중금속 스트레스에 대한 수생식물 5종의 엽록소형광 반응

  • 중금속에 대한 식물의 내성여부와 중금속 스트레스하에서 식물의 생리적 특성을 파악하기 위해 좀개구리밥, 생이가래, 은행이끼, 수련, 부들 등 5종의 식물을 대상으로 카드뮴(Cd$^{2+}$)의 농도를 달리하여 처리한 후 시간별로 식물의 광합성효율을 분석하였다. 그 결과 좀개구리밥는 카드뮴에 가장 민감하게, 생이가래와 은행이끼 등은 조금 민감하게 반응하였으며, 수련 등은 다른 식물종에 비해 덜 민감한 것으로 나타나고 있다. 좀개구리밥은 카드뮴 농도가 증가할수록 정상적인 O-J-I-P 곡선에서 J, I, P의 값이 큰 폭으로 떨어지고 있으며, 이는 카드뮴 농도가 증가할수록 명반응 중심이 손상을 입거나 어떤 원인에 의해서 불활성됨으로써 광합성 효율 즉 Fv/Fm의 감소를 일으키기 때문이다.

A Study on Nitrogen Metabolism of Lemnaceae: Assimilation of Nitrate and Ammonia in Spirodela polyrhiza and Lemna aequinoctialis (개구리밥과 식물의 질소대사에 관한 연구: 개구리밥(Spirodela polyrhiza)과 좀개구리밥(Lemna aequinoctialis)의 NO3-와 NH4-의 동화작용)

  • Spirodela polyrhiza and Lemna aequinoctialis often occurred at the sites of high ammonium concentration and at the sites of high nitrate concentration, respectively. We investigated the different distribution between two species in relation to the type of nitrogen sources and their concentrations. Our experiments showed that L. aequinoctialis grew faster than S. polyrhiza in nitrate media with lower than 15 mM concentration. The nitrate uptake was also faster in L. aequinoctialis than in S. polyrhiza. However, neither differences in growth nor in uptake patterns between these two species were observed in ammonium media. Glutamine synthetase (GS), glutamate dehydrogenase (GDH) and glutamate synthetase (GOGAT) activities were higher in L. aequinoctialis. In particular, nitrate reductase activity (NRA) in L. aequinoctialis was 12.1 times as high as that in S. polyrhiza. These results showed that the two species responded varyingly to the types of nitrogen sources and their concentrations. Therefore, the difference in geographic distribution between the two species appeared to reflect the interspecific differences in enzyme activities and, subsequently, nitrogen absorption abilities.

Steroidogenic Shift and Acquisition of Meiotic competence by Cultured Ovarian follicles of Rana at Breeding Season (번식기에 개구리 여포의 스테로이드 생성변화와 난자의 성숙능 획득)

  • 3 종의 한국산개구리를 사용하여 여포들이 생성하는 스테로이드와 난자의 성숙능이 번식기 근처에서 어떻게 변하는가를 조사하였다. 개구리의 여포를 생체외 배양하면서 이들이 뇌하수체추출물(frog pituitary homogenate, FPH) 혹은 CAMP의 자극으로 생성하는 progesterone(Pa)과 testosterone(T)의 양을 측정하여 이로부터 P4yT의 비율을 구하였으며 여포난자들이 FPH. P4 혹은 12-0-tetrsdecanoyl phorbol-13-acetate(TPA)에 반응하여 성숙을 일으키는 시기를 조사하였다. 북방산개구리의 여포난자는 11월 부터 호르몬에 반응하여 성숙을 일으켰으며 2월에 PJT의 비율이 급격히 증가하였다 참개구리는 5월에 가서of PJT의 비율이 현저히 증가하고 FPH에 반응하여 성숙을 일으켰다. 그러나 P4에 의한 난자성숙은 3월부터 시작하였다 옴개구리는 7월에 P4/T 비율의 증가가 있었으며 이때부터 또한 TPA에 반응하여 난자의 성숙을 일으켰다 따라서 3종의 개구리에서 모두 각각의 번식기 근처에서 PJT의 비가 급격히 증가하고 또한 성숙능이 생긴다는 것을 알 수있었다.

Electrophoresis of the Hemoglobins and the Serum Proteins of Korean Anuran (개구리目 혈색소와 혈청단백질의 전기영동)

  • Serum protein and hemoglobin patterns were obtained by cellulose acetate electrophoresis for several anurans. Differences were found both in the number of components from individual animal hemoglobins and in the mobility of these components. Under the conditions employed, toad and R. plancyichosenica had a single component. Other frogs showed 2-component pattern. Sera of all anurans examined contain albumin but no preablumin. Relatively few assayable proteins are found in serum from B. buro gragraizans and differ thereby from most serum patterns examined for frogs, which show complex serum protein bands. Different species have dissimilar patterns although some of bands are apparently homologous between species.

A Taxonomic Reconsideration of the Genus Lemna L. (Lemnaceae) in Korea (한국산 좀개구리밥속(개구리밥과)의 분류학적 실체에 대한 재고)

  • Duckweed family (Lemnaceae Martinov), including the genus Lemna L., is a typical floating aquatic perennial plant, and about five genera and 40 species in the family are in wide distribution around the world except the polar regions. The genus Lemna is the smallest and the simplest plant among the angiosperms. It has a characteristic of doubling every three days with fast vegetative propagation, which helps the organisms to increase in rapid growth. As such, the plant is ideal for environmental pollution assessment and toxicity test. Although taxonomists and scholars have used different scientific names for the species, many of them have agreed that there is only one member of species of the genus Lemna in Korea. Paying attention to the external morphological variation observed in the Korean genus Lemna, we conducted a molecular phylogenetic analysis to identify the entity of the Korean Lemna species and to investigate the possibility of two or more members of the species existing in Korea. We determined and aligned the DNA sequences of the atpF-H region of the chloroplast DNA in 37 populations of the nationally distributed Lemna species. The results showed that the sequence length of the cp DNA atpF-H region was 463-483 bp, the length of the aligned sequences was 488 bp, and the number of variation site in nucleotide sequences was 47. There were two types of aligned sequences of the cp DNA atpF-H region from 37 populations of Lemna species in Korea. The maximum parsimony analysis revealed that the Korean Lemna consists of two clades, and one of them had two subclades. The results suggest that, contrary to the general understanding, at least two taxa (L.aequinoctialis, L.minor) exist in Korea.

