Leaf Quality and Fatty Acid Composition of Collected Perilla Related Genus and Species Germplasm (들깨류사 종.속 수집 유전자원의 잎품질 및 지방산 조성)

  • Leaf quality and fatty acid compositions of collected perilla related genus and species germplasms such as Perilla frutescens var. japonica Hara, Perilla frutescens var. acuta Kudo, Perilla frutescens var. crispa Decaisne, Perilla frutescens var. for viridis Makino, Mosla punctulata Nakai, Mosla japonica Maxim, Mosla dianthera Maxim were analysized. The number of leaves per tiller and leaf size of perilla germplasms were more and bigger than those of mosla germplasms. Aroma degree of mosla germplasms was higher than aroma degree of perilla germplasms. Mosla germplasms could be utilized in the breeding for high aroma perilla lines. Otherwise, the softness of perilla germplasms was higher than that of mosla germplasms. In case of oil and protein contents, perilla germplasms was higher than mosla germplasms, however compositions ratio of fatty acid, especially linolenic acid of mosla germplasms was higher than that of perilla germplasms, therefore mosla germplasms could be utilized as breeding materials with high linolenic acid for industrial oil. The linolenic acid with excellent quality and unsaturated fatty acid showed negative correlation with oil content, protein content and saturated fatty acids.

Changes in oxidative stability of the oil extracted from perilla seed roasted at different roasting conditions (들깨의 볶음 조건에 따른 들기름의 산화 안정성 변화)

  • The oxidative stabilities of perilla oil increased as roasting temperature and time increased. Induction period of the perilla oil from unroasted perilla seed was 3.9 days, but that of the oil from perilla seed roasted at $210^{\circ}C$ for 30 min was 55 days. The electron donating ability(EDA) on DPPH by perilla oils increased as the roasting temperature and time increased. EDA of the unroasted perilla oil was 24% but that of the perilla oil roasted at $210^{\circ}C$ for 30 min was 64%. These results indicated that the reducing compounds were formed during the roasting process. The fluorescence intensity in perilla oil increased as the roasting temperature and time were increased. This result indicated that Maillard reaction has occurred during the roasting process and the reaction products seemed to provide stability to perilla oil.

Variation in Seed Germination According to the Storage Period after Harvest in the Accessions of Perilla Species (들깨, 차조기 작물의 수확 후 저장 기간에 따른 종자 발아 변이)

  • The present study aimed to understand the variation in seed germination according to the storage period after harvest in the accessions of cultivated and weedy types of Perilla species in Korea. In this milieu, we investigated the germination rate and germination energy of 59 Perilla accessions (15 cultivated var. frutescens type I, 4 cultivated var. frutescens type II, 20 weedy var. frutescens, and 20 weedy var. crispa), which were harvested in autumn 2016. The accessions of cultivated var. frutescens (type I) showed an average germination rate of 80.8% 6 months after harvest; however, the accessions of weedy var. frutescens and var. crispa showed an average germination rate of 0.9% and 8.7%, respectively. Furthermore, the accessions of cultivated var. frutescens (type II) showed an average germination rate of 18.2%. The accessions of cultivated var. frutescens (type I) showed an average germination energy of 75.8%, while the accessions of weedy var. frutescens and var. crispa showed an average germination energy of 0.6% and 6.9%, respectively. In addition, the accessions of cultivated var. frutescens (type II) showed an average germination energy of 14.3%. The germination rate and germination energy for the accessions of cultivated and weedy types of Perilla species increased marginally at 2 months from the first month after harvest. However, it did not significantly increase until six months after that. According to our results, there are two types of cultivated var. frutescens, namely, type I, which showed high germination rate and germination energy, and type II, which showed low germination rate and germination energy. The results of this study will provide basic information to understand variations in the germination of seeds during 6 months of storage period after harvest in the accessions of cultivated and weedy types of Perilla species in Korea.

