Effect of Lipid and Salt Contents on the Mutagenicity of Charcoal-Broiled Meats and Fishes (숯불구이시 육류의 지방질 및 소금 함량이 돌연변이 유발능에 미치는 영향)

  • The effect of lipid content and saline seasoning on the mutagenicity during the charcoal-broiling Process of beef, Pork and fish samples was examined by Ames test using Salmonella typhimurium TA98 and TA100 strains. Chloroform; methanol(1:1) extract of broiled samples showed a higher sensitive response toward TA98 strain than TA100 strain, indicating a frameshift mutation. The three samples of low lipid content demonstrated a slightly higher mutagenic activity, and the beef and pork samples treated with 20% saline solution showed a remarkable reduction in mutagenicity than the untreated samples.

  • Mutagenic action of p-phenylenediamine(PA) and nitro-p-phenylenediamine(NPA) has been investigated using auxotroph mutants of S.typhimurium LT-2 strain. PA, the major component of hair day in South and East Asia and South America, was proved as potent frams-shift mutagen only after activation system. On the contrary, NAP was directly mutagenic in this system.

  • The ethanol-and water-extracts of curled tops and stalks of young bracken (Pteridium aquilinum) fronds in raw and cooked states were examined for their mutagenicity by Ames test using Salmonella typhymurium mutants. Even a small amount of water extract from raw bracken was mutagenic to TA 1538 strain without addition of $S_9$ mix whereas a strong toxicity appeared at higher levels of the extractive. With $S_9$ mix, the water-extract from raw bracken was not mutagenic at all and not toxic to all strains tested, The amount of water-extract from cooked bracken was one-tenth of the amount from raw bracken and the water-extract showed no mutagenic effect to all strains tested. Ethanol-extract of bracken showed no mutagenicity in any case.

  • This study was undertaken to observe the mutagenic occurrence in urine excreted after the ingestion of roast beef. Two healthy nonsmoker persons of both sex were selected for this test, employing two strains(TA98, TA100) of Salmonella typhimurium according to Ames' method. The mutagenic activity began to appear in urine of both sex three hours after ingestion of 300g of roast beef, gradually increasing until 6 hours and declining thereafter.

  • In spite of high incidences in gastrointestinal cancers in Korea, there have been few studies so far, which tried to detect or isolate the mutagenic, namely the carcinogenic substances in the Korean taste traditional foods. Among the many plausible dietary sources of carcinogens, the pyrolytic products of proteins or amino acids would be the major ones. And it is more noteworthy that the food pattern analysis showed the increasing consumption of protein foods by Korean as pyrolytic products, such as Bulgogi and Kalbi, etc.

Antioxidative and Antimutagenic Effects of Agaricus bisporus Ethanol Extracts (양송이버섯 추출물의 산화적 스트레스 억제효과 및 항돌연변이 효과)

  • This study was performed to investigate the antioxidative effects such as the inhibition of malondialdehyde (MDA) and bovine serum albumin (BSA) conjugation reaction, inhibition of $Fe^{2+}$-induced lipid peroxidation and the scavenging effect on 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical, as well as antimutagenic capacities as Ames test in ethanol extracts of Agaricus bisporus. Agaricus bisporus ethanol extracts inhibited $Fe^{2+}$-induced lipid peroxidation and scavenged DPPH radical. The $IC_{50}$ of Agaricus bisporus ethanol extracts were 78.63 mg/assay for inhibition of MDA with BSA conjugation reaction, 4.06 mg/ assay for inhibition of $Fe^{2+}$-induced lipid peroxidation and 1.08 mg/assay for scavenging effect on DPPH radical. So, among the methods used in this study, the most effective antioxidative capacity in ethanol extracts of Agaricus bisporus was the scavenging effect on DPPH radical. The indirect and direct antimutagenic effects of ethanol extracts of Agaricus bisporus were examined by Ames test using Salmonella Typhimurium TA98 and TA100. The inhibitory effects on direct mutagenicity mediated by sodium azide in Salmonella Typhimurium TA100 and 2-nitrofluorene in Salmonella Typhimurium TA98 were 100%. The inhibition rates on indirect mutagenicity mediated by 2-anthramine were 86.09% in the Salmonella Typhimurium TA98 and 81.93% in the Salmonella Typhimurium TA100. The ethanol extracts of Agaricus bisporus showed considerable antioxidative activity and strong antimutagenic capacity.

