The effects of Lycii fructus and Lycii folium on osteoporosis and serum cholesterol levels were tested in ovariectomized (OVX) rats. Twenty-four female Sprague-Dawley rats were divided into four groups: Sham group (sham-operated), Control group (OVX, ovariectomized model), LCF group (Ovx+Lycii fructus extract), and LCL group (OVX+Lycium folium extract). After 8 weeks, the OVX ($330{\pm}5.39$ g), LCF ($315{\pm}2.99$ g), and LCL ($318{\pm}2.06$ g) groups showed increased body weight compared with sham group ($281{\pm}1.71$ g). The levels of serum osteocalcin (OC) also increased in the LCF ($444.6{\pm}26.9$ ng/mL) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($107{\pm}3.52$ ng/mL). The activities of serum alkaline phosphatase (ALP) increased in the LCF ($108{\pm}2.7$ U/L) and LCL ($407{\pm}18.9$ ng/mL) groups compared with the OVX group ($95{\pm}2.9$ U/L). Stereomicroscopy found that the low bone density that developed in the OVX group was significantly reversed in the LCF and LCL groups after 8 weeks. We also obtained molecular-based in vivo evidence that supports a mechanism of action involving novel estrogen receptor ($ER{\alpha}$) modulator in the uterus. We found that expression of ER${\alpha}$ mRNA in the OVX rat uterus was elevated by Lycium chinense. These results suggest that Lycii fructus and Lycii folium administered to rats during 8 weeks after oophorectomy may partially recover postmenopausal osteoporosis or delay the progression of osteoporotic changes.
This study was conducted to investigate the ovarian cycle changes of the mare according to the season. Twenty four Jeju crossbred horses(Thoroughbred ${\times}$ Jeju horse) raised in Subtropical Livestock Research Institute, National Institute of Animal Science, RDA were used to identify follicles and corpus luteum with ultrasonography once a week(May 2016~June 2017). Blood samples of experimental horses were collected twice a week for analysis of P4 hormone levels. The mares were considered to have resumed ovarian cyclicity on the day of ovulation if they followed by regular ovarian cycles. Only 13 cases(61.9%) of the total 21cases showed normal ovarian cycle, and 8 cases (38.1%) showed delayed ovarian cycle. Three cases(16.7%) in October, 5 cases(27.8%) in November and 5 cases(27.8%) in December(27.8%) ceased the heat and the remaining 5 cases(27.8%) showed that the estrus was maintained in winter. Horses that stopped estrus ceased the heat until March of next year, and 27.8% were continued the heat during non-breeding season. Eleven cases(61.1%) of 18 cases in April and 2 cases(11.1%) of 18 cases in May returned the estrus.
The nuclear maturation and developmental competence of immature, oocytes collected from donors at various morphology of corpus luteum (CL) and fertilized in vitro was investigated by comparing the meiotic activity and the yields of embryos. Ovaries were divided and classified into 4 groups as the following criteria : Group 1 ; ovaries showed evidence of recent ovulation (corpus hemorragicum). Group 2 ; apex of CL was red or brown. Vasculization was limited to periphery of CL. Group 3 ; apex of CL was orange or tan. Vasculization was covered over apex of CL. Group 4 ; CL was light yellow to white and firm in texture and the vascular network on the surface of CL had disappeared. Modified TCM 199 was used for maturation in vitro of immature oocytes and development was induced by using TLP-PVA as a basic medium. When oocytes collected from each group of donors had been matured for 4, 14, and 24 hours in vitro, the proportion of oocytes reaching metaphase I and metaphase II were not different among oocytes from 4 group of ovaries. Mature metaphase II stage of oocytes in each group was first observed at 14 hours, whereas completion of maturation of. oocytes in each group was at 24 hours. Luteal morphology of ovaries had little effect on the proportion of embryos reached 2 cells and 8 cell stage. However, the proportion of embryos cleaved to morula and blastocyst stage was significantly higher in the oocytes obtained from group 1 and 3 than in the oocytes from group 2 and 4 (p<0.05). This data suggest that reproductive status of the donor significantly influence the yield of in vitro embryos.
