Occurrence and Distribution of Selected Veterinary Antibiotics in Soils, Sediments and Water Adjacent to a Cattle Manure Composting Facility in Korea (국내 우분 퇴비화 시설 인근 농경지 및 수계 중 Tetracycline 및 Sulfonamide 계열 항생물질의 분포특성)

  • There has been increased concern regarding the release of antibiotics to different environmental compartments due to the possibility of the development of antibiotic resistant bacteria. However, limited information is available regarding the occurrence, fate, and transport of antibiotics in Korea in both the aqueous phase and in solid phases such as sediment and soil. Therefore, this study was conducted to monitor the concentration of released antibiotics in surface water, sediment, and soil adjacent to a cattle manure composting facility in Korea. Specifically, the following six antibiotics were monitored: tetracycline (TC), chlortetracycline (CTC), oxytetracycline (OTC), sulfamethazine (SMT), sulfamethoxazole (SMX), and sulfathiazole (STZ). To extract and quantify the antibiotics from different environmental compartments, solid phase extraction (SPE) and high performance liquid chromatography mass spectrometry (HPLC/MS) techniques were adopted. The concentration of the six antibiotics ranged from below the detection limit (BDL) to 0.71 ${\mu}g$/L in surface water, from BDL to 27.61 ${\mu}g$/L in sediment, and from 0.12 to 157.33 ${\mu}g$/L in soil. In addition, higher concentrations of antibiotics were observed in surface water and sediment at locations closer to the composting facility indicating that composting is the source of the antibiotics found in the environment. Furthermore, higher concentrations of antibiotics were observed in the solid phase (sediment and soil) than the aqueous phase. These findings indicate that the possibility of antibiotic resistant bacteria is increased because such bacteria are more stable in the solid phase. Overall, longterm monitoring of the aqueous phase and solid phase is necessary to gain a better understanding of the impact of antibiotics from source on the environment in Korea.

Enzymatic Characteristics of Water-Insoluble ${\alpha}-Amylase$ Immobilized on Dithiocarbamate Wool (Dithiocarbamate Wool에 고정(固定)된 불용성(不溶性) ${\alpha}-Amylase$의 특성(特性)에 대하여)

  • Yellowish modified wool, dithiocarbamate(DTC) wool, was synthesized by partial hydrolysis in 0.2 N-NaOH reacting with carbon disulfide to use as ${\alpha}-amylase$ immobilization matrix. ${\alpha}-amylase$ was immobilized reacting with sulfide group of DTC-wool by covalent binding within 1 hour. 0.5 gram of this preparation, $DTC-wool-{\alpha}-amylase$, contained 150 ug of enzyme protein and its specific activity was about 90% of the native one. General properties of $DTC-wool-{\alpha}-amylase$ were a little different from optimum temperature, optimum pH, heat stability, kinetic constants and activation energy. An apparent Michaelis constant and maximum velocity of $DTC-wool-{\alpha}-amylase$ were 5.56 mg/ml and 0.37 mg/ml. $min^{-1}$ respectively, while activation energy was 16.6 kcal/mole.

Estimations on the Water Purification of Forest by Analyzing Water Quality Variations in Forest Hydrological Processes (산지(山地) 물순환(循環) 소과정(素過程)에 있어서 수질변화(水質變化)의 추적분석(追跡分析)에 의한 산림(山林)의 환경적(環境的) 정화기능(淨化機能)의 계량화(計量化) 연구(硏究))

  • This study was carried out to evaluate the capacity of environment purification of forest and to reveal formation processes of stream water quality by evaluating water quality variations in forest hydrological processes. Water quantity, pH, electric conductivity(EC), dissolved oxygen(DO), and dissolved matter concentrations were monitored in open rainfall, throughfall, stemflow, litter flow and short-term stream flow for one unit storm, and also for those were monitored in long-term stream flow in Palgong, Yongsung, and Daedong catchments. The results were summarized as follows; 1. pH and DO values of stream flow were increased as the flux increased but EC values were decreased. 2. pH values of stemflow and throughfall were decreased with the lapse of rainfall time with lower values than open rainfall. Arid EC values were higher in initial rainfall period but lower gradually with the lapse of time than open rainfall. In litter flow, pH values were lower than open rainfall but EC values were higher. In stream flow, pH values of stream flow showed a high level in initial rainfall period and decreased remarkably with the lapse of time, but it recovered after the rainfall stopped. And however, the values of EC showed almost reverse tendency. DO values of litter flow and stream flow were decreased gradually with the lapse of time in litter flow and stream flow but there were no any tendency in open rainfall, stemflow and throughfall. 3. pH values of stemflow and throughfall in Quercus acutissima were higher than in Pinus densiflora, but EC values were lower. Soil CO2 Efflux Dynamics in Response to Fertilization in Pinus densiflora and Quercus variabilis Stands (소나무와 굴참나무 임분의 시비에 따른 토양 CO2 방출 동태)

