Discovery and Characterization of a Thermostable NADH Oxidase from Pyrococcus horikoshii OT3

  • A gene (PH0311) encoding a hypothetical protein from the genome sequence data of the hyperthermophilic archaeon Pyrococcus horikoshii OT3 was cloned and over-expressed in Escherichia coli. The purified recombinant protein was found to possess FAD-dependent NADH oxidase activity, although it lacked sequence homology to any other known general NADH oxidase family. The product of the PH0311 gene was thus designated PhNOX (NADH oxidase from Pyrococcus horikoshii), with an estimated molecular weight of 84 kDa by gel filtration and 22 kDa by SDS-PAGE, indicating it to be a homotetramer of 22 kDa subunits. PhNOX catalyzed the oxidation of reduced ${\beta}$-NADH with subsequent formation of $H_2O_2$ in the presence of FAD as a cofactor, but not ${\alpha}$-NADH, ${\alpha}$-NADPH, or ${\beta}$-NADPH. PhNOX showed high affinity for ${\beta}$-NADH with a Km value of 3.70 ${\mu}$M and exhibited optimum activity at pH 8.0 and 95$^{\circ}C$ as it is highly stable against high temperature.

The Effect of Repetitive Compression on the Fatigue Properties of Foam for Footwear Mid-sole (반복압축이 스포츠화용 발포체의 피로특성에 미치는 영향)

  • To study the fatigue properties of three type of foams for footwear midsole, polyurethane(PU), phylon(PH) and injection phylon(IP) foams were prepared with different hardnesses. Three types of foams were repetitively compressed for 50,000 cycles at 50 rpm. Cell shapes of foams were deformed with repetitive compression. The extent of cell deformation of IP was larger than those of PH and PU. Permanent strain of foam was made by repetitively compressing the foam, and the extent of IP was larger than those of PU and PH. Maximum compression forces of three types of the foams were decreased with the repetitive compression, and IP had the largest decrease in compression load of foam with compression. Decreases in maximum compression force of three types of foams were increased with increase of the hardness of foam.

  • To clarify of chemical contents for the Keumsan ginseng, two attempts have been done for two year: 2 year and 4 year-ginsengs coilected from the phyllite(Ph0 and granite area(Gr), and 1 year, 2 year and 3 year-ginsengs collected from the phyllite(Ph), shale(Sh) and granite (Gr), In the first attempt, the soil contents of the Ph are high in most of elements. In the comparisons between ages of the ginseng, regardless the area, most of the elements are high in the 4 year and these trends are distinctive in the granite area. In the comparisons of the areas, 2 year-ginsengs of the Ph show high contents in the most of the elements. Comparisons between upper part(leaf and branch) and root part of the ginsengs show that in the 2 year-ginseng, the Gr is high in the root part and the Ph is high in the upper part, while in the 4 year-ginseng, most of the elements are high in the root part. Comparisons between contents of soil and ginsengs show similar in crease and decrese trends. However, the 2 year-ginsengs show clear differences between two areas while in the 4 year-ginseng, differences between the Gr and Ph is not clear. It suggest that the ginsengs absorb eligible element contents with ages, despite of clear content differences of the different soils. In the second attempt, the Gr shows high W, Pb, Th, U, Sn and B contents wlile Ph and Sh show high Ni, Cr, Co, Sc, V, As, Cu and Zn contents. In the comparisons between ages of the ginseng, regardless the area, most of the elements are high in the 2 year, especially in the Gr and Ph. In the comparisons of the areas, 2 year-ginsengs of the Gr show low contents in the most of the elements, and the S1 show higher Ni, V, Th and U contents than the Ph. 3 year-ginsengs of the Gr show low contents in the most of the elements, while the S1 show high contents in the most of elements. relative to the Ph. Comparisons between upper part and root part of the ginsengs show that in the ginseng, regardless the ages, the upper part is high. Comparisons between contents of soil and ginsengs show similar increase and decrese trends, and soil is low than the ginsengs in the most of the elements

Kinematic Analysis of Accuracy Evaluation of Pyeongwon Poomsae (정확성 평가결과에 따른 태권도 평원 품새 동작의 운동학적 분석)

  • Objective : The purpose of this study was to investigate the relations between accuracy score and the motions which are performed in PyeongWon Poomsae, thereby developing objectivity in accuracy evaluations for Poomsae competitions. Method : The subjects were six male Poomsae players (age: $21.2{\pm}1.17yrs$, height: $173.4{\pm}3.95cm$, weight: $67.7{\pm}7.65kg$). A three-dimensional motion analysis was performed on the subjects using six high-speed cameras (60 frames/sec) and subjects' performed motions were evaluated by 5 evaluators. The entire Pyeong Poomsae was divided into 13 events and 9 phases; there were three pairs of symmetric phases among them: front kick & turning side kick phases (3PH, 3-1PH), arm motion & stance phases (4PH, 4-1PH), side kick with Hakdari-stance phases (5PH, 5-1PH). Performance time, change and range of COM, height of vertex, and foot of side kicks were analysed. The Data was analysed utilizing correlation analysis. Results : There was a positive correlation between accuracy score and the difference between right and left range of COM (X direction) at 4PH (r=0.921, p=0.009). Conclusion : The results of our study indicate that it is necessary to consider some of objective criterion such as performance time, COM range, and symmetrical movements in accuracy evaluations of Poomsae competitions.

