  • Seed separation from half-cut red pepper were investigated with free falling, up-and-down shaking and rotating collision methods. The separation rates were related with impact appiied and the rotating case was distinguished from the other two methods. Beside the impact effect, velocity of the pod and tumbling factors were involved. Momentum effectiveness of seed separation were calculated as $2.50{\times}10^{-6}$, $2.09{\times}10^{-6}$, and $3.94{\times}10^{-8}$ for free falling, shaking and rotating method on the same velocity basis, respectively. The tendency of separation rate was similar to that of red pepper drying rate against time.

Change of Rooting Potential as Affected by Rooting Promoter Treatment in Cuttings of Corylopsis coreana (히어리(Corylopsis coreana)의 삽목번식시 발근촉진제의 처리에 따른 발근력 변화)

  • This study was conducted to determine the effects of several rooting promoter treatments on rooting of Corylopsis coreana cutttings for mass production. In first year experiment, no effect of rooting promoters was observed in hardwood cuttings taken on March 20. However, In softwood cuttings of June 20, rooting potential was greatly increased by IBA $100mg{\cdot}L^{-1}$ dipping treatment for 24 hours. Particularly, IBA $100mg{\cdot}L^{-1}$ 24 hour dipping treatment greatly shortened the days required for the rooting. In second year experiment, the result of first year was reconfirmed. Almost of all the cuttings (97.8% of rooting rate) were well rooted after only 30 days after cutting by IBA $200mg{\cdot}L^{-1}$ 24 hour dipping treatment. In this treatment, numbers and length of root were also prominent compared with any other treatment results.

  • 전 국토의 산지 비율이 높은 우리나라는 산림지의 특성이 수질에도 많은 영향을 미치고 있다. 산림지역의 화재에 있어 산림이 갖는 수질, 토성의 변화와 발생한 연소 퇴적물, 유기물로 인해 수자원에도 악영향을 끼치게 된다. 산불은 단순한 산림지의 자원 유실뿐만 아니라 토양층의 물리적약화와 강수저류 효과를 떨어뜨려, 산불 후 유출의 증가를 보인다. 삼척시의 산불 선행연구에 따르면, 산불 발생 직후, 지역의 직접 유출이 기저 유출보다 높게 나타났다. 이에 따라, 모든 수질인자의 평균 농도는 비발생지역에 비해 발생 지역의 농도가 현저하게 증가하였으며, 이 중 T-N, T-P, SS의 농도가 특히 크게 나타났다. 또한, 계류 수질의 경시적 변화를 분석한 결과, BOD와 COD는 산불 발생 최소 4개월의 기간을 거친 후 점차 안정화하는 모습을 보였다. 다만, 산불로 인해 훼손된 물 환경의 상황을 복구하고자 하는 연구의 데이터가 부족한 실정이다. 따라서 본 연구는 2022년 4월 산불이 발생한 강원도 양구군 양구읍 송청리 비봉산 일대의 산불 유역을 대상으로 산불에 따른 물 환경의 변화를 밝힘으로써 산불로 훼손된 지역의 효과적인 수질 회복과 물 환경 보전 대책 수립을 위한 자료를 제공하고자 수행하였다. 2023년 4월 양구 비봉산의 산불 복구 작업이 본격적으로 시작되는데 복구 전 현재 수질의 상황을 살펴봤다. 조사 지역은 비봉산 일대의 산불 지역과 비발생 지역, 오봉산 일대까지의 유역 출구 총 7개의 지점을 선정하였다. 기본 수질항목은 현장에서 YSI PROPLUS (Mobile Multi Sensor Meter)를 이용하여 측정하였고 채취한 시료는 무균 채수 병에 담아 실험실로 운반하여 SS (Suspended Solids), TP(Total Phosphorus), TOC (Total Organic Carbon) 항목에 대해 수질오염공정시험기준에 의거하여 분석하였다. SS, TOC, t-p의 경우, 지정학적 특성, 산불에 의한 퇴적물들의 변화와 토양의 형태 및 식생의 변화로 인한 결과의 차이를 보였다. 1년이 지난 지금 산불 직후처럼 높은 수치는 보이지 않았다. 그러나 아직 복구 작업이 제대로 진행되지 않아 그 영향에 대해 무시할 정도로 안정화되지 않았다. 본 연구는 산불 발생 후 강우 및 토사유출에 대한 비 산불 지역과의 차이를 살펴봄으로써 수질 오염원으로 작용할 수 있는 가능성을 기초자료로 제공될 것이다. 또한, 수처리 기술개발을 포함한 예방책을 만들어 내는데, 기초자료로 활용할 수 있길 바란다.

