Preparation and Characterization of Poly(vinyl alcohol) Hydrogel Contain Metronidazole by Irradiation (감마선을 이용한 Metronidazole이 함유된 Poly(vinyl alcohol) 하이드로겔 제조 및 특성)

  • Periodontitis is disease of damaged gum tissue that is not removed the plaque onto teeth. In case that the symptoms of disease get pain worse, it will have to extract tooth because of tumefy or bleeding at gums so treatment of drug was required to periodontitis. In this study, the hydrogel was prepared by including superior viscous, excellent elastic, and biocompatibility of Poly(vinyl alcohol, PVA) and antimicrobial drug of Metronidazole (MD). The 15 wt% PVA was dissolved in deionized water and then prepared PVA solution was irradiated using gamma-ray at 25 kGy ($10kGy\;hr^{-1}$). In addition, PVA hydrogel was immersed in each 0.1, 0.25 and 0.5 wt% MD solution using stirrer for 24 hr. The result of the gelation, 0.5 wt% MD loaded PVA hydrogel(76%) was lower than PVA hydrogel (88.2%). The swelling ration of 0.5 wt% MD loaded PVA hydrogel (294.8%) was higher than PVA hydrogel (105.2%). The compressive strength and thermal properties of MD loaded PVA hydrogel was gradually lower. The drug release test of 0.5 wt% MD loaded PVA hydrogel (61%) was higher than 0.1 wt% MD loaded PVA hydrogel (12%). Therefore, MD loaded PVA hygrogel may be a promising tool for periodontitis medicine by gamma-ray.

  • This study was investigated the effect of tumor cell cytotoxicity of gamma irradiated Chaga mushroom extract (CME). CME was prepared by hot water extraction at $70^{\circ}C$ for 4 hours and lyophilized. $Ten\;mg\;ml^{-1}$ of lyophilized CME powder was dissolved with deionized water and then irradiated at the doses of 10, 50, 100, and 150kGy by cobalt 60 gamma irradiator. The gamma-irradiated and non-irradiated CME were treated into the cancer cell, including human stomach cancer and human colon cancer. Cytotoxicity against the cancer cell was increased in gamma-irradiated CME and antioxidant activity was also increased in gamma-irradiated CME, as irradiation dose increased. Therefore, it was considered that gamma irradiation was effective method for improvement of the cancer cell cytotoxicity and antioxidant activity of Chaga mushroom extract.

Genotoxicological Safety of Undaria pinnatifida Gamma-irradiated after Harvest (감마선 조사된 채취 미역의 유전독성학적 안전성 평가)

  • This study was conducted to evaluate the genotoxcity of gamma-irradiated Undaria pinnadifida after harvest for enhancing the storage and microbial safety during drying process. The harvested seaweed was irradiated with the doses of 1 and 10 kGy. The genotoxicity was evaluated by Ames test (Salmonella typhimurium reversion assay) and SOS chromotest. The results from all samples were negative in the bacterial reversion assay with S. typhimurium TA98 and TA100. No mutagenicity was detected in the assay, at both with and without metabolic activation. The SOS chromotest also indicated that the gamma-irradiated U. pinnadifida did not show any mutagenicity. Therefore, this study indicated that gamma irradiation could be used for extending the shelf-life of U. pinnadifida.

  • Cooking drips which were obtained as by-product after seafood processing in the food industries, still contain lots of proteins, carbohydrates, and other functional materials. This study was conducted to investigate the effect of gamma irradiation on the biological activities of seafood cooking drips. When the cooking drips of Hizikia fusiformis, Enteroctopus dofleini and Thunnus thynnus were irradiated, the antioxidant activities, whitening effect, and angiotensin I converting enzyme inhibition activity of the ethanol extract from seafood cooking drips were all increased by gamma irradiation. This was because of the increased extraction efficiency of available compounds by irradiation. These results suggested that the seafood cooking drips, wasted as by-products, can be used as functional compounds with gamma irradiation treatment.

Preparation of Polyethylene Foam Sheets Crosslinked by Gamma-ray Irradiation (감마선 조사에 의하여 가교된 폴리에틸렌 발포 시트의 제조)

  • In this study, crosslinked polyethylene (PE) foam sheets were prepared through gamma-ray irradiation. PE foam sheets were prepared through a foaming process using sodium bicarbonate as a blowing and foaming agent. The prepared PE foam sheets were then crosslinked through gamma-ray irradiation. The crosslinking degree was increased to 86.0% with an increase in the absorption dose. The tensile strength of the crosslinked PE foam sheets was increased with an increase in the absorption dose. However, the elongation-at-break of the crosslinked PE foam sheets was decreased. The thermal decomposition temperature of the crosslinked PE foam sheets was increased to $421.2^{\circ}C$ with an increase in the absorption dose. The SEM analysis revealed that the morphology was not changed significantly after the crosslinking through gamma-ray irradiation.