In this study, we compared patterns of hemolvmph with ovarian proteins during the yolk formation of Gewis paludum by using SDS/polyacrylamide gel electrophoresis and he dimen-tlonal electrophoresis. In addition, we examined patterns of glycoprotein which were composed of yolk substances. The results were as follows: The protein patterns in ovary of the 5th instar without eggs were similar to those of adult after ovulation. Protein amounts of the ovaw without developing eggs were less than those of the ovaw containing oocvtes or matured eggs at the molecular weights range from 66, 000 to 110.000 daltons. No glvcoproteins were observed in ovaw without eggs. But the glvcoproteins were gradually increased according to development of eggs in the 5th instar. After the ovulated ovaw of adult, no glvcoprotein bands urere occurred as the bands of the ovary without eggs in the 5th instar. Also, amounts of hemolvmph protein between 66, 000 and 110, 000 in molecular weight were increased during yolk formation of the 5th instal. The results suggest that ovarian protein substances may originate from hemolpnph. In the 5th instar, the amounts of glycoprotein of developing eggs was gradually increased in the hemolymph. The band of M.W. 29, 500 was occurred in the hemolpnph of the 5th instar and adult without eggs. This protein mal be precursor of glvcoprotein in the high molecular weight area in the ovary of more developed eggs. The number of spots and the amounts of protein in ovary without eggs were less than those of ovary containing eggs by two dimensional elec-trophoresis. The protein bands between 45, 000 and 110, 000 almost appeared in acidic field of he dimensional gel. Especially, the band, M.W. 109, 000, uras separated 3 spots, a1, a2, and as. The band, M.W. 102, 000, has spots which designated b1, b2, b3, and b4. Besides, proteins below M.W. 24, 000 were occurred less spots in the basic field than those of acidic field. The mechanism of intracellular organs (= cell organells) uras related to the yolk protein synthesis of oocyte in the process of yolk protein formation of derris pcfudum.
Purpose: The aim of this experimental study is to observe the effect of platelet-rich plasma (PRP) on early bone regeneration of rats both in normal condition and in osteoporosis induced by ovariectomy. Material and methods: Total 40 Sprague-Dawley female rats were divided into 4 groups. A 8-mm-diameter calvarial critical-sized defect (CSD) was made by drilling with trephine at the center of calvaria in cranium of every rat. Every CSD was augmented with an osteoconductive synthetic alloplastic substitute ($MBCP^{TM}$) and PRP as follows. Group A; 10 non-ovariectomized rats grafted with only $MBCP^{TM}$. Group B; 10 non-ovariectomized rats grafted with $MBCP^{TM}$ and PRP. Group C; 10 ovariectomized rats grafted with only $MBCP^{TM}$. Group D; 10 ovariectomized rats grafted with $MBCP^{TM}$ and PRP. At 4 weeks after graft, every rat was sacrificed. And histomorphometric analysis was performed by measuring calcified area of new bone formation within the CSD. Results: Comparing four groups, results were obtained as follows. 1. In non-ovariectomized groups, PRP showed a positive effect somewhat but not significant (P > .05). 2. In ovariectomized groups, PRP showed a positive effect significantly (P < .05). 3. In PRP untreated groups, ovariectomy diminished bone regeneration significantly (P < .05). 4. In PRP treated groups, ovariectomy diminished bone regeneration somewhat but not significant (P > .05). Conclusion: Within the limitation of this study, it can be concluded that PRP in combination with an osteoconductive synthetic alloplastic substitute has an effect on bone regeneration more significantly in ovariectomized osteoporotic rats than in normal rats.

This study was conducted to investigate the effect of abnormal ovarian cycle postpartum on subsequent reproductive performance of Holstein cows. The cows were considered to have resumed ovarian cyclicity on the day of ovulation if followed by regular ovarian cycles. Total 58.8 percentage of the cows(l14/194) had normal resumption of ovarian cyclicity(resumption within 40 days after calving), and 41.2%(80/194) had delayed resumption(resumption did not occur until >40 days after calving). Delayed resumption Type I(one or more ovarian cycles with luteal phase >20 days, i.e. prolonged luteal phase; 17.5%) and delayed resumption Type II(first ovulation did net occur until $\geq$40 days after calving, i.e. anovulation 22.7%) were the most common types of delayed resumptions. When compared with cows with a normal ovarian cycle, the cows of delayed resumption Type I had a lower 100 days AI submission conception and pregnancy rates(84.2% vs 40.0%; p<0.01, 24.0% vs 21.4% and 20.2% vs 11.1%, respectively), and longer intervals to first AI and to conception($64.7{\pm}2.79$ days vs $105.7{\pm}7.48$ days and $105.1{\pm}7.16$ days vs $133.7{\pm}11.17$ days, respectively; p<0.01). Similarly, when compared with cows with normal ovarian cycles, the cows of delayed resumption Type II had lower 100 days conception and pregnancy rates(24.0% vs 20.0% and 20.2% vs 16.3%, respectively), and longer intervals to first AI and to conception($64.7{\pm}2.79$ days vs $72.6{\pm}4.45$ days and $105.1{\pm}7.16$ days vs $120.8{\pm}12.33$ days, respectively). In conclusion, abnormal ovarian cycles postpartum adversely affected reproductive performance, including AI submission rate, pregnancy rate, interval to first AI, and calving to conception interval in Holstein cows.