Cytotoxicity of Paraquat or Bentazone and Compensatory Effects of 3-Methylcholanthrene on the Rat Liver (Paraquat 및 Bentazone의 세포독성과 흰쥐 간에서 3-Methylcholanchrene의 독성경감효과)

  • This study was carried out to investigate cytotoxicity of paraquat or bentazone on NIH 3T3 fibroblasts, toxicity of paraquat or bentazone, and compensatory effects of 3-Methylcholanthrene(3-MC) on the rat liver. In order to MTT assay, the $5.0{\times}10^4$ cell/mL of NIH 3T3 fibroblast in each well of 24 multidish were cultured. After 24 hours, the cells were treated with solution of paraquat or bentazone(1, 25, 50, 100 ${\mu}M$ respectively). After the NIH 3T3 fibroblast of all groups were cultured in same condition for 48 hours. MTT assay were performed to evaluate the cytotoxicity of cell organelles. Paraquat or bentazone $MTT_{50}$ were 1668.97 ${\mu}M$ and 1506.97 ${\mu}M$, respectively. These $IC_{50}$ of paraquat or bentazone were decided low cytotoxicity by Borenfreund. In order to observe the toxicity and compensatory effects of paraquat or bentazone on the rat liver, Sprague-Dawley male rats were used as experimental animals and divided into paraquat or bentazone only treated group and simultaneous application group of paraquat or bentazone and 3-MC. At 30 min and 1, 3, 6, 12, 24, 48 and 96 hrs interval after each treatment, the animals were sacrificed by decapitation and liver were immediately removed, immersed in fixatives, and processed with routine method for light microscopic study. Paraffin sections were stained with H-E, PAM and Best Carmine. Under the light microscope, degenerative changes of hepatic lobules were frequently observed in portal area from 3 hrs after paraquat or bentazone treatment. All hepatic cells were induced degenerative change at 12 hrs and more severe degenerative change at 48 hrs after paraquat or bentazone treatment. Especially, hepatic cells of bentazone only treated group were distinctly showed pyknotic. Glycogen granules were increased in portal area at 3 hrs, all hepatic cells at 12 hrs and remarkably increased at 48 hrs after paraquat or bentazone treated group. But hepatic cells of bentazone only treated group were regeneration at 48 hrs from portal area and glycogen granules of hepatic cells of paraquat or bentazone and 3-MC combination treated group showed in central area only at 48 hrs. The results indicate that 3-MC may be decrease paraquat or bentazone cytotoxicity on the rat liver.

  • The soil management practices before transplanting the ginseng plant were studied with two organic matter sources such as a traditional organic matter (wild grass) and commercial organic fertilizer (byproducts of amino acid fermantation) during the late spring to late autumn. During the soil management practices, the soil received 40kg N/10a from five different combination treatments with above two organic matter sources, a wild grass and a commercial organic fertilizer. After the application of the treatments, the soil were ploughed regularly at the interval of 20 days and the changes of physicochemical properties during the soil management practices were investigated. The next year after soil management practices, ginseng plants were transplanted to each treatment, growth and the content of some organic components of ginseng plant were measured for comparision of the different treatments. 1. The decrease in bulk density observed during the first 40 days of management was considered to be the effect of the improved physical conditions caused by ploughing, The decrease in bulk density observed after 40 days of management was considered to be the effect of organic matter. Similar results were observed in particle density, however porosity increased with time. 2. Soil pH tended to decrease during the first 40 days of management, after which period the pH increased and was stabilized. However, CEC increased with organic matter treatment and the exchangeable $NH^+_4-N$ and $NO^-_3-N$ increased in 20 and 40 days after the management practices, respectively, and after that period it became steady. 3. The decomposition rate of treated organic matter was measured by the incubation test in laboratory conditions. The rate of decomposition was rapid during the first 20 days of management, after which period it showed slight changes. 4. The weight of ginseng root significantly increased in the treatment of 10kg N/10a organic fertilizer and 30kg N/10a wild grass. 5. The saponin content of ginseng root was highest in the 40kg N/10a wild grass treatment. The addition of organic fertilizer at the rate of more than 20kg N/10a caused the decrease in the saponin content.

