Anticancer Activity of Extracts from Mistletoe Cultivated on Prunus mume (매실나무(Prunus mume) 겨우살이 추출물의 항암활성)

  • 인공재배한 매실나무 겨우살이(PM)의 동결건조시료와 자연산 굴참나무겨우살이(QM)의 열풍건조시료의 80% 에탄올 및 물 초음파추출물을 4종의 세포주(HEK 293, HepG2, AGS, MCF-7)배지에 첨가하여 MTT assay로 농도에 따른 세포생존율을 조사하였다. 시료의 HEK 293(인간신장 정상세포)에 대한 세포 독성은 $100{\mu}g/ml$에서 PM의 80% 에탄올 추출물 및 물 추출물 처리군의 생존율은 각각 $86.30{\pm}2.87%$, $89.27{\pm}0.86%$, QM의 80% 에탄올 추출물 및 물 추출물의 생존율은 각각 $80.76{\pm}1.67%$, $78.07{\pm}0.67%$이었다. HepG2(인간 간암세포)에 대한 항암활성을 측정한 결과 PM과 QM 모두 80% 에탄올 추출물이 물 추출물보다 비교적 높은 항암활성을 나타내었으며 $100{\mu}g/ml$에서 QM 80% 에탄올 추출물이 $57.33{\pm}1.30%$의 생존율을 나타내어 항암활성이 가장 높았고, PM 물 추출물이 $76.45{\pm}2.62%$의 생존율을 나타내어 항암활성이 가장 낮았다. AGS(인간 위암세포)에 대한 독성을 측정한 결과 모든 겨우살이에서 80% 에탄올추출물이 더 높은 독성을 나타내었으며, $100{\mu}g/ml$에서 QM 80% 에탄올 추출물의 생존률이 $60.94{\pm}2.44%$로 비교적 항암활성이 높았고, PM 물 추출물이 $80.10{\pm}1.96%$의 생존율을 나타내어 항암활성이 낮았다. MCF-7(인간 유방암세포)는 $100{\mu}g/ml$에서 QM 80% 에탄올 추출물이 $69.44{\pm}1.56$의 생존율로 비교적 높은 항암활성을 나타내었으며, PM 80% 에탄올 추출물이 $88.30{\pm}4.12%$로 낮은 항암활성을 나타내었다. PM 물 추출물이 $73.23{\pm}3.16$으로 PM 80% 에탄올 추출물보다 비교적 높은 항암활성을 나타내었다. 결론적으로, HepG2(인간 간암세포)와 AGS(인간 위암세포)에 대해서 굴참나무겨우살이 80% 에탄올 추출물의 $100{\mu}g/ml$ 농도가 적합하였고, 매실나무겨우살이는 물 추출물 $100{\mu}g/ml$ 농도에서 MCF-7(인간 유방암세포)에 대한 항암소재로 적합하였다.

Screening of Antibacterial Activity from Hot Water Extracts of Indigenous Plants (자생식물 열수추출액의 항균효능 검색)

  • Hot water extracts of 141 indigenous plants were screened for antibacterial activity against 7 Gram (+) and 12 Gram (-) bacteria by agar diffusion method. Of the 141 plants tested, 55 showed antibacterial activity against one or more species of Gram (+) bacteria and especially extracts of Oenothera stricta Ledebour (Onagraceae), Quercus variabilis Blume (Fagaceae), Alnus firma (Betulaceae), Geranium thunbergii (Geraniaceae), Lagerstroemia indica (Lythraceae) showed broad spectrum antibacterial activity on over 6 strains. Of the 141 plants tested, 45 showed antibacterial activity against one or more species of Gram (-) bacteria and especially extracts of Oenothera stricta Ledebour (Onagraceae), Punica granatum (Lythraceae), Quercus variabilis Blume (Fagaceae), Schizandra chinensis Baillon (Magnoliaceae), Alnus firma (Betulaceae), Alnus hirsuta (Betulaceae), Magnolia kobus (Magnoliaceae) showed broad spectrum antibacterial activity on over 8 strains. The most active antibacterial plants were Oenothera stricta Ledebour (Onagraceae), Punica granatum (Lythraceae), Quercus variabilis Blume (Fagaceae), Schizandra chinensis Baillon (Magnoliaceae) which are edible as permitted by Korea Food & Drug Adminstration. With in-depth research, the hot water extracts of these plants can be applied as food preservatives and alternatives of antimicrobials for livestock feeds.