Browning and Mutagenicity of Roasted Barley and Sesame Seeds (볶은 보리 및 참깨의 갈색도와 돌연변이 유발성)

  • This study was undertaken to measure the degree of browning and mutagenicity by Ames test using Salmonella typhimurium TA 98 and TA 100 strains for roasted barley and sesame seeds used as food materials. The degree of browning of roasted barley for barley tea on the market showed a wide variation; barley for restaurant-use was heavily roasted (5 times) in comparison with homeuse barley. Sesame seeds for oil extraction were more roasted (4 times) than those for seasoning. Water-, ethanol- and ether-soluble fractions from roasted barley and sesame seeds did not show any signs of mutagenicity, even at the extremely high concentrations of the extracts.

유전독성물질의 평가방법과 그 기작에 관한 연구

  • 유전독성물절외 검출과 평가에 용이하게 사용할 수 있는 모델 시스템의 개발 및 DNA 회복기작을 규명할 목적으로 수종의 돌연변이, 발암원을 이용하여 배양 포유동물세포 및 어류세포에서 세포생존률, DNA 합성 및 복제억제의 양상 등을 비교 분석하였다. MMS및 MNNG 와 같은 알칼라제는 CHO 세포에서 유의한 DNA 합성저해, DNA 복재억제, DNA 단사절단 및 비주기성 DNA 합성률의 증가를 유발하였다. Benzo(a)pyrene과 3-methylcholanthrene좌 같은 DNA 상해 전구물질의 경우 유전독성 여부의 판정에는 반드시 S-9/15과 같은 대사활성계 또는 mouse embryonic fibroblast와 같은 대사 활성능이 있는 세포와의 co-culture system들이 필요함을 확인하였으며, 이들에 의한 DNA 상해와 복재억제 유도의 작용양상은 자외선의 작용양상과 유사하였다. 배양 어류세포에서 자외선에 의한 세포생존율의 측정, 광재활성능의 분석 및 자외선에 의해 유발된 피리미딘 이량체 절제능 검토 및 DNA 합성 저해능의 결과를 분석함으로써 유전독성 평가를 위한 모델 시스템 구축의 기초결과를 얻었다.

Antioxidative and Antimutagenic Activity of Ethanol Extracts from Cuscutae Semen (토사자(Cuscutae Semen) 에탄올 추출물의 항산화 효과 및 항돌연변이 활성)

  • In this study, the antioxidant and antimutagenic activities of Cuscutae semen ethanol extract were evaluated. Antioxidant activity was measured by the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method and the Ames test was employed to determine the inhibition effect on mutagenicity in Salmonella typhimurium TA100. The extract showed significant free radical-scavenging activity towards the DPPH radical, and at a concentration of 400 ppm, its free radical-scavenging activity was similar to that of BHT. The $IC_{50}$ value of the extract was 89 ppm, indicating good antioxidant capacity. Moreover, at 5 mg/mL, the extract presented inhibitions of approximately 98.0% and 49.2% on mutagenicity induced by 4-nitroquinoline 1-oxide and sodium azide, respectively. The total polyphenols and flavonoid contents of the extract were 20.1 mg/g and 1.9 mg/g, respectively. Therefore, this study indicates that the ethanolic extract of Cuscutae semen has excellent antioxidative and antimutagenic potential.

신규 항암제 DA-125의 급성 및 특수독성 연구

  • 1) 급성독성시험: 랫드와 마우스 공히 투여후 5~6일에 농도의존적의 강한 독성이 발현 되었으며 표적장기로는 소화기계, 임파계, 조혈기계 등으로 나타났다. 랫드에서의 반수 치사량은 암수 각각 771.9(505.6~1178.4)mg/kg과 479.8(302.7~760.4)mg/kg이었으며 마우스에서는 암수 각각 527.4(393.7~701.1)mg/kg과 463.0(308.8~694.2)mg/kg 이었다. 2) 유전독성: S. typhimurium을 이용한 복귀돌연변이시험, CHL세포를 이용한 염색체이상 시험, 마우스에서의 소핵시험결과 DA-125는 돌연변이 유발능을 갖는 것으로 나타났다. 3) 국소자극성: 혈관주위주사에 의한 피부자극성 시험결과 DA-125는 adriamycin의 3배 용량에서 궤양, 괴사등 유사한 병변이 유발되었으나, 병변회복은 adriamycin에 비해 빠른 것으로 나타났다. 또한 토끼에서의 안점막자극시험 결과에서 DA-125는 실제적으로 자극이 없는 것으로 나타났다.
