Elctrophoretic Properties of Heat-Induced Bovine α-Lactalbumin

  • In order to study the reaction behavior of bovine holo- and apo-$\alpha$-lactalbumin ($\alpha$-La) during heat treatment at 65~10$0^{\circ}C$, the samples were analysed by first (ID)-and second-dimensional (2D) native-polyacrylamide gel electrophoresis (Native-PAGE) and sodium dodecylsulfate (SDS)-PAGE. When bolo-$\alpha$-La or apo- $\alpha$ -La were heated, they formed non-native, monomers, dimers and trimers. The apo-$\alpha$-La was more heat-sensitive than holo-$\alpha$-La. The monomers seemed to have the same composition as the native $\alpha$-La, but many of the disulfide bonds could be non-native.

  • Conjugated oligomers have been already used as active layers in field effect transistors, photodiodes and electroluminescent devices. Particularly thiophene oligomers such $\alpha$ -sexithiophene($\alpha$-6T) attract great interest for its prospective app1ications in large-area flexible displays. In this study, we investigated the contact properties between the organic semiconductor $\alpha$-6T and metals such as Au(Gold), Ag(Silver), Cr(Chromium), Al(Aluminum), Cr(Chromium). Using the Transmission Line Model(TLM) method, specific contact resistances of the metal lines in contact with the $\alpha$-6T were determined. From the current-voltage characteristics, electrical conductivity of the $\alpha$-6T films is found.

  • The radical copolymerization with propagation and depropagation is presented in order to estimate reactivity rate of monomers and $K_{11}$(the equilibrium constant for propagation and depropagation) in the copolymerization of ${\alpha}-methylstyrene-co-methylmethacrylate$ and ${\alpha}-methylstyrene-acrylonitrile$. The value of ${\alpha}-methylstyrene$ and methylmethacrylate and $K_{11}$ are found to be 0.48, 0.47 and 5.0 respectively. The value of ${\alpha}-methylstyrene$ and acrylonitrile and the $K_{11}$ are found to be 0.1251, 0.0577 and 23.8 respectively. The treatment rate constant of ${\alpha}-methylstyrene-co-methylmethacrylate$ and ${\alpha}-methylstyrene-co-acrylonitrile$ in the copolymerization is estimated to be 2.5, 80.72 regardless of monomer feed composition.

Inhibitory Effects of Steppogenin and Oxyresveratrol from Morus alba L. against Yeast ${\alpha}$-Glucosidase (뽕나무에서 분리한 Steppogenin과 Oxyresveratrol의 효모 ${\alpha}$-Glucosidase의 억제효과)

  • [ ${\alpha}$ ]Glucosidase inhibitor is a target in the treatment of type II diabetes through the mainly inhibition of glucose levels after meals. In this study, we purified steppogenin and oxyresveratrol from the stem of Morus alba L. and examined their inhibitory activity against yeast ${\alpha}$-glucosidase. Steppogenin and oxyresveratrol were inhibited yeast ${\alpha}$-glucosidase in a dose dependent manner. The $IC_{50}$ activities (50% inhibition) were 34.4 and 9.3 ${\mu}M$, respectively. The kinetic inhibition of steppogenin showed noncompetitive inhibition ($K_m:1.1{\times}10^{-3}M$; $K_i:1{\times}10^{-5}M$), meanwhile oxyresveratrol showed competitive inhibition ($K_m:4.3{\times}10^{-3}M$; $K_i:3.4{\times}10^{-6}M$) against yeast ${\alpha}$-glucosidase. These results indicate that steppogenin and oxyresveratrol are noncompetitive and competitive inhibitors, respectively, against yeast ${\alpha}$-glucosidase.

