  • 예로부터 인간은 단맛에 강한 욕구를 가지고 있었다. 따라서 설탕, 포도당, 과당 등 단맛이 강한 감미료를 선호하여 왔고, 현재까지도 식품 제조시 가장 널리 사용되고 있다. 그러나 최근에는 국민의 건강에 대한 관심이 고조 되고 설탕으로 대표되는 전통적인 일반 감미료의 충치, 비만, 당뇨병 유발 등의 부정적인 측면이 부각되고 있을 뿐만 아니라, 식품에 대한 건강 기능성이 강조되면서 기능성 감미료가 주목 받고 있다. 특히 고도의 경제성장 및 국민 소득의 향상에 따라 차별화된 기능성 감미료의 생산을 요구하는 실정이다. 이와 관련하여, 현재 식품 산업계에서는 올리고당류, 당알콜류, 고감미 감미료류 등에 많은 관심을 가지고 연구 개발에 집중하고 있다. 올리고당류의 대표적인 제품은 프락토올리고당, 갈락토올리고당, 이소말토올리고당 등이다. 현재 올리고단 시장은 성숙기로서 성장속도가 둔화되고 있으나, 최근 American Association of Cereal Chemists(AACC)등 세계적인 협회에서 수용성 식이섬유를 다당류, 올리고당, 리그닌 등을 포함하여 새롭게 정의하였고 이에 따라 해외에서는 프락토올리고당을 중심으로 수용성 식이섬유로 용도 사용이 가능하게 되었을 뿐만 아니라, 식이섬유로 표기도 가능하게 되어 수요가 증가할 것으로 예상된다. 국내에서는 아직 올리고당을 식이섬유로 사용하지는 못하고 있으나, 식품 공전 개정을 통해 식이섬유로 표기 가능하도록 수정될 전망이다. 당알콜류는 솔비톨 및 자일리톨이 주류를 이루고 있다. 솔비톨은 시장에 가장 먼저 소개된 당알콜류인데 저가여서 그동안 가장 큰 시장을 형성하고 있었으나 최근 3년 동안 자일리톨껌이 최대의 히트 상품이 되면서 자일리톨이 급성장하였다. 그 외에는 만니톨, 에리스리톨, 말티톨, 이소말트 등이 있다. 최근에는 말티톨을 중심으로 다양한 응용 연구 및 이소말트 및 에리스리톨 등을 생산 판매하고 있는 다국적 기업의 판촉 활동 등으로 당알콜류 시장의 새로운 변화가 예상된다. 고감미 감미료류는 스테비오사이드와 아스파탐이 주류를 이루고 있었지만, 최근 아세설팜 K 및 수크랄로스가 잇따라 식품으로써 사용 허가를 받으면서 시장이 재편되고 있다. 하지만, 해외에 비하여 국내의 고감미 감미료 시장은 매우 작고 성장률도 낮은 편인데 그 이유로는 국민 정서상 천연이 아닌 합성에 대한 부정적 인식과 맛에 대한 거부감 때문인 것으로 생각된다. 이와 관련하여 최근에는 다양한 감미질 및 Multi sweetener system 연구를 통해 설탕과 유사한 맛을 내는 혼합 기술 개발에 관한 연구가 진행되고 있다. 따라서 향후에는 해외의 선진국과 같이 고감미 감미료 시장도 점차적으로 확대될 것으로 예상된다. 기타 대체 감미료로는 타가토스, 트레할로스, 아라비노스 등이 있다. 이중에서 특히 해외에서 크게 기대를 모으고 있는 타가토스는 최근 국내 인증을 받아 시장 진출을 준비 중에 있다. 아라비노스는 양질의 감미를 갖는 천연 펜토오스인데 연구용 시약 이외에는 지금까지 거의 이용되지 않았다. 그러나 최근 아라비노스가 설탕의 소화흡수를 선택적으로 억제한다는 흥미 있는 결과가 임상 및 동물실험에서 확인되어 향후 제품으로써 개발을 기대할 수 있게 되었다. 이들 감미료는 기존 설탕이 가지는 문제점을 많은 부분 극복할 것으로 여겨지며, 이로 인해 수요는 점차 증가할 것으로 생각되며, 높은 경제적 부가 가치를 창출할 것으로 예상되어 진다. 생산 기술 적인 측면에서 건강 지향적 천연소재 감미료에 대한 선호가 날로 증가하는 추세에 따라 기존에 주로 사용되었던 화학합성법보다는 천연 이미지의 생물공학적인 방법에 의한 감미료 생산이 각광을 받을 것으로 예상된다. 생물공학적인 방법을 감미료 소재 개발에 응용하게 되면, 기존에 사용되었던 화학합성법 등이 가지는 환경적인 문제, 반응 부산물 생성에 따른 분리, 정제에서의 비용증가 등의 문제점을 해결할 수 있을 것으로 여겨지며, 고정화 방법의 도입 등을 통해 생산성 향상을 도모할 수 있다. 본 장에서는 감미료 소재 개발에 건강 지향적 국민 정서를 반영하고, 국내외 감미료 시장의 흐름을 파악하여 당류 산업의 발전적인 변화에 대처하고자 하였으며, 이를 통해 당류산업의 고부가가치 첨단산업으로서의 육성방안을 논의하고자 하였다.

