Studies on Specific Genes Related to the Regulation of Muscle Cell Differentiation (근세포 분화 조절에 관련된 특이 유전자에 관한 연구)

  • 배양 중의 골격근 세포는 증식을 거쳐 세포융합을 통해 다핵세포로 분화되므로 세포분화의 연구에 좋은 모델로서 이용되고 있다. 이전 실험에서 근원세포 융합을 억제하는 단일클론항체(MII-3J31)가 제작되었으며(Kim et al., 1992)이 항체에 대한 항원은 분자량이 약 35 kDa인 세포막 단백질로 추정되었다. 본 실험에서는 13일 계배와 성체의 근섬유 mRNA에서 CDNA라이브러리를 제작하여 근원세포 분화에 특이적으로 나타나는 유전자를 추적하였다. 근원세포 융합에 관여하는 단백질에 대한 CDNA는 계배 13일 째의 근원세포 CDNA라이브러리에서 단일클론항체를 사용한 immunoscreening 방법을 이용하여 확인하였다. 이 CDNA의 크기는 약 1.5 kb였다. 한편 13일 계배와성체 근섬유 CDNA 라이브러 리를 이용하여 13일 계배에만 특이하게 유전자 발현 이 일어 나고 성체에서는 나타나지 않는 약 0.8 kb의 CDNA플 찾았다.

  • Inflammatory myofibroblastic tumor (IMT) is a rare benign tumor, that is composed of myofibroblastic spindle cells with inflammatory cells. IMTs usually occur in lungs, intestine organs, orbits and paranasal sinuses, however, it may rarely be seen in the larynx. We present two cases of patients with laryngeal IMT that had different causes and prognosis.

  • Myofibroma is a rare spindle cell neoplasm that consists of myofibroblsasts. Although it was originally described as a multicentric tumor process affecting infants and young children, it is now recognized that most cases of the tumor are soliatary and that it can occur at any age. This case is a myofibroma involving the right mandibular body in a 9-year-old boy. Radiographic examination revealed a relatively well-demarcated radiolucent lesion surrounding the impacted right mandibular canine and first premolar crown. The histologic sections composed of interlacing bundles of spindle cells with tapered or blunt-ended nuclei admixed with a foci of hemangiopericytoma-like appearance. Immunohistochemical staining demonstrated a positive reaction to vimentin and smooth muscle actin, but they are negative for desmin and S-100. The presence of well circumscribed radiolucent solid lesion in children as observed in this case, differential diagnosis of myofibroma must be included. In this case, there was no recurrence 6 months after enucleation and eruption guide of the impacted tooth is ongoing now.

Influence of NaCl and pH on Hydrolysis of Chicken Myofibrillar Proteins by Leukocyte Lysosomal Proteinases (Leucocyte lysosomal proteinase에 의한 닭의 근섬유(筋纖維) 단백질(蛋白質) 분해(分解)에 미치는 NaCl과 pH의 영향(影響))

  • The influence of NaCl and pH on degradation of chicken breast muscle myofibrillar proteins by porcine leukocyte lysosomal proteinases was investigated. The degradation reactions were carried out at $38^{\circ}C$ for 24hours under different conditions. The degradation of myofibrillar proteins by leukocyte lysosomal enzymes at various pH values was limited to partial hydrolysis. Reactions at higher pH values resulted in lower molecular weight degradation products while reactions at lower pH resulted in higher molecular weight degradation products. When NaCl was added into the reaction mixture, enzyme activities of degradation were increased at all pH values studied, as evidenced by NPN-analysis and SDS-PAGE. More severe degradation was observed with higher salt concentration. The concentration of 0.5M NaCl in the reaction mixture gave more degradation of myosin heavy chain by enzyme than that of 0.1M NaCl.

