Growth Response and Absorption Capacity of Poplars on Livestock Waste Water (축산폐수(畜産廢水) 처리(處理)에 따른 포플러류(類)의 생육반응(生育反應) 및 축산폐수(畜産廢水) 흡수능력(吸收能力))

  • The two-month-old rooted-cuttings of Populus alba ${\times}$ glandulosa, P. euramericana and P. nigra ${\times}$ maximowiczii clones were exposed to livestock waste water - one of major water pollutants, and ground water in order to determine the effects of livestock waste water on growth response and absorption capacity of the species. For this purpose, 5 clones of each species were used. In all the species, the height growth of rooted-cuttings was better in livestock waste water treatment than in ground water. Of all the poplar species compared, the height growth was best in P. alba ${\times}$ glandulosa. In the cases of P. euramericana and P. nigra ${\times}$ maximowiczii, the height growth in the livestock waste water treatment was statistically different among clones, whereas there was no significant difference among P. alba ${\times}$ glandulosa clones. Aboveground biomass such as leaf and shoot dry weight of all the species increased in the livestock waste water treatment, while root dry weight decreased. In addition, chlorophyll contents in leaf of all the poplar species increased in the livestock waste water treatment. All the poplar trees showed temporal variation in the absorption amount of livestock waste water during the experimental period. In all the poplar species, the absorption amount of livestock waste water was less than that of ground water. Of 3 poplar species, P. alba ${\times}$ glandulosa was best in the absorption capacity of livestock waste water. Of all 15 poplar clones compared in this study, the 72-16 clone of P. alba ${\times}$ glandulosa showed the best absorption capacity.

Medium-chain fatty acid enriched-diacylglycerol (MCE-DAG) accelerated cholesterol uptake and synthesis without impact on intracellular cholesterol level in HepG2 (중쇄지방산 강화 디아실글리세롤(MCE-DAG)이 간세포 내 콜레스테롤 흡수 및 합성 기전에 미치는 영향)

  • The effects of medium-chain enriched diacylglycerol (MCE-DAG) oil on hepatic cholesterol homeostasis were investigated. HepG2 hepatocytes were treated with either 0.5, 1.0, or $1.5{\mu}g/mL$ of MCE-DAG for 48 h. There was no evidence of cytotoxicity by MCE-DAG up to $1.5{\mu}g/mL$. The level of proteins for cholesterol uptake including CLATHRIN and LDL receptor increased by MCE-DAG in a dose-dependent manner (p<0.05). Furthermore, proprotein convertase subtilisin/kexin type 9, an inhibitor of LDLR, was dose-dependently diminished (p<0.05), indicating cholesterol clearance raised. MCE-DAG significantly increased 3-hydroxy-3-methylglutaryl-coenzyme A reductase and acetyl-CoA acetyltransferase2 (p<0.05), required for cholesterol synthesis, and their transcriptional regulator sterol regulatory element-binding protein2 (p<0.05). These findings suggest that given conditions of prolonged sterol fasting in the current study activated both hepatic cholesterol synthesis and clearance by MCE-DAG. However, total intracellular level of cholesterol was not altered by MCE-DAG. Taken together, MCE-DAG has the potential to prevent hypercholesterolemia by increasing hepatic cholesterol uptake without affecting intracellular cholesterol level.

Betulinic Acid Stimulates Glucose Uptake through the Activation of PI3K and AMPK in 3T3-L1 Adipocytes (Betulinic acid의 PI3K와 AMPK경로 활성화를 통한 3T3-L1 지방세포에서 포도당 흡수 촉진 효과)

  • Hyperglycemia in type 2 diabetes can be alleviated by promoting cellular glucose uptake. Betulinic acid (3β,-3-hydroxy-lup-20(29)-en-28-oic acid) is a pentacyclic lupane-type triterpenoid compound. Although there have been studies on the antidiabetic activity of betulinic acid, studies on cellular glucose uptake are lacking. We investigated the effects of betulinic acid on glucose uptake and its mechanism of action in 3T3-L1 adipocytes. Betulinic acid significantly stimulated glucose uptake in 3T3-L1 adipocytes by increasing the phosphorylation of the insulin receptor substrate 1-tyrosine (IRS-1tyr) in the insulin signaling pathway, which in turn stimulated the activation of phosphoinositide 3-kinase (PI3K) and the phosphorylation of protein kinase B (Akt). The activation of PI3K and Akt by betulinic acid translocated glucose transporter 4 to the plasma membrane (PM-GLUT4), thereby increasing the expression of PM-GLUT4 and thus stimulating cellular glucose uptake. Betulinic acid also significantly increased the phosphorylation/activation of AMP-activated protein kinase (AMPK) and acetyl-CoA carboxylase. The activation of PI3K and AMPK by betulinic acid was confirmed using the PI3K inhibitor wortmannin and the AMPK inhibitor compound C. The increase in glucose uptake induced by betulinic acid was significantly decreased by wortmannin and compound C in the 3T3-L1 adipocytes. These results suggest that betulinic acid stimulates glucose uptake by activating PI3K and AMPK in 3T3-L1 adipocytes.