  • Filtering rates of Crassostrea gigas were experimentally investigated with reference to effects of water temperature and size. Absorptiometric determinations of filtering rates with oysters being fed diatom Chaetoceros calcirtans were carried out in a closed system. Optical density of 675nm in path length 100mm cell used as the indication of food particles absorption was appeared directly In proportion with the concentration of diatom pigment $chlorophyll-\alpha$. In the closed system where $C_0$ is $OD_{675}$ at initial time 0, $C_t$, at time t, and Z is the decreasing coefficient of OD as meaning of instantaneous removal speed, then $C_t=C_0{\cdot} e^{-2t}$, $Z=In(C_t/C_0)/t$. On the assumption that the filtering rate is constant, then removal rate per unit time (d) is $d=-e^{-z}$. If t is used to time unit of hour (hr), the filtering rate (FR) in I/hr is given by $FR=V{\cdot}d=V(1-e^{-z})$, where V is the water volume (I) of the experimental vessel. Filtering rate increased as exponential function with increasing temperature while not over critical limit. The critical temperature for filtering rate was assumed to be between $28^{\circ}C$ and $29^{\circ}C$. And the weight exponent for filtering rate is 0.223. The model formula derived from the results as FR, $Ihr^{-1}$ = $Exp(0.208{\cdot}T-4.324){\cdot} (DW)^{0.223}$ (T<29 $^{\circ}C)$ where T is water temperature $(^{\circ}C

Preparation and Keeping Quality of Intermediate Moisture Food from Oyster and Sea Mussel (굴, 홍합의 중간수분 식품제조 및 저장 안정성에 관한 연구)

  • An attempt was made in this study to investigate the possibility of processing oyster and sea mussel into an intermediate moisture food. To obtain a palatable and instant product, shucked samples were heated in soy sauce for 5 min., heated in seasoning solution for 10 min., and then dried by the method of hot-air blowing for 4-6 hours at $40^{\circ}C$. Optimum seasoning solutions consisted of 2% monosodium glutamate, 3.5% sodium chloride, 15% sorbitol, 2% propylene glycol, 5% glycine and 0.02% rosemary oleoresin in 72.48% water. Judging from water activity, thiobarbituric acid, viable cell count, volatile basic nitrogen, surface color and sensory evaluation, vacuum-packaging method in nylon/PE $(20{\mu}m/40{\mu}m)$ or polyester/viniliden chloride/polypropylene $(12{\mu}m/15{\mu}m/50{\mu}m)$ film bag did hardly damage to the quality of intermediate moisture products, but air-packaging method in polyethylene(0.06m/m) bag did considerable damage to the quality during storage for 60 days in an incubator with 49-51% relative humidity at $30^{\circ}C$.

Tyrosinase Inhibition Effect of extracts from Mistletoe Cultivated on Prunus mume (매실나무(Prunus mume) 겨우살이 추출물의 tyrosinase 저해활성)