Influence of Mg(II), Ca(II), Perilla Oil and Korean Ginseng on the Plasma Cholesterol Concentration and HMG-CoA reductase Activity of the Rabbits (마그네슘, 칼슘 및 들깨 기름이 인상 첨가 식이로 사육한 토끼의 혈액 성분 변화에 미치는 영향)

  • 인삼, 들깨 기름, 칼슘 및 마그네슘이 함유되어 있는 식이로 사육한 토끼의 혈장 콜레스테롤과 HMG-CoA reductase의 활성을 조사하였다. 간에 포함되어 있는 HMG-CoA reductase의 활성은 인삼과 들깨 기름 및 마그네슘 첨가 식이군에 있어서 증가되었으며 혈장 콜레스테롤의 농도는 감소되었다. 혈장 LDL-콜레스테롤은 대조군보다 감소하였으나 HDL-콜레스테롤은 증가되었다. 또한 베타 글로블린의 분포도 인삼과 마그네슘 및 들깨 기름을 첨가 급여한 실험군에 있어서 가장 낮았으며, ${\alpha}$-2-글로블린은 대조군에 비하여 가장 높은 값을 보였다. 전해질은 $K^+$은 인삼 첨가로 감소 되었으나 $Na^+$은 증가되었다. 혈장 마그네슘의 농도는 인삼의 첨가로 변화가 없었지만 칼슘과 들깨 기름 및 인삼 첨가식이로 마그네슘의 농도가 더 증가되었다. 혈장 칼슘의 농도는 마그네슘과 인삼을 첨가한 식이군에 있어서 감소하였다.

Effect of Carrot Powder on Anti-Oxidative and Quality Characteristics of Perilla-Dasik (당근가루를 첨가한 들깨다식의 품질 특성 연구)

  • Carrot powder was added at different levels (0, 5, 10, 15, and 20%) to Perilla-Dasik, and its effects on the physical and antioxidant properties of Dasik were measured. For texture characteristics, hardness, cohesiveness, and chewiness of Dasik slightly increased as carrot powder content increased. For color properties, both $a^*$ (redness) and $b^*$ (yellowness) values increased as carrot powder content increased. Anti-oxidative effects, as measured by acid value, peroxide value, and free radical scavenging activity, were greater in samples containing carrot powder compared to control. In the sensory test, Dasik with carrot powder was more preferred than control (without carrot powder). Based on these results, 20% addition of carrot powder in the formulation was the most acceptable for Dasik.

Effects of Seed Germination on Oil Oxidation and Tocopherol Stability of Perilla Oil (들깨의 발아가 들깨유의 산화 및 토코페롤 안정성에 미치는 영향)

  • Auto- and photo-oxidative stability of oil extracted from germinated perilla seeds during storage at $60^{\circ}C$ for 4 days was studied by determining peroxide and conjugated dienoic acid values. Tocopherol contents during oil oxidation were also monitored by high performance liquid chromatography. Perilla oil was oxidized and tocopherols were degraded during storage at $60^{\circ}C$ regardless of the presence of light. Light increased oil oxidation and tocopherol degradation. Seeds germinated for 12 h had increased tocopherol contents in the oil and improved the auto- and photo-oxidative stability of the perilla oil. Tocopherol played a more important role as an antioxidant in the presence of light than in the absence of light.

  • For the development of a qualitative PCR detection method for genetically modified perilla (Perilla frutescens), perilla species-specific gene, KAS-I (Beta-ketoacyl-ACP synthase I), was selected and validated as suitable for the use as an endogenous reference gene in perilla. Primer specificity was first tested by the means of qualitative PCR analysis. The primer pair Pfru3-F/R amplifying the perilla endogenous gene, KAS-I, gave rise to an amplicon 95 bp. No amplified product was observed when DNA samples from 15 different plants were used as templates. Qualitative PCR detection method was assayed with vitamin E-enriched GM Perilla developed in Korea. For the qualitative PCR detection method, the construct-specific detection primer pairs were constructed. The primer pair TMTO-F/R amplifying the junction region of TMT (${\gamma}$-tocopherol methyltransferase) gene and OCS (Octopine synthase) terminator introduced in GM perilla gave rise to an amplicon 148 bp.

Effect of Sowing time on Growth and Yield of Perilla in Central Region (중부지역에서 들깨의 파종시기가 생육과 수량에 미치는 영향)