A Survey on Sugar Intake in Meals from Nursery Schools in Gyeonggi-Do (경기도지역 어린이집 단체급식에서 당 섭취량 조사연구)

  • Excess sugar intake by food consumption may contribute to the development of diabetes, dental caries, attention deficit hyperactivity disorder (ADHD), and cardiovascular disease. The objective of this study was to investigate the sugar intake from meals at nursery schools in Gyeonggi-Do, and to construct a database for reduction policies sugar intake. A total of 601 samples were analyzed for sugar intake in summer and winter, respectively. The average sugar intake from food service was 2.22 g/meal, which was 5% of the Recommended Daily Intake (RDI). Furthermore, the analysis results of sugar content were in the decreasing order of fruits (5.05 g/100 g), fried food (2.92 g/100 g), and braised food (2.31 g/100 g). The habit of excessive sugar intake in childhood can be threaten health in adulthood. Thus, it is necessary for nursery school children to consume schoolmeals that contain less sugar.

  • This experiment was carried out to investigate the effects of alcohols and carbohyrates upon heat resistance of the amylase produced by Bacillus subtilis var. M-181. The results obtained are as follows: 1) Alcohols showed the effects of increasing heat resistance in the order of glycerin, propylene glycol, ethylene glycol, ethanol and methanol, while higher monohydroxy alcohol and dipropylene glycol affected reversely. 2) Inositol affected on heat resistance of the amlase better than sugar alcohols which effects were in the order of mannitol, dulcitol and sorbitol. 3) Monosaccharides affected on heat resistance of the amylase in the order of mannose, galactose, xylose, glucose, arabinose, rhamnose and fructose. 4) Among oligosaccharides, the effects of increasing heat resistance were recognized in the order of melibiose, lactose, maltose and sucrose. 5) Polysaccharides showed the effects of increasing heat resistance in the order of glycogen, soluble starch and dextrin, while the effects of inulin were not recognized.

  • Parenchyma cells of Streptanthus tortus suspension cultures possessed the different transport system for aldose-formed D-glucose and for ketose-formed D-fructose. $K_{m}$ value for D-glucose and D-fructose were 0.28mM and 15.02mM, respectively. $K_{m}$ value of D-mannose was 0.44 mM which is similar to the D-glucose transport system, but D-mannose was transported also through its own special uptake system. Parenchyma cells possessed the transport system of L-glucose, but the function of L-glucose was not known at all. Protoplast of parenchyma cells possessed only the monosugars transport system, but didn't possess the disugars, sucrose transport system. Early developing phloem protoplasts possessed glucose and sucrose transport system at the same time. On the contrary, in the complete developed phloem cells disappeared preexisted glucose transport system in the parenchyma cells, only new induced sucrose transport system existed.ted.

