  • The Geyoungju Ca-bentonite with dry density of 1.6 g/$cm^3$ has been considered as a standard buffer material for the disposal of high level waste in KAERI disposal system design. But it had relatively lower thermal conductivity compared with other surrounding materials, that was one of key parameters to limit the increase of the disposal density in the disposal system. In this study, various additives were selected and mixed with the Ca-bentonite in different mixing methods in order to increase the thermal conductivity from 0.8 W/mK to 1.0 W/mK. As an additive, CNT (Cabon Nano Tube), graphite, alumina, CuO, and $Fe_2O_3$ were selected, which are chemically stable and have good thermal conductivity. As mixing methods, dry hand-mixer mixing, wet milling and dry ball mill mixing were applied for the mixing. Above all, the ball mill mixing was proved to be most effective since the produced mixture was most homogeneous and showed higher increase in the thermal conductivity. From this study, it was confirmed that the thermal conductivity for the Geyoungju Ca-bentonite could be improved by adding small amount of highly thermal conductive material to 1.0 W/mk. In conclusion, it was believed that the experimental results will be valuable in the disposal system design if the additive effects on the swelling and permeability on the compact bentonite are also approved in further studies.

  • The objective of this study was to identify the effect of zebularine, a DNA methylation inhibitor, on the chromosomal association between homoeologous chromosomes in the wheat genetic background. Zebularine at a final concentration of 10 µM was used to treat the spikes of the double monosomic wheat addition line (DMA) with one Leymus mollis chromosome and one Leymus racemosus chromosome, both of which were in a homoeologous relationship. In late prophase, zebularine led to chromosome breakage in the Leymus homoeologous chromosomes. Chromosome breakage caused an increase in the frequency of chromosomal associations between the Leymus homoeologous chromosomes. Ordinary DMA showed 65 cells (35.3%) with chromosomal associations and 119 cells (64.7%) with no association, whereas treated DMA showed 102 cells (60.0%) with chromosomal associations and 67 cells (39.4%) with no association. In diakinesis, the Leymus bivalent showed a chromosomal association in the whole euchromatic region. In metaphase, the Leymus bivalent showed association in the whole chromosomal region, unlike other Leymus bivalents with partial chromosomal association. Chromosomal association by chromosome breakage occurred not only between Leymus chromosomes but also between Leymus and wheat chromosomes. The frequency of other chromosomal association (such as fusion and insert) was increased. Chromosome breakage by zebularine treatment is a useful method at the chromosome level as the spores with others are hereditary stable, although the homologous index (h) was not significantly different between ordinary DMA and treated DMA. It is necessary to study how to control zebularine treatment with a more stable concentration for chromosome breakage during meiosis.

  • The aim of this research is to infer paleovegetation and paleoclimate in the Hwajeon archeological site of Gwangju city during mid-Holocene through the analysis of pollen, waterlogged woods, and sediments. Between 8200 ~ 6800 years ago, relatively dry climate resulted in a weakened dominance of oak and high diversity of tree genus. During the Holocene climatic optimum period (6800 ~ 5900 years ago), oak forests expanded while wetland areas diminished as warm/humid climate intensified. Between 5900 ~ 4700 years ago, the entire forest area as well as oak climax forests was reduced due to a relatively cool/dry climate. However at the end of this period, oak forests expanded since a favorable climate condition temporarily resumed. Lastly, between 4700 ~ 3300 years ago, oaks dominated but alders were weakened. The density of forest was low because of a relatively dry climate in this period.

  • The degree of compaction of embankments is generally measured using the sand replacement method or a soil density gauge. However, these methods include coarse particles, which are relatively large. The degree of compaction is overestimated if the in-situ soil density is simply compared with the density obtained from a Proctor compaction test (KS F 2312, 2001), because the density of coarse particles is higher than that of soil. However, there is no recommended correction for the coarse particle ratio in Korea, thus intentionally increasing the degree of compaction for structures to which large loads are applied or for which compaction is critical. Here, a correction considering the Korean Proctor compaction test and the difference between the maximum allowable particle sizes was recommended after corrections for coarse particle ratios in other countries were collected and analyzed. The degree of compaction was re-estimated by applying the recommended correction to the results of both Proctor compaction and sand replacement tests. The degree of compaction without the correction of coarse particle ratio was overestimated, because the re-estimated degree of compaction decreased as the coarse particle ratio increased. The relatively accurate results obtained from the field application of the correction will offer long-term cost savings due to reduced maintenance fees during operation.