Analysis of Absorbed Dose on the Nucleus Size Change of Single Cells using Therapeutic Radioisotopes (치료용 방사성동위원소 사용 시 단일 세포의 핵 크기 변화에 따른 흡수선량 분석)

  • Targeted radionuclide therapy (TRT) is a method of treating tumor cells using radiopharmaceuticals. Cells and nuclei constituting tissues of the human body are composed of spherical and oval shapes, but cancer cells are composed of various cell types. Therefore, this study analyzed the absorbed dose for each organelle according to the change in the size of the cell nucleus for beta-emitting nuclides during targeted radionuclide therapy through the Monte Carlo method. Cells were set in two sphere shapes, 5 ㎛ and 10 ㎛, and the internal structure was divided into cell nucleus, cytoplasm, and cell surface. Next, the absorbed dose according to the increase in the size of the cell nucleus was evaluated. As a result, 177Lu among the target radionuclides showed the highest dose in all cell compartments. As the ratio of the nucleus in the cell increased, the absorbed dose on the cell surface increased, but the absorbed dose in the cytoplasm and nucleus tended to decrease. Accordingly, it is judged that it is important to select a radionuclide considering the size of cancer cells and determine an appropriate amount of radioactivity during targeted radionuclide treatment.

Studies on skin whitening efficacy and skin permeation using O/W Nanoemulsion system with Resorcinol Dipentyl Ether (레조시놀다이펜틸에터를 함유한 O/W 나노에멀젼의 경피흡수 및 미백 효능 효과 연구)

  • In this study, we investigated the effect on the increase of In vitro skin permeation experiments and In-vivo skin whitening efficacy using a O/W nanoemulsion produced via PIC(Phase Inversion Composition) with 1,3-di(pentyloxyl)benzene. skin permeation experiments of RS-nanoemulsion formulated with selected condition was evaluated compared to mineral oil containing 1,3-di(pentyloxyl)benzene and normal O/W type RS-emulsion. Compared to mineral oil with 1,3-di(pentyloxyl)benzene and RS-emulsion. RS-Nanoemulsion has a statistically significant high percutaneous absorption in terms of index substance, which is 1,3-di(pentyloxyl)benzene. In vivo test were prepared in the system of O/W cream containing RS-nanoemulsion. There was no adverse reactions in both samples. After 8 weeks, the subjects was evaluated by a dermatologist's scoring and Chromameter. In conclusion, the testing product showed statistically improvement (p<0.05) compared to the controlled product and proved its whitening efficacy.

Decay Resistance and Effectiveness of CCA Preservative against Decay on the 4 Imported Softwoods (수입침엽수(輸入針葉樹) 4수종(樹種)의 내후성(耐朽性) 및 CCA계(系) 목재방부제(木材防腐劑) 처리(處理)에 따른 방부효과(防腐效果))

  • With the aim to investigation of decay resistance and optimum concentration in chrome-copper-arsenic(CCA) preservative treatment on the imported softwoods from Siberia and North america, preservative absorption after CCA impregnation, weight losses and degradation patterns by decay fungi were examined. The density and latewood rate of Siberia softwoods(Spruce and Larch) were higher than those of North america softwoods(Douglas-fir and Western hemlock), resulting in the decrease of the CCA preservative absorption in the Siberia softwoods. In the case of untreated softwoods, decay resistance against Coriolus versicolor was lower than against Tyromyces palustris. For CCA treated softwoods, preservative effectiveness increased with increase in concentration of CCA solution. When treated with 0.7% CCA solution, efficiency value was more than 80 and 90 for C. versicolor and T. palustris, respectively. From this results, in the CCA preservative treatment for imported softwoods, it can be concluded that optimum concentration of CCA solution is approximately 0.7%. The absorption of CCA preservative distributed in the range of 3.8 and $5.5kg/m^3$. After exposure to testing fungi, in the untreated softwoods, bore holes formed in the cell walls and bordered pits, moreover, bordered pit canals enlarged by the fungi. However, 0.5% CCA treated softwoods was almost no deterioration in the cell walls and bordered pits due to decay.

Study on Optimization and Skin Permeation of PIT Nanoemulsion Containing α-Bisabolol (α-Bisabolol을 함유한 PIT Nanoemulsion의 최적화 및 피부흡수연구)

  • The skin is divided into three parts: the epidermis, the dermis, and the subcutaneous fat, and the stratum corneum, which is located at the top of the epidermis, acts as a barrier that prevents drug delivery. Nanoemulsions are known to be effective in transdermal delivery of drugs through intercellular lipids because of their unique small particle size. In this study, phase inversion temperature (PIT) nanoemulsion containing α-bisabolol was optimized using response surface methodology (RSM) for effective skin absorption of α-bisabolol. As a preliminary experiment, the 25-2 fractional factorial design method and the 23 full factorial design method were performed. Box-Behnken design was performed based on the results of the factorial design method. The content of surfactant (6.3~12.6%), co-surfactant (5.2~7.8%) and α-bisabolol (0.5~5.0%) were used as factors, and the dependent variable was the particle size of the nanoemulsion. PIT nanoemulsion optimization was performed according to the RSM results, and as a result, the optimal nanoemulsion formulation conditions were predicted to be 10.4% surfactant content, 6.3% co-surfactant content, and 5.0% α-bisabolol content. As a result of the skin absorption test, the final skin absorption rate of the PIT nanoemulsion was 35.11±1.01%, and the final skin absorption rate of the general emulsion as a control was 28.25±1.69%, confirming that the skin absorption rate of the PIT nanoemulsion was better.

