Using both light and scanning electron microscopies, it was investigated on the visual cells as well as the eyes of Macropodus ocellatus (Pisces, Osphronemidae). This species had a circular lens and yellowish cornea. The eyes had $3.5{\pm}0.2mm$ which is $31.1{\pm}3.0%$ in a percentage of eye diameter relative to head length. The retina ($158.2{\pm}10.6{\mu}m$) was built of several layers, including the visual cell layer which consists of three types of cells: single cons ($27.8{\pm}1.6{\mu}m$) and equal double cone ($33.9{\pm}3.7{\mu}m$), and large rods ($57.3{\pm}1.3{\mu}m$). The visual cell layer then was classified into the correct pattern. All visual cells were clearly distinguished from two parts (inner and outer segments). The elongated rod cells were extend to the bottom of the retinal pigment epithelium. In scanning electron microscopy, the outer segment links to inner segment by so-called calyceal piles. The M. ocellatus single and double cones appearance form a flower-petal arrangement, which is a regular mosaic pattern that contains quadrilateral units by four double cones surrounding a single cone.
We report on a quantitative analysis of cone and rod photoreceptors in hamster retina. Cone and rod photoreceptors were counted in retinal whole mounts using differential interference contrast (DIC) optics microscopy after staining of cone photoreceptors were stained with peroxidase-labeled peanut lectin. Middle-to-long-wave-sensitive-(M/L-), and shortwave-sensitive-(S-) cone opsins were visualized by observed using confocal microscope after immunocytochemical procedure. The average cone density was 9,307 $cells/mm^2$, giving a total of cones of 293,060 cone cells per retina. The peak density of cone cells (12,857 $cells/mm^2$) was found 0.3 mm from the optic disk (OD) of the nasal retina. The average rod density was 300,082 $cells/mm^2$, giving a total number of rods of 9,448,150 cells. The peak density of rod cells was found 0.3 mm from the OD of the dorsal retina. Of all photoreceptors studied, the total percentage of rods and cones were 96.99% and cones 3.01%, respectively. The mean ratio of rod and cone was 32.24 : 1. The cone photoreceptors of hamster contained both M/L- and S-cone opsins. The present results suggest that the hamster retina is strongly rod-dominated with some photopic property of vision.
Two Korean Rhodeus bittering fishes are known as having a unique reproduction system that the female deposits its egg inside freshwater mussels. Among them, R. ocellatus and R. uyekii show an ecological difference in feeding items and micro-habitat. Through a light microscope, the visual cells of the retina in the two species were investigated to get information between vision and environment related with habitat. The two fishes' eyes are spherical and R. ocellatus has relatively larger eyes. For the two species, the visual cells of the retina consisted of long single cones, short single cones, unequal double cones and rods. In the cones, all show a row mosaic pattern. Although the two species have a different micro-habitat in the wild, the component of the visual cells was very similar to each other.
The visual cells in the retina of Iksookimia longicorpa (Pisces, Cobitidae) were investigated by light and scanning electron microscopes. The retina ($216.42{\pm}13.36{\mu}m$) has several layers, and the visual cell layer consists of unequal double cones and large rods. In a double cone, two members are unequal such that one cone is longer than the other (long element $26.42{\pm}1.7{\mu}m$, short element $16.82{\pm}1.1{\mu}m$). The cones form a row mosaic pattern in which the partners of double cones are linearly oriented with a large rod. The visual cells observed have an outer segment (hematophilic), inner segment (eosinophilic). In scanning electron microscopy, the outer segment links to inner segment by so-called calyceal piles (calyceal processes) of membrane discs surrounded by double membranes.
Two strains of actinomycetes producing extracellular adenosine deaminase, strain J-845S and strain J-326TK, were isolated from soil. Strain J-845S was gram-positive and non-acid-fast. This strain formed whitish, rod-shaped, smooth and non-motile spores on the aerial mycelium, and the spore chain was spiral. The hyphae of the mycelium branched abundantly. Cell wall chemotypes of the strain were of type I containing LL-diaminopimelic acids, and of phospholipid type II, and then strain J-845S was designated as Streptomyces sp.. Strain J-326TK was gram-positive and non-acid-fast. The hyphae of primary and aerial mycelium fragmented into irregular rod of coccus-like elements. The aerial mycelium either did not branch or sparsely branched. Cell wall composition was of type I and phospholipid type I. Thus, strain J-326TK was identified as Nocardioides sp.
