Chinese Radish Juice as a Growth Substrate for the Production of Candida utilis ATCC 42416 Biomass (무즙을 이용한 Candida utilis ATCC 42416균체 생산)

  • Chinese radish juice(CRJ) was used as a culture medium for the production of Candida utilis ATCC 42416 biomass. Soluble solid and total sugar contents of Chinese radishes were in the range between 5.5 and $8.8^{\circ}$Brix and 3.5 and 6.5%, respectively. Since sugar in radishes are in readily usable forms, pretreatm ent procedures were not necessary after the extraction of juice from fresh radishes. In shake flask experimetns, C. utilis ATCC 42416 grew well in CRJ and completed growth in 24 hrs at $30^{\circ}C$ and 200 rpm. Maximum cell dry weight obtainable from a liter of CRJ(1.0% sugar $DCRJ{\times}5$) was 21.5g, when the yeast was grown on CRJ diluted 5 times or more with tap water to make sugar content to be eual to or less than 1.0%. Supplementation of 5-fold diluted CRJ with some nutrients did not greatly influence the growth rate, yeast biomass production, or cell protein content significantly, indicating that CRJ itself was a good substrate for the production of biomass by C. utilis ATCC 42416.

  • Seventeen extracellular protein producing bacteria were isolated from soil samples, among which T219 strain having a strong capability of producing the protein was selected and identified for investigation of biological characteristics. The factors which affect the protein production were investigated and the results are summarized as follows. T219 strain which produces the most extracellular protein was identified as Bacillus sp. Optimum temperature and pH for production of the extracellular protein by T219 strain were $25^{\circ}C$ and 7.5 respectively. Almost no activities of protease and amylase were observed in the protein produced by the protein producing bacteria. In the medium containing yeast extract, the cell growth was moderately high, but almost no accumulation of protein was observed. However, polypeptone had significant effects on both the cell growth and the protein accumulation. The addition of glycine and L-isoleucine to the medium containing polypeptone, yeast extract and meat extract had a great effect on the protein production; 4mg/ml of protein accumulation was observed.

  • The possibility of using Chinese cabbage juice as a substrate for the production of Candida utilis cell mass was explored. Dry cell weight production and cell yield coefficient were 1.35-1.45 g/100 ml undiluted juice and 47-50%, respectively, when C. utilis was grown by shake flask culture at $30^{\circ}C$ for 24 hr on more than three-fold diluted Chinese cabbage juice to make the final sugar content be equal to or less than 1.0%. Supplementation of glucose(2%), $KH_2PO_4(0.2%)$ and $(NH_4)_2SO_4(0.2%)$ to three-fold diluted Chinese cabbage juice did not enhance the dry cell weight yield or the protein content of the yeast cell, while supplementation of yeast extract(0.2%) and peptone(0.2%) increased dry cell weight production and protein content but not as much as the amount of each nutrient added. It was found that Chinese cabbage juice was an excellent substrate for the cultivation of C. utilis.

  • Cultural and nutritional conditions for Lyophyllum decastes and its chemical composition in a synthetic medium were investigated. The optimal temperature and pH for the production of mycelium were $25^{\circ}C$ and pH 7.5. The mycelium yield was the highest on 15th day. Among the carbon sources, glucose and CMC were the best for the production of mycelium and their optimal concentrations were 3 and 6%, respectively. As an organic nitrogen, proteose peptone was the best and $(NH_4)_2HPO_4$ as an inorganic nitrogen was good. The optimal concentration of proteose peptone and $(NH_4)_2HPO_4$ were 2 and 0.2%, respectively. The optimal ratio of glucose to proteose peptone for production of mycelium was 10 : 1. Also, the optimal concentrations of $K_2HPO_4$ and $MgSO_4$ were 0.2 and 0.06%, respectively and that of $CaCl_2$ was 0.1%. Among the bioextracts, yeast extract was the most effective and its optimal concentration was 1.5%. In chemical components of the mycelium of Lyophyllum decastes, total sugar, crude protein and crude fat were 34.80, 28.35 and 2.50%, respectively. Its ash was 7.57% and crude fiber 11.99%.

  • In order to maximize the cell growth and the polysaccharide production in Agaricus blazei, two kinds of fed-batch fermentation processes were performed with varying the feeding medium compositions and the feeding process. The relationship between dissolved oxygen and polysaccharide production in batch fermentation was applied to fed-batch fermentation. The biomasss concentration was 18.2 g/L and the polysaccharide production was 10.4 g/L.

