DISEASES OF THE CULTTVATED PORPHYRA AT CULTURE BEDS WITH SPECIAL REFERENCE TO THE EFFECTS OF FERTILIZER PLANT EFFLUENTS (낙동강 하구 부근(용원리) 김 어장의 갯병 특히, 공장 폐수의 영향에 관하여)

  • Considerable damages of the cultivated Porphyra by Porphyra diseases were reported from the Porphyra culture bed along the coast of Yongwon-ri, Changwon, Kyungnam during the years of 1969 and 1970. The present study deals with the effects of fertilizer plant effluents on the Porphyra diseases, and the results are summarized as follows : 1. By the result of current observation, the polluted water of Haeng-am Bay which has an inflow of pollutants from the fertilizer plant was affecting the bed with tides. The results of pollution studies in Chinhae Bay and adjacent waters conducted by Won and Park(1971) also show that Chinhae Bay and adjacent waters are contaminated with the plant effluents. It seems that the effect increases due to the wind drift current when north-westerly or westerly winds prevail. Accordingly, effects of the Porphyra diseases in the culture bed seem to originate from the pollutants, since there are more damages when the north-westerly or westerly winds prevail, and also spring tide develops. 2. As compared to the photosynthetic activity of the Porphyra suborbiculata in uncontaminated seawater, it decreases up to $4\%$ in 200pmm, $20\%$ in 300ppm and $43\%$ in 1,000ppm of contaminated seawater which contains dilluted pollutants from the fertilizer plant. The results of the measurements using the water collected in the polluted area of Chinhae Bay and adjacent waters revealed that the photosynthesis was depressed about 21 to $34\%$ near the plant, and in the area of the Porphyra beds, $15\%$ in the portion where tide is weak and $5\%$ where the tide is strong, in comparison with the area of unpolluted water. 3. Although the present results do not indicate the exact level of harmful pollutants, it is evident that the pollutants of the plant effluents interfere photosynthetic activity of the Porphyra, even in the water containing pollutants as low as 200 ppm.

Changes of Leaf Area Index, Physiological Activities and Soil Water in Tricholoma matsutake Producing Pine Forest Ecosystem (송이산 소나무림 생태계에서 엽면적지수와 생리적활동 및 토양수분의 변화)

  • The purpose of this study was to understand the ecosystem structure and function and soil water changes in Tricholoma matsutake producing pine stands. The investigated stands were pine forest in Sogrisan National Park in Chungbuk-do of Korea. For the purpose we investigated main vegetation, leaf area index(LAI) as ecosystem structural factors and measured photosynthesis, transpiration, xylem water potential, and soil water changes as ecosystem functional factors. Vertical vegetation structure of the site was composed of Pinus densiflora as a overstory species, Quercus mongolica as midstory, Rhododendron mucronulatum, R. schlippenbachii and Fraxinus sieboldiana as understory ones. In the stands LAI was 3.8 during June to September, 2.6 in October and 2.1 during November to April. Photosyntheses of the trees were 6.0 to $7.0{\mu}mol\;CO_2/m^2/s$ in August, and for P. densiflora about $4.0{\mu}mol\;CO_2/m^2/s$ and for Q. mongolica $2.0{\mu}mol\;CO_2/m^2/s$ in mid October. However, R. mucronulatum stopped fixing $CO_2$ and F. sieboldiana shed off the leaves already in mid October. Transpirations were 2.5 to $3.5mmol\;H_2O/m^2/s$ in late August and about $1.0mmol/H_2O/m^2/s$ in mid October. Plant water potentials were -10 to -22 bars for P. densiflora and -5 to -12 bars for the other woody species. The lowest potentials was in late August and highest in late October. Soil water in the stand was closely related to topography. Soil water contents were 7 to 11% at the ridge, 8 to 15% at the hillside and 11 to 19% at the base. Soil temperatures were 0.2 to $0.4^{\circ}C$ higher in T. matustuake colony than noncolony. Mid September soil temperature decreased to $19^{\circ}C$ at which T. matsutake forms primordia. In T. matsutake colony soil moisture was 0.5 to 2.0% lower due to metabolism for consuming water. We suggest that the complicate relationships between ecosystem structure and function in Tricholoma matsutake producing pine stand need to be further investigated.

  • The role of L-glutamic acid, a precursor of $C_5$ ALA biosynthetic pathway, on the production of 5-aminolevulinic acid (ALA) has been described in cells of Rhodospirillum rubrum N-1. To the Lascelles basal medium the addition of both 30 mM L-glutamicacid and 20 mM levulinic acid (LA) provided to increase the extracellular ALA yield up to 40 fold (76 mg/l). By the addition of both 60 mM glycine and succinic acid, precursorsof $C_4$ ALA biosynthetic pathway, at middle log phase of cell growth ALA yield was increased 27 fold (52 mg/l) although the celt growth was inhibited to a certain extent.

