Estimation of the Genetic Parameters on Egg Components and Egg Qualities in Korean Native Ogol Fowl (한국재래오골계(韓國在來烏骨鷄)의 난구성분(卵構成分) 및 卵質(卵질)의 유전모수추정(遺傳母數推定))

  • This study was conducted to estimate heritabilities and genetic correlations on egg compositions and egg qualities in Korean Native Ogol fowl. The data were gathered from a total of 58,320 eggs in 450 pullets produced from 150 dams and 20 sires of Korean Native Ogol fowl raised at Chungnam National University from June 18, 1987 to April 6, 1989. The results obtained are summarized as follows : 1. The heritability estimates of egg compositions based on the variance of sires, dams and combined components were 0.620 - 0.723, 0206 - 0.300 and 0.413 - 0.511 for albumen weight: 0.439 - 0.737, 0.484 - 0.544 and 0.492 - 0.615 for yolk weight: 0.172 - 0.187, 0.412 - 0.642 and 0.309 - 0.503 for shell weight, respectively. 2. The heritability estimates of egg qualities based on the variance of sires, dams and combined components were 0.202 - 0279, 0.557 - 0.819 and 0.428 - 0.508 for shell thickness : 0.202 - 0.394, 0.119 - 0.678 and 0.256 - 0.440 for albumen height : 0.108 - 0.443, 0237 - 0.698 and 0244 - 0.399 for Haught units, respectively. 3. The genetic and phenotypic correlations of egg compositions were as follows; The coefficients between albumen weight and yolk weight were 0.089 - 0.564 and 0.084 - 0.235; between albumen weight and shell weight were 0.396 - 0.925 and 0225 - 0.544; between yolk weight and shell weight were 0.220 - 0.357 and 0.098 - 0.358, respectively. 4. The genetic and phenotypic correlations of egg qualities as follows; between shell thickness and albumen height were 0.082 - 0.356 and - 0.163 - 0.060; between shell thickness and Haught units were - 0.076 - 0.167 and - 0.185 - 0.010; between albumen height and Haught units were 0.338 - 0.604 and 0.154 - 0285, respectively. 5. The genetic correlations of egg compositions and egg qualities were as follows: between albumen weight and shell thickness, albumen height. Haught units were - 0.380 - - 0.002, 0239 - 0.387, and - 0279 - - 0.127; between yolk weight and shell thickness, albumen height, Haught units were - 0.294 - - 0.133, - 0.049 - 0.133 and - 0.196 - - 0.136; between shell weight and shell thickness, albumen height, Haught units were 0.127 - 0.476, 0.140 - 0273 and 0.038 - 0223, respectively.

The Effect of Hen Age on Egg Quality in Commercial Layer (실용산란계의 산란연령이 계란의 품질에 미치는 영향)

  • Egg quality is a very important factor for both consumers and producers. Factors affecting egg quality include strain and age of hens; egg storage temperature, time, and humidity; laying season; and feeding. This study aimed to determine the effect of hen age and egg storage time on egg quality. A total of 700 eggs obtained from Hyline Brown commercial layers were used for this experiment, and they were separated into two hen age groups (30 vs. 60 weeks) with eight treatments and four storage times (day 0, 10, 20, and 30). The egg weight; shell color, thickness, and density; albumen height; Haugh unit (HU); yolk color; and the yolk and albumen pH and viscosity were measured for the egg quality assessment. The results showed that the age of the hen and egg storage time significantly affected almost all parameters of the internal and external egg quality. The shell thickness, albumen height, HU, yolk color, pH of yolk and albumen, and yolk viscosity significantly decreased with increasing hen age. The egg shell color was significantly lighter in eggs from 60-week-old hens than in those from 30 weeks-old hens. The egg weight; shell weight, thickness, and density; albumen height, HU; and albumen viscosity significantly decreased, but the yolk color and pH of the yolk and albumen increased with increasing egg storage time. The interaction effects between the storage time and hen age were significant in shell thickness, albumen height, yolk color, and yolk and albumen pH and viscosity. The eggs obtained from 60-week-old hens showed significantly lower shell thickness, albumen height, and HU values, which are considered typical egg quality measurements, than values of eggs from 30-week-old hens. Therefore, increasing hen age and egg storage time caused the deterioration of egg quality. In conclusion, this study demonstrated that hen age is the major factor affecting the quality of fresh eggs, whereas the storage time is the determinant factor affecting the quality of stored eggs.

