Development of transgenic sweet potato producing human lactoferrin (인체 락토페린 생산 형질전환 고구마 개발)

  • Human lactoferrin is an iron-binding glycoprotein with many biological activities, including the protection against microbial and virus infection and stimulation of the immune system. We introduced a human lactoferrin (hLf) cDNA under the control of 35S promoter into sweet potato by particle bombardment. Transgenic plants were regenerated via somatic embryogenesis. Transgenic plants were produced typical tuberous roots in soil. PCR, Southern and northern analyses confirmed that the hLf cDNA was incorporated into the plant genome and was properly expressed in plants. Western blot analysis showed that the 80 kDa full length hLf protein was produced in transgenic tuberous roots. Overall results indicated that sweet potato would be an excellent host to produce human therapeutic proteins.

  • 전세계 많은 나라들은 조제분유를 특수 영양식품으로 분류하여 관리에 각별히 신경을 쓰고 있으며, CODEX에서는 첨가되는 거의 모든 성분을 모유에 들어 있는 것이어야 하며, 또한 과학적으로 안전성이 입증된 것만을 넣을 수 있도록 규정하고 있다. 이 때문에 분유 메이커들은 제품 개발을 위해 많은 모유 및 우유 성분에 관한 연구를 바탕으로 하여 모유의 조성에 맞추는 노력을 하고 있다. 국내산 분유는 외국 제품에 비해 면역 기능 향상, 항-알레르기 능력 향상, 뇌와 소화기관의 발달 등을 돕는 성분이 강화되어 있다. 현재 생명과학기술의 진보와 각종 신기술의 도입으로 모유 및 우유속의 극미량 성분까지 분리 정제가 가능함에 따라 다양한 기능성을 갖는 성분들이 상품화되고 있으며, 이를 이용해 조제분유를 개발하고 있다. 두뇌 발달 성분인 DHA, 아라키돈산 등과 면역기능 강화 성분인 뉴클레오타이드, 락토페린 등의 첨가 및 정장기능을 보이는 올리고당 등의 성분강화에서 이제는 뼈 성장 ${\cdot}$ 발달인자, 아토피 및 알레르기의 감소를 위해 가수분해물의 이용 등의 기능을 보강한 다양한 제품이 개발되어지고 있는 중이다.

Expression of Human Lactoferrin in the Mammary Glands of Transgenic Mice using Regulatory Elements of Rat $\beta$-Casein Gene (흰쥐 베타-카제인 유전자의 발현조절 부위를 이용하여 유선에서 사람 락토페린을 발현하는 형질전환 생쥐의 개발)

  • Two human lactoferrin expression vectors(pCChcLf and pCChcLf-1) were constructed using rat $\beta$-casein gene and human lactoferrin cDNA. The recombinant DNAs containing human lactoferrin cDNA were microinjected into the fertilized eggs of hybrid mice (BDF1 : C57BL$\times$DBA) and the DNA-injected eggs were treansferred into the oviducts of foster mothers. Genomic DNAs were isolated from the tails of mice born from the microinjected eggs and analyzed by Southern blot analysis. As a result, 5 and 9 transgenic mice with CChcLf and CChcLf-1 gene were produced, respectively. To determine tissue-specificity of transgene expression, Northern blot analysis was performed. Female transgenic mice were killed at day 10 of lactation and total RNAs from various tissues were isolated. Based on Northern blot analysis, it was shown that transgene was mainly expressed in the mammary glands of transgenic mice. In addition, the human lactoferrin in milk was detected by enzyme-linked immunosorbent assay. For this study, milk was obtained from the mammary glands of the transgenic mice at day 10 of lactation. In line #2 of CChcLf and line #7 of CChcLf-1 transgenic mice, human lactoferrin was secreted into the milk at concentration levels of 340ng/ml and 60ng/ml, respectively.