  • The carrier materials used for the development of bacterial inoculants to be effective in field were made with various carrier materials of two major forms, alginate bead and powder inoculants. Inoculants were prepared after mixing those carrier materials with Pseudomonas fluorescens SSL3 and Bacillus subtilis B5, and the treatment effects of each inoculants was investigated on cucumber, tomato, pepper and potato. Survival density of SSL3 and B5 in various carrier materials for duration of storage and the bead inoculants were better than the powder. In the powders, survival rate increased in carrier materials treated 5% skimilk. The growth condition of microorganisms in carrier materials is good at powder. When they were preserved in the long period, contamination is problem. Scanning(200 to 600nm) of the P. fluorescens SSL3 supernatant in centrifuged MKB broth incubated for 48h had two main peaks, pyochelin(300nm) and pyoverdin(400nm). The potato yield in field experiments of spring, treated with bead formulas showed increase of 22~29% in whole potato breeds as compared with control, because the bead formulas degraded, and released the antibiotic microorganisms in slow and constant rate. In the pot experiment, there were significant difference in soil, wheatbran, and bead formed wheatbran.

Effect of Polymer, Calcium, Perlite and Chitosan in Soil Organic Amendment on Growth in Perennial Ryegrass (유기질 토양개량재에서 고분자 중합체, 칼슘, 펄라이트 및 키토산이 퍼레니얼 라이그래스의 생장에 미치는 효과)

  • The study was carried out to investigate the effects of polymer, calcium, perlite and chitosan on the growth of perennial ryegrass (Lolium perenne L., PR) and to provide a basic information needed for their practical application when establishing garden, parks, athletic field and golf courses with these materials. A total of 24 treatment combinations were applied in the study. Treatments were made of water-swelling polymer (WSP), calcium, perlite and chitosan mixed in soil organic amendment (SOA). Germination rate, turfgrass coverage, turfgrass density and top growth were evaluated in PR under greenhouse conditions. Significant differences were observed for these growth characteristics among the treatments. Turfgrass density and plant height, evaluated on a weekly basis, varied with time after seeding. A proper mixing rate of WSP was considered to be lower 3% for the growth of PR with an exception of being below 6% for turfgrass density. Germination rate and early survival capacity were greatly influenced by calcium and chitosan among the elements of calcium, perlite, and chitosan. But there was little effect by perlite. Calcium and chitosan were most effective one for turfgrass density and coverage, respectively. Top leaf-growth was influenced by all three elements, but the greatest effect was highly linked with calcium. Chitosan was very effective in early germination and vertical leaf growth, as compared with the others. Future studies are required for measuring the effect of WSP, calcium, perlite and chitosan on the turf growth characteristics in root zone mixtures of sand+SOA before a practical field use.

Studies on the Indigenous Vesicular-Arbuscular Mycorrhizal Fungi(VAMF) in Horticultural Crops Grown Under Greenhouse -III. Effects of the Indigenous VAMF Inoculation on the Early Growth and the Subsquent Growth after Transplanting of Greenhouse Grown Crops (시설원예(施設園藝) 작물(作物)에서 토착(土着) VA균근균(菌根菌)에 관한 연구(硏究) -III. 토착(土着) VA 균근균(菌根菌) 접종효과)

  • Effects of the indigenous Vesicular-arbuscular mycurrhizal fungi(VAMF) on early growth response of greenhouse grown crops were experimented. This study was done to evaluate the benefit of indigenous VAMF inoculation on the early growth and the subsequent growth after transplanting of some crops such as cucumber, tomato, hot pepper, eggplant, and melon. Leaf area, shoot dry weight, and plant length of mycorrhizal greenhouse crops showed the tendency of significant or no significant increase over control plants receiving no inoculation. The levels of VA mycorrhizal colonization were increased with plant growth, and infection rates of horticultural crop except hot pepper around one week after transplanting were decreased, while that of 8 weeks after emergence of mycorrhizal seedlings were increased again and infected by around 50% at harvesting time. In spore densities in the rhizosphere soil of craps experimented, the number of spore ranged from $72.7{\pm}26.3$ to $100{\pm}10.3g^1$ on dried soil basis and high density showed in both cucumber and tomato. Total nitrogen contents in shoots were lower in the mycorrhizal plants than non-mycorrhizal one, whereas P uptake in mycorrhizal hot pepper and tomato were highly ramarkable. The K contents in the shoots of mycorrhizal cucumber and eggplent were highly enhanced. Inoculation of the indigenous VAMF enhanced shoot Ca and Mg in both tomoto and melon. The contents of Fe, Zn, Mn and Cu in shoots of mycorrhizal crops were higher than non-mycorrhizal plants and vice versa in case of eggplent. Inoculation of the indigenous VAMF to horticultural crops were effective for alleviation of transplanting shock, and pretransplanting infection improved subsequent growth by reducing the time required for establishment of a functional mycorrhizal symbiosis following transplanting.