  • This study was carried out of to determine effects of high voltage pulsed galvanic stimulation on the soleus. target muscle of the sciatic nerve, of white rat two condition. The coditions included normal, and denervated muscle. The histochemical, ultrastructural observations were made. The following results were obtains. 1. The histochemical observations found the inflammatory cells between muscle bundle and muscle fiber since 1-week control group. In addition, nucleus located in the muscle fiber was frequently observed. 2. The experimental group showed a similar phenomenom to the normal muscles in terms of glycogen granules in the 1-week group, where as fiber were not distinguishable in4-weeks group which indicated that the degenerative changes had occured. 3. The NADH-TR reaction showed that the red muscle slightly increased in the 2-weeks group, and the distinguished was impossible the red fiber 4. The ultrastructures of the muscles in both groups were severely bend, and a number of vacuoles were observed due to the destruction of mitochondria..

  • A 34-year old man was admitted our hospital because he wished to evaluate the pulmonary mass that was incidentally detected on healthy examination. Bronchoscopy and chest CT showed endobronchial and peribronchial mass of the left lower lobe of the lung. Open thoracotomy and left lower lobectomy of the lung was done. Pulmonary mass was confirmed as a pulmonary inflammatory myofibroblastic tumor with bronchus invasion pathologically. Pulmonary inflammatory myofibroblastic tumor with bronchus invasion is a vary rare. Herein we report a case of pulmonary inflammatory myofibroblastic tumor with bronchus invasion.

Comparison of Pork Quality and Muscle Fiber Characteristics between Jeju Black Pig and Domesticated Pig Breeds (제주 개량 흑돼지와 국내에 도입된 주요 돼지 품종의 육질 특성 및 근섬유 특성 비교)

  • This study compared the pork quality and muscle fiber characteristics between the Jeju black pig (JBP) and domesticated pig breeds, including three-way crossbred (Landrace ${\times}$ Yorkshire ${\times}$ Duroc, LYD), Berkshire (B), Duroc (D), Landrace (L), Meishan (M), and Yorkshire (Y) pigs. JBP had the lowest carcass weight among the pig breeds (p<0.05) and its NPPC marbling score was lower than the other domesticated pig breeds (p<0.05), except for M. M had the highest value for backfat thickness among the pig breeds (p<0.05), whereas its NPPC marbling score was the lowest (p<0.05). The pH values were higher in JBP and B at 45 min and 24 h postmortem, respectively, but LYD and M had the lowest pH values at 45 min and 24 h postmortem, respectively (p<0.05). M had a higher lightness and lower redness than the other breeds (p<0.05). Moreover, M had the highest drip loss among the pig breeds (p<0.05), whereas D had the lowest drip loss and the highest redness (p<0.05). In terms of muscle fiber characteristics, JBP and LYD had the largest type IIB fibers (p<0.05). The fiber number and area composition of type I fibers were higher in JBP than the others (p<0.05) however type IIA fibers were lowest in JBP among the pig breeds (p<0.05). Overall, our results indicate that JBP provided better pork quality compared with M, but the carcass trait and pork quality of JBP were not better than LYD and D, respectively.

  • Electron micrographs of muscle of plaice killed instantly by spiking at the head clearly showed the A-band, I-band, Z-line and M-line in muscle strips, whereas these bands could not be distinguished from each other in electrically stimulated plaice muscle strips. As the electrical stimulation time increased, the continuity of Z-line disrupted rapidly. Electron microscopic observation showed that sarcoplasmic reticulum (SR) in unstimulated plaice had natural triad structures between Z-line. However these structures were disrupted in the electrically stimulated sample. These structures were clearly observed after storage for 10 hrs at $5^{\circ}C$ from all the samples. In association with the $Ca^{2+}$ translocation, $Ca^{2+}$-pyroantimonate deposits were observed at the inner part of SR immediately after spiked the plaice, However, $Ca^{2+}$-pyroantimonate deposits in electrically stimulated plaices were observed in the muscle strips and this phenomenon was clearly observed when electrical stimulation was prolonged.
