Protective Effect of the Ethyl Acetate-fraction of Methanol Extract of Ophiophogon japonicus on Amyloid beta Peptide-induced Cytotoxicity in PC12 Cells (소엽맥문동-에틸아세테이트 분획물의 아밀로이드 베타단백질-유발 세포독성에 대한 억제 효능)

  • Amyloid ${\beta}$-protein ($A{\beta}$) is the principal component of senile plaques characteristic of Alzheimer's disease (AD) and elicits a toxic effect on neurons in vitro and in vivo. Many environmental factors, including antioxidants and proteoglycans, modify $A{\beta}$ toxicity. It is worthwhile to isolate novel natural compounds that could prove therapeutic for patients with AD without causing detrimental side effects. In this study, we investigated the in vitro neuroprotective effects of the ethyl acetate fraction of methanol extract of Ophiophogon japonicas (OJEA fraction). We used an MTT reduction assay to detect protective effects of the OJEA fraction on $A{\beta}_{25-35}$-induced cytotoxicity to PC12 cells. We also used a cell-based ${\beta}$-secretase assay system to investigate the inhibitory effect of the OJEA fraction on ${\beta}$-secretase activity. In addition, we performed an in vitro lipid peroxidation assay to evaluate the protective effect of the OJEA fraction against oxidative stress induced by $A{\beta}_{25-35}$ in PC12 cells. The OJEA fraction had strong protective effects against $A{\beta}_{25-35}$-induced cytotoxicity to PC12 cells and was strongly inhibitory to ${\beta}$-secretase activity, which resulted in the attenuation of $A{\beta}$ generation. In addition, the OJEA fraction significantly decreased malondialdehyde (MDA) content, which is induced by the exposure of PC12 cells to $A{\beta}_{25-35}$. Our results suggested that the OJEA fraction contained active compounds exhibiting a neuroprotective effect on $A{\beta}$ toxicity.

Development of Analytical Method for Ergot Alkaloids in Foods Using Liquid Chromatoraphy-Tandem Mass Spectrometry (LC-MS/MS를 이용한 식품 중 맥각 알칼로이드 시험법 개발)

  • Ergot alkaloids are mycotoxin produced by fungi of the Claviceps genus, mainly by Claviceps purpurea in EU. Recently obtained informations indicates necessity for control the ergot in imported grains. Recent occurrence data of ergot alkaloids from EU countries indicate the necessities of management and control these toxins from the imported grains like rye, wheat, oat etc. The aim of this study is to optimize the liquid chromatography-tandem mass spectrometry method for determination of ergot alkaloids (ergometrine, ergosine, ergotamine, ergocornine, ergocryptine, ergocristine and their epimers (-inines) from grain and grain-based food. The test method was optimized by extracting the sample with acetonitrile containing 2 mM ammonium carbonate, purification with Mycosep cartridge, and instrumental analysis by LC-MS/MS using Syncronis C18 column. The standard calibration curves showed linearity with correlation coefficents; $R^2$ >0.99. Mean recoveries ranged from 72.0 to 111.3% at three different fortified levels (20, 50, and $100{\mu}g/kg$). The correlation coefficient expressed as precision was within the range of 1.9-12.9%. The limit or quantifications (LOQ) ranged from 0.012 to $0.058{\mu}g/kg$. The developed analytical method met the criteria of AOAC Int. and CAC validation parameters like accuracy and sensitivity. As a result, it was confirmed that the test method developed in this study is suitable for the simultaneous analysis of six species of ergot alkaloid from grains and grain products.

Analysis of esthetic factors and evaluation of esthetic perception for maxillary anteriors of dental students (치과대학생의 상악 전치부의 심미적 요소 분석 및 인식도 평가)

  • Purpose: This research is to investigate factors that affect dental aesthetics and analyze whether individual perception affects and difference of genders affects the esthetic factors. Materials and methods: Seventy dental students of Chonnam University aged from 25 to 35 years old without periodontal problems were included. Maxillary dental cast for participants were made, and standardized photo was taken with a digital camera. Maximum visual width and the position of gingival zenith of maxillary anterior teeth and maximum height of the maxillary central incisors were measured by a measurement program and the shape of central incisor was categorized. Questionnaire was conducted to evaluate esthetic perception. SPSS program was used to perform statistical analysis. Results: The average visual width ratio of right maxillary anteriors was 1.38:1:0.78 and 1.41:1:0.81 for the left which differ from the golden ratio. The width to height ratio for right and left central incisor was 0.84 and 0.83 respectively. The gingival zenith position was determined to be: mostly, central incisors and canines were located in the distal side, lateral incisors were located in the center. The visual width ratio of right maxillary anterior teeth, ratio of width to height of central incisor, gingival zenith position, crown morphology and amounts of gingival exposure upon smiling were not significantly different between genders, and facial patterns and these dental esthetic factors were not related to aesthetic perception. Conclusion: Even though participants had the visual width ratio of maxillary anteriors or ideal width to ideal height of central incisors that did not agree with the golden ratio, they had high satisfaction for dental esthetics. Esthetic perception depends more on subjective judgements of participants than objective indices.