  • Six herbicides were evaluated to investigate the phytotoxicity of rice plant and the weeding efficacy influenced by the time of application in mechically transplanted paddy field. The amount of each chemical applied was 3 kg, a. i, /ha. Chlormethoxynil : Rotala indica KOEHNE and Lindernia pyxuiaria PHILCOX were effectively controlled when applied on the 12th day after transplanting (12 DAT) and this herbicide was excellent for the control of Echinochloa crusgalli P. BEAUV, Monochoria vaginalis PRESL and Sagittaria pygmaea MIQ, when applied early (7 days after puddling) but its weeding effect for these weeds decreased greatly as the application time became later. It had a controlling effect for Potamogeton distinctus A. BENN, Cyperus serotinus ROTTB and Scirpus hotarui ROXB at the initial period at the earlier application time. Butachlor was effective in controlling E. crusgalli, R. indica and L. pyxidaria at 12 days after transplanting (DAT) but was not effective in controlling P. distinctus and S. pygmaea even at the early application time. M. vaginalis, C. serotinus and S. hotarui were effectively controlled by the butachlor treatment at 7 days after final puddling (2 DBT-SDAT) but this weeding effect decreased at the late application time. A combination of butachlor and naproanilide excellently controlled E. crusgalli, R. indica, L. pyxidaria and S. pygmaea regardless of the application time. For the control of M. vaginalis, C. serotinus and P. distinctus, the weeding effect of this mixtures was much greater than that of the single treatment of butachlor. Perfluidone was excellent for the control of E. crusgalli, R. indica, L. pyxidaria, M. vaginalis and S. pygmaea at either application time tested. P. distinctus, C. serorinus and S. hotarui could be controlled by this chemicals until the time of first observation (23 DAT) but the effect for these weeds somewhat decreased as time passed. The effect of pyrazolate on E. crusgalli, M. vaginalis, S. hotarui and P. distinctus was very excellent regardless of the application time but R. indica and L. pyxidaria could not be completely eliminated by this chemical. This chemical was effective in controlling C. serotinus when applied at 7-9 days after final puddling and showed a controlling effect for S. hotaruionly at the initial period. Piperophos + dimethametryn was very excellent for the control of all the annual weeds and P. distinctus. It showed a controlling effect on S. pygmaea, C. serotinus and S. hotarui only at the initial period. There was no difference in the effects on phytotoxicity and yield between chlormethoxynil and pyrazolate at either times of application tested. The later the application time was, the less the phytotoxicity of butachlor and piperophos+dimethametryne was. The phytotoxicity of butachlor + naproanilide and perfluidone decrease in the plots treated at the later application time. When the last two chemicals were treated at 2 days before transplanting (DBT) the yield decreased as compared with the hand weeded plot.

An Experimental Study on Strength Properties of Concrete Using Blast-Furnace Slag Subjected to Freezing at Early Age (초기재령에서 동결을 받은 고로슬래그 콘크리트의 강도발현특성에 관한 실험적 연구)

  • Recently, to consider financial and constructive aspect usage of Admixture such as Blast-Furnace Slag and Fly-Ash, are increased. Also the use of cold-weather-concrete is increased. Blast-Furnace Slag, a by-product of steel industry, have many advantage to reduce the heat of hydration, increase in ultimate strength and etc. But it also reduces early-age strength, so it is prevented from using of Blast-Furnace Slag at cold-weather-concrete. In this study, for the purpose of increasing usage of Blast-Furnace Slag at cold-weather-concrete, it is investigated the strength properties of concrete subjected to frost damage for the cause of early age curing. The factors of this experience to give early frost damaged were Freezing temperature(-1, -10, $-15^{\circ}C$), Early curing age(0, 12, 24, 48hour), Freezing times(0, 12, 24, 48hour). According to this study, if early curing is carried out before haying frost damage, the strength of concrete used admixture, subjected to frost damage, is recovered. And that properties are considered, the effect of using admixture like blast-furnace-slag, is very high