Removal of Nitrogenous Compounds by Immobilized Mixed Microorganisms Including Photosynthetic Bacteria (광합성 세균을 포함한 고정화 복합미생물에 의한 질소성분 제거)

  • For efficient removal of nitrogenous compounds produced in recirculating aquaculture system, the N removal characteristics of immobilized mixed microorganisms were investigated at various mixing ratios of photosynthetic bacteria (PSB) immobilized in PVA beads or CTA cubes and ammonium utilizing bacteria (AUB) immobilized in PVA beads. On the optimal medium of AUB, the maxium gas production rate was obtained at the mixing ratio of 10:40 (PSB:AUB), and the gas production rate increased as the portion of AUB beads in the mixed beads increased. When the mixing ratios of PSB:AUB beads were 50:0, 40:10, 25:25 and 10:40, the final pHs were measured to be 6.29, 6.01, 5.69 and 5.13, respectively. On the optimal medium of PSB, however, the volume and the rate of gas production decreased remarkably as the portion of AUB beads in the mixed beads increased. The final pH was measured to be approximately 6.5, regardless of the mixing ratio. In the reactions by the mixed culture of PSB cubes and AUB beads, all results showed the same tendency of those by the mixed culture of PSB and AUB beads, but the volume and the rate of gas production decreased remarkably, even with 0.2ml of gas production in control. From all the results, the use of mixed PSB and AUB beads at the ratio of 10:40 seems to be efficient to remove nitrogenous compounds in wastewater from recirculating aquaculture system.

  • A photosynthetic bacterium strain KUP-74 producing high amount of S-amino-levulinic acid(ALA) was isolated from soils, which was identified as Rhodocyclus gelatinosus. After 10 days cultivation under anaerobic-light condition at $30^{\circ}C$, 4 Klux and pH 6.8, 5 mg/l of ALA was formed extracellularly. ALA productions were increased up to 8 mg/l and 12 mg/l in cell cultivations either by the addition of 0.5% glycerol (v/v) or 10 mM of glycine and succinic acid, respectively, using Lascelles basal medium eliminated L-glutamic acid. By cultivation in the presence of 30 mM each D,L-glutamic acids and D,L-glutamines the yield of ALA showing a late induction phenomenon was reached the maximum value of 21 mg/l. Different culture times were needed to generate maximum ALA yields by the addition of initial precursors of $C_4$ and $C_5$ pathways in basal medium, as being 107 h and 262 h, respectively. 40 mg/l yield of ALA was observed by cell cultivation with the basal medium containing each 10 mM levulinic acid(LA) and glycine simultaneously.

Starter culture production of Rhodospirillum rubrum P17 for use in treatment of organic waste water (유기폐수처리를 위한 Rhodospirillum rubrum P17의 종균생산)

  • A photosynthetic bacterium strain P17 having high growth rate and assimilating ability of organic acids was isolated from several soil samples, which was identified as Rhodospirillum rubrum. Cultural conditions of the strain P17 were examined for the production of starter culture used in the treatment of organic waste water. The addition of organic acids mixture as carbon source containing 0.2% Na-acetate, 0.1% Na-propionate and 0.2% Na-lactate and 0.1% of yeast extract as growth factor stimulated the cell growth. The maximal cell production was obtained at $30^{\circ}C$, pH 7.0, 2,500 lux of illumination and $50{\sim}100\;rpm$ of agitation. Under the optimal conditions of batch and fed-batch culture systems in a Jar fermentor, 5.17 g/l and 7.93 g/l of cells were obtained after S days of cultivation, respectively. In continuous culture system, the cell productivity was 0.206 g/l/h at a dilution rate of 0.21 $h^{-1}$. When R. rubrum P17 was cultivated in a soybean curd waste water, initial COD level(3,240 mg/l) of the waste water was reduced to 250 mg/l after 4 days of cultivation.

Effect of Methionine Sulfoximine in nitrogenase activity by ammonia and glutamine in Rhodopseudomonas sphaeroides (암모니아와 glutamine에 의한 Rhodopseudomonas sphaeroides의 질소 고정 효소활성에 미치는 Methionine Sulfoximine의 영향)

  • The effect of ammonia and glutamine on nitrogenase activity of Rhodopseudomonas sphaeroides was examined. The nitrogenase activity of this strain was inhibited by ammonia and glutamine. When ammonia and glutamine were exhausted, nitrogenase activity promptly resumed at its original rate. Methionine sulfoximine (MSX), irreversible glutamine synthetase (GS) inhibitor, is a structural analogue of glutamate. MSX was used in order to know whether the nitrogenase activity was inhibited by ammonia and glutamine directly or not. The ability of MSX to prevent nitrogenase switch-off by ammonia was found to be dependent upon the phase of culture. When the cells were sampled after 12 hour culture, $500{\mu}M$ MSX would not prevent the nitrogenase switch-off by ammonia. Twenty one percents of GS actibity was inhibited by $500{\mu}M$ of MSX and concentration of released ammonia decreased. But nitrogenase activiy was still inhibited by ammonia. However, nitrogenase switch-off after 20 hours would be prevented by $100{\mu}M$ of MSX. On the other hand, GS activity was ingibited completely by $100{\mu}M$ MSX and concentration of released ammonia somewhat increased. But nitrogenase activity was not inhibited. The data indicated that the inhibition of in vivo nitrogenase actibity of Rp. sphaeroides by ammonia seemed to be mediated by products of ammonia assimilation rather than by ammonia itself.