Hydration Properties of $\alpha$-Tricalcium Phosphate in Tris. Solution ($\alpha$-Tricalcium Phosphate의 Tris. Solution에서의 수화특성)

  • $\alpha$-tricalcium phosphate($\alpha$-TCP) powders were synthesized and their hydration properties were investigated in Tris. solution. Two kinds of $\alpha$-TCP powder samples were prepared; the one is reaction product of CaHPO4.2H2O and CaCO3, and another is that of hydroxyapatite(HAp) and $\beta$-Ca2P2O7. They were satisfied with Ca/P mole ratio 1.5 and were heated at 150$0^{\circ}C$ for 5 hours. In the hydration of $\alpha$-TCP samples the powder which was synthesized from HAp and $\beta$-Ca2P2O7 was hydrated faster than that from CaHPO4.2H2O and CaCO3. The hydration reaction of $\alpha$-TCP powder transformed rapidly into HAp accompanying setting and hardening. It was realized that the hydration reaction of $\alpha$-TCP was due to the solution-precipitation mechanism and the hydrates from the reaction were Ca-deficient HAp having funtional group HPO42-.

  • The powder mixture of Si3N4-AlN-Y2O3, Si3N4-AlN-CeO2 and Si3N4-AlN-Y2O3-CeO2 system was hot-pressed at 175$0^{\circ}C$ for 2h in N2 to prepare $\alpha$-Sialon ceramics. The mechanical property and oxidation behaviour of the prepared $\alpha$-Sialon ceramics were investigated. At 120$0^{\circ}C$, oxidation resistance was best for the Y2O3 added $\alpha$-Sialon ceramics and oxidation rate increased when the amount of CeO2 increased. But when the mixture of Y2O3 and CeO2 added $\alpha$-Sialon ceramics showed a good oxidation resistance. Fracture toughness of (Y2O3+CeO2) added $\alpha$-Sialon ceramics was higher than Y2O3 added $\alpha$-Sialon ceramics.

  • A new in-line method for sampling neutrons emitted in (${\alpha}$,n) reactions based on alpha particle source information has been developed for continuous-energy Monte Carlo simulations. The new method uses a continuous-slowing-down model coupled with (${\alpha}$,n) cross section data to precompute the expected neutron yield over the alpha particle lifetime. This eliminates the complexity and computational cost associated with explicit charged particle transport. When combined with an integrated alpha particle decay source sampling capability, the proposed method provides an efficient and accurate method for sampling (${\alpha}$,n) neutrons based solely on nuclide inventories in the problem, with no additional user input required. Results from several example calculations show that the proposed method reproduces the (${\alpha}$,n) neutron yields and energy spectra from reference experiments and calculations.

  • A novel preparative method for hexaprofen, which is a potent antiinflammatory agent, is described. Friedel-Crafts reaction of cyclohexylbenzene with ethyl $\alpha$-chloro-$\alpha$-(methylthio) acetate 1 and $\alpha$-chloro-$\alpha$-(methylthio) acetonitrile 2 afforded ethyl 2-(methylthio)-2-(4-cyclohexylphenyl) acetate 7 and 2-methylthio-2-(4-cyclohexylphenyl) acetonitrile 8, respectively. Compounds 7 and 8 were converted into the corresponding ethyl 2-methylthio-2-(4-cyclohexylphenyl) propionate 9 and 2-methylthio-2-(4-cyclohexylphenyl) propionitrile 10 by methylation with sodium hydride and methyl iodide. Hexaprofen 13 was prepard by hydrolysis of ethyl 2-(4-cyclohexylphenyl) propionate 11 and of 2-(4-cyclohexylphenyl) propionitrile 12 followed by desulfurization of compounds 9 and 10.