  • This study compared soil CO2 efflux rates after fertilization, in Pinus densiflora and Quercus variabilis stands. Compound fertilizers were applied to the forest floor in March 2016, following a one-year calibration period (from March 2015 to February 2016). In situ soil CO2 efflux rates were measured every month during the two-year study periods, using an infrared gas analyzer with a closed chamber system. Mean annual soil CO2 efflux rates were higher following fertilizer application in the P. densiflora and Q. variabilis stands (P. densiflora: 2.180 μmol m-2 s-1; Q. variabilis: 1.977 μmol m-2 s-1) as compared with the rates measured during the calibration period (P. densiflora: 1.620 μmol m-2 s-1; Q. variabilis: 1.557 μmol m-2 s-1). The mean annual soil CO2 efflux rates in the unfertilized treatments of both stands were not significantly different between the two-year study periods. The Q10 values of fertilized treatments in Q. variabilis stands were higher in the fertilization period (3.41) than in the calibration period (3.14), whereas the Q10 values in P. densiflora stands did not change between the fertilization and calibration periods. The Q10 values of unfertilized treatments in the Q. variabilis stands were lower during the 2016-2017 period (3.69), than in the 2015-2016 period (3.85), whereas the Q10 values in P. densiflora stands were higher during the 2016-2017 period (3.65), than in the 2015-2016 period (3.15). These results indicate that the increase in soil CO2 efflux rates in P. densiflora stands could be more sensitive to fertilization compared with the rates in Q. variabilis stands.

Changes of Major Components and Microorganisms during the Fermentation of Korean Ordinary Kochujang (한국재래식(韓國在來式)고추장숙성중(熟成中)의 주요성분(主要成分) 및 미생물(微生物)의 변화(變化))

  • The present study was attempted to obtain the basic data concerning a reasonable preparing method and the optimum fermentation conditions of Kochujang (Red pepper paste). To establish the standard qualify of Kochujang, changes of the chemical composition and the numbers of bacteria and feasts in Kochujang during fermentation were observed. Moisture, salts and crude ash contents of Kochujang were not changed significantly during fermentation. Titrable acidity and amino nitrogen gradually increased with the time-passed, whereas crude fat gradually decreased with the time-elapsed. And reducing sugar and total nitrogen increased until 40 days, but slightly decreased after this period. The numbers of bacteria and yeasts in the ingrients for the preparation of Kochujang were $3.9{\times}10^7/g$, $1.5{\times}10^3/g$ in red pepper powder, $7.6{\times}10^4/g$, $2.8{\times}10^2/g$ in salts. respectively, but those of sugar and malt were not more than 100/g. Microbial counts in Kochujang during fermentation increased until 40 days, but those are gradually decreased after that.

Changes of Leaf Area Index, Physiological Activities and Soil Water in Tricholoma matsutake Producing Pine Forest Ecosystem (송이산 소나무림 생태계에서 엽면적지수와 생리적활동 및 토양수분의 변화)

  • The purpose of this study was to understand the ecosystem structure and function and soil water changes in Tricholoma matsutake producing pine stands. The investigated stands were pine forest in Sogrisan National Park in Chungbuk-do of Korea. For the purpose we investigated main vegetation, leaf area index(LAI) as ecosystem structural factors and measured photosynthesis, transpiration, xylem water potential, and soil water changes as ecosystem functional factors. Vertical vegetation structure of the site was composed of Pinus densiflora as a overstory species, Quercus mongolica as midstory, Rhododendron mucronulatum, R. schlippenbachii and Fraxinus sieboldiana as understory ones. In the stands LAI was 3.8 during June to September, 2.6 in October and 2.1 during November to April. Photosyntheses of the trees were 6.0 to $7.0{\mu}mol\;CO_2/m^2/s$ in August, and for P. densiflora about $4.0{\mu}mol\;CO_2/m^2/s$ and for Q. mongolica $2.0{\mu}mol\;CO_2/m^2/s$ in mid October. However, R. mucronulatum stopped fixing $CO_2$ and F. sieboldiana shed off the leaves already in mid October. Transpirations were 2.5 to $3.5mmol\;H_2O/m^2/s$ in late August and about $1.0mmol/H_2O/m^2/s$ in mid October. Plant water potentials were -10 to -22 bars for P. densiflora and -5 to -12 bars for the other woody species. The lowest potentials was in late August and highest in late October. Soil water in the stand was closely related to topography. Soil water contents were 7 to 11% at the ridge, 8 to 15% at the hillside and 11 to 19% at the base. Soil temperatures were 0.2 to $0.4^{\circ}C$ higher in T. matustuake colony than noncolony. Mid September soil temperature decreased to $19^{\circ}C$ at which T. matsutake forms primordia. In T. matsutake colony soil moisture was 0.5 to 2.0% lower due to metabolism for consuming water. We suggest that the complicate relationships between ecosystem structure and function in Tricholoma matsutake producing pine stand need to be further investigated.

