An analytical system detecting DNA particularly utilizing a concept of membrane strip chromatography initially applied to home-version tests for, such as, pregnancy and ovulation has been developed. We have chosen S. typhimurium as model analyte among food-contaminating microorganisms that occurred in high frequencies, and invA gene, as a detection target, specific to Salmonella species. This gene was able to be amplified by PCR under optimal conditions employing newly designed primers in our laboratory. The PCR product was specifically measured via hybridization between the analyte and a DNA probe, which was a totally different feature from the conventional gel electrophoresis detecting the products based only on the molecular size. It is notable thar the DNA probe sequence was specially designed such that no separation of excess primers present after PCR was required. This was immobilized on a nitrocellulose (NC) membrane via streptavidin-biotin linkage minimizing a steric effect when the hybridization with the amplified DNA took place. The analyrical system detected the microorganism in a concentration of minimum $10^3$ cfu/mL (i.e., 10 cells per system), estimated from the standard curve, 20 to 40 minutes after adding the sample. This sneitivity was approximately 10 times higher than that of gel electrophoresis as an analytical tool conventionally used. Furthermore, the assay was able to be run at room temperature, which would ofter an extra advantage to users.
This study was performed to clarify various conditions on the test of lipoxygenase-l and to establish the application of new test method for varietal improvement of soybeans in order to decrease beany flavors. Potassium borate and Tris were used as buffer and O.1M potassium borate solution showed the best result for the lipoxygenase-l test. In the range of pH 8.5~9.0 of the buffer, 2mM linoleic acid as substrate was effective. For color development, 100$\mu$l of two solutions(KI and starch) were added to the half soybean seed, successively. The substrate solution included linoleic acid was stored safely for 10 days at 4$^{\circ}C$ in refrigerator and for 4 days at room temperature. The best result was as follows; the 1ml of substrate solution[0.1M potassium borate(pH 9.0), 0.1% Tween-20, 2mM linoleic acid] was added to the chipped half soybean seed in l.5ml plastic tube, waited for 15 minutes, and 100$\mu$l of color development solutions(5% saturated KI in 15% acetic acid, 1% starch) were added to the tube, successively. After 4 hours, the purple color was observed in the upper phase of the plastic tube in the presence of lipoxygenase-1 and milky color in absence of lipoxygenase-1. The purple color was stable from 4 to 24 hours. There was no interfering effect by lipoxygenase-2 and -3. The plastic tube should be placed in the tube stand without shaking during the lipoxygenase-l test.

The measurement of radium ($^{226}Ra$) in the groundwater was established using ${\gamma}$-ray spectroscopy without sample preparation. The background interference by air borne radon daughter nuclides was reduced by $N_2$ gas flow into the counting chamber. Leakage of radon gas produced in the radioactive equilibrium with radium and its daughter nuclides was prevented by use of the air-tighted aluminium container. We investigated the effect of air layer in the counting container. Radioactivity variation due to emanation of radon into the air layer was within the counting error range 5%. When the nitrogen gas was flowed around the detector, peak counts of ${\gamma}$-rays from the daughters of airborne radon was decreased and detection limit was decreased to 0.02 Bq/L. The detection limit of detector was lower than 0.74 Bq/L, the $^{226}Ra$ Maximum Contaminant Level (MCL) in the groundwater proposed by US Environmental Protection Agency (EPA). It was confirmed that $^{226}Ra$ radioactivity in the groundwater could be determined by the ${\gamma}$-ray spectroscopy.

This paper presents and analyzes laboratory test results of distributed translator (DTxR) for distributed frequency network (DFN) in the ATSC (Advanced Television Systems Committee) terrestrial digital TV broadcasting system. The DTxR laboratory test is classified to receiving part test and transmitting part test. The receiving part test includes dynamic range, random noise, single echo, and adjacent channel interference. The transmitting part test includes quality of output signal (out-of channel emission, quality of transmitting signal, and phase noise), frequency synchronization among output signals, and TxID (Transmitter Identification) signal's affect to the legacy receiver. By the laboratory test results, the receiving part of DTxR eliminates average -2.5 dB of single echo and has average 17.5 dB at TOV (Threshold of Visibility) under random noise environment. In addition, the transmitting part of DTxR satisfies the specification of US FCC (Federal Communications Commission), and frequency difference among DTxR output signals is less than 0.001 Hz.

This study was carried out to give basic information about the environmental conservation in Masan District. It included the actual vegetation the degree of human disturbance of vegetation, plant biomass and biomass production. The natural vegetation was nonexistent and the major plant communities of the secondary forest was P. thunbergii-Q. acutissima and Quercus forest. P. thunbergii-Q. acutissima community area was 29.2% of Masan District and the secondary forest and the afforestation area was 48% and 13% of Masan District respectively. The ecological diversity of the plant community was high value in Q. acutissima- P. thunbergii, Q.aliena and Carpinus laxiflora-Q.variabilis communicty. As the investigation of the ecological succession in Masan Forest, P. thunbergii community is edaphic climax and mixed forest will be changed gradually dominant species of Quercus species. It was implied that Genus Quercus hadnever beenso easily taken ever by the Genus Pinus which forms the edaphic climax at vast areas of forest land. In the degree of human disturbance of vegetation, the degree 7 (secondary forest 1) was 52.8% and the green spaces in Masan cover 65%, which nongreen spaces 35 %. Total amount of plant biomass produced from Masan District was estimated to be 160, 470.95 tons. Annual Production of dry matter biomass amounts to be 32,940.64 tons. Estimated amount for O$_2$produced annually from the vegetated area in Masan was 34,856 tons.
In this paper, the 30 W power amplifier for an IMT-2000 repeater was developed a gain flatness and the third IMD (Intermodulation distortion) by microwave absorber. The absorption ability of the absorber is shown up to -10 dB and -4 dB at 3.6 GHz, 2.3 GHz band, respectively. The power amplifier without absorber has the gain over 57 dB, the gain flatness of $\pm$0.33 dB and the third IMD of 27 dBc at 33.3 W output. Otherwise, the power amplifier with absorber has the gain over 58 dB, the gain flatness of less than $\pm$0.9, the third IMD over 29 dBc at the same output power. As a result, the characteristic of the different type shows improvement of 1 dB in gain, 0.3 dB in gain flatness and 1.77 dBc in IMD.