Location Environment and Vegetation Structure of the Aconitum austrokoreense Habitat (세뿔투구꽃 서식지의 입지환경 및 식생구조)

  • Owing to the lack of consistent research on endangered plant species in Korea, there are insufficient data to preservespecies and expand habitats. This study analyzed the preferred habitat and threats to the survival of Aconitum austrokoreense, found on Baekwun Mountain in Gurye-gun, Gwangyang-si, Jeollanam-do Province, and classified as a level two endangered wild plant by the Ministry of Environment, by investigating major environmental factors such as climate, location, soil, and stand structure. By examining five selected sites inhabited by Aconitum austrokoreense on BaekwunMountain, this study found that the habitat had an altitude of 420 to 675 m above sea level and showed a northeast tendency, spreading over a range of inclination angles between 15° and 37°. The average number of plants across the five sites was 156. Site 4 (550 m) had the highest density of 372 plants, with an average height of 0.6 m. The average soil moisture and relative light intensity were 20.48% and 7.34%, respectively. Layer soil was presumed to be sandy loam, characterized by high sand content and good drainage. The habitat had average soil pH of 5.2, average organic matter of 16.46%, average nitrogen of 0.86%, average available phosphate of 11.86 mg/kg, average electrical conductivity of 0.44 dS/m, and average cation exchange capacity of 37.04 cmolc/kg. The total carbon in soil averaged 10.68%. From the analysis of the vegetation structure of sites inhabited by Aconitum austrokoreense, the dominant populations were Pinus koraiensis and Lindera erythrocarpa in Site 1, Magnolia obovata and Carpinus laxiflora in Site 2, Zelkova serrate and Quercus variabilis in Site 3, Staphylea bumalda and Lindera erythrocarpa in Site 4, and Morus bombycis,Styrax japonicus, and Carpinus laxiflora in Site 5. With most habitats located near trails and sap collection sites of Acer pictum, the species were exposed to artificial damage and interference threats.

Application of Terrestrial LiDAR for Reconstructing 3D Images of Fault Trench Sites and Web-based Visualization Platform for Large Point Clouds (지상 라이다를 활용한 트렌치 단층 단면 3차원 영상 생성과 웹 기반 대용량 점군 자료 가시화 플랫폼 활용 사례)

  • For disaster management and mitigation of earthquakes in Korea Peninsula, active fault investigation has been conducted for the past 5 years. In particular, investigation of sediment-covered active faults integrates geomorphological analysis on airborne LiDAR data, surface geological survey, and geophysical exploration, and unearths subsurface active faults by trench survey. However, the fault traces revealed by trench surveys are only available for investigation during a limited time and restored to the previous condition. Thus, the geological data describing the fault trench sites remain as the qualitative data in terms of research articles and reports. To extend the limitations due to temporal nature of geological studies, we utilized a terrestrial LiDAR to produce 3D point clouds for the fault trench sites and restored them in a digital space. The terrestrial LiDAR scanning was conducted at two trench sites located near the Yangsan Fault and acquired amplitude and reflectance from the surveyed area as well as color information by combining photogrammetry with the LiDAR system. The scanned data were merged to form the 3D point clouds having the average geometric error of 0.003 m, which exhibited the sufficient accuracy to restore the details of the surveyed trench sites. However, we found more post-processing on the scanned data would be necessary because the amplitudes and reflectances of the point clouds varied depending on the scan positions and the colors of the trench surfaces were captured differently depending on the light exposures available at the time. Such point clouds are pretty large in size and visualized through a limited set of softwares, which limits data sharing among researchers. As an alternative, we suggested Potree, an open-source web-based platform, to visualize the point clouds of the trench sites. In this study, as a result, we identified that terrestrial LiDAR data can be practical to increase reproducibility of geological field studies and easily accessible by researchers and students in Earth Sciences.

