This experiment was conducted to identify the optimal in vitro propagation condition of Cnidii rhizoma (Cnidium officinale Makino). It was effective to reduce contamination and improve regeneration of shoot when shoot tips taken in July were cultured in 1/2 strength Murashige and Skoog medium supplemented with 500 mg/L carbenicillin disodium 1.0 mg/L BA and 1.0mg/L $GA_3$followed by surface sterilization of explant source in solution of 1% sodium hypochlorite for 20 minutes. When shoot tips were 쳐cultured in 1/2 strength MS medium with 0.5 mg/L BA and 60 g/L sucrose, shoot elogation and subsequent multiplication of the formed shoot were favorable than in other media. Regenerants were well rooted in 1/2 strength MS medium containing 3.0 mg/L NAA.
This study was carried out to develop reliable systems for somatic embryogenesis in oil palm tree (Elaeis guineensis Jacq.), and to verify the somaclonal variants by RAPD analysis. Embryogenic callus was induced successfully on modified half-strength MS medium containing $NaH_2PO_4{\cdot}2H_2O$ and casein. Embryogenic callus was further developed to somatic embryo mass (SEM), which is very hard and bonded tightly each other. Plantlets were proliferated when SEM was cultured on modified MS medium containing half strength $NH_4NO_3$, casein and L-ascorbic acid. Plantlets were transplanted into pots containing artificial soils. When RAPD analysis was conducted using randomly selected 95 in vitro plantlets and 19 random primers, somaclonal variation was detected using BNR35 primer. There was missing band around 1 kb in #22, #28, #35, and #77 plantlets. In addition, bands obtained from #28, #35, and #77 was much stronger than other normal bands. The blast results at NCBI revealed that somaclonal variation observed in this study was related to chloroplast genome of oil palm. The results also revealed that oil palm reproduction system through somatic embryogenesis is quite reliable and early detection of somaclonal variants seem to be possible at in vitro stage by RAPD analysis.
This study was conducted to determine the effects of 3 cytokinins (BA,2iP and kinetin) and their concentrations (0, 0.1, 0.5, 1.0, and 2.0 mg/L) on multiple shoot production of Citrus spp. 'Sambokam' and 'Byungkyool' by nodal culture. Nodal explants were obtained from in vitro germinated seedlings of both cultivars. 'Sambokam' produced more multiple shoots than did 'Byungkyool' by nodal culture. Among the 3 cytokinins tested in this study BA supplemented in semi-solid MS basal medium was the most effective stimulator for multiple shoot production, and an optimal concentration was determined to be 1.0 mg/L. Shoot elongation and root formation were inhibited by increasing cytokinin concentration, regardless of cytokinin types. BA at 1.0 mg/L produced the most multiple shoots and the highest number of leaves in 'Sambokam', whereas any cytokinin and concentration studied in this experiment did not affect any scored variables such as shoot and leaf numbers, etc. in 'Byungkyool'.
Chinese foxglove (Rehmannia glutinosa) is receiving much attention as one of the principal medicinal crops and the crude drug damand expands rapidly.This study was conducted to obtain the basic breeding information of Chinese foxglove. Effects of supplemental plant growth regulators were investigated on leaf tissue for proliferation. 100% callus formation, 31% plantlet regeneration and 6% root differentiation were obtained by adding 0.5 mg/L NAA and 2.0 mg/L BA. 2,4-D and Zeatin treatment also resulted in 95% increase in callus formation, but shoot was not formed. During the subculture, callus propagation rate recorded 15.4% with 0.2 mg/L NAA and 1.0 mg/L BA and plant regeneration improved on MS medium supplemented with 0.2 mg/L NAA and 0.5 mg/L kinetin. The number of shoot formed ranged from 1.7 on WPM medium to 3.4 on MS medium with 0.1 mg/L NAA and 0.5 mg/L BA. Supplementation of 1.0 g/L activated charcoal improved the In vitro plant growth.