In order to prevent the spread of non-indigenous aquatic species through the ballast water in commercial ships, International Maritime Organization (IMO) adopted in 2004 the International Convention for Control and Management of Ship's Ballast Water and Sediments. The Convention mandates treatment of ballast water for most transoceanic voyages and its confirmation of treatment is made with plankton live/dead assay. Fluorescein diacetate assay (FDA), which produces bright green light for live phytoplankton, has been a de facto standard method to determine the survival of marine plankton, but its staining efficacy has been in dispute. In the present study, we examined the limitation of FDA, and compared its efficacy with Neutral red (NR) staining, another promising assay and widely used especially for zooplankton mortality. For all phytoplankton species studied in the present study, except Ditylum brightwellii, the staining efficiency was <50% with FDA. The green FDA fluorescence interfered with phytoplankton autofluorescence in most samples. In contrast, NR assay stained over 90% of both phytoplankton and zooplankton species tested in this study. FDA assay also showed that green FDA fluorescence rapidly faded when phytoplankton cells were exposed to microscope light. Both FDA and NR assay were negative on formalin-killed individuals of both phytoplankton and zooplankton species. Our results suggest that NR assay is more effective for determining the survival of marine plankton and can be applied to test the efficacy of ballast water treatment.

In this paper, we propose a band rejection filter (BRF) with a state variable filter (SVF) structure to effectively remove the influence of 60 Hz line frequency noise introduced into the sensor system. The conventional BRF of the SVF structure uses an additional operational amplifier (OPAMP) to add a low pass filter (LPF) output and a high pass filter (HPF) output or an input signal and a band pass filter. Therefore, the notch frequency and the notch depth that determine the signal attenuation of the BRF greatly depend on the tolerance of the resistors used to obtain the sum or difference of the signals. On the other hand, in the proposed BRF, since the BRF output is formed naturally within the SVF structure, there is no need for a combination between each port. The notch frequency of the proposed BRF is 59.99 Hz, and it can be confirmed that it is not affected at all by the tolerance of the resistor through the Monte Carlo simulation results. The notch depth also has an average of -42.54dB and a standard deviation of 0.63dB, confirming that normal operation as a BRF is possible. Also, with the proposed BRF, noise filtering was applied to the electrocardiogram (ECG) signal that interfered with 60 Hz noise, and it was confirmed that the 60 Hz noise was appropriately suppressed.

Bunhwangsa Temple, Baekryulsa Temple and Sambulsa Temple are not famous cultural heritage area, but they are visited by many tourists and are located in Gyeongju National Park and Gyeongju Historic Areas, so environmental and cultural preservations are required. The purpose of this study is to manage the unique environment and landscape of cultural assets by analysing the invasive alien plant of traditional temple of Gyeongju. The whole flora were summarized as 188 taxa including 73 families, 136 genera, 154 species, 3 subspecies, 11 varieties, 4 forms, 5 hybrids and 12 cultivars., and the landscape plants were 163 taxa and 38 taxa of the invasive alien plants. The 13 taxa of invasive alien plants were planted in three temples. The ecosystem disturbance species were 3 taxa including Rumex acetosella, Lactuca seriola and Symphyotrichum pilosum. Invasive alien plants have artificial causes such as landscape planting, but there are also those that are introduced naturally from outside. The parking lot of the temple is expected to be a major propagation path for invasive alien plants due to the large amount of interference and disturbance. Based on the results of this study, the management plans are suggested as follows. First, it is necessary to use native species suitable for the natural environment and traditional landscape of Korea for landscape planting of traditional temples, and development of planting guidelines centered on cultural properties is required. Second, it is necessary to refrain from planting invasive alien plants because traditional temples are located in an important environment and historically. Third, for the preservation of the temple environment, it is necessary to promptly remove the ecosystem disturbance species. Fourth, in order to express the landscape characteristics of traditional temples, a unique planting plan should be established in consideration of the location environment and historicity.
Antarctic Environmental Monitoring Handbook' was published by COMNAP/SCAR in 2000. The standardized method described in this handbook is recommended for monitoring of antarctic environment. High pressure bomb technique in this guide was used to decompose soil samples. In compliance with this guide book, high pressure bomb technique was applied to decompose the antarctic soil sampled at the King Sejong Station. An Isotope Dilution-Inductively Coupled Plasma-Mass Spectrometry (ID-ICP-MS) was applied to determine mass concentrations of Pb, Cu and Zn in the soil. The accuracy in this method was verified by the analysis of certified reference materials (CRM) of NIST 2702 (marine sediment). The analytical results agreed with certified value within the range from 99.5~100.8%. Matrix separation was necessitated for the determination of Cu and Zn by Chelex 100 ion exchange resin. As a result, the average mass concentrations of Pb, Cu and Zn which are suspected to be caused by anthropogenic pollution were 332.9 mg/kg, 95.6 mg/kg and 115.3 mg/kg, respectively. Those for the metals sampled in the soils of the remote regions from the station were 28.1 mg/kg, 101.8 mg/kg and 115.6 mg/kg, respectively.
