Isolation and Cultural Characteristics of a Phosphate-Solubilizing Bacterium, Aeromonas hydrophila DA57 (인산가용화균 Aeromonas hydrophila DA 57의 분리와 배양 중 가용화특성)

  • To develop biofertilizer solubilizing inorganic phosphate, a bacterium having high abilities to solubilize inorganic phosphate were isolated from cultivated soils. The strain was identified to Aeromonas hydrophila DA57, based on the physiological and biochemical properties. The optimum temperature and initial pH to solubilize insoluvle phosphate in sucrose minimal medium were $30^{\circ}C$ and pH 7.0, respectively. In these conditions phosphate solubilizing activities of the strain against three types of insoluble phosphate were quantitatively determined. It was possivle to distinguish between solubilization through release of gluconic acid and still unknown mechanism. Aemmonas hydrophila DA57 harbored a 4.5 kb cryptic plasmid.

Characteristics of Insoluble Phosphates Solubilizing by Klebsiella sp. DA71-1/pLYJ (Klebsiella sp. DA71-1/pLYJ의 난용성 인산염 가용화 특성)

  • To develop high efficiency biofertilizer solubilizing insoluble phosphates, lactate dehydrogenase (ldh) gene was isolated from Staphylococcus sp. LJ2. Genetic constructions were carried out using the pGEM-T-easy vector and pHSG398. Recombinant DNA plasmids containing the ldh gene were transferred to Klebsiella sp. DA71-1 by electroporation. The selected transformant was named as a DA71-1/pLYJ. The insoluble phosphates solubilization activity of DA71-1/pLYJ was higher than that of DA71-1 at various culture conditions. Glucose was the best carbon source for insoluble phosphates solubilization among the used carbon sources. Maximal insoluble phosphates solubilizing was found in sucrose minimal (SM) medium containing 3% glucose. The solubilizing activity of DA71-1/pLYJ against three types of insoluble phosphates, such as tri-calcium phosphate, hydroxyapatite, aluminium phosphate, were quantitatively determined. The optimal temperature and initial pH to solubilize insoluble phosphates in the SM medium was $37^{\circ}C$ and pH 5.0, respectively.

  • Sudan IV는 2시간 관류시키는 동안 약 30%의 농도감소를 보였다 Sudan IV자체는 물에 녹지않고 리포솜에 의해서 가용화 되는 물질이기 때문에 관류액중에서 리포솜으로부터 release out되지 않는다. 따라서 situation I의 가능성은 희박하며 또 그 자체로는 물에 거의 녹지않아 흡수되지 않을 것으로 예상되는 물질이므로 Sudan IV의 관류액으로부터의 소실은 곧바로 situation II를 반영할 것이다. 또 리포솜의 구성물질인 PC역시 관류액중에서 농도감소를 나타내었는데 이로부터 난용성약물을 봉입한 리포솜이 어떻게 장관으로 흡수되는 지 예상할 수 있었다.

  • Organogels are semi-solid systems that consist of an apolar solvent as the liquid phase within a three-dimensional networked structure. In this study, we developed a stable and skin penetration-enhanced Lecithin Organogel (LO) containing genistein, which is one of the poorly soluble active ingredients in both polar and apolar phase. After screening of various components (type of gelators, organic and aqueous phase), hydrogenated lecithin (HL), sunflower oil (SO), dipropylene glycol (DPG), and polyethylene glycol (PEG) were mainly used in this formulation. Phase ternary diagram was employed for optimization of the composition in the LO. The formulated LO were evaluated for its organoleptic characteristics, stability, pH, rheology, phase transition temperatures, microscopic analysis and skin penetration. The optimized stable LO system can be utilized as an effective and stable cosmetic formulation that can carry poorly soluble active ingredients at high concentration for topical dermal delivery.

  • This study was to investigate beneficial effects of microbial mixture on red pepper which was capable of promoting plant growth by solubilizing insoluble phosphate as well as protecting plants from pathogenic attack. Saccharomyces sp. L13 was isolated for phosphate solubilizing activity on aluminium phosphate, tricalcium phosphate, calcium hydrophosphate, and magnesium hydrophosphate. On the other hand, Bacillus sp. L32 was isolated for antagonistic activity against Phytophthora capsisi and Colletotrichum gloeosporioides, causing Phytophthora blight and Anthracnose disease in pepper, respectively. The strain L32 exhibited antagonistic activities both under dual culture assays and detached leaves assays. The each strain under the condition of mixed cultivation exhibited the same growth rates as one under pure cultivation. In greenhouse study, the mixed culture showed the both effect of plant growth promotion and reduction of disease symptom development against P. capsisi and C. gloeosporioides providing a potential as effective microbial agent for plant husbandry.

A Study of Stabilization for Insoluble Active Ingredients Using Swollen Micelles (Swollen Micelle을 이용한 난용성 효능물질의 안정화 연구)

  • Micelles, which are called solubilization formulation, have been widely used in skin lotion, mist or various formulations for the purpose of solubilizing the fragrance rather than the role of the active ingredient carrier because the particles are very small. In this study, we developed the swollen micelle having a transparent appearance to deliver ${\beta}$-sitosterol. When preparing the swollen micelle, solubility parameter was considered with active ingredient. This method allowed insoluble active ingredient to be safely entrapped inside the micelles. Stability of micelle was evaluated by dynamic light scattering (DLS). The transparency and shape of the micelles were confirmed by cryo-TEM. In addition, through the thermal analysis using DSC, ${\beta}$-sitosterol was found to be stably present in the swollen micelles. These results indicate that swollen micelles considered solubility parameter could be used as a new carrier for the insoluble active ingredients.