  • Lactoferrin was first identified 60 years ago as a "red protein" in bovine milk. Lactoferrin, one of the transferrin family proteins, is an iron-binding glycoprotein found in milk and various mucosal secretions; it is also released from activated neutrophils. Human lactoferrin has a molecular weight of 82.4 kDa and is composed of 702 or 692 amino acid residues. Bovine lactoferrin has a molecular weight of 83.1 kDa and is composed of 689 amino acid residues. Both lactoferrin and transferrin have the ability to bind two $Fe^{3+}$ ions, together with two ${CO_3}^{2-}$ ions with extremely high affinity; these proteins also have the ability to release this iron at low pH levels. The polypeptide chain in lactoferrin is folded into two globular lobes, representing the N-terminal and C-terminal halves. Both lobes have similar folding and 40% sequence identity. This protein is capable of multiple functions as described in various review papers, including antimicrobial, antiviral, antiinflammatory, anticancer, antioxidant, and cell growth-promoting activities. Lactoferrin also exhibits immunomodulating effects and plays an active role in the regulation of myelopoiesis and the inhibition of bacterial translocation.

Effect of Bovine and Human Lactoferrin on MA 104 Cell Infected with Human Rotavirus (락토페린이 국내분리 유아 로타바이러스의 MA 104세포 감염에 미치는 영향)

  • It has long been known that lactoferrin prevents human beings from infection of virus. To prove this activity of lactoferrin, we evaluated the activities of different lactoferrins to an isolate human rotavirus K-21. Bovine lactoferrin inhibited infection of K-21 to MA-104 cell at the concentration of $25.9\;{\mu}M$ whereas bovine hydrolysed lactoferrin prevented rotavirus infection at $103.8\;{\mu}M$. However human lactoferrin prevented infection of K-21 at the concentration of $217.5\;{\mu}M$. These data suggested that lactoferrin activity may be unaffected by the intestinal digestive enzymes and bovine lactoferrin is more active than human lactoferrin with respect to prevention of rotavirus infection.

Age-Related Changes of Flow Rate, Immunoglobulins, Lactoferrin and Electrolytes in Human Whole Saliva (증령에 따른 인체내 전타액의 타액분비율, 면역글로불린, 락토페인 및 전해질의 변화에 관한 연구)

  • 타액은 분비율과 그 구성성분으로 인해 구내환경을 조절하는데 있어 가장 중요한 요인으로 여겨진다. 타액 분비율과 성분에 관한 많은 연구가 이루어 졌지만, 증령이 타액과 그 성분에 미치는 영향에 대한 연구는 상반된 결과를 보고하고 있으며 현재까지도 논란의 여지가 많다. 또한 증령에 따른 lactoferrin과 전해질의 변화는 거의 보고되지 않은 실정이다. 이에 저자는 증령이 타액분비량과 타액성분에 미치는 영향을 연구하기 위하여 59명의 투약력이 없고 건강한 사람을 대상으로 연구를 시행하였다. 연구대상을 그들의 나이에 따라 A군, 10~15세 (남자7명, 여자7명); B군,20~30세 (남자8명, 여자7명 ); C군,40~50세 (남자7명, 여자7명 ); D군,60세 이상 (남자7명, 여자9명 ) 등의 4군으로 구분하여 각각의 비자극성 전타액을 표준화된 방법으로 채취한후 타액분비량과 immunoglobulin, lactoferin 및 전해질의 변화를 측정하였다. 이와 같은 실험을 통해 다음과 같은 결론을 얻었다. 1. 비자극성 타액분비량은 각 연령군간의 유의한 차이가 관찰되지 않았으며, 20-30세 군(B군)에서만 남성에 비해 여성에서 유의하게 낮았다. 2. 인체 전타액내 IgA와 lactoferin 농도는 연령이나 성별에 따른 뚜렷한 변화는 없었지만, 10-l5세 군(A군) 남성에서 유의하게 낮았다. 3. 인체 전타액내 IgG의 농도는 연령이나 성별에 따른 차이가 관찰되지 않았다. 4. 인체 전타액내 IgM의 농도는 60세이상 군(D군) 남성에서 유의하게 낮은 농도를 보였다. 5. 인체 전타액내 전해질(sodium, chloride, potassium, magnesium)의 농도는 증령에 따라 증가하는 경향을 보였다. magnesium과 chloride는 60세이상 군(D군)에서, sodium과 potassium은 40-50세 군(C관)에서 최대치를 보였다 성별간의 유의성 있는 차이는 발견되지 않았다.

