Optimization of Roasting Conditions for High-Quality Polygonatum odoratum Tea (둥굴레차의 고품질화를 위한 볶음조건의 최적화)

  • Response surface methodology was applied to determine the optimum roasting conditions (roasting temperature and time) for the high-quality Polygonatum odoratum tea which has been roasting with a traditional means. As quality criteria of Polygonatum odoratum tea, water-soluble solids, browning color, total phenolic compounds and electron-donating ability were proportionally increased with increased temperature and time of roasting conditions up to around $145^{\circ}C$ and 55 min, respectively, while they were decreased under the extended-roasting conditions. The optimum roasting temperature and time based on the organoleptic overall acceptability were $146^{\circ}C$ and 52 min, respectively. On the basis of superimposed contour maps for the tea characteristics, the optimum range of roasting conditions were $135{\sim}140^{\circ}C$ and $58{\sim}64$ min. Predicted values at the optimum conditions $(137^{\circ}C,\;60\;min)$ were in good agreement with experimental values.

  • 비짜루과 둥굴레속(Asparagaceae: Polygonatum)은 전 세계적으로 약 90여 종이 알려져 있으며, 유럽, 북아메리카, 아시아 등 북반구 온대 지역에 집중적으로 분포한다. 국내 둥굴레속 분류군은 총 16분류군이며, 이중 잎이 호생하고, 난형에서 타원형 모양의 엽질성 포를 가지며, 화피통 내부에 털이 없고, 수술대 표면에 돌기가 나있는 분류군들은 용둥굴레열(series. Bracteata)에 속한다. 그러나 이들은 종간 교잡 또는 주요 기관의 형질 변이가 다양하여 중간형질을 보이는 개체군들에 대한 종 식별에 많은 어려움이 있었다. 경남 창원시에서 채집된 목포용둥굴레(P. cryptanthum) 변이 개체집단는 기존의 목포용둥굴레와 달리 식물체 높이와 화경·소화경이 길며, 포 부착위치의 변이 폭이 넓으며, 포가 타원형이고 밖으로 말리는 습성으로 형태적 차이가 나타났다. 따라서 본 연구에서는 명확한 분류학적 실체를 구명하고자 분자생물학적 계통분석(nrDNA ITS + cpDNA matK, trnK-rps16, rps16, rbcL) 연구를 진행중에 있다.

Reexamination of plant names in the literature published during the Japanese Imperialism Period with special reference to Hwangjeong and Wiyu (일제 강점기 문헌에 나오는 식물명의 재검토: 황정(黃精)과 위유(萎蕤)를 중심으로)

  • The plant names Hwangjeong and Wiyu were written in Chinese characters in Hyang-yak-jib-seong-bang during the early Chosen dynasty. However, soon after, Hwangjeong and Wiyu were written in Korean characters as Jukdae and Dung-gul-re, respectively. However, since under the Japanese imperialist period in Korea, the taxonomic identities of these two Korean names have been incorrectly understood, with scientific names incorrectly assigned as well to these two names thus far. The results of the present study prove that Hwangjeong is Polygonatum sibiricum and that its Korean name should be Jukdae, its initial Korean name, and not Cheung-cheung-gal-go-ri-dung-gul-re, as used recently. Meanwhile, during the Japanese imperialist period, Wiyu was termed P. officinale or P. japonicum with the Korean name of Dung-gul-re. However, the correct scientific names were shown to be synonyms of P. odoratum.

Comparison of Flavor Compounds in Steamed- and Nonsteamed-Roasted Polygonatum odoratum Roots by Solid-Phase Microextraction (Solid-Phase Microextraction(SPME)을 이용한 둥굴레차의 증자 여부에 따른 향기성분 특성 비교)