Hyperlipidemic Inhibitory Effects of Phellinus pini in Rats Fed with a High Fat and Cholesterol Diet

  • This study evaluated the in vitro and in vivo hypolipidemic effects of the medicinal mushroom Phellinus pini. The methanol extract (ME) of the fruiting body of Ph. pini was active against pancreatic lipase and cholesterol esterase with 99.14% and 67.23% inhibited activity at 1.0 mg/mL, respectively. It also inhibited 81.81% and 55.33% of ${\alpha}$-glucosidase and ${\alpha}$-amylase activities, respectively, at 2.0 mg/mL. Hyperlipidemia as induced by feeding rats with a high fat and cholesterol diet (HFC). HFC supplemented with a 5% fruiting body powder of Ph. pini (HFC + PhP) significantly reduced plasma total cholesterol, low-density lipoprotein cholesterol, and triglycerides in rats compared with HFC. The reduced levels were comparable to rats fed the normal control diet (NC). The atherogenic index of HFC + PhP rats was significantly lower than that of the HFC rats. The excretion of fecal total lipid and cholesterol in the HFC + PhP rats was significantly higher than those in the NC and HFC rats. Histopathological examinations demonstrated scant deposition of lipids in the liver of rats fed HFC + PhP. The dietary supplementation with the fruiting body powder provided natural plasma lipid and glucose lowering effects in experimental rats without adverse effects on the plasma biochemical parameters and liver function related enzyme activities. Therefore, the hypolipidemic effects of Ph. pini may be due to the inhibitory effects on pancreatic lipase, cholesterol esterase, ${\alpha}$-glucosidase, and ${\alpha}$-amylase, and excretion of excess lipids and cholesterol in the feces.

Adsorption of selected endocrine disrupting compounds (EDCs)/pharmaceutical active compounds (PhACs) onto granular activated carbon (GAC) : effect of single and multiple solutes (EDCs/PhACs의 단일,복합 조건에서의 GAC에 대한 흡착 연구)

  • The widespread occurrence of dissolved endocrine disrupting compounds(EDCs) and pharmaceutical active compounds(PhACs) in water sources is of concern due to their adverse effects. To remove these chemicals, adsorption of EDCs/PhACs on granular activated carbon(GAC) was investigated, and bisphenol A, carbamazepine, diclofenac, ibuprofen, and sulfamethoxazole were selected as commonly occurring EDCs/PhACs in the aquatic environment. Various adsorption isotherms were applied to evaluate compatability with each adsorption in the condition of single-solute. Removal difference between individual and competitive adsorption were investigated from the physicochemical properties of each adsorbate. Hydrophobicity interaction was the main adsorption mechanism in the single-solute adsorption with order of maximum adsorption capacity as bisphenol A > carbamazepine > sulfamethoxazole > diclofenac > ibuprofen, while both hydrophobicity and molecular size play significant roles in competitive adsorption. Adsorption kinetic was also controled by hydrophobicity of each adsorbate resulting in higher hydrophobicity allowed faster adsorption on available adsorption site on GAC. EDCs/PhACs adsorption on GAC was determined as an endothermic reaction resulting in better adsorption at higher temperature ($40^{\circ}C$) than lower temperature ($10^{\circ}C$).

Effects of high glucose with or without other metabolic substrates on alpha-adrenergic contractions in rat mesenteric and femoral arteries

  • Hyperglycemia is associated with an increased risk of cardiovascular diseases. It has been demonstrated that chronic exposure to high glucose impaired endothelial functions. However, specific effects of short-term exposure to high glucose on vascular reactivity are controversial. Moreover, the combined effects of other metabolic substrates such as free fatty acids (FFA) on vascular reactivity remain poorly understood. Here we investigate the effects of short-term exposure to high glucose with or without other metabolic substrates including FFAs termed "nutrition full" (NF) solution, on mesenteric (MA) and deep femoral arteries (DFA) of rats. Arterial ring segments were mounted in a double-wire myograph. Contraction in response to phenylephrine (PhE) was determined in control (5 mM) and high glucose (23 mM, HG) environments over a 30 min period. In both arteries, PhE-inducedvasocontraction was enhanced by pre-incubation of HG solution. A combined incubation with HG and palmitic acid ($100{\mu}M$) induced similar sensitization of PhE-contractions in both arteries. In contrast, high $K^+$-induced contractions were not affected by HG. Interestingly, pre-incubation with NF solution decreased PhE-induced contraction in MA but increased the contraction in DFA. In NF solution, the HG-induced facilitation of PhE-contraction was not observed in MA. Furthermore, the PhE-induced contraction of DFA was attenuated by HG in NF solution. Our results demonstrate that the sensitization of PhE-induced arterial contraction by HG is differentially affected by other metabolic substrates. The conversation of skeletal arterial contractility by HG in NF solution requires careful interpretation of the previous in vitro studies where only glucose is included in physiological salt solutions. Further studies are required to elucidate the mechanism underlying the inconsistent effect of NF solution on MA and DFA.