Biological Treatment of Processed-Leachate from Landfills by Reed (Phragmites australis)-Bed in a Continuous Flow System (갈대-상(床)을 이용한 쓰레기 매립지 침출수의 생물학적 연속흐름 처리)

  • We investigate the biological treatment of processed-leachate from SUDOKWON landfill site in Korea by the reed (Phragmites australis)-bed with a continuous flow system. The reed individuals showing superior growth in processed-leachate experiment were selected among the reeds of thirteen natural habitats by means of the comparisons between the removal rates of nutrient salts, eco-physiological responses and growth. The reed-beds (combination of the reed individuals showing superior growth with helper microorganisms) were continuously supplied with processed-leachate. We monitored the effluents that passing through the reed-beds during the experiment period. After five weeks, analysis results of effluent from each reed-bed were as follows; chromaticity, total nitrogen, total phosphorus, biological oxygen demand, chemical oxygen demand, total dissolved solid and salinity decreased $29.5{\sim}36.9\%,\;49.4{\sim}67.2\%,\;42.1{\sim}94.6\%,\;74.5{\sim}88.8\%,\;15.6{\sim}20.8\%,\;17.5{\sim}35.4\%\;and\;15.3{\sim}34.7\%$, respectively. These results represented the substantial improvement of water-quality after passing through reed-bed in a continuous flow system.

General characters and applications of photosynthetic bacteria (광합성세균의 생리 및 이용에 관한 최근의 동향)

  • In order to investigate the production of aflatoxin in various conditions such as pH, moisture and temperature, 27 smaples were inoculated with Aspergillus flavus, and in addition 3 smaples were inoculated with the mixture of Aspergillus flavus and Bacillus subtilis and cultured under the conditions such as 20.deg.C and 30% moisture contents. The following results were obtained : 1) Aflatoxin production was the highest at pH 5.0 and relatively high at pH 7.0. Its production was decreased significantly when pH reached 9.0. 2) The yield of aflatoxin was shown comparatively high level at 30% moisture contens. The higher moisture contents was, the lower aflatoxin production was. 3) The highest level of aflatoxin production was at 20.deg,C, and comparatively high level was at 30.deg.C. However, its production was fairly low at 40.deg.C. 4) The highest crude aflatoxin production was 5,093ppm (B$_{1}$, 1.912ppm ; B$_{2}$, and the lowest one 2.197 ppm (B$_{1}$, 0.793 ppm : B$_{2}$, 0.185 ppm : G$_{1}$ ,0.102 ppm G$_{2}$, 0.381ppm) at 63% moisture, pH 9.0 and 40.deg.C. 5) When Aspergillus flavus and Bacillus subilis were cultured together under the conditions such as 20.deg.C and 30% moisture, aflatoxin production was decreased by 27% comparing with the culture of Aspergillus flavus alone.

Nitrogen Efficiency and its Relation to Various Physiological Characteristics among Rice Varieties (수도품종간(水稻品種間) 질소효율(窒素効率) 및 수종(數種) 생리적특성(生理的特性)과의 관계(關係))

  • Nitrogen efficiency for grain yield(E) and its relation to grain yield(Y), harvesting index(HI), percent translocation of nitrogen from straw to grain(T), nitrogen uptake amount(N), concentration in grain (GN%) or straw (SN%) and total dry matter yield (TY) among rice varieties (Oryza sativa, old and new varieties) were investigated at four nitrogen nutritional status (high and low fertilizer levels in high and low fertility fields) by simple correlation analysis. Relation between any two of above parameters or total dry matter yield (TY) and nitrogen efficiency for total dry matter yield (TE) was also investigated. 1. E is significantly and positively correlated with T, Y, HI but negatively with SN%, N, GN% and in negative trend with TY. 2. T is significantly and positively with GN% or Y, but negatively with SN%. 3. TE is significantly and positively correlated with TY but negatively with N. 4. The order of E among varieties showed consistency among different nitrogen nutritional environments. 5. From the above facts it was concluded that high yielding varieties have high nitrogen efficiency due to high percent translocation of nitrogen from straw to grain, subsequent low nitrogen concentration in straw and that translocated nitrogen in grain is greatly diluted with photosynthates. 6. Reported physiological characteristics of newly bred high yielding IR lines are well accordance with their high nitrogen efficiency and rice breeding was a selection on the basis of nitrogen efficiency. 7. It is postulated that high nitrogen efficiency varieties for yields have high nitrogen efficiency for root growth in early stage so that uptake more efficient soil nitrogen in later growth stage.
