Conditions for the Production of Amylase and Protease in Making Wheat Flour Nuluk by Aspergillus oryzue L2 (Aspergillus oryzae L2에 의한 밀가루 누룩 제조시 Amylase와 Pretense의 생산조건)

  • A Nuluk, a Korean traditional Koji for brewing, was made with wheat flour and Aspergillus oryzae L2 which had a good aroma and strong abilities In producing saccharogenic and dextrogenic enzymes. The cultural conditions for the production of saccharogenic and proteolytic enzymes were tested. The productivity of dextrogenic enzyme was improved when Nuluk was made with unsteamed wheat flour as compared with steamed one, but that of proteolytic enzyme was reduced. The addition of water containing 0.5% hydrochloric acid was unfavorable for the production of those two enzymes. The optimum ratio of water added to wheat flour for the production of those two enzymes was 28$^{\circ}C$ on the basis of wheat flour, The productivity of saccharogenic enzyme was enhanced when the Nuluk was molded after 20 hours of precultivation, but that of proteolytic enzyme was reduced as compared with no molding. The optimum temperatures for the production of saccharogenic enzyme and proteolytic enzyme were 36$^{\circ}C$ and 28$^{\circ}C$, respectively.

  • From Nuruks, a traditional Korean starter for rice wine, collected from 42 different areas in Korea, 111 fungal strains were isolated. These isolates were identified as 25 species belonging to seven genera of Rhizopus oryzae(14 strains), R. oligosporus(8 strains), R. nigricans(5 strains), R. arrhizus (5 strains), Aspergillus oryzae(12 strains),Asp. parasiticus(8 strains), Asp. fumigatus(3 strains), Asp. ochraceus(7 strains), Asp. wentii(5 strains), Asp. parasiticus(8 strains), Asp. penicilloides(3 strains), Asp. clavatus(4 strains), Penicillium purpurogenum(2 strains), Pen. glabrum(1 strain), Pen. granulaturm (1 strsin), Pen. fellutanum(1 strain), Geotrichum candidum(2 strains), Absidia corymbifera(12 strains), Mucor racemosus(2 strains), M. plumbeus(2 strains) and Curvularia lunatus(3 strains).

Genetic Variation in Natural Populations of the Cat-Snake (Elaphe dome) (누룩뱀(Elaphe diane) 자연집단의 체전적 변리에 관하여)

  • Starch gel electrophoresis was used to estimate the degree of genic variation and genetic relatedness among twelve populations of the Cat-Sanke(Elaphe diene) in Korea. Out of the 17 loci examined 9 loci (Mdh-2, Ldh-1, Ldh-2, Idh-1, Idh-2, Adh, Aco-3, ep, Got-1) were monomorphic with identical mobility in all populations, 4 loci (Pgm, Mpi, Pept-1, Got-2) were nearly identical with minor allele frequency differences, and 4 loci (Mdh-1, Fgi, Est-2, 6psd) showed moderate variation. Levels of genic variation were similar to those found for other reptilian species. The mean genetic similarity value(S) among twelve populations is 0.966. Chechu population was slightly remote genetically from the mainland populations.

  • In this paper a review of literature on Nuruk(yeast) between 1907 and 1945 was made, which showed that barley, rice bran, oat, rye, and other ingredients were originally used according to region and production quantity. Yeast can be classified into rough (Chokuk) and powder (Bunkuk) types depending on the degree of grinding. Yeast also had seasonal names, being called " Choonkuk", Hakuk", Jeolkuk", and "Dongkuk" in the spring, summer, autumn and wither respectively. The form of yeast in terms of quantity, size, and shape varied greatly by region, Production facilities were composed of plant structures to suit each process, enabling continuous output. The production process included shaping, placement in the fermenting chamber, piling by turns, risk-sifting and final output. Testing procudures were divided into visual inspection, physical testing, and chemical analyses.

