  • This work was carried out to evaluate the effects of preharvest 1-methylcyclopropene (1-MCP, $Harvista^{TM}$) and postharvest 1-MCP ($SmartFresh^{TM}$) treatments on the fruit quality attributes of cold-stored 'Fuji' ( Malus domestica Borkh.) apples. Fruits were exposed to 0, 95, 125, or $250 mg{\cdot}L^{-1}$ $Harvista^{TM}$ at 3, 2, 1 weeks before harvest (WBH), and treated with 0 or $1{\mu}{\cdot}L^{-1}$ $SmartFresh^{TM}$ at harvest. Fruit was then stored for up to 180 days at $0{\pm}1^{\circ}C$. Fruit fresh weight, Hunter's value a, internal ethylene concentration (IEC), flesh firmness, titratable acidity (TA), and soluble solids content (SSC) in fruit treated with $Harvista^{TM}$ were not different from those of control fruit at harvest. During cold storage, flesh firmness and TA were higher in fruit treated with $250mg{\cdot}L^{-1}$ $Harvista^{TM}$ at 2 and 3 WBH than in control fruit. IEC was 5.5-10.0% lower in fruit treated with $250mg{\cdot}L^{-1}$ $Harvista^{TM}$ at 2 and 3 WBH compared with control fruit as storage duration progressed, while SSC was not affected. Furthermore, flesh firmness, TA, and IEC were affected neither by $Harvista^{TM}$ nor $Harvista^{TM}+SmartFresh^{TM}$ treatments, compared with those fruit quality attributes at harvest. The correlation maps indicated that IEC was negatively correlated with firmness and TA, regardless of $Harvista^{TM}$ application levels. In addition, positive correlations between fruit quality attributes were detected in treatments with $250mg{\cdot}L^{-1}$ $Harvista^{TM}+SmartFresh^{TM}$. Therefore, the results suggest that with a single application of $SmartFresh^{TM}$, a higher level of $Harvista^{TM}$ application would help in retention of fruit quality attributes during cold storage.

  • Cell elongation is known to be promoted by ethylene in petioles of Ranunculus sceleratus. Treatment of petiole segments with spermine resulted in an inhibition of cell elongation and of ethylene biosynthesis in the presence of applied auxin. Dose response curve for the spermine inhibition of auxin-induced ethylene production appeared similar to that of ACC-based ethylene production suggesting that the polyamine inhibits ethylene biosynthesis by blocking the conversion of ACC to etylene. Auxin-induced ethylene production was significantly promoted by treatment of the tissue with either DFMA or DFMO. specific inhibitors of polyamine biosynthesis. Increased level of ethylene production by DFMA was found to be completely abolished by application of exogenous spermine at a high concentration. These results indicate that endogenous spermine plays a regulatory role in the growth response of Ranunculus petioles to auxin and ethylene.hylene.

  • This work was carried out to evaluate the effect of preharvest sprayable 1-methylcyclopropene (1-MCP: $Harvista^{TM}$) treatment on the fruit quality attributes in cold stored 'Gamhong' (Malus domestica Borkh) apples. Preharvest sprayable 1-MCP treatment did not affect fruit qualities at harvest but titratable acidity (TA) was higher in fruit treated 10 days before harvest (DBH) than in control. During cold storage, fruit firmness and TA were higher in 1-MCP treated fruit than in control fruit as storage duration progressed, while SSC was not affected. Internal ethylene concentration (IEC) levels were not different among treatments at harvest. While the IEC level in control fruit reached to $58.3{\mu}L{\cdot}L^{-1}$ after 6 month cold storage, the IEC levels in 1-MCP treated fruit ranged from 18.2 to $25.0{\mu}L{\cdot}L^{-1}$. Therefore, this result indicates that preharvest sprayable 1-MCP treatment should be another promising approach to retain fruit quality attributes in long-term cold stored 'Gamhong' apples.

