich source of these chemicals. Significant differences in the content and composition of epicatechin were found to depend on processing procedures, country of origin, and cultivar. This information may be useful to grape breeders and manufacturers of wine or juice.

  • Resveratrol, a phytoalexin found in grapes, berries, and peanuts, is one of the most promising agents for cancer prevention. Recent studies show that the antitumor activity of resveratrol occurs through p53-mediated apoptosis. This study demonstrated the mechanism that resveratrol induced apoptosis in human osteosarcoma Saos-2 cells lacking p53. Treatment of osteosarcoma Saos-2 cells with resveratrol resulted in decrease of cell viability, which was revealed as apoptosis characterized by activation of caspase-3 protease as well as cleavage of poly(ADP-ribose) polymerase (PARP) with change of mitochondrial membrane potential transition. These results suggest that resveratrol may be potentially useful to treat osteosarcoma via activation of caspase protease and mitochondrial dysfunction.

  • The ethanol solvent extracts obtained from the pulp, skin, seeds, leaves, fruit stems, and pruning stems of four Korean new grape varieties ("Dunoori", "Narsha", "Cheongsoo", and "Heukbosek"), as well as "Campbell Early", and "Muscat Bailey A (MBA)" were evaluated for their total phenolic and anthocyanin contents. The concentrations of four phenols of biological interest, catechin, epicatechin, quercetin, and resveratrol in the different parts were quantified by high-performance liquid chromatography-ultraviolet analyses. The skin of "Narsha" and "Heukbosek", the leaves of "Narsha", the fruit stems of "MBA", and the pruning stems of "Campbell Early" and "Heukbosek" had the highest resveratrol content. In particular, the resveratrol in the fruit stems of "MBA" had the highest concentration as compared to the other varieties in the different parts. The seeds of "MBA", and the fruit stems of "MBA" and "Heukbosek" had the highest catechin content. Epicatechin was detected in the seeds, fruit stems, and pruning stems. Quercetin was detected only in the leaves. In summary, the catechin and epicatechin contents were significantly higher than the quercetin and resveratrol contents. The concentrations of the physiologically active components present in the grapes were high in the non-edible parts than in the edible parts; therefore, they could be useful in industrial applications.

  • Polyphenols are naturally occurring antioxidants and play many important roles in human health. Grape leaves have not yet been reported to contain the antioxidants. In this study, the physicochemical properties of powders from young grape leaves, obtained after hot-air or vacuum freeze drying, were analyzed. Weight loss rangedfrom 76-80%, but no difference was found between samples prepared by hot-air orvacuum freeze drying. The L-, a-, and b- values of grape powder all fell upon hot-air drying. Quercetin, resveratrol, catechin, and epicatechin were detected in all cultivars. The contents of the four polyphenols were generally higher in powders obtained by vacuum freeze dryingthan in those prepared by hot-air drying, indicating that vacuum freeze drying is better for optimal preservation of polyphenols. None of three polyphenols assayed was found in some cultivars or after certain drying treatments. Based on physiochemical properties, our findings suggest that powders obtained after vacuum freeze drying are optimal for industrial use.

  • This study was accomplished to investigate the effect of harvest time on the contents of polyphenolic compounds such as epicatechin, catechin, resveratrol and quercetin in grapes (Vitis spp.). The souble solids contents were increased with harvest time delay, while the titratable acidity continuously decreased in all cultivars. Brightness (L) was higher in 'Campbell Early' and 'Shigyoku', Hunter a value was high in 'Shigyoku' which was harvested later. Hunter b value seemed to be different in 'Shigyoku' depending on harvesting time. The contents of EC which was extracted 'Campbell Early, 'Shigyoku' and 'M.B.A' were higher late harvest time than early harvest time, reapectively, In 'Shigyoku which requires the direct light, the contents of EGC and EC showed distinct differences depending on harvesting time. The compositions of epicatechin were different depending on cultivars. The contents of catechin and resveratrol were higher in 'Campbell Early' and 'M.B.A' which were harvested later, but 'Shigyoku' which was harvested earlier. The contents of quercetin was higher in 'Campbell Early' which was harvested earlier, but 'Shigyoku' and 'M.B.A' which were harvested later. In comparison with 'Shigyoku' and 'M.B.A', catechin and resveratrol contents were seen to be higher in 'Campbell Early' irrespective of harvesting time.

