Studies on the Availability of Marine Bacteria and the Environmental Factors for the Mass Culture of the High Quality of Rotifer and Artemia 1.   • To develop a beneficial microbial feed for the cultivation of rotifer, Brachionus plicatilis, an aerobic photosynthetic bacterium, Erythrobacter sp. $S\;\pi-I$ was isolated from marine structure at Haeundae beach in Pusan, Korea. Feeding effects of Erythrobacter sp. $S\;\pi-I$ on the growth of rotifer were analyzed comparing to other feeds such as PSB (purple nonsulfur bacteria), Chlorella sp. and baker's yeast. Erythrobacter sp. $S\;\pi-I$ contained more linoleic acid $(C_{18:3\omega3})$ and oleic acid $(C_{18:1\omega9})$ and amino acids than PSB (purple nonsulfur bacteria), Chlorella sp. and baker's yeast. The rotifer fed on Erythrobacter sp. $S\;\pi-I$ showed better effects than those fed on other feeds in the individual growth, size and weight. Also, the rotifer especially contained more eicosapentaenoic acid $(C_{20:5\omega3})$ and docosahexaenoic acid $(C_{22:6\omega3})$ in case of Erythrobacter sp. $S\;\pi-I$ feeding than the other feeds. In case of the feed of PSB and baker's yeast docosahexaenoic acid $(C_{22:6\omega3})$ did not show. In amino acid analysis, the rotifer fed on Erthrobacter sp, $S\;\pi-I$ showed more amino acid content comparing to those fed on other diets. Especially, arginine, isoleucine, histidine, lysine, methionine, phenylalanine, threonine, which are essential amino acid for fish growth, showed high contents. These results suggested that the aerobic photosynthetic bacterium, Erythrobacter sp. $S\;\pi-I$ would be a beneficial microbial teed for the cultivation of rotifer.

Flavor Components in Mash of Takju Prepared by Different Raw Materials (원료를 달리하여 담금한 탁주 술덧의 향기성분)

  • Flavor components in mash of Takju prepared by different raw materials such as nonglutinous rice, glutinous rice, barley and wheat flour were detected by GC and GC-MS method using non-polar column. Seven alcohols, 15 esters, 10 organic acids, 1 aldehyde, 4 benzenes, 3 phenols, 8 alkans, 2 ketones and 5 others were found in takju after 16 day of fermentation. takju by wheat flour had the most various components of volatile flavor. Treatment with addition starter had less flavor component than that without addition starter in takju by nonglutinous rice. Nine kinds of flavor components including acetic acid ethyl ester, 3- methyl-1-butanol, acetic acid, ethyl benzene, acetic acid 3-methyl butyl ester, 2-phenylethanol, 2,6-di-tert-butyl-4-methyl phenol. plumbagic acid and 1,2-benzenedicarboxylic acid dibutyl ester were commonly detected in all the treatments. Especially, 2,4,0-trimethyl-1,3-benzenediamine was isolated in takju that was made of nonglutinous rice without addition starter. Diethyl sulfide, 4-methoxy benzaldehyde, docosane and 2-methyl propyl octadecanoic acid were isolated from takju by nonglutinous rice with addition starter. Propionic acid ethyl ester, acetic acid butyl ester, 2-methyl butane and 3-methyl pentane were isolated from takju glutinous rice. 2-Hydroxy-4-methyl pentanoic acid and 2-methyl tridecane were isolated from akju by barley 3-(Methylthio)-1-propanol. hexanoic acid ethyl ester, butanoic acid monomethyl ester, tridecanoic acid, ethyl tetramethyl cyclopentadiene and 1,5-diaza-2,9-diketocyclotetradecane were isolated from takju by wheat flour. Major volatile flavor components were acetic acid ethyl ester, 3-methyl-1-butanol, acetic acid and 2-phenylethanol.