Aptitudes for Takju Brewing of Wheat Flour-Nuluks Made with Different Mold Species (곰팡이 균종을 달리한 밀가루 누룩의 탁주양조 적성)

  • The purpose of this study was to examine the aptitude of wheat flour-Nuluks, for Takju brewing, which were made with Aspergillus oryzae, Rhizopus japonicus or Aspergillus usamii mut. Shirousamii, and to know the way of effective use the Aspergillus oryzae-Nuluk showed high saccharogenic activity, and Rhizopus japonicus-Nuluk high proteolytic activity. When these two Nuluks were combined, the activities of saccharogenic amylase and protease were more balanced. The Takju mash of Aspergillus oryzae-Nuluk was high in acidity and in amino acidity, and that of Rhizopus japonicus-Nuluk high in the content of free sugar, but, that of Aspergillus usamii mut. Shirousamii-Nuluk was low in amino acidity, free sugar and ethanol content. The Takju of Rhizopus japonicus-Nuluk was high in the content of suspended solid and slow in its sedimenting rate, but that of Aspergillus usamii mut. Shirousamii-Nuluk was not. The Takju which was made with combined use of Aspergillus oryzae-Nuluk and Rhizopus japonicus-Nuluk was more balanced in the composition of flavoring ingredients, and was slow in sedimenting rate of suspended solid. Moreover, it gained good result in sunsory evaluation test.

Effects of Sweet Potato Cultivars and Koji Types on General Properties and Volatile Flavor Compounds in Sweet Potato Soju (고구마 품종과 국의 종류를 달리하여 제조한 고구마 소주의 특성 및 향기성분)

  • Alcoholic fermentations with rice koji [Aspergillus awamori Nakazawa KCCM 60246 (black), Aspergillus kawachii KCCM 32819 (white), Aspergillus oryzae KCCM 11372 (yellow)] and improved nuruk were carried out for the preparation of sweet potato soju using two different potato cultivars (Jinhongmi and Hobak). The Jinhongmi mashes showed $9.2-11.4^{\circ}Brix$, 195.6-260.5 mg glucose/100 mL, pH 4.6-4.9, 0.53-0.83% acidity and 13.2-16.2% alcohol content. The Hobak mashes showed $7.0-8.4^{\circ}Brix$, 31.9-47.4 mg glucose/100 mL, pH 4.4-4.7, 0.22-0.24% acidity, and 9.6-11.2% alcohol content. The alcohol yield of the Jinhongmi mashes using black, white, yellow koji and improved nuruk were 229.2, 194.5, 238.6 and 229.3 L/ton, respectively. The alcohol yields of Hobak mashes using black, white, yellow koji and improved nuruk were 132.8, 144.4, 141.6 and 167.4 L/ton, respectively. All types of sweet potato soju showed stronger flavor and taste than Kurokirishima (Japanese sweet potato soju). Especially, soju made from Jinhongmi with white koji and Jinhongmi with improved nuruk showed the strongest levels. Flavor components of sweet potato soju included decanoic acid ethylester, dodecanoic acid ethylester, tetradecanoic acid ethylester, hexadecanoic acid ethylester, 9-octadecanoic acid ethylester, and octadecanoic acid ethylester. Although the flavor profiles of Jinhongmi soju, Hobak soju, and Kurokirishima were very similar, the flavor content of Kurokirishima soju was lower. The results of the GC volatile analysis were in good correlation with flavor and taste.

The Comparison of Size and Morphology of Scales in Nine Korean Snake Species (6 in Colubridae, 3 in Viperidae) (국내 뱀류 9종의 비늘 크기와 형태 비교)

  • Body scales directly exposed to external environments can be an important factor to understand various characteristics of a species such as habitat features, life history and basic ecology. In this study, we compared size and morphology of dorsal, outermost dorsal, keeled dorsal and ventral scales of total nine snake species in Korea; Oocatochus rufodorsatus, Elaphe dione, Rhabdophis tigrinus, Amphiesma vibakari, Dinodon rufozonatum, Hierophis spinalis in the Colubridae and Gloydius ussuriensis, G. brevicaudus, G. saxatilis in the Viperidae. The morphological characteristics of the scales seem to well reflect foraging modes and moving activity of both families. Uniquely D. rufozonatum had a diamond shape dorsal scale and had the greatest and smallest value of the ratio of width/length of dorsal and ventral scales, respectively. O. rufodorsatus, D. rufozonatum and H. spinalis did not have keeled dorsal scales and E. dione had keel on the few of dorsal scales. In addition, morphological characteristics of scales of three viper species were closely consistent with previously known phylogenetic relationships.