  • This study investigated the effect of AVG treatment on lipid material changes in the 'Hongro' apple epidermis. The internal ethylene concentration in 'Hongro' apples treated with $200mL{\cdot}L^{-1}$ AVG and stored at room temperature after harvest was significantly lower than that in 'Hongro' apples treated with $20mL{\cdot}L^{-1}$ Ethephon and the controls. During the same period, the emission of wax after the AVG treatment was also significantly lower than that after the other treatments. In conclusion, treatment with $220mL{\cdot}L^{-1}$ AVG two weeks before harvesting was found to be effective in improving and maintaining the quality of the 'Hongro' apples. Reducing the level of internal ethylene can delay the aging of fruit and inhibit the accumulation of wax in the fruit epidermis.

  • Root gravitropism was investigated in the pea (Pisum sativum L.) mutant ageotropum lacking normal gravitropic response. Exogenous ethylene treatment inhibited gravitropic response in the normal (wild type) pea rook, but had no significant effect to restore the unresponsiveness in the mutant Neither inhibitors of ethylene biosynthesis nor antagonists of ethylene action were able to bring about the development of gravioopic curvature in the ageotropum roots. Auxin action seems to be normal since asymmetric application of agar blocks containing auxin to the mutant roots caused normal gravitropic response to occur. Endogenous as well as auxin-induced ethylene production in tissue segments of the mutant root was about equal to that of the wild type. However no appreciable lateral transport of labeled auxin was observed in glavistimulated mutant roots whereas typical auxin asymetry was apparent in the wild type roots under the same conditions. It is concluded that the mutant has a defect in either gravity perception or its transduction, but not in the effector system involving auxin action.

  • The effect of methylglyoxal-bis (guanylhydrazone) (MGBG) and ethylene synthesis inhibitors on adventitious root formation from soybean cotyledon in relation to ethylene production and endogenous polyamine content was investigated. Cotyledon explants cultured on rooting medium formed numerous adventitious rook on the cut surfaces after 2 weeks of culture. However when cultured on rooting medium supplemented with MGBG, the root formation was strongly inhibited, its inhibitory effect was reserved when cultured on medium with MGBG + spermine, MGBG + CoCl$_2$ and MGBG + spermine+CoC1$_2$. A slight reversion of the rooting inhibition was observed in 10$^{-3}$ M MGBG +10$^{-5}$ M spermine treatment, whereas it caused a significant effect in 10$^{-3}$ M MGBG +10$^{-4}$ M treatment .Ethylene production and endogenous polymine content was investgated in 10$^{-3}$ M MGBG , 10$^{-3}$ M MGBG +10$^{-5}$ Mspermine, 10$^{-3}$ M MGBG +10$^{-4}$ M CoCl$_2$and 10$^{-3}$ M MGBG +10$^{-5}$ M spermine +10$^{-4}$ M CoCl$_2$treatments. Ethylene production highest in 10$^{-3}$ M MGBG +10$^{-5}$ M spermine treatment was higher than control. In 10$^{-3}$ M MGBG +10$^{-5}$ M spermine + 10$^{-4}$ M CoCl$_2$ treatment, ethilene production was lowest, whereas polyamine level was highest.

  • Vie investigated the effect of indole acetic acid (IAA) on diamine oxidase (DAO; EC 1.4.3.6) in the seedling of soybean (Glycine max L. forma Paldalkong). DAO activity was not detected in the resting soybean seeds. During germination it appeared in the elongating zone on day 3, increased up to day 4 and decreased thereafter. Endogenous IAA content has been shown the same pattern as DAO activity. However, cadaverine content was reduced on day 4. To investigate the effect of IAA on DAO activity, the segments of hypocotyl on day 3 were soaked in various concentration of IAA. The activity increased at low concentrations ($10^{-7}-10^{-6}M$) of 1M as compared to that of control but not at high concentrations ($10^{-5}-10^{-4}M$) of IAA. Differing from DAO activity, cadaverine content increased as concentrations of IAA increased. On the other hand, ethylene was induced by IAA at high concentrations. To study the effect of ethylene on DAO activity, we cotreated IAA and 2,S-Norbonadiene (NDE). In this case, DAO activity was not affected.fected.

