현미의 방사선 처리여부 판별을 위한 점도측정법의 검증 (Verification of Viscosity Measurement for Identifying Irradiated Brown Rice)

  • 국산 및 중국산 현미를 대상으로 방사선 조사여부 판별방법의 하나인 점도측정법 검증을 위하여 0-15 kGy 범위의 감마선을 조사하고 점도, 전분함량 및 specific parameter 측정에 의한 미지시료의 조사여부 판별을 시도하였다. 현미의 전분함량은 국산과 중국산 각각 71.52와 64.58%이었고, 국산 현미가 중국산 보다 상대적으로 높은 점도를 나타내었다. 방사선 조사된 국산 및 중국산 현미의 조사선량에 따른 점도 및 specific parameter는 유의적으로 감소하였으며, 전단속도(50, 150, 300 rpm)에 영향을 받아 낮은 속도에서 조사선량의 영향이 크게 나타났다. 점도와 specific parameter의 변화에 대한 조사선량의 상관성을 나타낸 결정계수($R^2$)는 국산 현미는 0.9423-0.9567, 중국산 현미는 0.9119-0.9387을 보였다. 또한 상기 점도측정법에 의한 미지시료(n=30)의 조사여부 판별시험에서는 90%의 정확도를 보였다. 이로써 전분질 식품의 방사선 조사여부 확인에서 점도측정법은 screening 방법의 하나로써 보조적 역할의 가능성을 보여주었다.

광자극발광법(PSL)에 의한 방사선 조사 건조향신료의 검지 특성 (Analysis of the Detection Characteristics of Irradiated Dried Spices and Herbs by Photostimulated Luminescence (PSL))

  • 최근 방사선 조사는 식품의 발아억제, 살충, 살균 및 숙도조절을 위한 화학약품 처리 등의 대체방안 뿐만 아니라 식품의 저장 및 가공기술로서 인정받고 있으며, 우리나라를 비롯한 약 40개국에서 상업적인 조사식품을 생산하고 있어 이에 대한 정확한 정보를 제공하기 위한 신뢰성 있는 검지기술이 요구되고 있다. 방사선 조사식품 검지법 대상 식품 중 건조향신료 19품목에 방사선 조사 최대 허용선량인 10 kGy까지 $^{60}Co$ 감마선을 단계별로 조사하고 이를 광자극발광법(Photostimulated Luminescence, PSL)을 이용하여 검지특성을 확인하였다. PSL 측정 결과, 건조향신료 19품목 중 14품목은 1 kGy 조사된 시료에서, 1 품목은 5 kGy에서 조사시료(positive)의 threshold value인 5000 이상의 photon count/60 sec로 측정되어 방사선 조사여부의 확인이 가능하였다. 하지만 후추류, 육두구, 계피의 경우는 특이적으로 PSL 측정에 낮은 감도를 보이거나 미네랄 함량이 낮아서 최대 허용선량인 10 kGy 조사시료에서조차 방사선 조사여부의 확인이 불가능하였으며, 이 품목의 경우 열발광분석법(Thermoluminescence, TL) 등을 이용한 방사선 조사여부의 판정이 필요하였다. 또한 식물의 잎을 이용하는 향신료는 비조사 시료와 1 kGy 및 10 kGy 조사시료를 혼합하였을 때, 1 kGy 선량으로 조사된 시료가 1% 혼합된 시료에서도 방사선 조사여부의 확인이 가능하였다.

감마선 조사된 감자분말 첨가가 요구르트의 발효특성에 미치는 영향 (The Effects of Freeze-Dried Potato Flour Addition on the Fermentation Characteristics of Yogurt)