Quality Evaluation of Gamma-Irradiated Tarakjuk Powder, Korean Milk Porridge (감마선 조사에 따른 분말 타락죽의 품질 평가)

  • This study was conducted to investigate bacterial growth, viscosity, color, and sensory properties of gamma-irradiated Tarakjuk powder, a Korean milk porridge powder, at 1, 3, 5, 7, and 10 kGy. The total aerobic bacteria in non-irradiated Tarakjuk powder was $2.56{\log}\;CFU{\cdot}g^{-1}$, whereas it was not observed within the detection limit of $2{\log}\;CFU{\cdot}g^{-1}$ in samples irradiated at more than 1 kGy. Spore-forming bacteria, however, were not observed in all samples within the detection limit of $1{\log}\;CFU{\cdot}g^{-1}$. The viscosity of rehydrated Tarakjuk after gamma irradiation significantly decreased from 16,770 cP to 4,060 cP when irradiated at 10 kGy. The redness ($a^*$ value) and yellowness ($b^*$ value) evaluated using a colorimeter were significantly increased according to the increase in irradiation dose (p<0.05), while there was no difference in color evaluation conducted by panels. The overall acceptance decreased as the irradiation dose increased, and the 5 kGy sample was 4.0 (normal) on a 7-point scale. As a result, it is considered that a gamma irradiation of 5 kGy is enough to sterilize Tarakjuk powder with a acceptable sensory quality.

Genetic Variation Analysis of Arabidopsis (Arabidopsis thaliana L.) Plants Induced by Acute and Chronic Gamma Irradiation (감마선 완·급조사에 따른 애기장대의 유전적 유연관계 분석)

  • In order to identify the genetic relationship analysis by acute and chronic gamma irradiation, Arabidopsis (Arabidopsis thaliana L.) were irradiated with 200 Gy of gamma-rays using gamma-irradiator (3,000 Ci; Nordion, Canada) and gamma-phytotron (400 Ci; Nordion, Canada) for acute and chronic irradiation, respectively. Genetic relationship among two acute gamma-irradiated plants (A1 and A24) and three chronic gamma-irradiated plants (C1W, C2W, C3W) were analyzed using the amplified fragment length polymorphism (AFLP) technique compared with each non-irradiated plant. A total of 28 EcoRI and MseI primer combinations were used to screen 8 treatments by the ABI3130 capillary electrophoresis system. Amplified products by 28 primer sets showed 1,679 bands with an average of 51 bands per primer combination. Out of the total bands scored, 1,164 fragments were polymorphic bands, with different alleles existing among the treatments. The cluster analysis was performed using the UPGMA (Unweighted Pair Group Method using Arithmetic) in the computer program NTSYS-pc. In clustery analysis, acute gamma-irradiation showed higher genetic variation compared with chronic gamma-irradiation.

Sensory Evaluation of Korean Seaweed Soup, Miyukgook, with Gamma-irradiated Undaria pinnatifida (감마선 조사된 미역으로 제조한 미역국의 관능 연구)

  • This study was conducted to evaluated the sensory properties of Miyukgook, Korean seaweed soup, with gamma-irradiated Undaria pinnadifida for enhancing the storage of the harvested seaweed. The harvested seaweed was irradiated at the doses of 1, 3, 5, and 10 kGy. The Miyukgook was made by following the general recipe. The values of pH and acidity of the Miyukgook were not changed by the irradiation of harvested Undaria. The sensory evaluation result showed that the preference scores in all the sensory properties was a little decreased when it was irradiated, but sensory score of less than 5 kGy samples was similar in all terms. Therefore, it was considered that gamma irradiation with less than 5 kGy was effective for improvement of the storage stability of U. pinnatifida without the changes in the sensory of its major cooked food, Miyukgook.

  • Acetylsalicylic acid (ASA) has been issued recently in contaminated water environments because of potential impacts on ecosystem and public health. This study was aimed at investigating the possibility of ASA degradation using gamma ray irradiation. In addition, the use of sodium persulfate, hydrogen peroxide, ferrous sulfate were tested in order to examine a synergistic effect with gamma ray. The absorbed dose was ranged from 0.2 to 10 kGy and the concentration of oxidants were from 0.1 to 10 mM in this study. The concentration of ASA was gradually decreased corresponding to the increase of the absorbed dose. When soudium persulfate was simultaneously applied, most of the parent compound was completely degraded even at a low dose of 0.8 kGy. The removal efficiency of total organic carbon was 90% even at the highest dose of 10 kGy without sodium persulfate. However, the efficiency was dramatically enhanced up to 98% at the same dose by adding 10 mM of oxidants. It was suggested that hydroxyl radical ($OH{\cdot}$) and sulfate radical ($SO{_4}^-{\cdot}$) were formed in the system and made roles in degrading ASA at the same time.

Selection of Male-sterile and Dwarfism Genetically Modified Zoysia japonica through Gamma Irradiation (감마선 처리에 의한 웅성불임 및 왜성형질의 유전자변형 들잔디(Zoysia japonica Steud.) 선발)

  • The aim of this study is selection of the male-sterile plant for inhibiting transgene flow through gamma-irradiation ($^{60}Co$) at the pollination and fertilization cycle of herbicide-tolerant genetically modified (GM) zoysiagrass (Zoysia japonica Steud.). High frequencies of plant mutations were obtained about 18% from $M_1$ generation at the doses (10 to 50 Gy). We also found that some $M_1$ plants showed male-sterile plants using de-husked seeds and comparison of stainable pollen using $KI-I_2$ solution. Besides the effects of irradiation on pollination and fertilization cycle, various other mutations like dwarf, cold tolerance, increasing grains and mass were observed. Four of dwarfism plants were selected through comparison of morphological characteristic between control and mutants during 4 years. These results demonstrated that the gamma-irradiation on pollination and fertilization cycle is very effective to induce the various mutations, and the male-sterile mutants are useful for controlling transgene flow and developing of high quality turfgasses.