Mineralogical and Geochemical Characteristics of the Precipitates in Acid Mine Drainage of the Heungjin-Taemaek Coal Mine (흥진태맥 석탄광 산성광산배수 침전물의 광물학적 및 지구화학적 특성)

  • Fe(II) released from mining activities is precipitated as various Fe(III)-oxyhydroxides when exposed to an oxidizing environment including mine drainage. Ferrihydrite, one of the representative precipitated Fe(III) minerals, is easy to adsorb heavy metals and other pollutants due to the large specific surface area caused by very low crystallinity. Ferrihydrite is transformed to thermodynamically more stable goethite in the natural environment. Hence, information on the transformation of ferrihydrite to goethite and the related mobility of heavy metals in the acid mine drainage is important to predict the behaviors of those elements during ferrihydrite to goethite transition. The behaviors of heavy metals during the transformation of ferrihydrite to goethite were investigated for core samples collected from an AMD treatment system in the Heungjin-Taemaek coal mine by using X-ray diffraction (XRD), chemical analysis, and statistical analysis. XRD results showed that ferrihydrite gradually transformed to goethite from the top to the bottom of the core samples. Chemical analysis showed that the relative concentration of As was significantly high in the core samples compared with that in the drainage, indicating that As was likely to be adsorbed strongly on or coprecipitated with iron oxyhydroxide. Correlation analysis also indicated that As can be easily removed from mine drainage during iron mineral precipitation due to its high affinity to Fe. The concentration ratio of As, Cd, Co, Ni, and Zn to Fe generally decreased with depth in the core samples, suggesting that mineral transformation can increase those concentrations in the drainage. In contrast, the concentration ratio of Cr to Fe increased with depth, which can be explained by the chemical bond of iron oxide and chromate, and surface charge of ferrihydrite and goethite.

Comparative Analysis of Detection Methods for Food-borne Pathogens in Fresh-cut Agricultural Materials (신선 농산물내 식중독균 검출 방법의 비교 분석)

  • The consumption of fresh-cut agricultural materials is increasing due to increased public interest in health and the increase of single-person households. Most fresh-cut agricultural materials can be eaten without heating, thus easily exposing the consumer to food-borne pathogens. As a result, food-borne diseases are increasing worldwide. In the analysis of food-borne pathogens, it is important to detect the strains, but this is time consuming and laborious. Alternative detection methods that have been introduced, include polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE), which is performed without prior culturing. Samples of fresh-cut agricultural materials, such as vegetables, were analyzed by the culture-based method. In 129 samples, non-pathogenic Escherichia coli (3.9%), Bacillus cereus (31.8%), Clostridium perfringens (5.4%), Yersinia enterocolitica (0.8%), and enterohemorrhagic E. coli (0.8%) were detected. Eight samples contaminated with bacteria were randomly selected, further analyzed by PCR-DGGE, and compared with the culture-based method. Two cases detected non-pathogenic E. coli by PCR-DGGE only, despite a lack of detection by the culture method. It was supposed there was possibility of sample loss during its 10-fold dilution for appropriate cultivation. In the detection of high-risk food-borne pathogens, it was found that the detection limit was lower in PCR-DGGE than in the culture-based method (10 CFU/g). This suggests that PCR-DGGE can be alternatively used to detect strains. On the other hand, low-risk food-borne pathogens seem to have higher detection limits in PCR-DGGE. Consequentially, this study contributes to the improvement of food-borne pathogen detection and the prevention of its related-diseases in fresh-cut agricultural materials.

Determination of Mycotoxins in Agricultural Products Used for Food and Medicine Using Liquid Chromatography Triple Quadrupole Mass Spectrometry and Their Risk Assessment (LC-MS/MS를 이용한 식·약 공용 농산물의 곰팡이독소 분석 및 위해평가)

  • For this study, we surveyed concentrations of 8 mycotoxins (aflatoxin B1, B2, G1, G2, ochratoxin A, fumonisin B1, B2 and zearalenone) in agricultural products used for food and medicine by liquid chromatography-tandem mass spectrometry and conducted a risk assessment. Samples were collected at the Yangnyeong Market in Seoul, Korea, between January and November 2019. Mycotoxins were extracted from these samples by adding 0.1% formic acid in 50% acetonitrile and cleaned up by using an ISOLUTE Myco cartridge. The method was validated by assessing its matrix effects, linearity, limit of detection (LOD), limit of quantification (LOQ), recovery and precision using four representative matrices. Matrix-matched standard calibration was used for quantification and the calibration curves of all analytes showed good linearity (r2>0.9999). LODs and LOQs were in the range of 0.02-0.11 ㎍/kg and 0.06-0.26 ㎍/kg, respectively. Sample recoveries were from 81.2 to 118.7% and relative standard deviations lower than 8.90%. The method developed in this study was applied to analyze a total of 187 samples, and aflatoxin B1 was detected at the range of 1.18-7.29 ㎍/kg (below the maximum allowable limit set by the Ministry of Food and Drug Safety, MFDS), whereas aflatoxin B2, G1 and G2 were not detected. Mycotoxins that are not regulated presently in Korea were also detected: fumonisin (0.84-14.25 ㎍/kg), ochratoxin A (0.76-17.42 ㎍/kg), and zearalenone (1.73-15.96 ㎍/kg). Risk assessment was evaluated by using estimated daily intake (EDI) and specific guideline values. These results indicate that the overall exposure level of Koreans to mycotoxins due to the intake of agricultural products used for food and medicine is unlikely to be a major risk factor for their health.