  • 우유에 감자분말을 첨가하여 2종의 Lactobacillus bulgaricus, Streptococus thermophillus 젖산균을 혼합하여 배양할 경우 젖산발효가 어떤 메카니즘에 의해 촉진되며 요구르트의 품질에 미치는 영향을 조사코자 젖산균의 산생성 능력과, 요구르트의 점도, 유리아미노산 및 관능검사를 하였다. 0.5%(w/w) 감자분말을 첨가하여 배양한 젖산발효의 경우 무 첨가군에 비하여 산도는 높았고, pH는 낮았으며 점도는 증가되었다. Streptococus thermophillus와 Lactobacillus bulgaricus를 혼합배양한 경우 산의 생성은 가장 높았으며, 각각의 균을 단독 배양 시에는 S. thermophillus가 L. bulgaricus보다 산 생성도가 높았다. 감자분말을 첨가하여 각각의 균을 배양한 경우 L. bulgaricus와 S. thermophiilus 균주에서 모두 lysine의 이용률은 높았으며, L. bulgaricus는 glutamic acid를 다량 생성한 반면, S. thermophillus는 glutamic acid를 소모하였다. 감자분말을 첨가한 요구르트 관능평가에서 조직감은 향상되었고, 색, 맛, 후미 및 전체적인 기호도는 다소 저하되었다. 이상의 결과로부터 L. bulgaricus는 S. thermophillus 균주가 요구하는 유리 glutamic acid을 공급함으로써 S. thermophiilus의 생육이 촉진되어 감자분말 첨가 요구르트의 발효기간을 단축시킨 것으로 사료된다.

방사선(放射線)에 의(依)한 밤나무의 돌연변이(突然變異) 육종(育種)에 관(關)한 연구(硏究) (Studies on the Mutation Breeding in Castanea SP.)

  • 밤나무와 약밤나무의 종자(種子), 접수(接穗) 및 생체(生體)에 감마선(線)을 총선량별(總線量別), 총량률별(總量率別)로 조사(照射)하여 방사선(放射線)의 생물학적(生物學的) 효과(効果) 및 돌연변이(突然變異)의 유기(誘起)를 조사(調査)한 결과(結果)를 다음과 같이 요약(要約)한다. 1. 밤나무의 ${\gamma}$선조사(線照射)에 의(依)한 종자발아율(種子發芽率) 반감선량(半減線量)은 4.6 kR 인데 접목활착율(接木活着率) 반감선량(半減線量)은 3.1 kR으로 더욱 높은 감수성(感受性)을 보였다. 2. 종자조사(種子照射) 5 kR구(區)에서 낮은 빈도(頻度)(0.84%로 albino 개체(個體)가 나타났으며 선량(線量)의 증가(增加)에 따라 묘장(苗長)이 유의성(有意性)있게 감소(減少)하였다. 3. 접수조사(接穗照射)에 의(依)한 접목활착율(接木活着率) 반감선량(半減線量)은 급조사(急照射)에서 약밤나무 및 밤나무가 3.2 kR 및 3.1 kR인데 반급조사(半急照射)에서는 10.2 kR으로서 3배(倍)의 높은 누적선량(累積線量)의 조사(照射)가 가능(可能)하였다. 4. 약밤나무의 휴면기(休眠期) 생체(生體) 완조사(緩照射)의 총선량(總線量) 7.5-4.9 kR, 선량율(線量率) 10-98 R/day구(區)에서 대형엽아조변이(大形葉芽條變異)가 높은 빈도(頻度)(16.6%)로 나타났으며 평균엽면적(平均葉面積) $96.36cm^2$로서 정상엽(正常葉) $26.28cm^2$보다 매우 컸다.

감마선 조사가 렌틸(Lens culinaris L.)의 감수성에 미치는 영향 (Radiosensitivity of Lentil Bean (Lens culinaris L.) to Gamma-irradiation)

  • We examined damages from gamma-irradiaion and determined the optimal gamma-ray dose for mutation breeding in lentil (Lens culinaris L.) bean. Four individual lines (L-C, L-2, L-8 and L-9), that have remarkable adaptability in South Korea were gamma-irradiated at doses of 50, 70, 100, 200, 300, 400, and 500 Gy. The germination rate of seed decreased as the dose increased over 50 Gy in all lines. However, $LD_{50}$ and $RD_{50}$ were different among lines. The median lethal doses($LD_{50}$) were approximately 127 (L-C), 74 (L-2), 95 (L-8), and 144 (L-9) Gy. The median reduction doses($RD_{50}$) for plant height, number of leaves, root length, and flash weight were 156, 176, 150, and 180 Gy for L-C, 253, 198, 127, and 142 Gy for L-2, 188, 175, 200, and 190 Gy for L-8, and 162, 210, 224, and 184 for L-9, respectively. The growth characteristics of the $M_1$ generation decreased as the dose increased over 70 Gy. The optimal doses of gamma irradiation for mutation breeding of lentil were determined to be 70 Gy (L-2, L-8) and 100 Gy (L-C, L-9). We performed the comet assay to observe nuclear DNA damage induced by gamma-irradiation. In comet assay, a clear difference was identified over 100 Gy treatments. With increasing doses of gamma-ray in the range of 50 to 500 Gy, the rate of head DNA was decreased significantly from 97.5% to 81.6%. Tail length was consecutively increased from $1.9{\mu}m$ to $17.4{\mu}m$. Our result provides basic information for construction of mutant pools in lentils.