Effect of High Dose ${\gamma}-irradiation$ on the Physicochemical Properties of Shell Eggs during Storage (고선량 감마선 조사가 신선란의 저장 중 이화학적 특성에 미치는 영향)

  • To determine the quality change of the irradiated eggs during storage, fresh shell eggs were irradiated using $^{60}Co$ at 0, 1, 5, 10, 30 kGy and stored for 30 days. The york index, color, pH, viscosity, egg weight, and SDS-PAGE profile of the irradiated eggs were examined. During storage, york index values of the irradiated eggs and the control were decreased and the increase of dose decreased yolk index. However, the yolk index values were increased temporarily at 10 kGy and 30 kGy. The yolk color had a bright yellow with increases in dose level and there was no significant change during storage. The albumen viscosities were decreased with increases in dosage and were decreased during storage. Also, the albumen pH values of the irradiated eggs were higher than that of the control and were increased during storage. The weight losses of eggs were increased during storage and there were no significant changes by dose level. SDS-PAGE profile of the egg white proteins of the shell eggs showed the change in molecular weight distribution and had aggregation pattern as well as degradation. CD and fluorescence spectroscopy study showed changes in the secondary and tertiary structure of egg white proteins by ${\gamma}-irradiation$. Therefore, this study clearly indicates that irradiation dose of eggs should be appropriate to prevent the loss of egg qualities.

  • In vitro 소화시 계란 성분이 anti-Y ruckeri IgY 항세 활성을 안정화시키는데 어떡해 관여하는지를 SDS-PAGE와 ELISA로 조사한 결과는 다음과 같다. Anti-Y ruckeris IgY 항체와 난황 및 난백으로 혼합 후 펩신으로소화시킨 후 경우 1시간 후 난황 및 난백이 혼합된 시료에서 모두 anti-Y ruckeri IgY 항체의 heavy chain과 light chain이 분해는 되었지만 어느 정도 밴드를 관찰할 수있었으며, anti-Y, ruckeri IgY 항체 활성 측정한 결과 난황시료는 35%, 난백시료는 61%의 항체 활성을 유지시켰다. Anti-Y ruckeri IgY 항체와 오보알부민, 오보뮤우신, 라이소자임 및 오보뮤코이드를 혼합 한 후 펩신으로 소화시킨 경우 1시간 우에는 anti-Y, ruckeri IgY항체의 heavy chain의 밴드르 fdjsm 정도 볼 수 있는 형태로 나타났으며, anti-Y, ruckeri IgY 항체의 light chain는 오보뮤우신 및 오보뮤코이드가 홉합되어 펩신으로 소화시킨 경우 1시간 후에는 밴드를 거의 볼수 없었으나 오보알부민 오보트란스훼린 및 라이소자임에 혼합된 시료에서 밴드를 관찰할수 있었다. 특히 라이소자임과 오보트란스훼린의 경우 펩신 호화 2시간 우에도 anti-Y, ruckeri IgY 항체의 light 밴드가 관찰되었다. 펩신소화 1시간 후에 난백성분이오보알부민, 오보뮤우신, 라이소자임, 오보트란스웨린 및 오부뮤코이드 중에서 단지 오보트란스훼린만이 38%의 anti-Y, ruckeri IgY 항체 활서을 보인후 2시간 후에도 15% 정동의 활성을 나타내었다. anti-Y, ruckeri IgY항체와 난백 및 전란을 혼합한 다음 무지개송어 위 추출액으로 소화시 2시간 후에 난백은 14%, 전란은 69%로 anti-Y, ruckeri IgY 항체 활성을 유지시킨 것으로 나타났다.X> $e_{I}$ WPi_BE_QE]]]]]로 상징하며 WLWQ에 적용되는 몇 가지 제약을 관찰하고 이를 일반적인 언어원리로 설명한다. 첫째, XP는 주어로만 해석되는데 그 이유는 XP가 목적어 혹은 부가어 등 다른 기능을 할 경우 생략 부위가 생략의 복원 가능선 원리 (the deletion-up-to recoverability principle)를 위배하기 때문이다. 둘째, WLWQ가 내용 의문문으로만 해석되는데 그 이유는 양의 공리(the maxim of quantity: Grice 1975) 때문이다. 평서문으로 해석될 경우 WP에 들어갈 부분이 XP의 자질의 부분집합에 불과하므로 명제가 아무런 정보제공을 하지 못한다. 반면 의문문 자체는 정보제공을 추구하지 않으므로 앞에서 언급한 양의 공리로부터 자유롭다. 셋째, WLWQ의 XP는 주제어 표지 ‘는/-은’을 취하나 주어표지 ‘가/-이’는 취하지 못한다(XP-는/-은 vs. XP-가/-이). 이는 IP내부 에 비공범주의 존재 여부에 따라 C의 음운형태(PF)가 시성이 정해진다는 가설로 설명하고자 했다. WLWQ에 대한 우리의 논의가 옳다면, 본 논문은 다음과 같은 이론적 함의를 기닌다. 첫째, WLWQ의 존재는 생략에 대한 두 이론 즉 LF 복사 이론과 PF 삭제 이론 중 전자의 입장을 지지한다. 둘째, WP를 XP로부터 복원할 때 부분 자질만 복사된다. 이는 어휘가 통사층위로 들어온 이후에도 어휘 자질들이 완전히 동결되는 것이 아니라 계속 지시될 수 있다는 가설을 지지한다.