  • The headspace flavors of roasted tea, prepared with steamed and nonsteamed polygonatum roots, were absorbed in solid-phase microextraction(SPME) fiber coated with $65\;{\mu}m$ of carbowax/divinylbenzene(CW/DVB) and analysed by GC-MS. The absorption conditions of SPME fiber for equilibrated headspace were selected as $60^{\circ}C$ and 30 min. In a comparison for both samples roasted at $130^{\circ}C$ for 15 min, gas chromatograms showed a similar pattern in overall profiles between steamed and nonsteamed samples before roasting, but some differences were observed in peak characteristics. From 40 separated peaks, 25 compounds were identified with both GC-MS and retention time comparison. The pyrazines including 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one, 2,5-dimethyl-4-hydroxy-3(2H)-furanone, 2-acetyl-1-pyrroline, etc. were higher in their contents in nonsteamed-roasted sample than steamed-roasted one. In particular, steamed-roasted polygonatum showed higher contents of acetic acid(8.17%) and hexanoic acid(5.43%) than the corresponding compounds of nonsteamed-roasted one, 2.40% and 2.00%.

Sensory Evaluation and Electronic Nose Analysis for the Development of Mixed Eucommia ulmoides Leaf Tea (두충혼합차 개발을 위한 관능검사 및 전자코 분석)

  • The leaves of Duchung(Eucommia ulmoides), an oriental medicinal plant, have a peculiar aroma of Chinese medicine and astringent taste, which make the consumer be reluctant to Duchung leaf tea. Therefore, the purpose of this study was to develop a mild flavored Duchung leaf tea by mixing with other plants. The flavor patterns of developed tea were analyzed using an electronic nose. Polygonatum odoratum and Elsholtzia splendens were used for improving the flavor of Duchung leaf tea. The addition of 20, 30 and 40% of Polygonatum odoratum improved the overall acceptance in hedonic sensory evaluation. The flavor pattern of the tea was described by principal component analysis(PCA) and the resistance ratio(R/cub gas/R$\_$air/) of sensors. The PCA plot was also used to explain the mild flavor of the tea, which was extended from the right side(positive value of the first principal component) to the left side(negative value). Analysis by using an electronic nose with metal oxide sensors could be applied to detect whether mixed Duchung leaf tea was acceptable or not.

Interspecific Relationship of Polygonatum Species Collected from Gyeongnam Area Using Cluster Analysis (경남지역 둥굴레속의 Cluster 분석에 의한 종간 유연관계)

  • Polygonatum species are a useful medical crop in Korea but basic study in the species was not well conducted. The study was carried out to analyse genetic diversity and intraspecific relationship of 47 Polygonatum species collected from Gyeongnam province. Their analysis was done through principle component analysis and average linkage cluster analysis with their twelve morphological traits. The result of principle component analysis showed the Prin 1, Prin 2 and Prin 3 represented 79% of total variation. By the 0.7 average distance of the cluster analysis and the calculated Euclidian distance, the 47 collected species were grouped into five groups. Group I included 22 collected species representing P. ordoratum var. pluriflorum, group II did 5 ones representing P. involucratum, group III was divided into two subclasses, 2 species including P. inflatum and 7 species including P. thunbergii, group IV also consisted of 2 subclasses, a species similar to P. thunbergii and P.involucratum, respectively, and finally group V included 8 species representing P.lasianthum var. coreanum. meaning that the useful germplasms can be collected from relatively small area.

Genetic Analysis of Polygonati Rhizoma and Polygonati odorati Rhizoma using Random Amplified Microsatellite Polymorphism (RAMP를 이용한 황정과 위유의 유전적 분석)

  • Two herbal medicines of the Polygonatum genus, namely Polygonati Rhizoma and Polygonati Odorati Rhizoma, are difficult to distinguish from each other through exterior morphologic aspects. Furthermore, because the standard components for physiochemical distinction have not yet been standardized, the identification of these medicines through botanic taxology is based on genetic methods of random amplified microsatellite polymorphism (RAMP). For the RAMP evaluation, we used five sets of UBC microsatellite primers 811, 818, 834, 836, 842 and random primer M1. Although no specific band that could clearly distinguish Polygonati Rhizoma from Polygonati Odorati Rhizoma was found, 11 Polygonatum plants could be divided into two groups by this method. P. sibiricum and P. stenophyllum were classified to group I and the others were to group II. As P. sibiricum and P. stenophyllum were very similar in genetic and morphologic perspective, the results suggest that P. stenophyllum belongs to the Polygonati Rhizoma family.