Volcanic Activity of the Volcanoes in the Hallasan Natural Reserve, Jeju Island, Korea (한라산천연보호구역 소화산들의 화산활동 기록)

  • This study reports the Ar-Ar dating results for the volcanic rocks from small volcanoes(oreum) of the Hallasan Nature Reserve. According to the age of 40Ar/39Ar, the volcanic activity of the Hallasan Natural Reserve was started from about 192 ka ago. The basaltic trachyandesite and trachyte located in the Y valley near the Eorimok in the western part of the Hallasan Natural Reserve represent an age of about 191~192 ka, showing the oldest record of volcanic activity in the Hallasan Natural Reserve. In the Hallasan Natural Reserve, the small volcanoes older than 100 ka are Y Valley in Eorimok area (192±5 and 191±5 ka), Dongsu-Ak (184±19 ka), Mansedongsan (153±5 ka), Janggumok-Orum (135±6 ka), Eoseungsaengak (123±9 ka), Samgagbong (105±2 ka). And the small volcanoes younger than 100 ka are Witbangae-Oreum, Seongneol-Oreum, Muljangol, Yeongsil, Bori-Ak, Witsenueun-Oreum, Witsejokeun-Oreum, Heugbuleun-Oreum, Bangae-Oreum, Albangae-Oreum, Witsebuleun-Oreum, Baengnokdam, Nongo-Ak. According to the eruption of trachytes, the Hallasan Natural Reserve can be interpreted as having about 8 volcanic activities. Among them, 4 volcanic activities are related with the formation of trachyte dome, such as Wanggwanneung, Samgakbong, Yeongsil, and Baengnokdam, and 4 volcanic activities are related with flow or dyke of trachyte. The volcanic activity at the Hallasan Natural Reserve was started from northwest area, to in the southern area, and in the eastern area, and finally volcanic activity related to the formation of Baengnokdam.

Antibacterial, Antioxidative and Antiaging Effects of Allium cepa Peel Extracts (양파껍질 추출물의 항균, 항산화 및 항노화 효과에 관한 연구)

  • In this study, the antibacterial, antioxidative and inhibitory effects of Allium cepa peel extracts on tyrosinase and elastase were investigated. MIC values of the ethyl acetate fraction of Allium cepa peel on especially, S. aureus among the skin resident flora (Staphylococcus aureus, S. aureus; Propionibacterium acnes, P. acnes; Pityrosporum ovale, P. ovale; Escherichia coli, E. coli) were 0.06%. The aglycone fraction showed more excellent free radical (1,1-diphenyl-2-picrylhydrazyl radical, DPPH) scavenging activity ($FSC_{50}=5.05{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of the ethyl acetate fraction and aglycone fraction in the luminol-dependent $Fe^{3+}-EDTA/H_2O_2$ system were 0.05 and $0.03{\mu}g/mL$, respectively. The cellular protective effect of the aglycone fraction on the rose-bengal sensitized photohemolysis of human erythrocytes exhibited more prominent (${\tau}_{50}$, 480 min at $25{\mu}g/mL$). The inhibitory effects ($IC_{50}$) of the ethyl acetate fraction and aglycone fraction on tyrosinase were 9.16 and $8.68{\mu}g/mL$, the inhibitory effect ($IC_{50}$) of the aglycone fraction on elastase was $14.12{\mu}g/mL$ The transepidermal water loss of the cream containing 0.1% ethyl acetate fraction was decreased from $8.3g/m^2h$ in control to $6.8g/m^2h$ in the subjects applied with cream containing the ethyl acetate fraction. These results indicate that extract/fractions of Allium cepa peel can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS, and possibly as antiaging agents. Allium cepa peel extract could be used as a new cosmeceutical for whitening and anti-wrinkle products.