Preparation of Shikhae with Starch Hydrolysing Enzymes/Malt Mixture in Tea-bag (전분 분해효소 첨가와 종이봉지를 이용한 식혜의 제조 방법)

  • As an improved preparation method of Shikhae, a tea-bag system containing malt and amylolytic enzymes was developed in which extraction of malt enzymes and saccharification occured efficiently. The amylolytic activity of the malt was increased by adding the mixture of ${\alpha}-amylase$, glucoamylase and glucoisomerase. Malt and the mixture of enzymes were placed in tea-bag $(16{\times}20cm)$, extracted in water at $30-40^{\circ}C$ for 1-2 hours and followed by saccharification of the cooked rice at $60-70^{\circ}C$ for 3-4 hours. In the conventional Shikhae, content of maltose was about 50% and that of oligosaccharides larger than trisaccharides was about 40% of total sugar. The content of monosaccharides such as glucose and fructose was about 95% and this improved method would be effective for increasing the sweetness and the monosaccharide contents in the product.

Role of Sugars in Early Stage of Spore Germination in Filamentous Fungi, Aspergillus nidulans (사상균인 Aspergillus nidulans의 무성포자 발아와 당의 역할)

  • Initiation of spore germination in filamentous fungi such as Aspergillus nidulans and Botrytis cinerea requires the presence of nutrients. In this study, involvement of sugar sensing machinery was suggested in the germination of A. nidulans spores. Germination did not occur when the spores of A. nidulans were incubated in distilled water, whereas they were successfully germinated in the presence of 5% glucose with a germination rate of over 98% after 6hr incubation. Similar results were obtained when the spores were incubated in the presence of various sugars such as fructose, sucrose, and starch. Interestingly, spore germination was not observed in the presence of D-arabinose, whereas L-arabinose could induce germination as determined by the formation of germ tubes, indicating the presence of sugar sensing machinery that distinguish between the enantiomers of sugars. This inference was further supported by a decrease in germination rate (less than 25%) upon treatment of spores with trypsin. Subsequent MALDI-TOF mass spectrometry analysis of the surface proteins of spores identified ten proteins among which eight were involved in sugar metabolism. Taken together, our results suggest that spore germination in A. nidulans is initiated by the interaction of sugars with sugar binding proteins on the surface of spores.

  • Direct starch-utilizing microorganisms were isolated from 50 samples. Among them, Y-5 strain was selected as one of the potential microorganisms which could utilize starch directly to produce protein or lipid as food resources. The Y-5 strain was identified as a strain of Sporobolomyces holsaticus. It grew on starch or inulin better than on glucose of fructose and its composition was 45% of crude protein, 16% of crude lipid and 9.2% of ash.

Characterization of Chemical Composition in Poplar wood (Populus deltoides) by Suppression of CCoAOMT Gene Expression (CCoAOMT 유전자 발현 억제에 의한 현사시나무의 화학조성 변화)