Separation of Highly Purified Antimicrobial Lysozyme Using Ultrafiltration and Characteristics of Membrane Fouling (한외여과 공정을 이용한 고순도 향균 Lysozyme 의 분리 및 막 침착 특성)

  • The value of lysozyme as a natural food preservative is continuously increased due to its unique antimicrobial activity. To determine the optimum separation concentration among the various hen egg white protein (HEWP) concentrations (0.25, 0.5, 1.0, w/v), protein concentrations, lysozyme concentrations, specific activities (SA), and purification factors of prefiltered solution (PFS) and PM30 permeate solution (PMS) were compared. The purity of lysozyme separated at each step was analyzed and confirmed by gel permeation chromatography and electrophoresis. The fouling deposits on membrane were observed by SEM. The non-enzymatic proteins were removed over 99% by ultrafiltration (UF). The increased feed concentration did not contribute to the increase of SA. SA of PMS was 18 to 31 times higher than that of PFS. The optimum feed concentration was decided as 0.25% based on SA and purification factor. The non-enzymatic region of gel chromatogram was proved to be ovalbumin. The thickness of deposit on the UF membrane was approximately $0.9{\mu}m$ and removed by cleaning with 0.1 N NaOH. Therefore, UF using PM30 membrane was very effective to separate the antimicrobial lysozyme from various HEWPs.

Quality Factors Affecting Egg Freshness and Palatability During Storage (계란의 저장에 따른 물리화학적 특성 평가 및 품질 인자 설정)

  • This study investigated the effects of storage temperature and time on the physicochemical properties of eggs, and the corelationship of quality factors with egg freshness and the palatability during storage. As storage temperature and time increased, weight loss significantly increased (p<0.05), while the specific gravity, Haugh unit, yolk index, albumen index, and yolk viscosity decreased significantly (p<0.05). A multiple regression equation of $Y_1$ (freshness)=$106.50286-0.81196X_1$ (weight loss)-$77.51667X_5$ (specific gravity)-$2.27156X_8$ (yolk pH)+$0.00178X_{16}$ (yolk viscosity) with $r^2$=0.89 was obtained for the evaluation of egg freshness. Also, sensorial palatability through the multiple regression equation was determined as $Y_2$ (palatability)=$104.98174-0.81466X_1$ (weight loss)-$76.53434X_5$ (specific gravity)-$2.20338X_8$ (yolk pH)+$0.00178X_{16}$ (yolk viscosity) with $r^2$=0.89 indicating that weight loss, specific gravity, yolk pH and yolk viscosity are major factors that affect the freshness and palatability of eggs during storage.