  • Soil microorganisms including bacteria, fungi, and actinomycetes were seasonally isolated at depths (0.5~2, $10{\pm}1$, $50{\pm}1cm$) of field. The frequency of microbial isolates was employed for the determination of microbial population (CFU/g dry soil) and distribution ratio (%) in soil. Both bacteria (24-fold) and actinomycetes (7-fold) exhibited the biggest change at the depth of $50{\pm}1cm$, whereas fungi showed the maximum (13-fold) at $10{\pm}1cm$. On the whole, the bacterial population was high in spring soil, fungi in winter, and actinomycetes in autumn. Soil microorganisms also exhibited the seasonal variation on their distribution ratio (%). The maximum distribution ratio (85.7%) of bacteria was observed at the depth of $50{\pm}1cm$ in spring, whereas bacteria showed the minimum (35.2%) at the depth of $10{\pm}1cm$ in spring. The maximum distribution ratio (23.0%) of fungi was found at the depth of $50{\pm}1cm$ in spring, whereas its minimum (0.5%) at the depth of $10{\pm}1cm$ in spring. Actinomycetes exhibited the maximum distribution ratio (45.2%) at the depth of $10{\pm}1cm$ in spring, whereas its minimum (12.2%) was showed at the depth of $50{\pm}1cm$ in spring.

  • The aim of this study was to apply the quantitative light-induced fluorescence(QLF) and use of fluorescein-enhanced QLF method for quantitative assessment of early enamel demineralization in vitro, comparing effectiveness of light sources : argon laser, halogen lamp, light emitting diode (LED) and plasma arc lamp. Sixty extracted teeth were used, prepared by gentle pumicing and coating in an acid-resistant nail-varnish, except for an exposure to a demineralizing solution. Teeth were removed at regular intervals (24, 48 and 72hrs), air-dried and QLF image were taken. Mineral loss, as measured by difference of optical density, was recorded. For dyeenhanced QLF a 0.075% sodium fluorescein dye was applied after QLF examination and mineral loss was recorded. The following result were obtained : 1. Comparing with mean difference of optical density, plasma arc lamp was higher than other light sources in all group (p<0.05). 2. Comparing each group with mean difference of optical density, there was significant different using plasma are lamp and halogen lamp. 3. For use of dye-enhanced QLF, comparing with mean difference of optical density, plasma arc lamp were higher than other light sources in all group(p<0.05). 4. With this model dye-enhanced QLF compared with QLF in optical density, dye-enhanced QLF was higher than QLF in optical density. From the results presented in this paper, it was concluded that plasma light source was more effective compared with other light source for the detection and the quantification of early enamel caries.

  • Firing characteristics, transverse rupture strength, and heat capacity were studied of the heat-reservoir refractory materials made of red-firing clay, mill scale, and water glass. The firing shrinkage increased with increase of the clay proportion in samples. The volume of fired bodies showed shrinkage by drying up to $300^{\circ}C$, steady expansion in the 300-$700^{\circ}C$ range due to phase transition of iron oxides. and drastic expansion above $1200^{\circ}C$. Flexural strength decreased from 5.6 Mpa to 2.35 Mpa with the decrease of the ratio of clay to mill scale from 1:1 to 1:3 Heat capacities changed from 1.1 Joul/g$^{\circ}$C to 1.35 Joul/g$^{\circ}$C with the ratio of millscale to clay ratio from 1:1 to 1:3. Mill scale in the specimen appears to exist as liquid phase during firing. Firing the specimens in air leads to the eruption of the molten mill scale to the sample surfaces. Contrarily, firing samples in a refractory sagger with a cover suppressed the eruption of the molten mill scale to the surfaces. The addition of mill scale gave rise to porous sintered bodies which would delay cooling rates of heat-reservoir brick.

  • The field control efficacy of entomopathogenic nematode, Steinernema carpocapsae Weiser, was evaluated on the beet armyworm, Spodoptera exigua (Hiibner). The insect pest has been known to be a defoliator at the aerial part of the crop and difficult to be controlled effectively with most commercial chemical insecticides due to its insecticide resistance. To overcome the susceptibility of the nematodes to desiccation when they were applied by leaf spray in field condition, we screened several commercial antidesiccants (alkyl glucoside, CMC, glycerol, Keltrol-F, Kunipia-G, and Laponite LXG) optimal for survival of the nematodes. Keltrol-F (0.1 %) was selected as a candidate supplement for field application of the nematodes. Leaf spray of the nematodes at 5,000 infective juveniles/ml of distilled water containing 0.1 % Keltrol-F resulted in 87.7% control efficacy on the 3rd instar larvae of Sp. exigua.