The taxonomical properties of strain J-275L isolated from soil as a microorganism which produces extracellular adenine deaminase and cultural conditions for the enxyme production were studied. The hyphae of strain J-275L is fragmented into rod-or coccus-like elements. The elements of fragmented aerkal hyphae has smooth surfaces. The cell wall of the organism contains LL-diaminopimelic acid. Mycolic acid are not produced. As a result of taxonomical studies, strain J-275L is designated as Nocardioides sp. J-275L. The optimum medium for the enzyme production from Nocardioides sp.J-275L wascomposed of 0.5% peptone, 0.5% dextrin, 1% yeast extract, and 0.2% $K_{2}HPO_{4}$. The optimum initial pH of the medium was pH 7.5.
These experiments were performed in order to study histologically and histochemically on the epithelial cells of gall bladder in Carassius carassius, Bufo bufo gargarizans, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus. The results of the observation were as follows: 1. There were different cell types in the epithelium of gall bladder in each animal and it could not be supported histochemically that the epithelia cells of gall bladder were divided into two cell types of the rod-shaped and barrel-shaped ones. 2. The epithelium of gall bladder in Carassius carassius, Bufo bufo gargarizans, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus was simple columnar epithelium. 3. The eosinophilities of cytoplasm in the epithelial cells of gall bladder were in uniform stronger in the upper portion of nucleus in Carassius carassius, Bufo bufo gargarizans and Natrix tigrina lateralis than its other portions, and in Urloncha striata var. domesticus and Lepus cuniculus var. domesticus existed uniformly in all portions, but there were many non-eosinophilic cells in Bufo bufo gargarizans and many cells that weakly eosinophilic around nucleus in Bos taurus var. domesticus. 4. The periodic acid Schiff's reactivities in the epithelial cells of gall bladder were different in each other and the epithelial cells in PAS reaction were divided into two cell types of the dark and light ones. There presented the light cells of 6.4%, 4.3% and 3.7% of epithelial cell of gall bladder in Carassius carassius, Bufo bufo gargarizans and Urloncha striata var. domesticus for each other, but were not presented in Natrix tigrina lateralis and Bos taurus var. domesticus. 5. The ninhydrin-Schiff-active proteins were much in the epithelial cells of gall bladder in Bos taurus var. domesticus, Carassius carassius, Urloncha striata var. domesticus and Natrix tigrina lateralis in order and were much in epithelial cells in the upper portion of mucosal folds in Carassius carassius, Urloncha striata var. domesticus and Natrix tigrina lateralis, and the ninhydrin-Schiff-active protein of the epithelium of gall bladder in Bufo bufo gargarizans was uniformly distributed. 6. The epithelial cells of gall bladder in Carassius carassius, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus had no stain reactivity or weak stain reactivity to neutral fat and all epithelial cells in Bufo bufo gargarizans had strong stain reactivity, though they were different in quantity of epithelial cell portion. 7. The stain reactivities to RNA and DNA were stronger in the epithelial cells of the upper portion of mucosal fold than in those of other portions.
Ultrastructure of adult Drosophila ocellus was compared with conventional electron microscopic method and osmium impregnation. When osmium impregnation was applied, some organelles of cells were strongly stained. Especially, subrhabdomeric cisternae (SRC) were strongly stained and showed network-iike structure as in compound eye. Other organelles including SSC, ER, nuclear envelope, pigment granules and mitochondria were also strongly stained. These organelles are known as a general calcium ion reservoir. In conclusion, the strong effect of light and shade by osmium impregnation was regarded as a result of strong binding between calcium ion and osmium tetroxide. Thus, we agree to the opinion that osmium impregnation is very useful methods to the comparative morphology of cell organelles.
In order to understand fine structural and histochemical changes in the cyst cell and he interstitial cell in the testis of the spottybelly greenling Hexagrammos agrammus associated with the reproductive cycle from January to December, 1992, there cells were studied by electron microscopy and light microscopy. The cyst cells in the mature testis show a weak affinity to haematoxylin. while they become larger in size. At this time, these cells appear to be more functional than those on the growing stage because several mitochondria, endoplasmic reticulum, glycogen particles and a few lipid droplets appear in the cytoplasm of the cyst cell. It appears, therefore, that the cyst cell of this species has vital functions for nutrition, secretion and steroidogenesis. Well-developed interstitial cells contain large rod-shape or spherical mitochondria with tubular cristae and the large quantities of smooth endoplasmic recticulum and electron-dense materials in the vesicle at the mature and spawning stage. The interstitial cells of this species show characteristics of steroid interstitial cells having a vesicular nucleus, mitochondria with tubular cristae, and smooth endoplasmic reticulum. However, these interstitial cells of teleost give negative histochemical reactions for Sudan black B.
