Spermatogenesis and Chromosome Number of the Tobacco Budworm Helicoverpa assulta Guenee (Lepidoptera: Noctuidae) (담배나방(Helicoverpa assulta: 인시목 밤나방과)의 정자형성 과정과 염색체수)

  • The spermatogenesis and chromosome number were investigated in the pupal testes of Helicouerpa assulta Guenee by light microscopy. During the spermatogenesis, each bundle of P8(256) sperms developed by 6 mitotic and 2 meiotic spermatogonial divisions. From the early stage of spermatogenesis, it was distinguishable between two kinds of sperm differentiation, eupyrene and apyrene spermatogenesis, which are characteristic in Lepidoptera, by the differences in nuclear shape and cell distribution in immature spermatocyst. Through the followed spermiogenesis, the spermatocysts were developed into two kinds of mature cyst, a streamline-shaped eupyrene cyst with nucleated sperms of thready head or a long spindle-shaped apyrene cyst with anucleated sperms of cylindrical head. As the results off chromosomal analysis at metaphase of the spermatogonial mitosis and spermatocytic meiosis, the chromosome number were 2n=6a/n=31, respectively, and no variation between individuals.

Isolation and Activity of Insect Pathogenic Bacillus thuringiensis Strain from Soil (국내 토양으로부터 곤충병원성 세균인 Bacillus thuringiensis 균주의 분리 및 생물검정)

  • Bacillus thuringiensis strains were isolated from the domestic soil and a strain was selected that had a new host range and high toxicity against agriculture insect pest. The 142 samples of soil were sampled from the mountains, paddy fields and patches, in Daejon, Chungnam, Chungbuk and Jeonbuk and used for the investigation. Sixteen B. t strains were isolated from 12 samples among collected samples. There were 11 strains that showed toxical activity on Plutella xylostella (Lepidoptera: Yponomeutidae), 7 steins on Spodoptera litura (Lepidoptera: Noctuidae), 5 strains on Arete coerulea (Lepidoptera: Noctuidae), 5 strains on Culex pipiens pallens (Diptera: Culicidae) among the 16 isolated B. t strains. But there were not any strains that showed activity against Hyphanria cunea (Lepidoptera: Arctiidae) and Sitophilus oryzae (Coleoptera: Rhynchophoridae). And also some of B. thuringiensis strains showed insecticidal activity with 2, 3 or 4 kinds of insects. But there were also 3 strains that did not show any activities to the 6 insects which were used in the experiment. When examined with a phase-contrast microscope, the insecticidal crystal protein produced from 16 selected strains had 13 bipyramidal and 3 spherical shapes. The insecticidal bioactivity of the S. litura showed 100% mortality when there were $1.3{\times}10^{7}\;(cfu/ml)$ of CAB109 isolates.

Comparison of Gene Expression in Larval Fat Body of Helicoverpa assulta in Different Temperature Conditions (온도변화에 따른 담배나방 유충 지방체의 유전자 발현 비교 분석)

  • Insects are known to live at wide range of temperature, but can not survive when they are exposed to over $40^{\circ}C$ or below supercooling point. The larvae of Helicoverpa assulta have been reared at high ($35^{\circ}C$), low (3 to $10^{\circ}C$), and room temperature ($25^{\circ}C$; control). To identify stress-related genes, the transcriptomes of fat body have been analyzed. Genes such as cuticular proteins, fatty acyl ${\Delta}9$ desaturase and glycerol 3 phosphate dehydrogenase were up-regulated whereas chitin synthase, catalase, and UDP-glycosyltransferase were down-regulated at low temperature. Superoxide dismutase, metallothionein 2, phosphoenolpyruvate carboxykinase and trehalose transporter have been up-regulated at high temperature. In addition, expressions of heat shock protein and glutathione peroxidase were increased at high temperature, but decreased at low temperature. These temperature-specific expressed genes can be available as markers for climate change of insect pests.

  • Embryonic developmental rates of the Oriental tobacco budworm, Heliothis assulta Guenee, were compared at various constant temperatures and 16 hours of light, and detailed embryogenesis was also studied at $25^{\circ}C$. The egg was nearly globular in form and had an average equatorial diameter of 0.53mm. A single micropyle was in the center of the circular area at the anterior pole of the egg. Durations of embryogenesis at 20, 25, 30, and $35^{\circ}C$ were 147, 81, 61, and 67hrs, respectively. Embryonic death was especially higher at $35^{\circ}C$ than any other temperatures investigated. Embryogenesis progressed with changes in color and pattern, which are quite characteristic at each developmental stage of the embryo. At $25^{\circ}C$, organogenesis began in 14hrs after oviposition, formation of gut completed in 44hrs and eclosion occurred in 80hrs. The embryo formed along the long axis at early developmental stage, moved towards the equatorial plane in 24hrs, and made a half-turn on the plane in 36hrs. In 40hrs, head was oriented to the anterior pole of the egg until eclosion.