Intra-, Inter-specific Variation of Korean Rana (Amphibia: Ranidae) Based on the Partial Sequence of Mitochondrial 16S rDNA (미토콘드리아 16S rDNA부분 염기서열을 이용한 한국산 개구리 속(Amphibia: Ranidae)의 종간, 종내 변이에 대한 연구)

  • In order to clarify intra-and inter-specific variation of Korean Rana species, the partial DNA sequences of mitochondrial 16S rDNA gene were determined from 6 Korean and 1 Japanese Rana species, DNA sequences from Korean and Japanese species were comparison-analysed within, and also with the sequences from three species of Japanese brown frogs. DNA similarities were calculated as 91.3∼97.3% among brown frog (R. amurensis coreana, R. dybowskii and R. huanrenensis), as 96.11∼97.26% among pond frogs (R. nigromaculata and R. planeyi chosenica). Genetic distance of pond frog and wrinkle fyog (R. rugosa) were near than that of pond frog and brown frog. Two clusters were formed brown frogs and the other group by neigh-bor-joining and maximum-likelihood analysis, also the populations of R. nigromaculata were well distinguished between Korean peninsula and Korean island. But result from maximum-likelihood analysis slightly differed from neighbor-joining to cluster of R. rugosa. Further analyses for their population will be necessary to study the phylogenetic status.

Features of Plastids within Reduced Spirodela polyrhiza (축소된 개구리밥 식물체 내 색소체 특성)

  • Reduced plants of Spirodela polyrhiza consisting only of fronds, stalks and roots form turions during dormancy. In development, mature fronds produce offspring fronds by vegetative reproduction, and turions arise laterally from the mother frond before dormancy. The turion primordium is derived from the frond, while the frond primordium forms within the turion tissue. In the present study, cellular features, especially those of the plastids, of the above four tissue types have been examined and compared using electron microscopy. Proplastids, found to be numerous in the frond and turion primordia, differentiated into chloroplasts rapidly upon growth. The proplastids were small and the thylakoidal membrane system was rudimentary, howerver the chloroplasts exhibited variation by cell type. Chloroplasts were found within cells of the frond, stalk and root tissue. The thylakoidal membrane system, which formed grana stacks, was moderately developed within frond chloroplasts, while only a few were present in those of the stalk and root cortical cells. One to two starch grains were accumulated within frond chloroplasts, but little to none were found in stalk and root cortical chloroplasts. Contrary to other types of root chloroplasts, those found in the root cap cells developed chloroplasts similar to the frond type. Unlike proplastids of the turion primordia, numerous large amyloplasts occupied most of the turion cell volume. Moreover, the turion cell produced quite large starch grain (s) within the amyloplasts. Accumulation of the starch grains continued until they occupied the most of the stroma and in some cases, individual starch grains reached up to $9.0{\mu}m$ in length. None to little, if any, thylakoidal or internal membranous systems were seldom detected in these amyloplasts. Although the degree of cellular and tissue differentiation was rather minimal within their reduced body, the functional differentiation of Spirodela polyrhiza was very efficient, as is the case in other advanced species.

Monitoring Extensive Breeding Populations and Daily Call Activity of the Gold-spotted Pond Frog, Rana chosenica in Chungju City and Chungwon Gun (청주시와 청원군에서 금개구리 (Rana chosenica) 번식 군집 모니터링 및 일중 울음소리 빈도)

  • To investigate the spatial distribution and daily calling pattern of the Gold-spotted pond frog, Rana chosenica, in Chungju city and Chungwon gun, Chungbuk province, Korea, we divided the study area into 226 plots with a $2{\times}2km^2$ plot on the map, of which we assigned 32 plots for monitoring populations. Call monitoring on whether the species are present or not were conducted for 5 minutes in between sunset and the midnight from at the end of May to July in 2006. Gold-spotted pond frogs were detected at least once four out of 32 plots. Using program Presence, we obtained site occupancy rate as 0.170 which was quite low compared with other species, while detection probability was 0.66 that propose at least three times to visit the monitoring site to confirm the absence of the frogs. The frogs were actively calling from 21:00 to 02:00, and the number of calling male was significantly and highly correlated with water temperature and humidity. This study shows the present status of the Gold-spotted pond frogs in Chungju city and Chungwon gun and we suggests various effective monitoring methods based on the this study.

붕어 및 횡소개구리 체내의 Organochlorine pesticides의 분석

  • 유기염소계농약류 25종을 분석한 결과 HCHs(4종, endosulfan(2종), octachlorostyrene은 모든 시료에서 검출되지 않았으며 trans-nonachlor, heptachlor는 각각 한 지점의 붕어에서 검출되었고 나머지는 검출되지 않았다. Chlordane의 경우 두 이성질체가 한 지점의 붕어에서 검출되었고, DDT는 6개 이성질체 중 p,p’-DDT와 p,p’-DDD가 한 지점의 붕어에서 검출되었으며 o,p’-DDT는 한 지점의 황소개구리에서 검출되었다. 그 외의 이성질체는 모든 시료에서 검출되지 않았다.
