Natural Dyeing with Aqueous Black Bean Seed Coat extract (검정팥 종피 추출물에 의한 천연염색)

  • 팥에 함유되어있는 색소와 관련된 연구 중에서는, 검정팥의 색소성분을 분리하여 delphinidin-3-glucoside(D-3-G)로 보고한 Sasanuma의 연구와 검정팥 껍질속에 포함된 안토시아닌의 함량을 평가한 Yoshida의 연구 결과 등이 있다. 본 연구에서는 검정팥 종피에서 색소를 추출하여, 적정 염색조건을 설정하였으며, 매염제별 색상변화 및 견뢰도를 측정하는 과정을 통해 염색 특성을 파악하여 새로운 천연염재로서의 활용 가능성을 진단하였을 뿐만 아니라 응용 연구를 위한 기초자료의 확보에 주안점을 두었다. pH에 따른 염색성에서는 견직물의 경우 pH 4에서 가장 염착량이 많았고, 면직물의 경우에는 주어진 범위내에서는 pH가 높을수록 염착량이 많았다. 염색온도와 시간에 따른 염색성은, 견직물의 경우, 염색온도 $80^{\circ}C$에서는 시간이 경과할수록 K/S 값이 크게 나타났으며, 면직물의 경우도 견직물과 비슷한 결과로 온도가 높아질수록 염색시간이 경과될수록 K/S 값이 크게 나타났다. 견직물에 대한 매염제 종류별 표면색의 변화에서는 Fe 매염포만 $YR{\rightarrow}Y$ 로 변화되었을 뿐 다른 매염제에서는 매염 후에도 색상 변화는 크지 않았다. 매염처리 전 견직물의 일광 견뢰도는 무매염이 4~6등급, Al 처리포는 4~5등급, Cu와 Sn은 3~4등급으로 나타났고 Fe는 2~3등급으로 가장 낮게 나타났으며, 세탁견뢰도에서는 무매염 2등급, 매염제 처리의 경우 2~3등급으로 나타났다. 무매염 면직물의 일광견뢰도는 1~2 등급, Fe 매염은 2~3등급, Cu 2등급 Al과 Sn은 1~2등급으로 매염처리에 의해 전혀 개선되지 않았으며, 세탁견뢰도의 경우 Cu매염이 4등급으로 양호하게 나타났으며 무매염, Al, Sn과 Fe는 3등급으로 나타났다.

Physicochemical Properties of Aqueous Extracts in Small Red Bean, Mung Bean and Black Soybean (두류(팥, 녹두, 검정콩) 물추출액의 이화학적 특성)

  • In order to compare physicochemical properties of aqueous extract in 3 beans (small red bean, mung bean, black soybean), proximate composition, free sugars, free amino acids, minerals, absorbance and surface refractance color were investigated. Regardless of raw materials composition, the 3 beans extracts had similar proximate compositions. Free sugar analysis showed that fructose, glucose, sucrose, maltose, raffinose and stachyose were varied among the extracts. Raffinose and stachyose were the major sugar and fructose was measured only in black soybean and glucose was merely found in small red bean. Seventeen free amino acids in beans extract were analysed in the extracts. Among the free amino acids, arginine, aspartic acid, glutamic acid and serine were the main amino acids. Black soybean extract had maximum absorbance at 460 nm and 540 nm. L value of black soybean extract was lower than those of small red bean and mung bean.

Colorant Extracting and Its Storage Stability from Red bean and Black bean Seed Coat (팥 종피에서의 색소 추출과 저장 안정성)

  • Extraction efficiency of colorants from red bean and black bean seed coat and its storage stability were examined according to the various extracting and storage conditions in this study. The obtained results were as follows ; The absorbance values of extracted colorants were increased with increasing extract time and temperature. pH 4 extracting was most efficient among the various pH conditions. The absorbance values of extracted colorant from red bean were increased with increasing extract time and temperature, when the extraction process was kept at $30^{\circ}C$ and $50^{\circ}C$, but it were decreased with increasing extract time at $70^{\circ}C$. In the case of black bean, it were also increased with increasing extract time and temperature. pH 5 extract was most efficient among the various pH conditions. In juice extractor system from red bean, the pH 5 of the pre-treatment was more efficient than the distilled water of pre-treatment. In black beans of the short-term pre-treatment with distilled water, the colorant was extracted better than at pH 5, but above 4 hours pre-treatment was decrease in pH 5. The Methionine added as a stabilizer gave the more effect to the stability of colorant from red bean the more Methionine addition. In the case of black bean 10mmol addition of methionine was more efficient than 20mmol.