Estimating Greenhouse Gas (GHG) Removal by Cryptomeria japonica and Chamaecyparis obtusa Stands Using New Stem Volume Tables (신규 입목수간재적표를 활용한 삼나무 및 편백 임분의 온실가스 흡수량 추정)

  • The aim of this study was to quantitatively evaluate a new stem volume table for estimating the growth, carbon storage, and greenhouse gas (GHG) absorption in Cryptomeria japonica and Chamaecyparis obtusa stands and to provide suggestions for improving the domestic GHG inventory. Carbon storage and GHG absorption were estimated using growing stock data obtained from invariable sub-sample plots between the 6th and 7th national forest inventories. We assessed changes in growing stock using the parameters employed by Kozak (1988) and Versions 1 and 2 of the stem volume table. Version 2 has new stem tables for 16 species, including Cryptomeria japonica, which were unavailable in Version 1. Version 2 also includes new data for trees with diameters at breast height equal to or greater than 30 cm. We found greater growing stock values using Version 2 than Version 1 for both stands, and the differences were statistically significant (p<0.001). Applying the new stem volume table increased GHG absorption by 22% for the Cryptomeria japonica stand and 13% for the Chamaecyparis obtusa stand. The growing stock estimation method used in this study should therefore be applied to re-estimate GHG absorptions in the forestry sector to produce accurate statistics for the IPCC guidelines.

Sorghum Extract Lowers Lymphatic Absorption of Trans Fat and Cholesterol in Rats (흰쥐에서 수수추출물이 트랜스지방산이 함유된 지방과 콜레스테롤의 흡수에 미치는 영향)

  • Excessive intake of trans fats is a risk factor for cardiovascular diseases. Previously, we showed that green tea extract lowers intestinal absorption of cholesterol and elaidic acid in rats. This study was conducted to investigate a possible role for sorghum extract on lymphatic absorption of trans fat and cholesterol in rats. Adult male rats with lymph cannulae were infused at a rate of 3.0 mL/h for 8 h via a duodenal catheter with a lipid emulsion containing $146.4{\mu}mol$ of trielaidin, $36.8{\mu}mol$ of trilinoelaidin, $452.0{\mu}mol$ of triolein, $1.0{\mu}Ci$ cholesterol labeled with $^{14}C$ ($^{14}C-cholesterol$), $20.7{\mu}mol$ of cholesterol, and $396.0{\mu}mol$ of Na-taurocholate without or with 100.0 mg of sorghum extract in phosphate-buffered saline buffer (pH 6.4). Lymph was collected hourly for 8 h. No significant difference was noted in lymph flow. However, the lymphatic absorption of elaidic acid and linoelaidic acid for 8 h was significantly lower in rats infused with sorghum extract than in those infused with no sorghum extract. Further, lymphatic absorption of $^{14}C-cholesterol$ was reduced by sorghum extract, which was observed previously. These data indicate that sorghum extract has an inhibitory effect on the intestinal absorption of trans fat and cholesterol. The mechanism(s) by which sorghum extract lowers intestinal absorption of trans fat warrants further study.

Soil Moisture Influence on Growth and Nutrient Uptake of Seedlings in Denuded Forest Land (황폐지(荒廢地) 토양수분(土壤水分)이 묘목(苗木)의 생장(生長) 및 양분흡수(養分吸水)에 미치는 영향(影響))

  • In afforestation of denuded forest land soil conditions play a very important role in early growth of cover vegetations. This study was designed for understanding the effect of soil moisture regime on growth and nutrient uptake of some seedlings. Cover vegetations such as Pinus rigida Miller, Robinia pseudoacacia L. and Lespedeza bicolor Turcz. were planted in pot with the soil transported from denuded forest land in Musu-ri Sannae-Myeon. Daedeog-Kun, Chungnam Province. There were 3 moisture treatments and 4 fertility levels in $P_2O_5$ with 4 replications. Influence on growth was observed by the variation in dry weight and nutrient uptake was studied in nitrogen, phosphate and kalium. Results are as follows: 1. For Pinus rigida seedlings decrease in soil moisture tension increases growth of dry weight and enhance the uptake of kalium. Increase in $P_2O_5$ fertility level tends to decrease the uptake of kalium rather than increase in $P_2O_5$ uptake. 2. In Robinia pseudoacacia increase in soil moisture content stimulates the uptake of nitrogen and kalium. Increase in $P_2O_5$ level enhances the uptake of $P_2O_5$ and increases growth of dry weight. 3. In Lespedeza bicolor increase in soil moisture content has a tendency of decrease in nitrogen uptake. Increase in $P_2O_5$ level increases the growth of dry weight as well as the uptake of $P_2O_5$.