Effect of NMDA Receptor on Analgesic Effect of Bovine Milk-derived Lactoferrin (BLF) (우유속 락토페린의 NMDA 수용체를 통한 진통효과)

  • Lactoferrin is a multifunctional protein that is found in milk, neutrophils, and other biological fluids, and its receptors have also been identified in the central nervous system. Recently, it was reported that bovine milk-derived lacto­ferrin (BLF) produced analgesia via a $\mu$-opioid receptor-mediated response in the spinal cord. However the precise mech­anism of this analgesic effect is remains unclear. In Randall-Selitto paw pressure study, each single administration of morphine (10 mg/kg) and BLF (50, 100 and 200 mg/kg) induced analgesia, however, NMDA receptor antagonist MK-801 (0.1, 0.2 and 0.3 mg/kg), inhibited analgesia induced by BLF (100 mg/kg). Intracerebroventricular infusion (I.C.V.) of N­methyl-D-aspartic acid (NMDA) ($0.3\;{\mu}g/8.0\;{\mu}l/hr/day$), as a NMDA receptor agonist, reversed inhibition of MK-801 (0.3 mg/kg) on analgesia induced by BLF (100 mg/kg). These results suggest that BLF have analgesic effect, through NMDA recep­tor activation.

  • Many countries divide infant formula into special nutrient food and they are in control of thorn very carefully. CODEX prescribes that the components which can be added to infant formula must be exist in the breast milk and the additives must be scientifically proved to be safe. Therefore infant formula manufacturers make efforts to develop infant formula to be similar to human milk based on research of human and bovine milk components. Domestic infant formulas are rich with functional components in comparison with foreign formulas ; such as immunity, anti-allergy, growth factor, developments of brain and digestion organs etc. At this point, progress of bioscience technology and introduction of various new technologies lead micro components in human and bovine milk to be separated and purified. Therefore many functional components are commercialized and infant formula companies use these. Various functional components are added to infant formula ; DHA and arachidonic acid as brain growth components, nucleotide and lactoferrin as immunity increment and oligosaccharides fur digestion. And new infant formula is being developed by using bone growth factor and dehydration components f3r atopy and allergy decrement.

Analysis of Changes in Colostrum Proteins by Mammalian Species (포유류의 종에 따른 초유 단백질의 변화에 대한 분석)

  • There have been numerous reports indicating that milk proteins influence immune functions. Colostrum refers to the breast milk of mammals, secreted starting from the fourth or fifth day after delivery. It has abundant nutrition for the survival of newborn infants. Most importantly, it contains bioactive substances with growth-stimulating and antibiotic, functions. Thus, the colostrum has various physiological roles. This study measured the differences in the composition of colostrum derived from dairy cattle, hanwoo, porcine, and goat sources. The results showed that immunoglobulin, lactoferrin, lactoperoxidase, serum albumin, IgG heavy chain, and IgG light chain were significantly higher in the colostrum of dairy cattle, hanwoo, and goats, but low in porcine colostrum. There was no significant difference in ${\alpha}_{S2}$-casein, ${\alpha}_{S1}$-casein, ${\beta}$-casein, ${\kappa}$-casein, ${\beta}$-lactoglobulin, and ${\alpha}$-lactalbumin contents until seven days after birth. However, porcine colostrum showed high contents of all proteins from the first day to the second day after delivery.

  • To study and validate tissue-specific promoters and vectors, it is important to develop cell culture systems that retain the tissue and species specificity. Such systems are attractive alternatives to transgenic animal models. This study established a line of porcine mammary gland epithelial cells (PMECs) from a primary culture based on the cellular morphology and mRNA levels of porcine beta-casein (CSN2). The selected PMECs were stained with the cytokeratin antibody, and were shown to express milk protein genes (CSN2, lactoferrin, and whey acidic protein). In addition, to confirm the acini structure of PMEC932-7 in 3D culture, live cells were stained with SYTO-13 dye, which binds to nucleic acid. The acini of these PMECs on matrigel were formed by the aggregation of peripheral cells and featured a hollow lumens. The system was demonstrated by testing the effects of the culture conditions to cell culture including cell density and matrigel methods of the PMECs. These results suggest that PMECs possess the genetic and structural features of mammary epithelial cells.