The study was carried out to elucidate the feedback mechanism on the hypothalamo-hypophyseal system from the functional changes of ovary in female rats. One hundred and forty-four mature female rats were lloted into the three groups; ovariectoimzed group, estradiol treated group and intact control group. The varies of 48 heads of rat were completely removed. Forty eight heads of rat were administered with $200{\mu}g$ of estradiol benzoate every 48 hours. Serum FSH, LH and prolactin levels were determined with radioimmunoassay method at 3,6,12,24 ours, and 5,10, and 15days after treatment. The rats were necropsied to measure the weights of hypophysis and to examin the histological changes in the organs. The results obtained were as follows: The weights of hypophysis were increased after ovariectomy and decreased after estradiol injection. The differences in hypophysis weights were significant between the group from 5 days after treatment. The histological changes in hypophysis were appeared from 5th day after ovariectomy. Proliferation and hypertrophy began to occur in basophilic from 10th day after ovariectomy, chromophobes were slightly hypertrophied and acidophilic cells were atrophied. In estradiol injected rats the histological findings were appeard to be contrary to those of ovariectomized rats. Serum FSH levels significantly changed after ovariectomy and estradiol injection and were higher in both the treated groups than in the intact control group. Within 18 hours after treatment the level was the highest in ovariectomized group, and thereafter the highest level was found in estradiol treated gorup. In ovariectomized rats the levels were rapidly increased 3 hours after treatment and maximum levels were found 18 hours after treatment. In estradiol treated rats the levels started to increase 18 hours after treatment and reached maximum levels 24 hours treatment. 4. Serum LH levels started to increase 3 hours after ovariectomy and estradiol injection and reached maximum levels 12 hours after ovariectomy and 24 hours after estradiol injection. There were significant differences in LH levels between the groups in each observation time. Up to 18 hours after treatment levels were higher in ovariectomized rats than in estradiol treated rats. but thereafter the levels were higher in estradiol treated rats than in ovariectomized rats. The multiple range test showed that a significant difference in LH levels was not found between ovariectomized group and estradiol treated group 18 hours and 5 days after treatment. 5. Serum prolactin levels were significantly changed after ovariectomy and estradiol injection. The levels were lower in ovariectomized rats than in intact control rats.

One hundred and forty-four mature female rats were allotted into the three groups ; ovariectomized group, estradiol treated group and intact control group. Serum estradiol-$17{\beta}$ and progesterone levels were determined with radioimmunoassay method at 3, 6, 12, 24 hours and 5, 10, 15 days after treatment. The rats were necropsied to measure weights of thyroid gland, adrenal gland and uterus and to examine the histological changes in the organs. The results obtained were as follows ; 1. Serum estradiol-$17{\beta}$ levels were rapidly decreased below 27.20pg/ml 18 hours after ovariectomy. In estradiol treated rats the levels were rapidly increased 18 hours after treatment, but thereafter slowly decreased. The significant differences in the estradiol level were found between the group at every observation time. 2. Serum progesterone levels were significantly decreased after ovariectomy and estradiol injection. The lowest level was found in the group of ovariectomized rats. 3. The weights of thyroid glands decreased in ovariectomized rats rather than in intact rats 5 days after treatment. The weights tended to increase after estradiol injection but significant differences between the groups were seen on 10th and 15th days. 4. In the histological findings of thyroid glands, follicular epithelial cells were changed to be squamous 5 days after ovariectomy and accompanied pyknosis 10 days and karyorrhexis 15 days after ovariectomy. On the contrary follicular epithelial cells were changed to be columnar with hypertrophy 10 days after estradiol injection. 5. The significant differences in adrenal gland weights were recognized between all the groups 5 and 15 days after treatment in ovariectomized rats were lighter than intact rats and the adrenal gland weights were rather heavier in estradiol treated rats. 6. The days after ovariectomy the adrenal glands were atrophied accompanying with pyknosis in the cortical cells of zona fasciculata. The cells in zona fasciculata and zona reticularis started to hypertrophy 5 days after estradiol injection, but no changes were found in the zona glomerulosa of adrenal cortex and in the adrenal medulla. 7. The significant differences in uterus weights were recognized between the groups at each observation time. After ovariectomy the uterus weights decreased rapidly but after estradiol injection they increased rapidly. 8. Through histological examination of uterus, the atrophy and degeneration started to occur in endometrium and lamina propria 12 hours after ovariectomy, and in myometrium one day after ovariectomy, and the changes progressed rapidly after that. On the contrary, the myometrium was proliferated and hypertrophied from 12 hours after estradiol-$17{\beta}$ injection.