Structure of sensilla on the Antenna and Mouthparts of the Oriental Tobacco Budworm(Heliothis assulta Guenee) Larvae (담배나방의 유충의 촉각 및 구기 감각기의 구조)

  • Studies were carried out to describe the structure and distribution of various sensilla on the antenna and mouthparts of the Oriental tobacco bud worm (H. assulta) larvae by light and scanning electron microscopy. The larval antenna has 3 segments. The second segment has 2 hairs and 3 pegs, while the third segment shows 3 regular pegs and a segmented peg (styloconicum type). Each mandible bears 2 aporous hairs on the lateral surface. The labrum bears 12 aporous hairs on its external surface. and 6 aporous hairs, 4 coeloconica-like sensilla and 2 campaniformia-like sensilla on its epipharynx. The stipes has 3 hairs, and the galea has 3 pegs. 2 papillae and 2 domes. The maxillary palpus has only a digitiform sensillum on its 1st segment but 11 sensilla of 4 different types on its 2nd segment. Maxillary sensilla represent almost 70% of total number of larval sensilla on the mouthparts. Labial pal pus has a single segment bearing a large segmented uniporous peg and a small peg. Finally their possible fun¬ctions were suggested.

Behavior and Circadian Rhythm of Emergence, Copulation and Oviposition in the Oriental Tobacco Budworm, Heliothis assulta Guenee (담배나방의 우화, 교미 및 산란의 행동과 일일리듬)

  • The behavior and circadian rhythm of emergence, copulation and oviposition in the Oriental tobacco budworm (Heliothis assulta Guenee) were studied to obtain the following results. More than three quarters of adults emerged within 3 hours after the light-off. It took about 33 min for an adult to expand and tan its wings after emergence. Adult male showed a sequential pattern of mating behavior, such as antennal movement, wing elevation and vibration, exten¬sion of hairpencils, and tapping of female ovipositor leading to a copulation. However, adult female revealed a rather simple behavior, such as protraction and retraction of her terminal abdomen and vibration of wings. All of the mating took place during the early part of the scotophase, with the peak from 1 hr to 2 hrs after the light-off. But the mating peak in virgin females tended to advance with age. Adult females showed the highest rate of mating among 24 hr-olds and the rate declined with age. The rate of first matings in males gave a very similar pattern to that of females. The duration of copulation was $77\pm$14min. Oviposition occurred throughout the entire scotophase, with the peak during the first 2 hrs. The most nu¬mber of eggs deposited daily was seen on the 3rd day after the mating. Mating rate, number of eggs laid and longevity we~e compared under different sex ratio. Unmated adults lived longer than mated adults.

Status of the Occurrence of Insect Pests and Their Natural Enemies in Soybean Fields in Honam Province (호남지역 콩 재배지 주요해충 및 천적류)

  • This survey was conducted to illustrate insect pests and their natural enemies in soybean fields in Honam area. A total of 23 pest species in 13 families and 13 species of natural enemy were observed during 2002 to 2003. Delia platura, Medythia nigrobilineata, Spodoptera exigua and Aphis glycines occurred relatively with high population densities during the early growth stage of soybean. Dolycoris baccarum, Riptortus clavatus, Nezara antennata, Piezodorus hybneri and Halyomorpha halys began to occur between mid July to early August and lasted until the harvest time. Lepidopteran insect pests, Leguminivora glycinivorella, Pleuroptya ruralis, Scopula missaria, Macdunnoughia purissima and Spodoptera litura occurred significantly in late August in soybean field. Natural enemies of soybean insect pests observed in this survey were seven parasitoids, five predators, and an entomopahogenic fungus. Two egg parasitoid species for R. clavatus, two parasitoid species for P. ruralis, and five natural enemy species (including parasite, predator and entomopathogenic fungi) for either S. litura or A. glycines were identified, respectively. Among them two egg parasitoid species, Gryon japonicum and Ooenyrtus nezarae for R. clavatus, for and one parasitoid species for P. ruralis, that belong to Bethylidae, were new to Korea.

The inhibition of chitin synthesis in Spodoptera litura by new insecticides of benzoylphenyl urea, DBI-1015 and DBI-3204 (담배거세미나방(Spodoptera litura)에서 benzoylphenyl urea계의 신규살충제 DBI-1015 및 DBI-3204의 키틴합성 저해 효과)

  • This study was conducted to investigate insecticidal mechanisms of the new insecticides DBI-1015 and DBI-3204 with label compounds of chitin precursors, [$^{14}C$] N-acetylglucosamine and [$^{14}C$] UDP-N-acetylglucosamine in Spodoptera litura. The concentrations of the insecticides for incorporation of chitin precursors into chitin were founded to be functional relationship. The result of in vivo test, $I_{50}$ (ppm) of the DBI-1015, DBI-3024 and diflubenzuron to [$^{14}C$] N-acetylglucosamine were 0.57, 0.89 and 0.26 ppm respectively, and to [$^{14}C$] UDP-N-acetylglucosaminen were 0.99, 0.53 and 0.45 ppm respectively. in vitro test of DBI-1015, DBI-3024 and diflubenzuron by integument fragments, the incorporation rate in the cuticle were low, however, $40{\sim}60%$ inhibitions were observed at $2{\mu}M$ when compared to the untreated control.