Sensory and Lipid Characterizations of Boiled-dried Pacific Herring Clupea pallasii as a Substitute for Boiled-dried Anchovies Engraulis japonicus (마른멸치(Engraulis japonicus) 대용으로서 마른청어(Clupea pallasii)의 관능 및 지질 특성)

  • The objective of this study was to investigate sensory and lipid characterizations of boiled-dried Pacific herring Clupea pallasii (PH) as a substitute for boiled-dried anchovies (A). The crude lipid content of commercial boiled-dried PH ranged from 1.3 to 5.1%, which was similar to that of commercial boiled-dried A (2.2-5.1%). The peroxide values (POVs) of commercial boiled-dried PH ranged from 20.5 to 129.1 meq/kg. The POVs of commercial boiled-dried PH suggest that boiled-dried PA of high quality should be controlled by a POV standard similar to that of boiled-dried A. The trichloroacetic acid soluble-nitrogen (TCA soluble-N) content of commercial boiled-dried PH ranged from 495.7 to 998.6 mg/100 g, which was high compared to that of commercial boiled-dried A (372.6-690.0 mg/100 g). Volatile basic nitrogen (VBN) content of commercial boiled-dried PH ranged from 8.4 to 28.1 mg/100 g, which was low compared to that of commercial boiled-dried A (16.8-33.0 mg/100 g). Results from the lipid, POV, TCA-soluble-N, and VBN content analyses suggest that boiled-dried PH could be used as a substitute for boiled-dried A.

Anti-inflammatory Effect of Portulacae Herba Water Extract on Lipopolysaccharide-activated RAW 264.7 Macrophages (마치현(馬齒莧) 물추출물의 항염효능에 관한 연구)

  • Objectives : The present study aimed to investigate the anti-inflammatory effects of the water extracts of Portulacae Herba (PH).Methods : We measured the effects of the water extracts of Portulacae Herba (PH) on the cell viability of mouse macrophage RAW 264.7 cells, the intracellular calcium production, and the proinflammatory mediators including nitric oxide (NO), vascular endothelial growth factor (VEGF), platelet-derived growth factor (PDGF)-BB, which are induced by the lipopolysaccharides (LPS), and obtained the results shown below.Results : After the cultivation of the PH extracts along with the mouse macrophages, the cell survival rate did not decrease with the MTT assay. However, the PH extracts did significantly suppress the production of NO by the mouse macrophages induced by LPS at the concentrations of 25, 50 and 100 ㎍/mL. The PH extract also significantly suppressed the VEGF, PDGF-BB and intracellular calcium production of the mouse macrophages by LPS at concentrations of 25, 50 and 100 ㎍/mL. As shown in the results above, the PH extracts do not have a toxic effect on the macrophages, but still have an anti-inflammatory effect that significantly reduces the intracellular calcium production as well as the production of NO, VEGF and PDGF-BB at concentrations above 25 ㎍/mL.Conclusions : In conclusion, the inhibitory anti-inflammatory effects of the PH extract can be used for a new treatment of anti-inflammatory diseases.

Alterations of papilla dimensions after orthodontic closure of the maxillary midline diastema: a retrospective longitudinal study

  • Purpose: The aim of this study was to evaluate alterations of papilla dimensions after orthodontic closure of the diastema between maxillary central incisors. Methods: Sixty patients who had a visible diastema between maxillary central incisors that had been closed by orthodontic approximation were selected for this study. Various papilla dimensions were assessed on clinical photographs and study models before the orthodontic treatment and at the follow-up examination after closure of the diastema. Influences of the variables assessed before orthodontic treatment on the alterations of papilla height (PH) and papilla base thickness (PBT) were evaluated by univariate regression analysis. To analyze potential influences of the 3-dimensional papilla dimensions before orthodontic treatment on the alterations of PH and PBT, a multiple regression model was formulated including the 3-dimensional papilla dimensions as predictor variables. Results: On average, PH decreased by 0.80 mm and PBT increased after orthodontic closure of the diastema (P<0.01). Univariate regression analysis revealed that the PH (P=0.002) and PBT (P=0.047) before orthodontic treatment influenced the alteration of PH. With respect to the alteration of PBT, the diastema width (P=0.045) and PBT (P=0.000) were found to be influential factors. PBT before the orthodontic treatment significantly influenced the alteration of PBT in the multiple regression model. Conclusions: PH decreased but PBT increased after orthodontic closure of the diastema. The papilla dimensions before orthodontic treatment influenced the alterations of PH and PBT after closure of the diastema. The PBT increased more when the diastema width before the orthodontic treatment was larger.
