The resveratrol contents in grapes showed a great variation according to the cultivars. In spite of it, a high resveratrol content in fruit was found generally in cultivars of Shigyoku, Pione, Muscat of Alexandria (black colored cultivars) and Muscatel Ruso, Gold Finger, Niunai (green colored cultivars). But in case included fruit stem and rachis, some cultivars, i.e. Shigyoku, Muscat of Alexandria and Baby Finger, showed relatively higher resveratrol content, and these cultivars could be advantageous for cultivation without chemical fungicide, because resveratrol was known as a phytoalexin. While resveratrol was not detected in fruit flesh, fruit stem and rachis contained considerably high content of resveratrol; that shows resveratrol may be synthesized in these parts.

In this study, the cellular protective effect of resveratrol on oxidative damage and its antioxidative activity were investigated. The free radical-scavenging activity ($FSC_{50}$) of resveratrol was measured to be $103{\mu}M$. The reactive oxygen species-scavenging activity ($OSC_{50}$) of resveratrol on the ROS generated in a $Fe^{3+}-EDTA/H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. Resveratrol displayed $0.042{\mu}M$ ROS scavenging activity, which is 9.6-fold higher than that of L-ascorbic acid ($0.405{\mu}M$) and had a more prominent cellular protective effect than (+)-${\alpha}$-tocopherol. When HaCaT cells were exposed to $800mJ/cm^2$ of UVB or treated with $30{\mu}M$ rose bengal, resveratrol protected the cells against oxidative stress in a concentration-dependent manner; however, it was unable to protect the cells when the damage was induced by 10 mM $H_2O_2$. These results indicate that resveratrol could be employed to improve and prevent the skin aging through its antioxidative and cellular protective activities.
Glioblastoma multiforme is the most common and most aggressive type of primary brain tumor in humans. Despite intensive treatment, including surgery, radiation, and chemotherapy, most patients die of the disease. Although the anti-cancer activity of resveratrol has been demonstrated in various cancer cell types, its underlying mechanism in glioma cells is not fully elucidated. The present study was undertaken to investigate the effect of resveratrol on cell viability and to determine the molecular mechanism in A172 human glioma cells. Resveratrol caused the generation of reactive oxygen species (ROS), and resveratrol-induced cell death was prevented by antioxidants (N-acetylcysteine and catalase), suggesting that an oxidative mechanism is responsible for resveratrol-induced cell death. Resveratrol-induced phosphorylation of extracellular signal-regulated kinase (ERK), p38 kinase, and c-Jun N-terminal kinase (JNK), and resveratrol-induced cell death were prevented by inhibitors of these kinases. Resveratrol-induced activation of caspase-3 and cell death were prevented by the caspase inhibitors. ERK activation and caspase-3 activation induced by resveratrol was blocked by N-acetylcysteine. Taken together, these results suggest that resveratrol causes a caspase-dependent cell death via activation of ERK, p38, and JNK, mediated by ROS generation, in human glioma cells.
Effects of resveratrol glucosylation on the immunomodulation properties of resveratrol and on the viability of macrophage cells have been studied by using murine macrophage RAW 264.7 cells. Nitric oxide (NO) and interleukin 6 (IL-6) expression in macrophages in vitro were studied after treatment with different concentrations of (E)-resveratrol, (E)-resveratrol 3-O-${\beta}$-${\small{D}}$-glucoside (R-3-G), or (E)-resveratrol 4'-O-${\beta}$-${\small{D}}$-glucoside (R-4'-G). In vitro viability of RAW 264.7 cells after treatment with the aforementioned three compounds was also studied. As demonstrated by macrophage cell viability assays, two different resveratrol monoglucosides, R-3-G and R-4'-G, exhibited 50-80% reduced cytotoxicity in comparison to (E)-resveratrol in A549 and HepG2 cells. Compared to the resveratrol aglycon, both glucosylated resveratrol derivatives positively modulated NO and IL-6 production in macrophages positively via transcriptionally up-regulating IL-6 and iNOS expression. Conjugation of a glucose moiety on resveratrol was found to enhance the immunomodulating activity of resveratrol and the viability of RAW 264.7 cells.
Contents of resveratrol in different parts of Gerbong, Campbell, and Sheridan grapes were measured through HPLC analysis. Resveratrol contents in the flesh, skin, seed, and fruit stem of each grape showed significant differences, whereas little difference was observed among the cultivars. In fruit stem, and skin and seed, resveratrol contents were 170 to 440 and 4 to $8\;{\mu}g/g-dry$ weight, respectively, an indication that the fruit stem may be a good resource for resveratrol production.