Identification of Virus from Azuki Bean Plant (팥에 발생하는 바이러스 분리 동정)

  • Three types of virus disease symptoms were observed in azuki bean plants: yellow mosaic; mosaic; severe mosaic with dwarf. The symptoms developed in the indicator plant inoculated with a virus- infected leaf of azuki bean showed similar host range with those of AMV, CMV and AzMV. In antiserum response, yellow mosaic symptom formed sediments with AMV antiserum, mosaic type with CMV antiserum, respectively, From the electron microscope observation, eclliptic particle (18~58$\times$18nm), isometric particle (30nm), and filamentous(730$\times$12nm) combined with inclusion body were observed in yellow mosaic, mosaic, and severe mosaic with leaf curling symptoms, respectively, The results demonstrate that yellow mosaic, mosaic, and severe mosaic with dwarf are caused by AMV, CMV and AzMV.

  • In this study the natural colorant was extracted from black bean seed coat in aqueous solution and used to dye silk and cotton fabrics. To obtain the optimal dyeing conditions it was examined at various dyeing condition (temperature, pH, time and liquor ratio). The dyeing behavior and the depths of shade which were evaluated in terms of K/S and CIELAB color difference values of the dyed and mordanted fabrics were also investigated. The dyeing fastness evaluated standard light and wash fastness tests. The obtained results were as follows ; The most K/S value of silk and cotton fabrics were obtained when the pH was 4.0 and 4.8, respectively and it increased slightly with dyeing time passes when the dyeing temperatures were at $40^{\circ}C$ and $60^{\circ}C$ but, it increased at $80^{\circ}C$. The color of silk fabrics changed from yellowish red to yellow at only Fe mordanting among various mordanting. Sn and Ti mordanting of silk fabric and only Ni mordanting of cotton fabric increased the $L^*$ values, but the others decreased. The light fastness of silk fabrics showed 4-6 grade without mordant, 4-5 grade with Al, 3-4 grade with Cu and Sn, and 2-3 grade with Fe as mordant, and that of cotton fabrics showed 1-2 grade without mordant, 2-3 grade with Fe, 2 grade with Cu, 1-2 grade with Al and Sn as mordants. All mordanting coluld not improve the light fastness of fabrics. Washing fastness(fade) of silk fabrics showed 2 grade without mordants and 2-3 grade with mordants and those of cotton fabrics showed 4 grade with Cu, 3 grade without mordant and with Al, Sn and Fe. All of the washing fastness(stain) of both fabrics showed 4-5 grade.

  • To investigated the anticarcinogenic activity of 70% ethanol extracts from various cereal in vitro, antimutagenic activity, inhibitory effect of DNA strand scission and tumor promotion were examined. The antimutagenic activity of the beans such as black bean and small red bean was generally higher than that of cereals examined. However inhibitory activity of 70% ethanolic extracts against DNA strand scission induced mitomycin C showed that millet, job's tear, black bean and soy bean among cereals and beans tested in this study inhibited effectively the DNA strand scission. Antioxidative activity of some cereal extracts determined by using linoleic acid model system showed that Job's tear, millet and black bean were higher antioxidative activity than other cereals and beans. Conventional short-term antipromoter assay system using activation of Epstein Barr virus (EBV) clearly demonstrated that sorghum, buckwheat, black bean and small red bean have inhibitory effects on promotion in cellular carcinogenesis.

