The Effect of Molecular Size and Degree of Phosphorylation on the Emulsion Stability of Microcrystalline Chitin (Chitin의 분자량과 Phosphorylation 정도가 Microcrystalline Chitin의 유화안정 능력에 미치는 영향)

  • This study was carried to investigate the emulsion stability of microcrystalline chitins (MCC) prepared from chitins of different molecular sizes and different degrees of phosphorylation in a model fatty food system. Chitins of low, medium and high molecular size prepared from crabshells were phosphorylated to 30-50% to make MCC. MCC prepared from chitin of medium molecular size revealed a high emulsion stability. The best emulsion stability was observed in MCC prepared from medium size chitin with 40% phosphorylation (M-40-MCC). The fat binding capacity of MCC was not significantly different among the samples. MCC with 50% phosphorylation had more fat binding capacity, ranging from 650-690%. When 2.46% of M-40-MCC was applied to a coconutoil-water system as an emulsifier, emulsion separation was observed from 10 min after emulsification, indicating that it could not be used as a sole emulsifier. When 50% of emulsifier (Span-60 &Tween-60) in a liquid coffee creamer, selected as a model fatty foods, was replaced by M-40-MCC, emulsion stability was as good as control. Consistency of liquid creamer was decreased and L value increased as the amount of MCC repacement increased. A liquid creamer with 50% emulsifier replaced by MCC had sensory characteristics equivalent to control.

  • New type of chitosan derivatives, chitosan-g-MAP, were synthesized by graft copolymerization of mono (2-methacryloyl oxyethyl) acid phosphate (MAP) into chitosan, in order to solubilize chitosan in water. Ceric ammonium nitrate was used as an initiator for graft copolymerization. The optimal conditions for graft copolymerization were determined on the basis of reaction temperature, time, and the concentration of initiator and monomer. The reaction conditions for the highest percentage of grafting were as follows: an initiator concentration, 3.5${\times}$10$\^$-3/ M; monomer concentration, 0.19 M; and reaction temperature, 40$^{\circ}C$ The reaction rate reached the maximum value after 4 hrs of reaction. Antifungal activity was tested against Candida albicans, Trichophyton rubrum and Trichophyton violaceum by using chitosan-g-MAP and two other chitosan samples which have degree of deacetylation of 70% (DA-7) and 90% (DA-90). Their antifungal activities were investigated in weak acidic range. Maximum antifungal activity of them was observed at pH 5.75. Chitosan-g-MAP inhibited thoroughly the growth of Candida albicans and Trichophyton violaceum. Howerver, DA-70 and DA-90 showed higher antifungal activities on Trichophyton rubrum than that of chitosan-g-MAP.

Suppressive Effects of Homemade Environment-friendly Materials on Alternaria Blight and Anthracnose of Ginseng (친환경자재를 이용한 인삼 점무늬병과 탄저병의 발병억제효과)

  • This study was performed to evaluate the suppressive effects of organic fungicides made using environment-friendly materials on leaf spot disease and anthracnose that infect ginseng. Anthracnose (Colletotrichum gloeosporioides) and leaf spot disease (Alternaria panax) are principal diseases that decrease the yield of ginseng by defoliation before root enlargement. Fermented eggs and oyster shells, water extract of green tea and ethanol extract of red ginseng dregs were significantly effective in suppressing leaf spot disease. Fermented crab and shrimp shells and fermented motherwort were also effective in suppressing the recurrence of ginseng anthracnose. The preventive effects of these environment-friendly materials were definitely superior to the therapeutic effects. Therefore, these materials could be used as alternatives to chemical pesticides, which can not be applied in organic ginseng cultivation field. These organic fungicides need to be applied before the incidence of ginseng anthracnose in order to maximize their suppressive effects.

  • Chitin was isolated from red snow crab (Chinonecetes japonicus) and deacetylated by boiling alkaline solution to produce chitosan. The physical properties of chitosan solution and its film properties was examined, As the molecular weights of chitosan was increased from 297Kpa to 319Kpa, the tensile strength, degree of elongation and water permeability of chitosan film were increased. As the degree of deacetylation of chitosan was increased, the tensile strength, the degree of elongation and water permeability of chitosan film were increased. As the concentration of chitosan solution was increased, the degree of elongation and water permeability of film were increased, whereas the tensile strength of film was decreased, As the pH of chitosan solution was increased, the tensile strength and water permeability of film were decreased, whereas the degree of elongation of film was increased.

