  • 우리나라는 계절적 특성으로 인하여 겨울에서 봄에 이르기까지 도로포장의 동결 융해가 반복되어 도로 파손을 유발 시키고 있다. 동결 융해로 인한 피해를 줄이기 위하여 도로포장 설계시 동상방지층을 별도로 설치를 하고 있으나 실효성 검증 없이 무분별하게 적용되고 있어 국가적으로 예산을 낭비하고 있다. 본 연구에서는 효과적인 동상방지층의 설계를 목적으로 계측된 온도 및 함수비 데이터를 효율적으로 관리하고 모니터링하는 기반을 마련하고자 한다.

Evaluation of Freeze-Thaw Effect on the Modulus of Subgrade Soils from Impact Resonance Test (충격공진시험을 이용한 동결.융해에 따른 노상토의 탄성계수 평가)

  • Anti-freezing layer does not used in case of non frost heaving in subgrade soils. In this case, the modulus of subgrade soils were varied with freezing and thaw cycles under non frost heaving. That effect should be properly considered in pavement design. Impact resonance test that is nondestructive testing method was used for continuously determining the modulus of subgrade soils during freezing and thaw cycle. The modulus of subgrade soils was identical with freezing and thaw cycles under closed freezing and thaw system which is no water supplement into specimen during testing. There was also no difference in the modulus of subgrade soil between before and after freezing-thaw cycles for all specimens with different water content and density. That is thaw-weakening of subgrade soils do not occur under closed freezing and thaw system. The moduli at freezing conditions are varied with water content and density, but it can be ignored in practical design sense.

  • This study was estimated on performance of deicers, corrosion inhibitors and low corrodible deicer used for removal of snow or ice on the road and on influence on structure. The weight loss rate after freezing and thawing of low corrodible deicer is lower than one of deicer, corrosion inhibitors and these combination. Relative dynamic elastic modulus of all except water, low corrodible deicer and NaCl+JF-1004 was radically reduced after freezing and thawing 150 cycles. And concretes after freezing and thawing were showed severe surface damage. It was found that individual use of low corrodible deicer and corrosion inhibitors had a problem of field application because of lack of early ice melting effect and considerably low durability. Products combined with NaCl was showed rapid weight loss by metal corrosion. Therefore, It will need to circumspectly select combination of deicers having low effect on concrete pavement and bridge if possible.

Classification of cold regions and analysis of the freeze-thaw repetition cycle based on heat transfer quantity by freezing test (실내동결시험을 통한 열류량 분석에 따른 동결-융해 조건 분석 및 한랭지역의 분류)

  • Tunnels constructed in cold regions can cause serious defects such as cracks and leaks due to external temperature changes in the portals and vents. In order to prevent the freezing damage of the tunnel, appropriate measures should be applied to the section where the freeze damage is concerned. However, the specific criteria and contents for judging whether or not the anti-freeze measures are applied are not presented. In this study, the laboratory freezing tests on the temperature changes of the concrete specimens under freezing conditions were carried out. And the freeze-thaw repetition cycle (F), which can judge the possibility of freezing damage, were presented based on the heat transfer quantity (W) by experimental results of case studies. Also, we propose a classification of cold regions considering the climatic characteristics of Korea for using it to efficient design and maintenance.

  • Structures in our surroundings deteriorate over time due to environmental and chemical factors, resulting in a decrease in their performance. The primary causes of degradation in concrete structures are carbonation, salt damage, and freeze-thaw cycles. Various maintenance methods exist to address these degradation issues. However, research and technological development for existing maintenance methods have been ongoing, but the accuracy and effectiveness of repair materials and techniques have not been extensively validated. Therefore, in this study, we conducted a material performance evaluation of various manufacturers' repair materials. Based on this evaluation, we applied corrosion inhibitors and epoxy, which are the methods most closely related to crack repair, to assess the durability performance against carbonation, salt damage, and freeze-thaw cycles. The results show approximately a two-fold performance improvement against carbonation and salt damage, and a 5% enhancement in repair performance against freeze-thaw cycles. Thus, it is considered effective in preventing rebar corrosion when appropriate maintenance is carried out according to environmental and chemical factors during structural repairs.

Motility of Rooster Spermatozoa under Different Thawing Conditions (닭 동결정액 융해방법이 정자 운동성에 미치는 영향)

  • In this study, to increase the survival rate of frozen/thaw rooster semen, standard protocols of semen thawing procedures were tested by computer-assisted sperm assay (CASA). We tested 4 different thawing protocols for frozen semen, $5^{\circ}C$ for 2 min, $35^{\circ}C$ for 30 s, $54^{\circ}C$ for 13 s, and $70^{\circ}C$ for 7 s. The pooled semen from 5 to 8 Ogye rooster line was diluted in the HS-1 diluent and frozen in 8% methylacetamide (MA) in liquid nitrogen vapors. To determine standard thawing method, straws were plunged into different temperatures and times. The resulting motilities were recorded by the CASA system. The results of this study showed that the best viability of the spermatozoa was shown by exposure at $5^{\circ}C$ for 2 min. Moreover, the longevity test of thawed sperm at $5^{\circ}C$ for 2 min also supported the higher viability under low temperature preservation of $17^{\circ}C$ for 1 hr. Further research is needed to increase the motility of thawed rooster semen for field application. In addition, the in vivo tests for different rooster lines are also needed for the establishment of avian genetic resource bank.