Resveratrol is naturally occurring phytoalexin compounds produced by grape berries, peanuts, pine tree, mulberry fruits and their products in response to stress such as fungal infection, heavy metal ions or UV irradiation. The objective of this study was to determine resveratrol contents in wild-type mulberry leaves (Morus alba L., M. bombycis Koidz, and M. Lhou (Ser.) Koidz) collected from Won-ju, In-je and Yang-yang regions, Gangwon province, Korea. Resveratrol contents in wild-type mulberry leaves were higher in Autumn's than in Spring's, and higher than 16 cultivar's. Among the samples, 'No. 3', collected from Won-ju region and 'No. 7' collected from In-je showed high resveratrol contents of 539 and $491{\mu}g$/100 g DW (dry weight; DW), respectively, through the collection time. Therefore, two mulberry trees might be promising as potent resources for mulberry breeding and functional material development.
Resveratrol is a non-flavonoid polyphenol which belongs to the stilbenes group and is naturally generated in several plants in response to damage or fungal invasion. It has been shown in published studies that resveratrol has an anti-adipogenic effect. A good consensus regarding the involvement of a down-regulation of $C/EBP{\alpha}$ and $PPAR{\gamma}$ in this effect has been reached. In addition, different metabolic pathways involved in triacylglycerol metabolism in white adipose tissue have been shown to be regulated by resveratrol. Concerning lipolysis, though this compound in itself seems to be unable to cause lipolysis, it increases lipid mobilization stimulated by ${\beta}-adrenergic$ agents. The increase in brown adipose tissue thermogenesis, and accordingly the associated energy dissipation, can attribute to accounting for the body-fat reducing effect of resveratrol. Besides its effects on adipose tissue, resveratrol can also acts on other organs and tissues. Therefore, it increases mitochondrial biogenesis and accordingly fatty acid oxidation in skeletal muscle and liver. This effect can also attribute to the body-fat reducing effect of this molecule. The present review purposes to collect the evidence concerning the potential mechanisms of action which underlie the anti-obesity effects of resveratrol, acquired either in cultured cells lines and animal models.
Resveratrol (RSV) exerts several beneficial effects on metabolism and metaflammation-related diseases, including diabetes and non-alcoholic fatty liver disease (NAFLD). The purpose of this study is to investigate whether RSV affects pathophysiology of diabetes and NAFLD as well as hepatic autophagy in a rodent model of diet induced obesity (DIO). DIO was induced in a subset of C57BL/6J mice fed a high fat (HF, 45% kcal fat) diet. After six weeks of HF diet treatment, RSV (8 mg/kg/day) was administered via an osmotic pump for a period of four weeks. Therefore, the experimental groups were as follows: 1) lean control (CON), 2) HF diet-induced obese control (HF), and 3) HF_RSV. Body weight and food intake were monitored daily. Fasting glucose, insulin, and adiponectin in serum and lipid profiles in serum and liver were analyzed. In addition, the autophagic process was investigated using transmission electron microscopy (TEM). Body weight and food intake were not affected by RSV treatment. Impaired glucose control accompanied by DIO was recovered with RSV as shown by lower levels of fasting serum glucose and insulin when compared with HF obese controls. In addition, RSV treatment resulted in increased levels of serum adiponectin, however, indices of lipid profile in serum and livers were reduced. Results of TEM analysis showed that a HF diet induced excessive autophagy with the presence of double-membrane autophagosomes, which was ameliorated by RSV. The regulatory effect of RSV on autophagy was confirmed by the altered LC3-II formation, which increased with a HF diet and was decreased by RSV treatment. These results suggest that RSV treatment improves glucose control and lipid profile and these beneficial effects may be mediated by an altered autophagic process.

This study was conducted to find out optimum cultivars, and growth temperature and stage in peanut sprout for resveratrol production. Morphological characteristic, days to 50% of the final germination percentage($T_{50}$) and early growth vigor were measured in 8 different peanut varieties. In result 'Jopyeong' appeared to be the most appropriate cultivar for peanut production because of its lower contamination rate, lesser lateral root formation, and thicker hypocotyl length. Optimum temperature for growing peanut sprouts was determined 27. Content of resveratrol was examined by high performance liquid chromatography(HPLC) to investigate appropriate growth stage of peanut sprouts for resveratrol production. Resveratrol was higher than $17.0{\mu}g/g$ in peanuts sprouts 9 days after plating. Considering peanut sprout's shapes and content of resveratrol into account, it was most appropriate to harvest in 9-day after germination.
The extraction conditions for resveratrol production from grape fruit stem, which is a by-product of grape processing, were optimized to develop high-functional grape-based products. Additionally, the bioefficacy of grape fruit stem extract (GFSE) as an antioxidant agent was evaluated. Resveratrol was extracted using various experimental conditions such as extractant type, extractant concentration, raw material-extractant ratio, extraction time and temperature, and the results were analyzed using a statistical program (SPSS). The resveratrol yield was the highest when 80% ethanol with a raw-material-extractant ratio of 1:10 (w/v) was used. In addition, the optimal temperature and time were selected as $60^{\circ}C$ and 90 min, respectively. When the antioxidant activity was analyzed and expressed as DPPH radical scavenging activity and SOD-like activity, the antioxidant activity of GFSE was higher than that of BHT, BHA and L-ascorbic acid. Finally, it was found that GFSE could be used as a raw material for the production of high antioxidant agents.