Volatile Flavor Compounds Identified from the Sauces Made with Waste of Shrimp, Crab and Lobster (새우, 게 및 바다가재의 부산물로 만든 소스의 휘발성 향기 성분)

  • Volatile flavor compounds of sauces made from shrimps, crabs or lobsters were analyzed by the combination of canister system, gas chromatography(GC) and mass selective detector(MSD). Of 72 total volatile compounds from 4 kinds of sauces, 45 compounds were identified from shrimp sauce(SS). Ten alkanes, 5 ketones, 3 aldehydes were obtained from SS. Especially, 3-methyl-2-butanone, 2-pentanamine, isobutane, 3-methyl-2-butanol, carbon disulfide and dimethyl sulfide were predominant compounds in SS. In crab sauce(CS), there were 18 compounds identified, including 4 alcohols, 4 alkanes, 3 aldehydes, 2 ketones, acid and amine. 2-Methoxy ethanol, trimethyloxirane and 3-buten-1-ol were special volatile compounds in CC. Volatile compounds from lobster head sauce(LHS) or lobster shell sauce(LSS) were 16 or 18 kinds respectively. The major volatile compounds of LHS were formic acid, 1-propanethiol, $\beta$-pinene and allyl sulfide, and those of LSS were acids, pentane, 3-methyl-1-butanol and 2,4-dimethyl-3-pentanone. It was thought that the volatile compounds identified from sauces as well as shrimps, crabs or lobsters might come from wine, onions, bay leaves or celery used as minor ingredients.

A Study on the Synthesis of Carboxymethyl Chitin and Separation of Alkali-Earth Metal ions by Adsorption (Carboxymethyl Chitin의 합성 및 알칼리 토금속 이온의 흡착분리에 관한 연구)

  • Carboxymethyl chitin(CM-chitin) was prepared by the reaction of alkali chitin with monochloroacetic acid in isopropyl alcohol. According to the pH variation, the adsorptivity of this chelating polymer to the alkali-earth metal ions such as $Ca^{2+},\;Mg^{2+}$, $Sr^{2+}$, $Ba^{2+}$ ions was determined by batch method. The adsorption tendency of this chelating polymer to most metal ions was increased with the increase of pH. The highest degree of adsorption was observed toward $Ca^{2+}$ ion among the alkali-earth metal ions. The selectivity adsorption property toward $Ca^{2+}$ ion was examined in the solution of $Ca^{2+}$ and $Mg^{2+}$ ions, and it was observed that CM-chitin showed excellent selectivity to $Ca^{2+}$ ion than $Mg^{2+}$ ion. $Mg^{2+}$ ion bound to CM-chitin molecule in the presence of $Ca^{2+}$ ion owing to low equilibrium constant. In the adsorption experiment of $Ca^{2+}$ and $Mg^{2+}$ ions to the CM-chitin under coexistence of $Na^+$ and $K^+$ ions, it observed that adsorptivity of only $Ca^{2+}$ ions was not affected by these monovalent cations.

The Effect on Sauce Selection and Preference of Teriyaki Sauce by Material (데리야끼 소스의 재료별 선호도와 소스 선택에 미치는 영향)

  • This study aims to provide basic data about the diversification and the development of sauces by making four kinds of Teriyaki sauce, and the findings are as follows. The satisfaction with Teriyaki sauce by age and cooking career and groups showed a significant difference in several items. According to the result of the preference analysis on professors and teachers, the preference for health/nutrition showed a significant difference; also, the result of the preference analysis on Teriyaki sauce ingredients among the culinary students showed a significant difference in awareness and health/nutrition. In addition, the result of the preference analysis on Teriyaki sauce ingredients among college students showed a significant difference in the order of taste, awareness, price, and health/nutrition. Lastly, the result of the preference analysis on Teriyaki sauce ingredients among all groups showed a significant difference in the order of awareness, price and health/nutrition. As the mindset that main ingredient of Teriyaki sauce is eel bone prevails and the sauce made by crab shells is highly preferred, sauces added with sea food products are considered to be possibly developed. The follow-up study will have to deal with a manufacturing method of sauce with high nutrition and economy efficiency resulting from cheap manufacturing unit price by making Teriyaki sauce with inedible but nutritions sea food products.