감마선 조사에 따른 동결건조과일의 품질 특성 평가 (Evaluation of Quality Properties of Gamma-irradiated Freeze-dried Fruits)

  • The quality properties of freeze-dried apples, pears, strawberries and pineapples gamma-irradiated at 0, 1, 2, 3, 4, and 5 kGy were evaluated to develop germ-free products for immuno-compromised patients. The initial count of total aerobic bacteria in non-irradiated apples, pears, strawberries and pineapple was 2.5, 3.1, 2.6, and $3.2{\log}\;CFU{\cdot}g^{-1}$, respectively. Microorganisms were not observed in apples after 1 kGy, in pears and strawberries after 4 kGy, and in pineapples at 5 kGy within a detection limit of $10{\log}\;CFU{\cdot}g^{-1}$. In addition, the sterilization of each sample was confirmed at the same dose. The score for the overall acceptance of freeze-dried fruit irradiated at a sterilization dose was 5.5 for apples, 4.1 for pineapples, and 4.0 for the other fruits, whereas that of non-irradiated control sample was 5.6 for apples, 5.2 for pears, and 5.8 for strawberries and pineapples with a 7-point scale. As a result, gamma irradiation of 1 kGy for apples, 4 kGy for strawberries and pears, and 5 kGy for pineapples is sufficient to sterilize each freeze-dried fruit with acceptable sensory properties.

감마선 조사가 열처리 겨우살이의 항당뇨 및 세포독성에 미치는 영향 (Effects of Gamma-irradiation on Anti-diabetic and Cytotoxic Activities of Heat-treated Mistletoe (Viscum album))

  • Mistletoe (Viscum album) has been widely used as a functional food material for various therapeutic purposes from ancient time. In this study, we examined anti-diabetic and cytotoxic activities of heated-treated mistletoe and the effects of gamma-irradiation on its activities. Heat-treated mistletoe extract was prepared by heating during different time (3, 6, 9 and 12 h) and gamma-irradiated with different doses of 0, 10, 30, 50, 70 and 100 kGy. Heat-treated mistletoe extracts showed a concentration-dependent cytotoxicity on rat insulinoma RINm5F cells and the effect was gradually decreased as heating time increased up to 12 h. 12 h heat-treated extract was no cytotoxic. Gamma-irradiation enhanced the reduction of heat-treated mistletoe-induced cytotoxicity and the decreasing effect was an irradiating dose-dependent. In particular, all of 70 kGy irradiated and heat-treated mistletoe extracts did not showed the cytotoxicity and the effect was comparable to 12 h heat-treated mistletoe extract. Among those extracts, 3 h heat-treated mistletoe extract gradually increased the insulin secreting activity by gamma-irradiation and the effect was the best at 70 kGy, whereas 12 heat-treated extract was no effect. On the test of ${\alpha}$-glucosidase inhibitory activity, 3 h heat-treated mistletoe extract showed the concentration dependent effects and gamma-irradiation induced more activity at 70 kGy, compared to non-irradiated 3 h and 12 h heated mistletoe extracts. These results suggest that the combination of heat treatment and gamma-irradiation might be more effective than only heat-treatment for improving the anti-diabetic activity of mistletoe extract and reducing its cytotoxicity.