Assessment of Microbiological Quality for Raw Materials and Cooked Foods in Elementary School Food Establishment (초등학교에 공급되는 급식용 식재료 및 조리식품의 미생물학적 품질평가)

  • This study was conducted to assess the microbiological quality of raw and cooked foods served in the elementary school food service. Raw and cooked food samples were collected from 11 selected elementary schools in both June to July and September to October of 2005. Petrifilm plates were used to determine (in duplicate) total aerobic colony counts (PAC), Enterobacteriaceae (PE), coliform counts (PCC), and E. coli counts (PEC). Heavy contamination of Enterobacteriaceae (from 0.08 to 7.40 log CFU/g) and total coliform (0.50 to 6.52 log CFU/g) were observed in raw materials and cooked foods. Escherichia coli (E. coli) were detected in the sample of currant tomato (3.70 log CFU/g), sesame leaf (3.59 log CFU/g), dropwort (0.20 log CFU/g), crown daisy (3.15 log CFU/g), parsley (3.00 log CFU/g), peeled green onion (1.74 log CFU/g), frozen pork (0.65 log CFU/g), frozen beef (0.20 or 1.50 log CFU/g), chicken (1.78 log CFU/g), and young radish leaf seasoned with soybean paste (1.24 log CFU/g). Multiplex PCR system was used to determine the food-borne pathogens: Salmonella spp., Bacillus cereus (B. cereus), E. coli O157:H7, Staphylococcus aureus, Listeria monocytogenes (L. monocytogenes), Vibrio parahaemolyticus, Campylobacter jejuni (C. jejuni), Shigella spp., B. cereus was detected in 19 samples of raw materials and 8 samples of cooked foods. With regard to quantitative analysis, B. cereus counts exceeded 5.46, 3.48 and 1.79 log CFU/g in sesame leaf, peeled green onion and seasoned mungbean jelly, respectively. E. coli O157:H7 was detected on 2 samples of frozen beefs, and its biochemical characteristics of one beef sample was confirmed with API 20E kit (93.7%). L. monocytogenes was detected in fried rice paper dumpling, but the presumptive colonies were not detected onto the conventional plate. C. jejuni was detected in peeled & washed onion.

Decay Rate and Nutrient Dynamics during Litter Decomposition of Quercus acutissima and Quercus mysinaefolia (상수리나무와 가시나무 낙엽의 분해율 및 분해과정에 따른 영양염류 함량 변화)

  • Decay rate and nutrient dynamics during leaf litter decomposition of deciduous Quercus acutissima and evergreen Quercus mysinaefolia were studied for 24 months from December 2008 to December 2010 in Gongju, Chungnam Province, Korea. Percent remaining weight of Q. acutissima and Q. mysinaefolia leaf litter after 24 months elapsed was $46.3{\pm}5.4%$ and $37.8{\pm}2.5%$, respectively. Decomposition of evergreen Q. mysinaefolia leaf litter was significantly faster than that of deciduous Quercus acutissima leaf litter. Decay constant(k) of Q. acutissima and Q. mysinaefolia leaf litter after 24 months elapsed was 0.38 and 0.49, respectively. Initial C/N and C/P ratio of Q. mysinaefolia leaf litter was significantly lower than those of Q. acutissima leaf litter. Initial C/N and C/P ratio of Q. acutissima leaf litter was 46.8 and 270.9, respectively. After 24 months elapsed, C/N and C/P ratio of decomposing Q. acutissima leaf litter decreased to 22.5 and 104.2, respectively. Initial C/N and C/P ratio of Q. mysinaefolia leaf litter was 22.4 and 41.7, respectively. After 24 months elapsed, C/N and C/P ratio of decomposing Q. mysinaefolia leaf litter decreased to 16.7 and 89.7, respectively. Initial concentration of N, P, K, Ca and Mg in leaf litter was 8.31, 0.44, 4.18, 9.38, 1.37 mg/g in Q. acutissima, and 19.88, 2.73, 7.06, 8.24, 2.61 mg/g in Q. mysinaefolia, respectively. Initial concentration of N and P in Q. mysinaefolia leaf litter was significantly higher than those in Q. acutissima. After 24 month elapsed, remaining N, P, K, Ca and Mg were 100.91, 114.75, 32.99, 50.63, 15.51% in Q. acutissima, and 43.22, 11.35, 12.98, 82.22, 44.23% in Q. mysinaefolia, respectively. N and P in decomposing leaf litter was immobilized in Q. acutissima, and mineralized in Q. mysinaefolia.