  • Resveratrol (3,5,4'-trihydroxy-trans-stilbene) has been considered to be as one of major antioxidants present in grapes responsible for beneficial effects of red wine consumption on coronary heart disease. This triphenolic stilbene has been suggested as a potential cancer chemopreventive agent based on its striking inhiitory effects on diverse cellular events associated with tumor initiation, promotion, and progression. The compound has strong antioxidative and anti-inflammatory activities which amy contribute to its chemopreventive/chemoprotective properties. In the present work, we have found that resveratrol reduces viability and DNA synthesis capability of cultured human promyelocytic leukemia (HL-60) cells. Likewise, the viability of human breast cancer cell line, MCF-7 was reduced by resveratrol treatment. The growth inhibitory and antiproliferative properties of resveratrol appear to be associated with its induction of apoptotic cell death as determined by morphological and ultrastructural changes, agarose gel electrphoretic analysis of internucleosomal DNA fragmentation, and in situ terminal end-labeling of fragmented DNA (TUNEL). This compound also inhibited the phorbol ester-induced expression of cyclooxygenase-2 (COX-2) protein in immortalized human mammary epithelial MCF-10A cells. These results suggest that resveratrol has the promising cancer therapeutic/chemopreventive potential.

  • In the present study, we have investigated whether methylated marliolide could induce NRF2 thereby exerting anti-oxidant effects. MTT assay showed that methylated marliolide did not exhibit cytotoxicity on HaCaT cells. Methylated marliolide induced a higher ARE-dependent luciferase activation in HaCaT ARE-GFP-luciferase cells, compared with resveratrol. In addition, exposure of methylated marliolide to HaCaT cells significantly induced NRF2 and transcriptionally activated HO-1 and NQO1, both of which are target genes of NRF2. Finally, methylated marliolide protected HaCaT cells against TPA-induced oxidative damages on nucleotides and lipids. Together, results shows that methylated marliolide could suppress oxidative damages through induction of NRF2 which implies that methylated marliolide might serve as a good candidate for novel cosmetic ingredient with anti-oxidant effects.

  • Resveratrol, a polyphenolic compound present in many fruits and vegetables such as grapes, mulberries, and peanuts, has been reported to have various biological effects. However, the molecular mechanisms underlying resveratrol-induced apoptosis in A549 ovarian cancer cells are not well understood. In this study, we investigated the effect of resveratrol on A549 lung cancer cells (expressing wild-type p53) and compared it with that observed for SKOV3 ovarian cancer cells (expressing null-type p53). Resveratrol significantly inhibited the viability and proliferation of A549 cells in a concentration- and time-dependent manner, compared with its effects on SKOV3 cells. It also induced A549 cell apoptosis, but did not affect anoikis resistance. Furthermore, the viability and proliferation of p53-knockdown A549 cells were unaffected by the presence of resveratrol. Therefore, we demonstrate that the anticancer effect of resveratrol against A549 lung cancer cells is dependent on the presence of functional p53.

  • The aim of this study was to investigate the effect of resveratrol on the pharmacokinetics of nifedipine in rats. The pharmacokinetic parameters of nifedipine were measured after the oral administration of nifenipine (6 mg/kg) in the presence or absence of resveratrol (0.5, 2.5 and 10 mg/kg, respectively). The effect of resveratrol on the P-glycoprotein (Pgp), CYP 3A4 activity was also evaluated. Resveratrol inhibited CYP3A4 enzyme activity in a concentration-dependent manner with 50% inhibition concentration ($IC_{50}$) of 0.94 ${\mu}M$. In addition, resveratrol significantly enhanced the cellular accumulation of rhodamine 123 in MCF-7/ADR cells overexpressing P-gp. Compared to the control groups, the presence of 2.5 mg/kg and 10 mg/kg of resveratrol significantly (p<0.05, p<0.01) increased the area under the plasma concentrationtime curve (AUC) of nifedipine by 49~75%, and the peak concentration ($C_{max}$) of nifedipine by 48~66%. The absolute bioavailability (AB%) of nifedipine was significantly (p<0.05) increased by 22.9-34.8% compared to the control (19.8%). The terminal half-life ($T_{1/2}$) of nifedipine was significantly (p<0.05) increased compared to the control. While there was no significant change in the time to reach the peak plasma concentration ($T_{max}$) of nifedipine in the presence of resveratrol. It might be suggested that resveratrol altered disposition of nifedipine by inhibition of both the CYP3A and P-glycoprotein efflux pump in the small intestine of rats. In conclusion, the presence of resveratrol significantly enhanced the oral bioavailability of nifedipine, suggesting that concurrent use of resveratrol or resveratrol-containing dietary supplenment with nifedipine should require close monitoring for potential drug interation.

  • Resveratrol, a polyphenolic compound with antioxidative property, was purified from the grape's twig to be used as functional additives of food and/or cosmetics. Extraction of the grape's twig was performed using 80% ethanol in ultrasonic extractor for 60 min. The crude extract was purified up to 99% after elution through silica gel open column chromatography. The stability of the purified resveratrol was as follows: a half life of 90 days at 40$^{\circ}C$ and 60 days at 25$^{\circ}C$. A sensory test of the commercial grape juice including the 1-10 ppm of purified resveratrol showed better preference than the grape juice without purified resveratrol additive. Color and smell test showed no difference between the samples. The grape twig can be used as a valuable resource for the extraction of resveratrol, which would be added to nutraceutical and cosmetic products.