Physiological Functionality and Cytotoxic Effect of Korean Traditional Noble Wine, Samhaeju, and Commercial Rice Wine on Various Tumor Cell Lines (삼해주와 시판 곡주의 생리 기능성 및 세포 독성 효과)

  • This study was conducted to investigate the antioxidant activity, fibrinolytic activity and cytotoxic effect of Korean traditional noble rice wine made using different methods (A-C) and commercial rice wine (D-H) on various tumor cell lines. The antioxidant activity of rice wine was measured by DPPH (2,2-dipicryl-1-picrylhydrazyl), ABTS [2,2'-azinobis(3-ethylbenzthiazoline-6-sulphonic acid)] and NO (nitric oxide) radical scavenging assay. In this study, Samhaeju showed the greatest fibrinolytic activity of 13-17U and exhibited the highest antioxidant activity. Among the different Samhaeju, the sample prepared using method C had the highest antioxidant activity. The cytotoxic effect of rice wine were also examined against the human cancer cell line (A549 cells and HeLa cells) based on the results of a WST-1 assay and morphological changes. Rice wine induced the inhibition of cell proliferation and morphological changes in tumor cell lines in a concentration-dependent manner, with Samhaeju diluted 10 fold having the strongest effect on these factors. These findings suggest that Korean rice wine has antioxidant activity and cytotoxic effect, and that these factors are influenced by the method of preparation.

Production of L-Lactic Acid from Soluble Starch by Enterococcus sp. JA-27. (Enterococcus sp. JA-27에 의한 가용성 전분으로부터 L형 젖산의 생산)

  • Lactic acid bacteria with amylolytic and acid producing activities can ferment starch directly to lactic acid thereby producing a monomer for the production of biodegradable poly lactic acid (PLA). In this study, the strain producing L-lactic acid from soluble starch was isolated from Nuruk. The isolated strain was identified as Enterococcus sp. through its morphological, cultural, biochemical characteristics as well as the 16S rDNA sequence analysis, and named Enterococcus sp. JA-27. Enterococcus sp. JA-27 produced exclusively L-lactic acid from soluble starch as a carbon source. The optimal conditions for the maximum production of L-lactic acid from Enterococcus sp. JA-27 were 30 C, pH 8, 1.5 % soluble starch as a substrate and 3.5 % tryptone as a nitrogen source, 0.1 % $K_2$$HPO_4$, 0.04 % $MgSO_4$. $7H_2$O, 0.014 % $MnSO_4$$.$4$H_2O$, 0.004% $FeSO_4$$.$$7H_2$O. Batch and fed batch culture were carried out and the former was more effective. L-Lactic acid production in the optimum medium was significantly increased in a 7 L jar fermenter, where the maximum L-lactic acid concentration was 3 g/L. For the purification of lactic acid in fermented broth, two stage ionexchange column chromatographies were employed and finally identified by HPLC.

Changes of Components in the Rice-porridge Fermented by Nuruk (누룩에 의한 쌀죽발효액 중의 성분변화)

  • Shindari, a traditional low alcoholic drink in Cheju island, was prepared with nuruk and the rice porridge. The rice porridge was fermented with 10% nuruk at a temperature of $25^{\circ}C,\;35^{\circ}C,\;45^{\circ}C$ for 24 hours. In all temperature, total acidity and oBrix were increased gradually and pH were decreased gradually and viscosity were considerably decreased after 2 hours and then slightly decreased. Alcohol contents were also increased during fermentation and especially at $45^{\circ}C$. The organic acids were mainly found phytic acid and succinic acid. The fermented liquids rice porridge contained maltose and glucose as free sugar, amount of them increased during fermentation. Lactic acid bacteria cell count increased at higher tem perature of fermentation and reached to $10^{7}\;cfu/ml\;at\;35^{\circ}C$ for 10hr. In conclusion, the optimal fermentation conditions Shindari preparation were at $35^{\circ}C$, 24hrs. pH, titratable acidity, viscosity and soluble solids in the optimal fermentation condition were 4.2, 0.18%, 190cp, $12.2^{\circ}Brix$, respectively.