Inhibitory Effect of the Hexane Extract of Saussurea lappa on the Growth of LNCaP Human Prostate Cancer Cells (목향 헥산추출물의 LNCaP 전립선암세포 증식 억제 효과)

  • Saussurea lappa (SL) has been used to reduce abdominal pain and tenesmus in traditional oriental medicine. SL and major compounds of SL, sesquiterpene lactones, have been suggested to possess various biological effects, including anti-tumor, anti-ulcer, anti-inflammatory, anti-viral and cardiotonic activities. Recently, it has been reported that ethanol extracts from roots of SL have antiproliferative effects on gastric cancer cells. To explore the possibility that SL has chemopreventive effects on prostate cancer, we examined whether the hexane extract of SL (HESL) inhibits the growth of LNCaP human prostate cancer cells. Cells were incubated with various concentrations ($0{\sim}4$ mg/L) of HESL in DMEM/F12 containing 5% charcoal stripped fetal bovine serum. HESL substantially decreased viable cell numbers and induced apoptosis of LNCaP cells in dose-dependent manners. HESL increased the levels of cleaved caspase-8, -9, -7 and -3, and poly (ADP-ribose) polymerase. HESL increased the levels of the pro-apoptotic Bak and truncated-Bid proteins whereas it had no effect on the anti-apoptotic Bcl-2, Bcl-xL, or Mcl-1. The present results indicate that HESL inhibits the growth of human prostate cancer cells by inducing apoptosis, which involves the activation of the caspase cascades.

Angiotensin Converting Enzyme Inhibition and Antihypertensive Effects of Phyllostachys pubescens Culm Extracts in Spontaneously Hypertensive Rats (맹종죽 줄기 추출물을 투여한 본태성 고혈압 쥐(SHR)의 ACE 저해 활성 및 혈압 강하 효과)

  • This study investigated the anti-hypertensive effect of Phyllostachys pubescens culm extract (PCE) by examining its effects on renal angiotensin-converting enzyme (ACE) inhibition and blood pressure using the spontaneously hypertensive rat (SHR) system. Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were measured weekly for 8 weeks. Also, total antioxidant capacity and protein oxidation of tissues were examined by plasma Trolox Equivalent Antioxidant Capacity assay (TEAC) and hepatic protein carbonyl values, respectively. Twenty male SHR were randomly divided into four groups: PCE50, PCE100, and PCE500 (50, 100, and 500 mg of PCE per kilogram bodyweight daily, respectively), and control group. At week 2, the SBP in all PCE groups appeared to be significantly lower than the control (p<0.05), whereas the DBP were not different until week 4 (p<0.05). At week 8, SBP in the PCE500 was lower by 20% than the control. PCE groups considerably suppressed ACE dose-dependently compared with the control. Plasma TEAC and hepatic protein carbonyl values indicated increased antioxidative activity due to the PCE feed. No adverse effect was observed on the liver of SHR as there was no difference for the GOT and GPT values among the groups. Results of this study suggest that ACE inhibition may be one possible mechanism for the blood pressure lowering effect of PCE; thus, long term consumption of PCE may be beneficial in preventing high blood pressure along with the increased antioxidative status.

Analysis of Characterization in Commercial Extra Virgin Olive Oils (유통 압착올리브유의 이화학적인 특성)

  • To analyze and differentiate volatile compounds of 13 extra virgin olive oils from market, solid-phase micro extraction (SPME) GC-MS and electronic nose (EN) equipped with metal oxide sensors were applied. The volatiles identified in extra virgin olive oils include hexanal, 4-hexen-1-ol, (Z)-3-hexen-1-ol, acetic acid, and 2,4-dimethyl-heptane, etc. Response from EN was analysed by the principal component analysis. Proportion of the first Principal component was 99.70%, suggesting that each aroma pattern of the 13 extra virgin olive oils could be discriminated by EN. Fatty acid compositions were oleic (61.1${\sim}$77.9 mole%), palmitic (11.7${\sim}$16.5 mole%), linoleic (4.7${\sim}$9.7 mole%), stearic (2.5${\sim}$2.9 mole%), Palmitoleic (0.8${\sim}$2.4 mole%), and linolenic acid (0.7${\sim}$1.2 mole%). In color study, extra virgin olive oil showed $L^{\ast}$ value of 81.7${\sim}$92.9, $a^{\ast}$ value of -28.3${\sim}$13.5 and $b^{\ast}$ value of 52.2${\sim}$139.0. Total phenol and ${\alpha}-tocopherol$ contents were 6.2${\sim}$24.9 mg/100 g and 5.5${\sim}$12.8 mg/100 g, respectively. In Rancimat test, the induction period of 13 extra virgin olive oils showed 31.76${\sim}$54.04 hr while their POV ranged from 13.5 to 22.9 meq/kg oil.