The Change of Seedling Emergence of Abies koreana and Altitudinal Species Composition in the Subalpine Area of Mt. Jiri over Short-Term(2015-2017) (지리산 아고산대의 단기간(2015-2017)에 걸친 구상나무 치수 발생 및 고도별 종구성 변화)

  • To investigate the changing patterns of sub-alpine forest vegetation due to climate change requires accumulation of contiguous reference data and continuous monitoring. Furthermore, it is crucial to monitor short-term ecological change of lower level vegetation to understand the trend of long-term vegetation change. Therefore, this study carried out a vegetation survey and tree diameter measurement in 36 plots of Mt. Jiri inhabited by Abies koreana species from 2015 to 2017 to examine the short-term dynamics of Abies koreana seedling and the change of vegetation distribution according to altitude. We analyzed the importance value and MIV (mean importance value) of major species by each stratum as well as the importance value and species diversity index of major species and the change of seedling population by altitude. The results showed that Abies koreana had the highest importance value on tree layer, Rhododendron schlippenbachii on shrub layer and Tripterygium regelii on herb layer. MIV was high in the order of Abies koreana, Rhododendron schlippenbachii and Acer pseudosieboldianum. Regarding the species composition and species diversity index (H') along the altitudinal gradient, Sasa borealis showed high MI and low H' in the elevation less than 1,500 m, and IV of Tripterygium regelii and H' of herb layer were high in the elevation of 1,700 - 1,800 m. Abies koreana seedling decreased by 22.4% from 1,250 n/ha in 2015 to 970 n/ha in 2017 (p <0.05) throughout the investigated area. The decline rate along seedling and sapling height were 22.9% in less than 10 cm, 3.4% in 10-30 cm, 8.9% in 30-50 cm, 39.3% in 50-100 cm, and 55.1% more than 100 cm. Few of A. koreana seedlings appeared due to the dominance of Sasa borealis in the elevation of 1,500 m or less and due to the dominance and high species diversity of Tripterygium regelii in the elevation of 1,700-1,800 m. On the other hand, many of A. koreana seedlings appeared in the elevation of 1,600-1,700 m due to no distribution of S. borealis and T. regelii species in that altitude range. Therefore, we concluded that those seedlings and saplings of A. koreana could be more stable in the altitude of 1,600-1,700 m.

Studies on the Preservation of Soy Sauce -Part I. The Periodical Change of Chemical Composition and Microflora- (제품 간장의 보존에 관한 연구 -제1보 일반성분 및 미생물의 경시적 변화-)

  • This experiment was carried out to investigate the chemical composition and microflora of soy sauce during storage under the different temperature. The results obtained are as follows. (1) Total nitrogen, color density, specific gravity and sodium chloride concentration of soy sauce showed a increasing tendency in the progress of storage period. Open-storage state at $30^{\circ}C\;and\;15^{\circ}C$ were responsible to the increase of components as compare with close-storage state at $5^{\circ}C$. (2) pH and buffer action were not almost changed during the storage. (3) Alcohol and sugar contents of soy sauce showed a decreasing tendency in the process of storage period, especially in the case of open state alcohol being almost disappeared within 11 months in all groups. (4) The number of common bacteria in one ml of soy sauce were counted as $96{\times}10^4$ before pasteurization and $10^3$ after pasteurization. The osmophilic bacteria was counted as $38{\times}10^4$, $10^2$ after pasteurization. (5) The spore number of mold in one ml of soy sauce were counted $32{\times}10^7$ before pasteurization, 58 after pasteurization and 10 to $10^2$ in the progress of storage period. (6) The bacteria number of soy sauce were somewhat decreased with the passing, of the time. The group of high temperature and open state were more notable than low temperature and close state.