  • This study was conducted to estimated the genetic and phenotypic correlations between economic traits and certain other traits in Korean Native Ogol fowl The data analysis were the record of 450 pullets bred from 150 dams and 20 sires of Korean Native Ogol fowl at Chungnam National University from June 18, 1987 to April 6, 1989. The results obtained are summarized as follows : 1 The genetic correlation coefficients of the economic traits and body shape components were as follows : between body weight and shank length, breast width. breast girth, tibia length were 0.210~0.788, 0.231~0.826, 0.610~0.995 and 0.096~0.503 between age at first egg and shank length, breast width, breast girth, tibia length were 0.555~0.626, 0.149~0.270, 0.370~0.445 and 0.014-0.124. between number of egg production and shank length, breast girth, tibia length were -0.446~-0.167, -0.162~-0.320, 0.076~0.336 and 0.203~0.312 : between egg weight and shank length, breast width, breast girth, tibia length were 0.132~0.498, 0.236~0.410, 0.148~0.775 and -0.019~0.593, respectively. 2. The genetic correlation coefficients of the economic traits and egg components were as follows : between body weight and albumen weight, yolk weight, shell weight were 0.083~0.591, 0 110~0.541 and 0.336~0.782 between age at the first egg and albumen weight, yolk weight, shell weight were 0.476-0.692, 0.265~0.631 and 0.420~0.519 between number of egg Production and albumen weight, yolk weight, shell weight were -0.578~-0.240, -0.255~-0.060, -0.477~-0.313. between egg weight and albumen weight, yolk weight, shell weight were 0.825~0.939, 0.382~0.564, 0.374~0.337, respectively. 3. The genetic correlation coefficients of the economic traits and egg qualifies were as follows : between body weight and egg shape index, shell thickness, albumen height, Haugh units were 0.215~0.367, 0.248~0.650, 0.161~0.624, 0.157~0.449. between number of egg production and egg shape index, shell thickness, albumen height, Haugh units were -0.384~-0.207, -0.557~-0.306, -0.555~-0.198, -0.582~-0.074 between egg weight and egg shape index, shell thickness, albumen height, Haugh units were 0.276~0.697, 0.290~0.627, 0.238~0.538, -0.207-0.020, respectively. 4. The genetic correlation coefficients of egg compositions and egg qualities were as follows : between albumen weight and egg shape index, shell thickness, albumen height and Haugh units were 0.110~0.584, -0.380~-0.002, 0.239~0.887 and -0.195~0.279 : between yolk weight and egg shape index, shell thickness, albumen height and Haugh units were -0.204~0.160, 0.294~0.133, -0.049~0.133 and -0.196~-0 136 : between shell weight and egg shape index, shell thickness, albumen height and Haugh units were 0.127~0.503, 0.127~0.476, 0.140~0.273 and -0.172~0.233, respectively.

Studies on the Egg Storage Technology (계란비축을 위한 저장기법연구)

  • This experiment was carried out to establish effective egg storage technology by studying the physico-chemical properties of albumen and yolk during the cold storage. The results obtained were summarized as follows:1. Egg Yolk was gelated and whipping quality and whipping stability of albumen was decreased by the freezing storage. 2. NaCl was the best anti-freezer to prevent the gelation of egg yolk among various anti-freezer (NaCl, glucose, sucrose, amides). 3. Viscosity of egg yolk and albumen was increased and color was changed but changes of colorin albumen was slight during the storage at $-5^{\circ}C$ and $-20^{\circ}C$. 4. To store the 5% NaC1 added egg yolk -at $-5^{\circ}C$ was the best way to prevent the gelation of egg yolk and save the energy. 5. Emulsion capacity of egg yolk was decreased by the cold storage with the addition of NaCl.

  • Separation and purification of lysozyme from chicken egg white was studied using ultrafiltration. We have obtained experimental data through the cellulose membranes with the molecular weight cut off (MWCO) of 10 kDa, 30 kDa and 100 kDa in a stirred ultrafiltration device. Certain amounts of egg white were dissolved into 20 mM phosphate buffers of pH 6, 7 and 8 to make protein solutions of $1\%,\;2\%,\;3\%\;and\;10\%$ concentration. Permeation flux increased with increasing MWCO of the membrane. Permeation flux increased with increasing transmembrane pressure (TMP) and decreasing the protein concentration. As the MWCO of membrane decreased, the selectivity increased. The selectivity increased with increasing TMP and protein concentration of the solution.