The Inhibitory Effects of Pogostemon cablin Bentham Extract on Melanogenesis (광곽향 추출물의 멜라닌 생성 저해 효과)

  • To develop a new natural whitening agent for cosmetics, we investigated the inhibitory effects of Pogostemon cablin Bentham extracts (PCE) and its active component on melanogenesis. PCE showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $24.2{\pm}2.85{\mu}g/mL$ and $IC_{50}=118{\pm}0.43{\mu}g/mL$, respectively. PCE reduced melanin contents of B16 melanoma cells in a dose-dependant manner and decreased to about 23 % at a concentration of $20{\mu}g/mL$ without cell cytotoxicity (below $100{\mu}g/mL$). And the PCE reduced intracellular tyrosinase activity about 18 % at concentration of $50{\mu}g/mL$. We purified one active compound from PCE and identified its structure. It was identified as patchouli alcohol, sesquiterpene family, by 1H-NMR, $13_C$-NMR, and Mass analysis. Patchouli alcohol also inhibited ROS scavenging activities in DPPH radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.14{\pm}0.12{\mu}g/mL$ and $49{\pm}3.24{\mu}g/mL$, respectively. Patchouli alcohol inhibited melanin synthesis in a dose dependent manner ($IC_{50}=3.9{\mu}g/mL$). And the patchouli alcohol reduced intracellular tyrosinase activity about 40 % at concentration of $10{\mu}g/mL$. Patchouli alcohol inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that PCE and patchouli alcohol reduced melanin formation by the inhibited of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that PCE could be used as a useful whitening agent.

The taxonomic consideration of leaf epidermal microstructure in Korean Thymelaeaceae Adans (한국산 팥꽃나무과 잎표피 미세구조의 분류학적 검토)

  • To examine the leaf epidermal microstructure, nine species in five genera (Daphne L. - 4 spp., Diarthron Turcz. - 1 sp., Edgewarthia Meisn. - 1 sp., Stellera L. - 1 sp., Wikstroemia Endl. - 2 spp.) of the Korean Thymelaeaceae were investigated by light microscopy (LM) and scanning electron microscopy (SEM). The stomata of stuo야ed taxa were 'hypostomatic type' and the size range of guard cell was $13.8-34.4{\times}8.7-22.9{\mu}m$: the smallest size of stomata was found in Diathran linifolium ($15.9{\pm}2.6{\times}10.0{\pm}1.3{\mu}m$), while the largest one was measured to Daphne adara ($32.8{\pm}1.6{\times}20.7{\pm}1.3{\mu}m$). The stomatal complex was anomocytic in the most studied taxa, except Daphne kiusiana by having combined with anisocytic together. The shapes of epidermal cells are undulate anticlinal wall. The size range of epidermal cell was $20.7-61.0{\mu}m$; the smallest size of epidermal cell was found in Stellera charnaejasme ($26.0{\pm}1.9{\mu}m$), on the other hand the largest one was found in Edgeworthia chrysantha ($53.6{\pm}3.1{\mu}m$). The well-developed flaky epicuticular waxes can be divided three kinds of pattern - (1) smooth in comparison, not entire platelets and scattered, (2) isolated flake-like platelets, mostly paralleled, sparsely, (3) flake-like platelets, flat, membraneous, protruding from the surfaces at varying angles and densely. Two types of trichome are recognized; (1) Type I: uniseriate trichome of striate surface (D. genkwa, Diarthron linifalium, E. chrysantha, W. ganpi and W. trichotama), (2) Type II: multicellular trichome of papillose surface, uncinated 3-4 nodes (Diathron linifolium). Finally, the systematics significance of the leaf micromorphological features in identification and elucidation of Korean Thymelaeaceae, especially between or within the genera including among the species is also briefly discussed.

The Temperature-Dependent Development of the Parasitoid Fly, Exorista Japonica (Townsend) (Diptera: Tachinidae) (항온조건에서 긴등기생파리 [Exorista japonica (Townsend)] (Diptera: Tachinidae) 온도별 발육)

