  • Microsporidian spores were isolated from yellow-hind winged arctiid, Eilema griseola (Hubner), and cutworm, Agrotis tokionis Butler, and their morphological characteristics and their indectivity to silkworms were investigated. The shape of the microsporidian isolates was oval and the sige of the spore isolated from yellow-hind winged was measured in 2.6$\times$1, 5$\mu$ and that of the isolated from cutworm was measured in 3.7$\times$20.$\mu$. Both the isolated microsporidians showed the infectivity to silkworms, but the isolates did transovarian transmission in silkworms and the silkworm moth infected with the isolate from yellow-hind winged laid eggs irregularly.

  • The survey of silkworm disease occurrence in autumn rearing season, 1984, was carried out and the results are as follows. 1. The percentage of diseased larvae in the current season was about 17%. 2. Among silkworm diseases, grassarie was predominant, as it took 38.5% of total diseased larvae, followed by muscardine which took 20.3% and flacherie was taken by 11.9% 3. The infection of silkworm larvae with pathogens occurred more at grown larva stages than at young larva stages. 4. The loss of cocoon yield was mainly caused by unsuitable climate conditions and agricultural insecticides applied in the rice paddy fields and fruit gardens near mulberry fields. The cocoon production also was reduced by the failure of silkworm disease control due to the incomplete disinfection of rearing rooms and tools.

  • For developing the therapeutic agents to be administered orally for treating certain silkworm diseases and applying them to clinical conditions, vehicles suitable for the agents are needed. The author presumed that Lactose, Casein, and Urea, respectively, have an effectiveness as the vehicle. A series of experiments were carried out on these substances for conforming the applicability of the vehicles at the silkworm rearing house of the College of Agriculture, at Seoul National University during the spring silkworm rearing season of 1974 and the spring and autumn seasons of 1975. The author recognized, Lactose, Casein, and Urea, when put on the mulberry leaves in a powdered form, are eaten along with the leaves by silkworm. Thus, the systemic administration of them to silkworms are possible. The author administered these individual substances orally to silkworm larvae in doses of l0mg., 30mg., 50mg., and 100mg. per gram of the body weight of the silkworm larvae at intervals of once a day, once every two days, and once every three days for 15 days. This was carried out from the first day of the fourth instar to just before mounting. The influences of these substances on the growth of silkworm larvae and the cocoons made by the larvae were observed. The author found out after completing the experiment that Lactose had no effect on growth and cocoon formation. This was observed in all cases. Casein inhibited the growth of silkworms except in the case of administered doses of once every third day in whick the growth and the cocoons were normal but the administration of the agent brought considerably large numbers of abnormal cocoons in every case. Urea inhibited both the growth and the formation of tile cocoons in most cases. For the confirmation of the clinical effectiveness as the vehicles for some therapeutic agents, the three substances, Lactose, Casein, and Urea, will be investigated more closely for the mixing amounts needed and other pertinent data.

  • Human Stem Cell Factor (hSCF) is a cytokine that binds to the c-Kit receptor and plays an important role in hematopoiesis, spermatogenesis, and melanogenesis. To produce the human Stem Cell Factor (hSCF) recombinant protein, we constructed a germline transgenic silkworm using the piggyback vector. The expression of the hSCF gene was driven by the Drosophila heat shock protein 70 (dHsp70) promoter. 3XP3 promotor-driven EGFP was used as a marker which allowed us to rapidly distinguish the transgenic silkworm. A mixture of the donor and helper vector was micro-injected into 1,020 eggs of bivoltin silkworms, Keomokjam. We obtained approximately 22 G1 broods that were EGFP-positive. The expression of the hSCF gene in the transgenic silkworm was analyzed by SDS-PAGE and immunoblotting. Also, analysis of insertion sites into the silkworm genome using inverse PCR showed that exogenous DNA was inserted into the transgenic silkworm genome. These results show that successfully constructed transgenic silkworm expresses the hSCF recombinant protein.

  • The molecular relationships have analyzed between the Bombyx mandarina(wild silkworm) and Bombyx mori strains (domesticated silkworm, geographical silkworms). A total of 166 polymorphic RAPD markers amplified from 35 different primers were used to analyze the molecular relationships among thirteen silkworm strains. The genetic similarity coefficient between Bombyx mandarina and Jam305 showed the lowest genetic similarity value with 0.451, Bombyx mandarina and Bibaekjam showed the highest genetic similarity value with 0.958. These strains were classified into Bombyx mandarina(a wild silkworm) and Bombyx mori(twelve domesticated silkworm) groups upon the genetic similary coefficient of 0.55. Further classificient of 0.60; the 1st sub-group (J111, Bibaekjam, $pnd^{ps}$), the 2nd sub-group (Galwon, C18, od yujam, JAM306, C108), the 3rd sub-group(R-hwang, p50), the 4th sub-group(zebra) and the 5th sub-group(JAM305). According to this study, RAPD markers seems to be a valuable tool for molecular relationships and classification among the silkworms.

  • In this study, silk-gland powder of silkworm was investigated for the usefulness as a potential agent for functional cosmetics, total content of amino acids and DPPH free radical scavenging assay, and clinical trial were done. The analysis of the amino acids of silk-gland powder showed that serine (26.77 %) content was the highest and aspartic acid, glycine, and glutamic acid followed that. DPPH free radical scavenging activity of silk-gland powder was lower than vitamin C (82.3 % v.s. 97 %), The moisture and elasticity were increased in silk-gland powder compared to control cosmetics by 257.2 % and 181.15 %, relatively. These results suggest that silk-gland powder of silkworm may have beneficial properties as a material for cosmeceuticals.