  • Cultural conditions for the production of extracellar $\beta$-galactosidase by Loctobacillus sporogenes, a spore forming lactic acid bacterium, were investigated with shaken flask and jar fermenter cultures. The fermentation medium giving maximum $\beta$-galactosidase yield was found to consist of 1 % lactose as a carbon source, 1.5% peptone as an organic nitrogen source. 0.2% ammonium sulfate as an inorganic nitrogen source, 0.8% ammonium phosphate dibasic as a phosphorus source, and 0.05% potassium chloride and 0.001% ferric chloride as mineral source. Optimal initial pH of the medium was 7.0 and the highest enzyme excretion was observed after 40 hours of cultivation at 37$^{\circ}C$. In this experiment, the 500$m\ell$ conical flask containing 50-200$m\ell$ of medium was shaken at 140 strokes per minute with 7cm amplitude in a reciprocating shaker. The maximum enzyme value attained was 38 U/$m\ell$ of the culture broth which was found to be slightly higher than the highest intermolecular enzyme activity (30 U/$m\ell$) observed after 24 hours of incubation. In the fermentor culture, the fermentation profile was shown to be similar to that observed in the shaken flask experiment. But the maximum extracellular enzyme activity was 45 U/$m\ell$ to be even higher than the value obtained with the shaken flask culture.

Cell Immobilization of Zyrnornonas rnobilis by Entrapment (포괄담체에 의한 Zymomonas mobilis 균체의 고정화)

  • The immobilization characteristics of Zymomonas mobilis for ethanol production were examined. Four different strains of Zymomonas mobilis have been used for ethanol production. Among those, Zymomonas mobilis KCTC 1534 has been selected as the best strain for the highest ethanol productivity from glucose and sucrose. The optimum temperature and pH of the selected strain for ethanol production were $37^{\circ}C$ and 5.0 respectively for both free and immobilized cells. When the cells were immobilized by the gel entrapment method, the immobilized cells could produce ethanol at a little higher temperature than free cells. Calcium alginate was selected as the best gel for immobilizing cells. The immobilized cells could maintain the viability of 80% in 10 weeks storage at $4^{\circ}C$ in the medium with 2% calcium chloride. 20-25 hours of preincubation in 10% glucose solution was required for the activation of immobilized cells entrapped within calcium alginate gel.

  • Experiments were carried out to find possibility of economic production of SCP in mixed culture by Cellulomonas sp. KL-6 and E. coli LI-10. The best cell growth was obtained at the ratio of 1 : 1(v/v) in mixed culture. When these strains were mixed culture, cell growth was increased to about 63%, compared with those of single culture of strain KL-6. It was found that the majority of the population during growth in mixed culture consisted of strain KL-6. $CaCO_3$ added to the medium as the ratio of 0.1% was enhanced medium pH. Cell growth increased in that circumstances. These strains produced much amounts of cellobiose, but glucose was not detected in filter paper medium. When these organisms were cultured under the optimal medium for 4 days, cell mass was produced $1.0\;g/{\ell}$. The results showed the increase of cell mass up to 53% than those produced in CMC medium.

  • L-Threonine fermentation process was constructed on batch and fed-batch culture by using Escherichia coli MT201. The production type of L-threonine was observed as growth-associated production in batch culture. In fed-batch culture studying optimal concentration of yeast extract in feeding media, when 600 g/l of glucose and 60 g/l of yeast extract were added in feeding media, 87 g/$\ell$ of L-threonine was produced. To improve cell growth and L-threonine production, the culture of high cell density was performed in fed-batch culture with oxygen enriched air and feeding media containing L-methionine and phosphate. Under the conditions, we could achieve the highest L-threonine production of98 g/$\ell$ at 60 h. The highest productivity of L-threonine was about 3.85 g/$\ell$/h.

  • Hansenula nonfermentans KYP-l was selected and identified from 19 methanol utilizing yeasts isolated from soil samples by the enrichment culture technique. This strain showed a high cell concentration and a high aldehyde production. Aldehyde production was carried out in a resting cell system using methanol utilizing yeast as a biocatalyst. The molar yield of acetaldehyde was the highest among the aldehyde investigated, and the maximum amount of aldehyde was produced by cells obtained from a 40 hours' culture.