  • 인공재배 매실나무겨우살이의 의약 화장품의 소재 및 산업화 가능성을 검토하기 위해 80% 에탄올 및 증류수 초음파추출물의 Tyrosinase 저해활성을 측정하였다. 인공재배한 매실나무겨우살이(PM, 동결건조) 및 자연산 굴참나무겨우살이(QM, 열풍건조, $30^{\circ}C$, 7day)의 80% 에탄올, 증류수 초음파추출물에 대한 tyrosinase 저해활성을 측정한 결과, 물 추출물의 경우 100 ppm의 농도에서 PM과 QM 모두 5분이 경과 하였을 때 PM은 88.37%, QM은 87.69%로 최대치를 나타내었고, 농도와 시간이 증가함에 따라 tyrosinase 저해활성이 감소하는 경향을 보여주었다. PM 80% 에탄올 추출물은 5분이 경과하였을 때 2,500ppm이 92.08%로 최대값을 보여주었고 이후 시간이 지남에 따라 저해활성이 감소되는 경향을 나타내었다. 반면 5,000ppm은 5분이 경과하였을 때 66.38%로 최소값을 나타낸 반면, 20분까지 저해활성이 증가하였고 10분이 경과한 이후로 거의 일정하게 유지되어 지속적으로 감소되는 1,000ppm보다 높은 저해효과를 보여 주었다. QM 80% 에탄올 추출물은 5분이 경과하였을 때 1,000ppm이 88.14%로 가장 높았고 5,000ppm은 63.49%로 가장 낮은 저해활성을 보여주었다. 15분까지 1,000ppm이 가장 높은 저해활성을 보여주었으나 꾸준히 감소한 반면, 2,500ppm의 경우 시간이 경과함에 따라 비교적 일정한 저해활성을 보여주어 15분이 경과한 이후로 2,500ppm의 저해활성이 가장 높게 나타났다. 25분이 경과하였을 때 2,500ppm은 70.27%로 가장 높은 저해활성을 나타내었고, 5,000ppm은 30.09%로 가장 낮은 저해활성을 보여주었다. 본 연구 결과에 따르면, 물 추출물의 경우 겨우살이의 농도가 낮을수록 더 높은 tyrosinase 저해활성을 나타내었고, 80% 에탄올 추출물의 경우 2,500ppm에서 가장 높은 저해활성을 나타내어 의약 화장품의 소재 및 산업화 가능성이 높음을 확인할 수 있었다.

  • Mortality patterns ad changes of vegetation in newly formed gaps were investigated to examine the succession process of Pinus densiflora forest located at Mt. Worak. The main findings from this study were as follows: ⑴ The forest was dominated by P. densiflora and Quercus variabilis in the oversotry, while Q. mongolica, Q. serrata and Fraxinus sieboldiana in the mid-story. ⑵ In all study areas, the standing dead type was the most common factor influencing gap-forming mortality. ⑶ DBH analysis showed that Pinus densiflora community was replaced by Quercus variabilis community first, and then by quercus mongolica and quercus serrata community. ⑷ Seedlings of P. densiflora were discovered only at dry sites of the gap.

Vegetation Structure of the Hyangjeokbong in the Deogyusan National Park (덕유산국립공원 향적봉 일대 식생구조 연구)

  • This study was carried out to classify vegetation structure of the Hyangjeokbong in the Deogyusan National Park using the gradient analysis and phytosociological method. The vegetation was classified into Quercus mongolica community(Rhododendron schlippenbachii subcommunity, typical subcommunity), Q. serrata community, Q. variabillis community, Pinus densiflora community, Cornus controversa community, Fraxinus mandshurica community and Taxus cuspidata community. Ecological characteristics such as species composition, layer structure, vegetation ratio, and the distribution of individual trees by DBH(diameter at breast height) were significantly different among communities. The order of important value of the forest community with DBH 2cm above plants was Q. mongolica(81.2), F. mandshurica, Q. serrata, P. densiflora, Acer pseudosieboldianum, Q. variabillis, Rhododendron schlippenbachii, C. controversa, T. cuspidata. Distribution of DBH of Q. mongolica and Q. serrata had suggesting a continuous domination of these species over the other species for the time being. In contrast, F. mandshurica appeared limited to the valley of the sheet and a higher frequency of young individuals, suggesting a continuous domination of these species the development of a climax forest terrain. P. densiflora and T. cuspidata had suggesting a continuous domination of these species over the other species for the time being in maintain. Q. variabilis and C. controversa had a formality distribution, suggesting a continuous domination of these species over the other species for the time being. This study examined the correlation between each community and the environment according to DCCA ordination. The Q. mongolica community and T. cuspidata community predominated in the highest elevation habitats which had few moisture, Ca and in the low pH. The Q. serrata community mainly occurred in the low elevation habitats which had many moisture. P. densiflora community predominated in the middle elevation and south-facing slope habitats which had few moisture, Ca and in the low pH. F. mandshurica community predominated in the low elevation habitats which had many moisture, Ca and pH.