장기 중선량률 감마선 피폭에 의한 마우스의 생존율 및 생물학적 영향 평가 (Survival Rate and Biological Effect of Chronic Medium-Dose-Rate Gamma Radiation Exposed to Mice)

  • Late effects of chronic exposure to gamma radiation are potential hazards to worker in radiation facilities as well as to the general public. Recently, chronic gamma radiation exposure effects have become a serious concern. Using a total of 60 mice, we studied the biological effects of medium-dose chronic exposure to gamma radiation. Sixty female 6-week-old specific pathogen free Balb/c mice were randomly divided into six groups (five groups irradiated and one non-irradiated control group). Irradiation was carried out for 7 days using gamma rays at dose rates of 119.65, 238.10, 357.14, 476.19 and $595.24mGy\;h^{-1}$ with total doses 20, 40, 60, 80 and 100 Gy. After irradiation, we determined survival rate of gamma radiation exposed mice during 1 week and 476.19 and $595.24mGy\;h^{-1}$ exposed group mice showed less 10% of survival rate. Otherwise, 119.65, 238.10 and $357.14mGy\;h^{-1}$ exposed group mice were survived each 100%, 80% and 70%. Half of survived mice after 1 week are immediately sacrifice and counted body and spleen weights. Compared with control non-irradiated group, total body weights and spleen weights isolated from 119.65, 238.10 and 357.14 irradiated group mice showed significant decreased. However, no significant alteration was observed between 119.65, 238.10 and $357.14mGy\;h^{-1}$ irradiated group. Overall, our results show for the first time that medium-dose chronic gamma radiation has the potential to stimulation of biological effects.

감마선 조사 유래 블랙베리(Rubus fructicosus L.) 돌연변이 계통의 농업부산물 사료가치 분석 (Forage Quality Evaluation of Mutant Lines derived from Gamma-ray Treatments in Rubus fructicosus L.)

  • We examined the forage quality to provide the basic information for the utilization of blackberry by-products (leaf and stem) among thirty-five mutant lines derived from gamma-ray. The leaves had higher crude protein (CP) contents than stems. In addition, the leaves showed broad ranges of variation in crude fat, crude fiber and crude ash. The overall forage quality of the stems were similar to those of the original variety. The mean of neutral detergent fiber (NDF) content of leaves and stems in mutant lines were 25.88% and 58.98%, respectively. Also, the mean of acid detergent fiber (ADF) content of leaves and stems in mutant lines were 15.43% and 49.27%, respectively. Forage quality grades of all blackberry leaf were special quality class. However, the blackberry stem ranked $1^{st}$ to $5^{th}$ in quality class. The stem diameter was negatively ($P{\leq}0.05$) correlated with the moisture and crude protein. Also, the fruit length was highly negatively ($P{\leq}0.01$) correlated with the RFV (relative feed value) in blackberry leaf. The one hundred fruit weight was highly negatively ($P{\leq}0.01$) correlated with the crude ash. Based on these results, the by-products of blackberry could provide high quality forage for feeding of live stock.

이종이식재 개발을 위한 감마선 조사 돼지 인대의 미생물학적 안전성 및 인장강도 (Microbiological Safety and Tensile Strength of Gamma Irradiated Porcine Tendon for the Development of Xenografts)

  • This study was to determine the microbiological safety and tensile strength of gammairradiated porcine tendon for the development of safe xenografts. Escherichia coli and Bacillus subtilis were used as model pathogens and inoculated as $10^6{\sim}10^7log$ colonies forming unit $(CFU)g^{-1}$. As model virus from porcine, porcine parvovirus (PPV), bovine viral diarrhoea virus (BVDV) and poliovirus were inoculated as $10^5{\sim}10^6$ tissue culture infectious dose $(TCID)_{50}g^{-1}$ into porcine skin. The $D_{10}$ value of E. coli and B. subtilis was measured as $0.32{\pm}0.082kGy$ and $4.0{\pm}0.312kGy$, respectively. Additionally, the $D_{10}$ values of PPV, BVDV and poliovirus were also shown as $1.75{\pm}0.131kGy$, $3.70{\pm}0.212kGy$ and $6.26{\pm}0.332kGy$, respectively. Gamma irradiation decreased the tensile strength of porcine tendon. Results indicate that microbiological safety of porcine tendon can be improved significantly by gamma irradiation. However, further studies are needed to improve the tensile strength of gamma-irradiated porcine tendon.