Analysis of Differentially Expressed Genes by Resveratrol Using Membrane Microarray (Membrane Microarray를 이용한 Resveratrol에 의해 차별적으로 발현되는 유전자 군의 분석)

  • In the present study, we investigated whether several phytochemicals (resveratrol, genistein, epicatechin gallate, dially disulfide, caffeic acid phenetyl ester) and sulindac sulfide could induce expression of tumor suppressor p53 protein in human colorectal HCT116 cells. We found that p53 was dramatically induced by all phytochemical treatments except sulindac sulfide. Among treated phytochemicals, we selected resveratrol for further experiments because it is one of the highest p53 inducer. Using a Western blot analysis, we found that resveratrol induced p53 in a dose- and time-dependent manner. Additionally, using membrane-based microarray analysis, we found that twenty-five genes were up-regulated and two genes were down-regulated by resveratrol treatment. Among the up-regulated genes, we selected 4 genes and performed reverse-transcription-PCR to confirm microarray data. The results of RT-PCR were highly accorded with those of membrane microarray. In addition, we found that thrombospondin-1 (TSP-1) expression was not dependent on p53 presence, whereas mammary serine protease inhibitor (MASPIN) expression was dependent on p53 expressed by resveratrol treatment. The results of this study may help to promote our understandings of the molecular mechanisms of chemoprevention that are mediated by resveratrol in human colorectal cancer.

A Study on Adsorption Characterics of Metallic Ions by Tannin Immobilized on Chitin and Chitosan (Chitin 및 Chitosan에의 고정화 탄닌의 금속이온 흡착특성에 관한 연구)

  • Epoxy-activated chitin was synthesized by the reaction of epichlorohydrin with chitin which was isolated from waste marine sources such as crab shell. Followed by the reaction of epoxy-activated chitin with hexamethylenediamine, the aminohexyl chitin was synthesized. The aminohexyl chitin was subsequently reacted with epichlorohydrin to prepare the epoxy-activated aminohexyl chitin. Finally, the tannin-immobilized chitin (Resin I) was synthsized by the reaction of tannin solution with epoxy-activated aminohexyl chitin. Using silane coupling agent, the tannin-immobilized chitosan(Resin II) was synthesized by the reaction of $\gamma$-glycidoxypropyltrimethoxy silane with chitosan which was prepared by the deacetylation of chitin. Upon the pH variation, adsorptivities of these immobilized tannins to the metal ions such as $Cu^{+2}$, $Ni^{+2}$, $Cr^{+6}$, $Co^{+2}$, $Ca^{+2}$, $Pb^{+2}$, $Ba^{+2}$, and $UO_2{^{+2}}$ ions were determined by batch method. The adsorptivity tendencies of these immobilized tannin to the most of metallic ions were increased with pH. Furthermore, the adsorptivities of Resin(I) and Resin(II) upon the variation of pH, contact time, amount of resin and concentration of metal ion were investigated. As a result, it was found that these immobilized tannin on both chitin and chitosan showed good adsorptivities for uranyl ion.

  • Coating with chitosan ($2\%$, pH: 5.0) had a beneficial effect such as less weight loss, flesh firmness and maintenance of sugar content of tomatoes stored at the normal temperature. The weight loss of chitosan-coated tomatoes was $4.8\%$ during the storage-period (18days) while that of control was $10.2\%$. When it was stored for 18 days, the tomatoes coated with chitosan showed higher in firmness (1.63 kg) and pectin content ($0,58g\%$) than the control (1.03 kg, $0.44g\%$). For the total soluble acid and pH, the tomatoes coated with chitosan were higher in oxalic acid ($0.17 g\%$) than the control ($0.15 g\%$) and the latter were higher in the pH than the former, when they were stored for 18 days.