Selection of Appropriate Herbicides for Establishment of Weed Control System in Adzukibean and Mungbean (팥과 녹두 잡초방제체계(雜草防除體系) 수립(樹立)을 위한 제초제(除草劑) 병발(迸拔)에 관한 연구)

  • To select appropriate herbicides for adzukibean and mungbean, a series of experiments was carried out in both field and pot from 1980 to 1983. Tolerance to the herbicides tested was highest in soybeans and followed by mungbean and adzukibean in the order. Pre-emergence herbicides showed relatively low phytotoxicity were chlorambem, linuron, and metribuzin for adzukibean and alachlor and butachlor for mungbean. Post-emergence herbicides showed no external phytotoxicity for adzukibean and mungbean were alloxydim, Dowco 453, fluazifop, etc.. For mungbean, terbutryn (1.5 kg, a.i./ha) which is pre-emergence herbicide showed the best weeding control efficacy but some possible phytotoxicity. Among post-emergence herbicides, acifluorfen (300 g, a.i./ha) showed the best weeding efficacy with no yield reduction though some phytotoxicity which recovered within 20 days. Compared to single herbicide application, the mixture or systemic treatments of herbicides showed much higher weeding control efficacy and seed yields: the systemic treatments of linuron (500 g, a.i./ha) / Dowco 453 (180 g, a.i./ha) or linuron (500 g, a.i./ha) / fluazufop (260 g, a.i./ha) for adzukibean and the mixture treatment of alachlor (1.31 kg, a.i./ha) / acifluorfen (150 g, a.i./ha) for mungbean, respectively.

Studies on Pathogenicity of Wheat Scab Fungus(Gibberella zeae) to Various Crop Seedlings

  • 1. Purpose of this experiment is to investigate the influence of wheat scab fungus(Gibberella zeae) remaining in soil or residues of infected plants to the growth of the seedlings of 14 different crops. 2. Susceptibility of these crops to the infection by G. zeae was investigated by planting these crops both to the artificially inoculated soils and naturally infected soils where both serious and light wheat scab epidemic have occurred previously. 3. In artifical inoculation tests, bailey, wheat, rye, soybean, rice, buckwheat, corn, cotton and greenbean has shown susceptible reaction, while indianbean, sesame, sorghum, chinese cabbage and radish has shown resistant reaction. 4. In artificial inoculation tests, there was significant difference in the germination ratio of the susceptible crops between the plants planted in inoculated soils and uninoculated soils with the exception of rice, com and cotton. Preemergence seedling blight was confined only to barley and corn, whereas postemergence seedling blight was confined to rye, wheat, rice buckwheat, barley and corn. la most of the susceptible crops secondary roots were almost rotted and the primary roots were either partially rotted or discolorated in inoculated soil. There was significant difference in the stem length of the plants grown in inoculated soils and uninoculated soils in susceptible crops. 5. No infection by G. zeae was observed when wheat, barley and rye plants were sown to the soils where both serious and light wheat scab epidemic occurred naturally.

  • We have established the quantitative method for assay antimutagenic activity from natural products using SOS chromotest technique. Establishment of the method in this study makes it possible to numerize antimutagenic activities from samples in term of the sample amount required for 50% inhibition to mutagenic activity induced by the chemical mutagen under the standard assay condition. Antimutagenic activities of rices from different cultivars as well as other cereals were assayed through this method. The results revealed that antimutagenic activities of mutant cultivar, Suwon 393(Hyangdo) and Sanghaehyanghyulla(Jado), were higher than Chuchung which mainly consumed for steamed rice, and also indicated that antimutagenic activities of cereals, such as job'tear, buckwheat, small red bean, black bean were generally higher than that of brown rice.

Effects of Acidic Treatments for Anthocyanin and Proanthocyanidin Extraction on Black Bean (Glycine max Merrill.) (검정콩 안토시아닌 및 프로안토시아니딘 추출에 대한 산 처리의 영향)

  • This study was performed to investigate the effects of various acidic treatment for anthocyanin and proanthocyanidin extraction from black bean. Extracts were prepared with 80% methanol solutions containing HCl, acetic, formic, phosphoric, and citric acids of different concentrations (0.1, 0.2, 0.3, 0.5, and 0.7%). Total anthocyanin content ranged from 0.74 mg/g in 0.7% citric acid to 1.74 mg/g in 0.3% HCl, depending on acid type and concentration. The major anthocyanins were delphinidin-3-glucoside (D3G), cyanidin-3-glucoside (C3G), and petunidin-3-glucoside (Pt3G). The highest C3G content was 1.12 mg/g in 0.3% HCl. Proanthocyanidin content ranged from 2.01 mg/g in 0.5% acetic acid to 5.29 mg/g in 0.3% HCl. Thus, acidic condition is a significant factor affecting anthocyanin and proanthocyanidin extraction from black bean, and the optimum extraction conditions were determined as 80% methanol containing 0.3% HCl.