The application of bioreactor culture techniques for plant micropropagation is regarded as one of the ways to reduce production cost by scaling-up and automation. In an attempt to optimize mass proliferation systems in Eucalyptus pellita, four types of bioreator systems including temporary immersion system with or without net were tested. Highest growth was achieved with 30-min flushes of medium at every 4-h intervals in TIN (temporary immersion with net) system. Results indicate over three-fold increase in shoot growth with the TIN system when compared with TIX (control: temporary immersion without net) system which is without net in bioreactor. Furthermore, plants produced from the TIN system increased total chlorophyll content, chlorophyll a/b and dry matter, giving higher yields of acclimatized plants. Our findings suggest that plantlet growth increases with appropriate exposure to media at correct intervals, as well as use of net for maintaining aerobic condition in the vessels. The TIN system thus has great potential for in vitro mass production of Eucalyptus clones commercially.
Guzmania was propagated through in vitro culture of lateral shoots. When new shoots grown in greenhouse were cut and cultured in vitro, contamination rate was very high at about 80% in the first stage of in vitro culture. Among cytokinin treatments for agar medium, 2.0 mg/L BA was most effective for shoot multiplication, and those with 0.5 mg/L kinetin and 0.5~1.0 mg/L BA were favorable for shoot multiplication. BA was more effective for shoot multiplication than kinetin, and shoot multiplication was more enhanced when 2.0 mg/L BA was combined with 0.1~0.5 mg/L IAA than 2.0 mg/L BA alone. The medium with 2.0 mg/L BA and 0.1 mg/L IAA showed the highest rate of shoot multiplication with about 8.7 in shoot number, and those with 2.0 mg/L BA and 0.5~1.0 mg/L IAA also resulted in high multiplication of shoots. Shoots were multiplicated more in liquid rotation culture(80 rpm) with the medium containing 0.5 mg/L BA and 0.1 mg/L IAA than liquid stagnating and solid cultures. Regenerated shoots formed roots very favorably in the medium supplemented with 2.0 mg/L IBA.

본 연구는 응달고사리[Cornopteris crenulatoserrulata (Makino) Nakai]의 전엽체 대량증식 및 포자체 형성을 위한 배지구성물질 및 배양토의 최적조건을 구명하고자 수행되었다. 실험재료는 기내에서 포자발아를 통해 획득한 전엽체를 계대배양하여 실험에 사용하였다. 배지구성물질이 전엽체의 증식 및 형태 형성에 미치는 영향을 알아보고자, 배지구성물질의 농도를 1/4, 1/2, 1배로 조절한 MS배지와 Knop배지를 조성하였다. 기내 배양된 전엽체 300mg을 메스로 다진 후 상기 5종의 배지에 배양하였다. 실험의 결과, 전엽체의 생체중은 Knop배지에서 3.93g으로 가장 많이 증가되었으며, 현미경으로 관찰한 결과 기관형성도 정상적으로 이루어졌다. 선행연구에서 선발된 Knop배지를 기준으로 sucrose(0, 0.5, 1.0, 2.0 및 3.0%) 및 활성탄(0, 0.1, 0.2 및 0.4%)의 농도를 달리 첨가하여 전엽체에 미치는 영향을 확인하였다. 실험결과, 0.5%의 sucrose를 첨가한 배지에서 생체중이 5.90g으로 증가하였으며, 활성탄은 첨가하지 않는 것이 효과적이었다. 포자체 형성을 위한 최적의 토양조건을 구명하고자, 원예상토와 피트모스, 펄라이트, 마사토 등 인공토양의 비율을 달리하여 8종류의 토양을 사각분에 충진하였으며, 기내에서 배양된 전엽체를 10초간 분쇄한 후 사각분에 1g씩 균일하게 분주하여 16주간 배양하였다. 배양결과, 원예상토를 단용으로 처리한 실험구에서 포자체가 171.5개로 가장 많이 발생하였으며, 원예상토와 마사토를 2:1(v:v)로 혼합한 토양에서 125.0개, 원예상토, 피트모스, 마사토를 1:1:1(v:v:v)로 혼합한 토양에서 113.3개 순으로 포자체가 발생하였다. 포자체의 생육은 포자체의 형성이 가장 많았던 원예상토를 단용한 토양에 비해 원예상토와 마사토를 2:1(v:v)로 혼합한 토양에서 생체중, 엽장, 엽폭, 근장 등이 우수하였다. 한편 피트모스가 혼합된 토양에서는 전엽체와 포자체 형성이 억제되는 경향을 보였다. 위 결과로 보아 응달고사리의 전엽체 기내배양에는 sucrose 0.5%가 첨가된 Knop배지가 적합하며, 포자체 대량생산을 위해서는 원예상토를 단용한 토양을 사용하는 것이 좋다고 판단되었다.