  • In this study, chemical compositions - holocellulose, lignin and monomeric sugars - were characterized with two poplar wood cell walls, one of which was grown at normal condition (CPW) and the other was genetically modified by antisence suppression of CCoAOMT gene expression (ACPW). Milled wood lignins were isolated from CPW and ACPW and subjected to methoxyl group, DFRC, Py-GC/MS, GPC, $^{13}C$-NMR analysis, respectively. There were few differences in holocellulose contents in both cell walls, which were determined to 81.6% in CPW and to 82.3% in ACPW. However, lignin contents in ACPW was clearly decreased by the suppression of CCoAOMT gene expression. In CPW 21.7% of lignin contents was determined, while lignin contents in ACPW was lowered to 18.3%. The relative poor solubility of ACPW in alkali solution could be attributed to the reduction of lignin content. The glucose contents of CPW and ACPW were measured to 511.0 mg/g and 584.8 mg/g and xylose contents 217.8 mg/g and 187.5 mg/g, respectively, indicating that suppression of CCoAOMT gene expression could be also influenced to the formation of monomeric sugar compositions. In depth investigation for milled wood lignin (MWL) isolated from both samples revealed that the methoxyl contents at ACPW was decreased by 7% in comparison to that of CPW, which were indirectly evidenced by $^{13}C$-NMR spectra and Py-GC/MS. According to the data from Py-GC/MS S/G ratios of lignin in CPW and ACPW were determined to 0.59 and 0.44, respectively. As conclusive remark, the biosynthesis of syringyl unit could be further influenced by antisense suppression of CCoAOMT during phenylpropanoid pathway in the plant cell wall rather than that of guaiacyl unit.

Changes of the Constituents in the Rehmanniae Radix Preparata during Processing (숙지황 제조과정에 따른 성분함량 변화)

  • This study was performed to obtain the good processing in the Relunanniae Radix Preparata. The contents of the Relunanniae Radix and the Relunanniae Radix Preparata produced through different processes were analyzed in the 5-hydroxymethyl-2-furaldehyde (5-HMF), sugar, total nitrogen, crude lipid and ash. 5-HMF was not detected in the Relunanniae Radix, but detected in the Relunanniae Radix Preparata. 5-HMF content was increased gradually with processing times (1∼9 times) and increased expressly in the Relunanniae Radix Preparata steamed for 7 times. Sucrose, fructose and glucose were contained in the Relunanniae Radix, but sucrose was not detected and fructose and glucose were increased largely in the Relunanniae Radix Preparata steamed for 1 time. Fructose and glucose were decreased gradually with processing times (2∼9 times), but the gap of decrease was insignificant. Total nitrogen was changed slightly and crude lipid was decreased slowly with processing times. The ash was suitable to KPⅧ rules (less than 6.0%). From this analysis we found out the content of 5-HMF from Relunanniae Radix Preparata steamed more than 7 times was suitable to KPⅧ rules (more than 0.1%).

Comparisons of growth characteristics, biological activities, nutritional contents, and sugar contents of Ganoderma spp. strains (영지 균주별 생육특성, 생리활성, 영양성분 및 당 성분 함량 비교)

  • This study was carried out to compare the growth characteristics, biological activities, β-glucan contents, sugar contents, and amino acid contents of 14 strains of Ganoderma spp. Among the 14 strains of Ganoderma spp., KMCC02960 (G. meredithae) and KMCC02932 (G. tropicum) showed excellent growth characteristics such as those with respect to the size and yield of fruiting bodies. The highest 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity was observed in KMCC02932 (G. tropicum). The nitrite scavenging activities of KMCC02824 (G. lucidum) and KMCC02852 (G. neo-japonicum) were higher than those of the other strains. The total polyphenol contents of the extracts from KMCC02824 (G. lucidum) and KMCC02852 (G. neo-japonicum) were higher than those of the other strains. KMCC03018 (G. lingzhi) showed the highest β-glucan content of 33.4%. In an analysis of the 4 types of monosaccharides, 2 types of disaccharides, and 4 types of sugar alcohols, only KMCC02996 (G. weberianum) and KMCC03018 (G. lingzhi) were commonly detected out of the 14 strains of Ganoderma spp. Eighteen amino acids, including eight essential amino acids, were identified: the highest total amino acids and total essential amino acids were found in KMCC02932 (G. tropicum), which had the highest levels of tyrosine and phenylalanine. Although the contents of amino acids differed by strain, cysteine, tyrosine, and phenylalanine were the most abundant amino acids in the analyzed extracts.