  • The possibility of circadian production of plant hormone gibberellin (GA0 was examined in phytochrome B mutant (plyB-1) and wild-type sorghum. GA$_{12}$, GA$_{20}$ and GA$_{1}$ levels were found to cycle circadianly in both phyB-1 and wild-type. The periods (33 h) of GA$_{20}$ and GA$_{1}$ cycling in constant light were longer than normal photoperiods in both genotypes and typical average free running periods in plants of 22 to 28 h. The biological clock was thus shown to function properly in phyB-1. However, circadian regulation of GAs productions were not clear as compared to circadian ethylene regulation reported by Lee (1996). Although, in sorghum, EOD FR treatment hasten floral inititation, the differences in GA concentrations between treatments and untreated control were generally less dramatic than expected. Thus, it can be concluded that FR does not act primarily by changing absolute levels of GAs but rather by increasing flowering responsiveness to GAs.

  • 'Jonathan' apples are relatively small size which contributes to enhancing fruit consumption and gaining popularity. Thus, this study was carried out to evaluate the effects of AVG (aminoethoxyvinylglycine, ReTain$^{(R)}$), sprayable 1-MCP (1-methylcyclopropene, Harvista$^{TM}$), and fumigation 1-MCP (SmartFresh$^{TM}$) applications on fruit quality attributes and storability in 'Jonathan' apple fruits during cold-stored. The Jonathan fruits were dipped with either ReTain (75 mg/L) or Harvista (125 mg/L) solutions for 5 min, or fumigated with SmartFresh (1 mg/L) for 18 hr before storage at $0{\pm}1^{\circ}C$ for 75 days. Flesh firmness and titratable acidity remained higher in all pre-treated apples than control ones during cold storage period. Flesh firmness was higher for apples treated with ReTain and SmartFresh than samples treated with Harvista, while soluble solid content and respiration rate were not affected by sample pretreatment. Internal ethylene concentration (IEC) of all pretreated apples remained below about $4.5{\mu}L/L$ during the entire storage period while that of control sample greatly increased to $10.29{\mu}L/L$. Ethylene production was much higher in control fruits than in treated ones during cold storage. These results indicated that ReTain and 1-MCP treatments would be considerably effective in retention of fruit quality attributes of 'Jonathan' apple during cold-stored.

  • This study aimed to evaluate the fruit quality characteristics and incidence of flesh browning in response to air storage at $0{\pm}1^{\circ}C$ a fter controlled atmosphere s torage (CA condition: $O_2$ $2.5{\pm}0.5$%, $CO_2$ $1.5{\pm}0.5$%) at $0{\pm}1^{\circ}C$ in 'Fuji' apples (Malus domestica Borkh.). The storage system was performed as followed: air storage for one month, CA storage 4 months + air storage 3 months ( CA 4M + A ir 3M), CA storage 5 m onths + air storage 2 months ( CA 5M + Air 2M) a nd C A storage 6 m onths + air storage 1months (CA 6M + Air 1M), while the control fruits were stored at CA storage for 8 months right after harvest. The incidence of flesh browning ranged from 17.1% to 30.2% during CA storage but not detected under the treatments of CA 4M + Air 3M and CA 5M + Air 2M. The respiration rate was not affected by storage treatments for 6M while the respiration rate was lower in the treatments of CA 4M + Air 3M and CA 5M + Air 2M than the other storage treatments after 7 months. Ethylene production and internal ethylene concentration were lowest in rapid CA storage and increased with a decreasing CA storage duration. Therefore, the results indicate that CA 5M + Air 2M storage treatment should be recommended to maintain the fruit quality and reduce the risk development of flesh browning rather than typical CA storage in 'Fuji' apples.