Cytotoxic and Apoptotic Effects of Saponins from Akebia quinata on HepG2 Hepatocarcinoma Cells (으름유래 사포닌의 HepG2 간암세포에 대한 세포독성 및 세포자살유도 효과)

  • Four saponins (1~4) were isolated from Akebia quinata pericarp through bioassay-guided fractionation. Pericarps of A. quinata were extracted with ethanol and sequentially fractionated with dichloromethane, ethyl acetate, butanol and water. Compounds 1~4 from the butanol fraction were identified as 3-O-${\alpha}$-L-arabinopyranosyl hederagenin (${\delta}$-hederin), 3-O-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$2) ${\alpha}$-L-arabinopyranoly oleanolic acid (${\beta}$-hederin), 3-O-${\beta}$-D-xylopyranosyl (1${\rightarrow}$3) ${\alpha}$-L-arabinopyranosyl hederagenin (saponin C), and 3-O ${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$2) ${\alpha}$-L-arabinopyranosyl hederagenin (${\alpha}$-hederin) based on the spectroscopic evidences, respectively. Oleanolic acid and hederagenin were identified as the corresponding sapogenins by acid-hydrolysis. These compounds exhibited strong cytotoxic activity in MTS [3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxy-methoxyphenyl)-2-(4-sulfophenyl)-2H- tetrazolium, inner salt] assay on HepG2 cells. ${\beta}$-Hederin obviously attenuated the expression of bcl-2, an anti-apoptotic protein. All of the compounds also induced the activity of caspase-3, an apoptotic enzyme, while ${\alpha}$-hederin was the most potent activator of the enzyme. Our data demonstrate for the first time the apoptosis-inducing activity of A. quinata. These results suggest that A. quinata could be used as a potential source of natural cancer chemopreventive agents.

Effect of Epidermal Growth Factor and Transforming Growth Factor-$\alpha$ on In Vitro Maturation of Porcine Oocytes (Epidermal Growth Factor(EGF)와 Transforming Growth Factor-$\alpha$(TGF-$\alpha$)가 돼지 난포란의 체외성숙에 미치는 영향)

  • The present study examined the effects of epidermal growth factor(EGF) and transforming growth factor-$\alpha$(TGF-$\alpha$) on in vitro maturation of porcine follicular oocytes. Basic medium used TCM-HEPES, and oocytes cultured for 42 hours in vitro. The results obtained are as follows; 1. The nuclear maturation rates of EGF-treated groups(10ng/ml, 75.9% ; 30ng/ml, 69.2% ; 50ng/ml, 67.2% ; 100ng/ml, 71.0%) on the porcine oocytes cultured in medium without pFF in vitro were significantly(P<0.01) higher than those of non-treated group(57.1%). When the oocytes were cultured in media with 10%(v/v) pFF, the nuclear maturation rates of 30ng EGF/ml(77.1%) treated group were significantly(P<0.01) higher than those of non-(59.2%) and EGF-treated groups(10ng/ml, 65.4% ; 50ng/ml, 65.5% ; 100ng/ml, 70.4%). 2. The nuclear maturation rates of 30ng TGF-$\alpha$/ml treated group(71.9%) in media without pFF in vitro were significatnly(P<0.01) higher than those of non-(57.1%) and TGF-$\alpha$ treated groups(10ng/ml, 60.4% ; 50ng/ml, 65.4% ; 100ng/ml, 60.0%). When the oocytes were cultured in media with 10%(v/v) pFF, the nuclear maturation rates of 30 and 50ng TGF-$\alpha$/ml(77.4% and 79.6%) treated groups(10ng/ml, 64.2% ; 100ng/ml, 61.6%). 3. On the effect of EGF(30ng/ml) and/or TGF-$\alpha$(30ng/ml) treated groups in medium without pFF in vitro, the nuclear maturation rates indicated 57.3, 60.4, 75.9 and 79.7% in media with no EGF & TFG-$\alpha$, TGF-$\alpha$ only, EGF only nad EGF+TGF-$\alpha$ treated groups, respectively. The nuclear maturation rates in medium with EGF only or EGF+TGF-$\alpha$ were significantly(P<0.01) higher than those non- and TGF-$\alpha$ treated groups. When the oocytes were cultured in media with 10%(v/v) pFF, the nuclear maturation ratesof EGF+TGF-$\alpha$ treated group(75.9%) were significantly(P<0.01) higher than those of non-(59.2%), TGF-$\alpha$ only (64.2%) and EGF only(69.4%) treated groups.