  • Filtering rates of two farming ascidians Styela clava and S. plicata, and of a farming mussel Mytilus edulis were experimentally investigated with reference to effects of water temperature and size. Optical density (OD) of 440 nm in path length 22 mm cell used as the indication of food particles absorption was appeared directly in proportion with the concentration of neutral red dyes. The filtering rate F is calculated by Kim's equation $F\;=\;V(1-e^{-z})$, where V is the water volume ($\ell$) in the experimental jar, and Z is the decreasing coefficient of OD as meaning of instantaneous removal speed as In $C_t\;=\;In\;C_{o}-Z{\cdot}t$, in this formula $C_t$ is OD at the time t. Filtering rate of S. clava increased as exponential function with increasing temperature while not over critical limit, and the critical temperature for filtering rate was assumed to be between $28^{\circ}C$ and $29^{\circ}C$. In case of S. plicata, the critical temperature was to be below $13^{\circ}C$, and through the temperature range $15\~25^{\circ}C$ appeared a little difference in level even though with significant. M. edulis was not appear any significant effects by water temperature less than $29^{\circ}C$. Development of Protocol for the Effective Detection of Feline Calicivirus as Norovirus Surrogate in Oyster and Lettuce (굴과 상추에서 노로바이러스의 대체모델 feline calicivirus의 효율적 검출법 개발)

  • Foodborne illness caused by Noroviruses (NVs) is increasing rapidly in Korea. This study developed an effective detection protocol for NVs found in contaminated oysters and lettuce through an investigation using the major steps of virus particle separation, concentration and RT-PCR. As a surrogate model for NVs, the cultivable feline calicivirus (FCV) that belongs to the same Caliciviridae family was used. Instead of using a time-consuming ultracentrifugation method, efficient methods based on solvent extraction and PEG precipitation procedure were applied. Direct homogenization of a 25g sample of whole oyster and lettuce in 175mL PBS provided the simplicity that would be needed in the actual field of food product examination. The overnight PEG precipitation step at $4^{\circ}C$ was reduced to 3 h by placing the reaction tube in ice and by adjusting the PEG concentrations. The application of the use of chloroform and 0.2 ${\mu}m$ syringe filtration together showed a better detection efficiency than the use of chloroform alone in removing PCR inhibitors for both oyster and lettuce samples. Also, dilution of the extracted RNA solution before PCR provided increased sensitivity. The improved detection protocol developed in this study could be efficiently applied to detect FCV and most likely NVs from oysters and lettuce.

Forest Vegetation Classification and Species Composition of Mt. Ilwol, Yeongyang-Gun, Korea (일월산 산림식생의 종구성적 특성)

  • Forest vegetation classification and species composition of Mt. Ilwol, Yeongyang-Gun, Korea, were studied combining the Braun-Blanquet approach with numerical syntaxonomical analyses (TWINSPAN). Vegetation types and various ecological characteristics such as flora, constancy classes, species ratio of life-form, species diversity and importance value were analyzed. Sixty-eight samples were taken from a $100m^2$ square plot each. Forest communities were identified as two great types: arid landform of mountainside (AM) and humid fertility of piedmont and valley (HP). The former was divided into 3 communities (Rhododendron mucronulatum, Quercus variabilis, Hosta capitat community) and 2groups, and the latter into 3 communities (Tilia amurensis, Vitis coignetiae, Philadelphus schrenckii community) and 2 groups. Vegetation was classified into 8 units. Floristically, the most represented family was Compositae with 26 species. Species with percentage constance degree of more than 61% was Quercus mongolica (72.1%, IV); Carex siderosticat (III) and Fraxinus rhynchophylla (III) were 50.0 and 41.1%, respectively. Life-forms species ratios for trees, subtrees, shrub, vines, grominoids, forbs and ferns were 18.5, 5.7, 14.9, 6.6, 8.8, 42.4 and 3.1%, respectively, PH type showed from $1.70{\pm}0.50\;to\;1.97{\pm}0.57$ and AM type was from $1.40{\pm}0.18\;to\;1.62{\pm}0.20$ in species diversity; therefore, the former type showed higher species diversity than the latter, According to importance value analysis, Pinus densiflora, Quercus mongolica and Q. variabilis were higher in the tree layer, Q. mongolica in the subtree layer, Fraxinus sieboldiana, R. schlippenbachii, etc. in the shrub layer and Carex siderosticta, Carex humilis, etc. in the herb layer.