Effect of Superoxide Dismutase and Cryoprotectants on Viability of Frozen-thawed Porcine Oocytes by Vitrification Method (Vitrification법에 의한 돼지 난자의 동결-응해 후 생존능력에 있어서 동해보호제와 Superoxide Dismutase의 영향)

  • This study was performed to investigate the effect of different cryoprotectants and superoxide dismutase(SOD) on viability of frozen-thawed oocytes by vitrification method in the pig. The proportions of oocytes matured to metaphase-I stage were higher in medium with ethylene glycol and DMSO(19.9%) than in medium with glycerol and DMSO(6.5%). When the oocytes were exposed in medium containing ethylene glycol, oocyte matured to prophase-I were not observed. On the other hand. significant differences were not observed between in medium with and without SOD( 1 unit/$m\ell$) during IVM of vitrified and thawed immature oocytes. However, the maturation rate from metaphase-I to metaphase-II were higher in medium with that than without SOD. The penetration rates after IVF of oocytes frozen-thawed were also higher in medium with that than without SOD. These results indicate that frozen-thawed oocytes treated with ethylene glycol and DMSO was more protective against freezing effect and that addition of 1 unit/$m\ell$ SOD in medium fur prevent of lipid peroxidation may play a positive role in improving of viability of frozen-thawed oocytes.

  • Korea has seasonal weathers which result in the frosting of soil in winter times, and the thawing of soil in spring. These climate characteristics result in the damaging of pavements, due to the repeated freezing and thawing of road pavements during winter and spring. In order to reduce these pavement damages, anti-frost heave layers are being specially installed, however it is being applied based on foreign researches, and therefore result in the waste of national budget. With this study, a database system was constructed for effective management and monitoring of measured temperatures and function data of 2 meters below the embankment, cut slope, and the cutting-embankment boundary, which are 15 regions picked by the frost index diagram. As the study result, an effective storage and management-purpose database was established for easy data searching and downloading for the pavement design engineers.

Developmental Capacity of Porcine Oocyte Frozen-Thawed at Immature, Maturing and Mature Stages (각 성숙단계에서 동결ㆍ융해한 돼지 난포란의 발달능력에 관한 연구)

  • These experiments were conducted to investigate the optimal maturation stage for cryopreservation of porcine oocyte when the oocytes were frozen-thawed and/or exposed in cryoprotect ant at immature, maturing and mature stages. The results of this research are as follows ; 1. When the oocytes matured for 0, 24 and 44h were exposed in media containing cryoprotectants or without in vitro, the rates of cultured oocytes developed to metaphase II were 44.0, 45.0, 50.3 or 55.0%, respectively. 2. When the oocytes matured for 0, 24 and 44h were exposed in media containing cryoprotectants or without in vitro, the cleavage rates of cultured oocytes were 18.6, 19.7, 47.6 or 50.9%, respectively. 3. When the oocytes matured for 0, 24 and 44h were frozen and thawed using vitrification or not in vitro, the rates of cultured oocytes developed to metaphase II were 4.3, 7.1, 46.7 or 62.4%, respectively. 4. When the oocytes matured for 0, 24 and 44h were frozen and thawed using vitrification or not in vitro, the cleavage rates of cultured oocytes were 2.5, 2.4, 10.2 or 49.6%, respectively.

여러 종류의 동해방지제를 이용한 소 체외수정란의 동결 및 일단계 융해후의 체외배양 생존율 및 수태율

  • 치밀 난구세포로 둘러싸인 소 난자를 $38^{circ}C$. 5% $CO_2$ 배양기에시 5% superovulated cow serum(SCS)이 첨가된 m-TCM 199 medium 으로 $20{\sim}22$ 시간 배양하였으며, 수정능이 획득된 정자와 체외수정하였다. 7일${\sim}$8일경의 수정란을 1.3M methyl cellosolve(MC), 1.1M diethylene glycol(DEG), 1.8M ethylene glycol(EG), 1.6M propylene glycol(PG) 및 1.1M 1,3-butylene glycol(BG) 용액에서 10분간 평형시킨 후 0.25 ml 스트로내에 장전하였다. 스트로를 $0^{\circ}C$의 alcohol bath freezer에 넣고 $-6^{\circ}C$까지 $-1^{\circ}C$/분 속도로 냉각, 식빙 후 10분간 정체시켰으며, $-0.3^{\circ}C$/분 또는 $-0.5^{\circ}C$/분으로 $-30^{\circ}C$까지 냉각 후 스트로를 액체질소에 침지하여 보관하였다. 수정란이 들어있는 스트로를 $30^{\circ}C$ 온수에서 융해하였으며, 수정란을 TCM 199 medium 으로 옮긴 후 5% SCS가 첨가된 TCM 199 medium 에서 48시간 배양하였다. 수정란이 양호한 형태를 유지하며 나중의 발육단계로 진행된 것을 생존한 것으로 간주하였다. 각 종류의 동해방지제에서 동결된 수정란의 일부는 융해 후 동해방지제를 제거하지 않고 직접 비외과적으로 이식하였다. 동결-융해 후 동해방지제의 종류에 따른 탈출배반포 발달율은 EG 50.0%, MC 53.6%, DEG 56.9%, PG 58.0% 그리고 BG 11.5%였다. $-0.3^{\circ}C$/분 또는 $-0.5^{\circ}C$/분 으로 냉각한 수정란의 생존율은 두 그룹간에 유의적인 차이가 없었으나 (P<0.05), 탈출배반포 발달율은 -0.5분 $^{\circ}C$/분(22.6%, 12/53)보다 $-0.3^{\circ}C$/분(64.6%, 31/48) 냉각시에 유의적으로 높았다(P<0.01). 동해방지제의 종류에 따른 수정란의 수태율은 MC 48%(10/21). DEG 30%(3/10), EG 74%(20/27) 및 PG 40%(4/10) 였다. 이러한 결과로 보아 MC, DEG, EG 그리고 PG는 소의 체외수정란의 동결을 위한 동해방지제로서 이용될 수 있음을 보여주었다.