  • Exorista japonica is one of the major natural enemies of noctuid larvae, Mythimna separata and Spodoptera litura. The examined parasitoid was obtained from host species M. separata, collected at Gimje city and identified by DNA sequences (partial cytochrome oxidase I, 16S, 18S, and 28S). For purposed of this study, laboratory reared S. litura served as the host species for the development of the E. japonica. The developmental period of E. japonica immature stages were investigated at seven constant temperatures (16, 19, 22, 25, 28, 31, $34{\pm}1^{\circ}C$, RH 20~30%). Temperature-dependent developmental rates and development completion models were developed. E. japonica was successfully developed from egg to adult in $16{\sim}31^{\circ}C$ temperature regimes. Developmental duration was the shortest at $34^{\circ}C$ (8.3 days) and the longest at $16^{\circ}C$ (23.4 days) from egg to pupa development. Pupal development duration was the shortest at $28^{\circ}C$ (7.3 days). Total immature-stage development duration decreased with increasing temperature, and was the shortest at $31^{\circ}C$ (16.3 days) and the longest at $16^{\circ}C$ (45.4 days). The lower developmental threshold was $7.8^{\circ}C$ and thermal constant required to complete total immature-stage development was 370.4 degree days. Among four non-linear temperature-dependent developmental rate models, Briere 1 model had the highest adjusted R-squared (0.96). The distribution model of development completion for total immature stage development of E. japonica was well described by all model ($r^2_{adj}=0.90$) based on the standardized development duration. These results of study would be necessary not only to develop population dynamics model but also to understand fundamental biology of E. japonica.

Protective Effects of Enzymatic Oyster Hydrolysate on Acetaminophen-induced HepG-2 Cell Damage (아세트아미노펜 유도 HepG-2 세포주 손상에 대한 굴 효소 가수분해물의 보호 효과)

  • This study investigated the detoxification effects of enzymatic hydrolysate from oyster on acetaminophen-induced toxicity using HepG-2 cells. Oyster hydrolysate was made with 1% Protamex and 1% Neutrase after treatment with transglutaminase (TGPN) or without (PN). Two types of oyster hydrolysate were added to human-derived HepG-2 hepatocytes damaged by acetaminophen, after which the survival rate of HepG-2 cell was measured. In addition, glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) activities in the culture media were evaluated. The survival rates of HepG-2 cells were $136.2{\pm}1.4%$ at $100{\mu}g/mL$ of TGPN and $179.6{\pm}3.8%$ at $200{\mu}g/mL$ of TGPN. These cell survival rates were higher compared to that of the negative control group ($60.7{\pm}3.2%$) treated only with acetaminophen. GOT activity was $38.3{\pm}0.2$ Karmen/mL in the negative control group, whereas it was $19.9{\pm}0.5$ for TGPN ($200{\mu}g/mL$) and $22.0{\pm}2.4$ Karmen/mL for PN ($200{\mu}g/mL$). GOT and GTP activities were shown to be dependent on TGPN concentration, and significant reduction in activities could be conformed. The detoxification efficacy of TGPN was higher compared to that of PN. These results suggest that oyster hydrolysate has potential as a healthy food or pro-drug for liver protection.

Non-Starch Polysaccharides of Cell Walls in Glutinous Rice, Rice and Black Rice (점미, 백미, 흑미 세포벽의 비전분성 다당류의 성분분석)

  • The non-starch polysaccharides in the cell wall of rice, glutinous rice, and black rice, were analyzed. They were fractionated into fractions; water-soluble, hot writer-soluble, ammonium oxalate-soluble, sodium hydroxide-soluble, potassium hydroxide-soluble, and the alkali-insoluble, according to the solvent solubility. The dietary fiber contents were 5.4% in glutinous rice, 4.2% in rice and 7.5% in black rice. The sodium hydroxide soluble fibers were abundant in each kind of rice, especially 4.01% in black rice. The water soluble fibers were 80% of dietary fiber in glutinous rice, 66% in rice, 86% in black rice. It was supposed that the content of the water soluble fibers in rice was increased by pounding. Pectic substances in water soluble fibers, hot water soluble fibers, and ammonium oxalate soluble fibers fraction, were 2.4% in glutinous rice fraction,1.59% in rice, and 1.12% in black rice. Alkali soluble fibers were considered as hemicellulose. Black rice contained 5.80% of hemicellulose, which was more than twice as much as glutinous rice(2.58%) and rice(2.22%). Alkali insoluble fibers were considered as cellulose, which showed no considerable difference. Among samples content of uronic acid in glutinous rice, rice and black rice were 0.9%, 0.66%, 1.8% respectively. Uronic acid of black rice was twice more than other samples tested. The fraction of black rice that uronic acid was extracted at most was the fraction of sodium hydroxide. Mono saccharides of the fraction was the glucose, the arabinose, the xylose.