  • 들깨는 식물성 오메가-3의 우수한 급원으로서 국내에서 그 수요가 크게 확대되고 있는 실정이다. 들깨 재배농가에서는 파종시기를 고려한 적합한 들깨 품종에 대한 요구가 많아 이를 해결하고자 중부지역에 적합한 품종을 선발하고자 시험을 수행하였다. 파종시기는 6월 상순, 6월 중순, 7월 초순 3시기로 구분하여 파종하였으며, 토양 검정시비량을 시비하고 완전임의배치법으로 3반복으로 시험구를 배치하였다. 시험품종으로 다유 등 10품종을 선정하여 시험하였다. 6월 초순 파종에서 개화기는 다실 품종이 8월 20일로 가장 빨랐으며, 다유, 대실, 들샘, 새엽실 품종이 9월 10일로 가장 늦었다. 6월 중순 파종에서 개화기는 다실 품종이 8월 27일로 가장 빨랐으며, 들샘 품종이 9월 11일로 가장 늦었다. 7월 초순 파종에서 개화기는 다실 품종이 8월 27일로 가장 빨랐으며, 들샘과 새엽실 품종이 9월 11일로 가장 늦었다. 들깨 천립중은 파종기가 늦어질수록 대체적으로 무거워지는 경향을 보였다. 6월 초순 파종에서 백진, 들향, 새엽실 다유 품종이 10a 당 110kg 이상의 수량을 나타냈으며, 6월 중순 파종에서 다유, 들향, 백진, 새엽실 품종이 10a 당 120kg 이상의 수량을 나타내었고, 7월 초순 파종에서는 다유, 들향, 새엽실 품종이 10a 당 110kg 이상의 수량을 나타내었다. 이상의 결과로부터 중부지역에서는 대체적으로 6월 중순 파종이 적합하였으며 전작물을 고려하여 들깨 파종시기를 빠르거나 늦게 할 경우에는 각 시기에 적합한 품종을 파종하면 안정적인 수량 확보가 가능하다. 중부지역에서 파종기에 따른 영향이 적고 안정적인 수량을 생산할 수 있는 품종으로는 다유, 들향, 새엽실 이었다. 이상의 결과로 부터 품종별 최대 수량을 얻기 위해서는 각 품종에 맞는 파종기를 준수하여야 할 것으로 사료되었다.

Antioxidant Activities of Perilla frutescens Britton Seed Extract and Its Inhibitory Effects against Major Characteristics of Cancer Cells (들깨 추출물의 항산화 활성과 암세포 기본 특성에 대한 억제 효과)

  • The aim of the study was to investigate the antioxidant activities of ethanol extract of perilla seed (PSE) and its inhibitory effects against major characteristics of cancer cells, such as unrestricted growth and activated metastasis in vitro. The total polyphenol and flavonoid levels of PSE were 222.6 mg gallic acid equivalent/100 g and 285.7 mg quercetin equivalent/100 g, respectively. The radical scavenging activity and ferric reducing antioxidant power of PSE at concentration of 87.5 to $350{\mu}g/mL$ were 24~45% and 28~62%, respectively. Treatment of HCT116 colorectal carcinoma cells and H1299 non-small cell lung carcinoma cells with PSE dose-dependently inhibited growth by 18~94% (at concentration range of 87.5 to $350{\mu}g/mL$) and completely abolished colony formation (at concentration of $175{\mu}g/mL$). PSE was also effective in inhibiting migration of H1299 cells (by 30~37% at concentration range of 87.5 to $350{\mu}g/mL$) and adhesion of both HCT116 and H1299 cells (by 14~16% at concentration of $350{\mu}g/mL$). These results indicate that PSE exerts antioxidant and anti-cancer activities in vitro. It needs to be determined whether or not similar effects can be reproduced in vivo.

Anti-inflammatory Activity of Perilla frutescens Britton Seed in RAW 264.7 Macrophages and an Ulcerative Colitis Mouse Model (RAW 264.7 대식세포와 궤양성 대장염 마우스 모델에서의 들깨의 항염증 효과)

  • This study aimed to investigate the anti-inflammatory activities of raw (P) and roasted (RP) Perilla frutescens Britton (perilla) seeds in RAW 264.7 macrophages and an ulcerative colitis mouse model. In lipopolysaccharide-treated RAW 264.7 cells, treatment with ethanol extract of P at the concentrations of 75 and $150{\mu}g/mL$ decreased nitric oxide (NO), interleukin-6 (IL-6), and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) levels to 48-85% of the control (p<0.01). Treatment with RP extract exhibited similar effects on NO, IL-6, and TNF-${\alpha}$, decreasing those levels to 51-84% of the control (p<0.01). In dextran sulfate sodium-treated ulcerative colitis mice, dietary treatment with 1% RP for 7 days decreased the colonic levels of prostaglandin $E_2$ and leukotriene $B_4$ to 34% and 58% of the control, respectively (p<0.05). Dietary P treatment, however, did not decrease those levels significantly. These results indicate that roasted perilla seed exerts anti-inflammatory activity both in vitro and in vivo.