In order to establish a micropropagation system of Naricissus, the ability of bulblet regeneration among propagation materials was compared, and the adequate growth regulators and concentrations for each cultivar were investigated. The inorganic components were also assayed in the parts of propagation materials. In propagation materials, scape with based plates showed hightest rate of bulblet formation and rapid growth of formed bulblets in vitro, comparing to other parts of it. In comparing of varieties, 'Dutch Master' and 'Golden Harvest' showed a high ability for bulblet regeneration. The ability of bulblet regeneration was most favorable in the medium, supplemented with 5.0 mg/L BA and 2.5 mg/L NAA in 'Dutch Master', and 5.0 mg/L BA and 1.0mg/L NAA in 'Golden Harvest', respectively. In inorganic component analysis of propagation materials, the White part of scape contained 1.18 mg/L$P_2O_5$, 2.57 me Ca, 0.94 me Mg and 3.20 mg/L total N. It showed higher levels in concentration of inorganic components as compared to those of the other part of scape. In addition, leaves and yellow part of scape contained significantly high levels of Ca and Mg while scales bulb showed considerably low levels in all inorganic compounds.
This study was conducted to investigate the effect of supporting material and ionic strength of the MS medium on growth of strawberry plantlets cultured in vitro for the rapid mass production. Explants of $Fragaria$$ananassa$ 'Houkouwase' in vitro were planted in the agar or Tosilee (commercial plug medium) medium as the supporting material and supplied with 1/4 MS, 1/2 MS or basal (as the control) MS medium in an autotrophic micropropagation. Plant height and root length were significantly greater when they were cultured in 1/2 MS medium as compared to those grown in the agar medium. Also, shoot fresh and dry weights, and leaf area in the 1/2 MS medium were greater than in 1/4 MS or basal MS medium. When plantlets were cultured in Tosilee medium and fed with the basal MS medium, plant height, root length, shoot fresh and dry weights, root fresh and dry weights, and leaf area were promoted and greater than those in plantlets cultured in the agar medium.

The purpose of this study was to investigate the suitable parts for callus induction and optimal concentrations of growth regulators contained in the medium affecting shoot and rooting for the in vitro mass production of Haworthia maughanii. To determine suitable parts of the plant for callus induction, the leaves, flower bloom and flower stalks were cultured in MS medium at different concentrations of $0{\sim}2mgL^{-1}$ NAA and $0{\sim}2mgL^{-1}$ TDZ, respectively. All of the parts showed 100% callus formation rate at $NAA\;1mgL^{-1}$ and $TDZ\;1mgL^{-1}$ treatment, $NAA\;2mgL^{-1}$ and $TDZ\;2mgL^{-1}$ treatment and NAA 1 to $2mgL^{-1}$, respectively. While the rate of callus formation was high in all parts, the leaves were the most efficient to obtain most culture parts. $NAA\;0.1mg\;L^{-1}$ and $BA\;0.1mg\;L^{-1}$ treatments were the most effective in shoot formation with 22.0 shoots. In addition, multiple shoot propagation showed 16.3 shoots, the highest, with $NAA\;0.1mg\;L^{-1}$ and $BA\;0.1mg\;L^{-1}$ treatments. These results led us to speculate that the optimization of culture conditions was responsible for the mass propagation for in vitro cultures of Haworthia maughanii.
