Evaluation of Efficacy of Bacteriophage CJø07 against Salmonella enteritidis Infection in the SPF Chicks (박테리오파아지 CJø07의 Salmonella enteritidis 감염에 대한 SPF 병아리에서의 효능 평가)

  • In the present study we report in vivo inhibitory potential of single strain of bacteriophage ($CJ{\phi}07$) in day-old SPF chicks experimentally infected with Salmonella enteritidis (SE). The bacteriophages prepared by feed additives and drinking water were given to chicks for 20 days starting prior 10 days before challenge with SE. Chicks were euthanized at 10 days after challenge for quantitative salmonella isolation from intestine and determination of environmental contamination level of salmonella. Bacteriophage therapy as additives in feed and drinking water resulted in significant inhibition of the SE replication in intestines of SPF chickens (P<0.05). In addition, environmental contamination by SE fecal shedding was decreased in bacteriophage-treated birds. Therefore, bacteriophage $CJ{\phi}07$ examined in this study may be a plausible alternative to antibiotics for the reduction of salmonella infection both in poultry.

Ceci ne viendrait-il pas $peut-\^{e}tre$ do notre $pens\'{e}e$ dans l' $\'{e}change$ culturelle entre le monde et nous qui s' appuie trop une $temporalit\'{e}$ et logique? Cette $the\`{s}e$ approche ce $probl\`{e}me$ $\`{a}$ travers one perception anthropologique do monde, en $\'{e}tudiant$ les arts tridimensionnels avec ses $\'{e}schelles$ qui deviennes plus on plus agrandis. Maurice Merleau-Ponty, dans son livre 'la $ph\'{e}nom\'{e}nologie$ de la perception', bien $r\'{e}sumait$ une $intersubjectivit\'{e}$ entre l' espace et nous. Selon lui, l' homme prend conscience do la forme des objets avec son environnement parce quil est un corps on interaction avec eux. Donc nous $r\'{e}f\'{e}rons$ ce livre pour notre set. Et on essayera ici d' $\'{e}tudier$ le travail do Constantin Brancusi, de Alberto Giacometti, do Richard Serra et do quelques d' autres travaux d' artistes L' oeuvre do Brancusi 'la Colonne sans fin', la taille plus de 29 m, produit one impression sur le spectateur relativement $\grave{a}$ la forme continue et $r\'{e}p\'{e}t\'{e}e$ $\`{a}$ linfini de $mani\'{e}re$ unifome. La pratique de Giacometti qui tenait compte do son envergure et de la longueur do son corps ainsi que do distance et de la mesure do sos bras, Il $d\'{e}formait$ aussi les silhouettes et transformait les objects en les $\'{e}tirant$ dans la perspective. La sculpture et l' architecture $r\'{e}ciproque$ l' espace tridimensionnel entre eux, mais selon l' argument do Herbert Read pour la distinction de ces deux arts originaux sont, deux saisies de l' espace, ici englobant, $l\`{a}$ $englob\'{e}$. On $\'{e}tudie$ dans co set de concept de l' espace et de l' $\'{e}chelle$ d' objet par rapport au corps humain.

Electrokinetic Remediation of Soil Contaminated with Zn, Ni and F (동전기 정화기술을 이용한 Zn, Ni, F 복합오염 토양의 정화)

  • The feasibility of electrokinetic remediation was investigated in the laboratory to treat contaminated soil with Zn, Ni and F. Electro-migration and electro-osmosis are the major removal mechanisms because fluorines desorbed from soil exist as an anionic form in soil pores, and Zn and Ni exist as a cationic form. Desorption of fluorine was enhanced under the alkaline condition, but that of Zn and Ni increased under the acidic condition. Sequential pH control was effective to control the mixed wastes from contaminated soil. 2 V/cm was applied to reactor to evaluate the effect of constant voltage gradient, after two weeks, the removal efficiency of Zn, Ni and F was 20.5%, 2.5% and 57.4%, respectively. Even though the removal of Zn and Ni was very low, the pH control enhanced transport of Zn and Ni significantly. As a result, sequential pH control is a effective method to remediate mixed waste-contaminated soil.

Enzymatic Properties of Pectin Esterase from Aspergillus sp. (Aspergillus속(屬)이 생성(生成)하는 Pectin Esterase의 효소학적(酵素學的) 성질(性質))

  • The enzymatic properties of pectin esterase from Aspergillus sp. were studied. The optimum conditions for the enzyme reaction were pH 4.2 and temperature of $45^{\circ}C$. The crude enzyme was very stable at pH range of $2.2{\sim}4.6$, but about 20 percent of activity was lost at the range of pH $5.4{\sim}8.0$. The crude enzyme was very stable at $50^{\circ}C$ for one hour, however almost 100 percent of enzyme activity was lost at $70^{\circ}C$ for 30 minutes. The pectin esterase activity of crude enzyme was greatly inhibited by addition of sodium chloride at lower pH range. That is, the inhibition rates of enzyme activity at pH 3.0 and 4.0 were 47% and 28% in concentration of 1M sodium chloride, respectively. The enzyme activity was not affected by sodium chloride at pH 7.0 at different concentration of sodium chloride. Although the enzyme activity was not affected by addition of sucrose, it was slightly inhibited at higher concentration of sucrose.

Studies on Heat Sensitivity of Egg Albumen II. Effects of pH and/or the Addition of Metal ions on Heat Sensitivity of Egg Albumen (난백의 숙감수성에 관한 연구 II. 금속염의 첨가와 pH가 난백의 열감수성에 미치는 영향)

  • In order to dull heat sensitivity of egg albumen, metal ions (aluminium, ferric, ferrous, copper) were added and functional properties or egg albumen were determined before and after heat treatment at $60^{\circ}C$ for 5 minutes. Effect of pH on heat sensitivity of aluminium salt added egg albumen was also determined. Addition of metal ions increased turbidity of egg albumen before and after the heat treatment. Changes of the turbidity were minimized by addition of aluminium salt. The foaming power was markedly increased by addition of ferric salt before the heat treatment and increased by addition of aluminium, ferric and copper salt after the heat treatment. Before the heat treatment the foam was stable by addition of ferric and ferrous salt but after the heat treatment it was stable by addition of aluminium and ferric salt. The turbidity and foaming property of the egg albumen with aluminium salt were not largely changed after the heat treatment at pH range 7 to 8.5. Over pH 9 the turbidity and foaming power were not decreased, but the foam stability was increased before and after the heat treatment. Salmonella typhimurium ATCC 14028 (10$^{6}$ cells/$m\ell$) inoculated in egg albumen at pH range 7 to 8.5 was destructed by the heat treatment.

Studies on the Hesperidinase of Aspergillus niger S-1 (Aspergillus niger S-1이 생산하는 Hesperidin 분해효소에 관한 연구)

  • Aspergillus niger S-1 was proved to be a good hesperidinase producer which have been selected for naringinase utilization. Enzyme of this strain had good characteristics and purified relative high degree with good recovery by ammonium sulfate or aceton treatment. Results obtained were summarized as follows (1) The enzyme was most active at 60$^{\circ}C$, when the reaction was performed in the pH 4.0 for 30min. Optimum pH for enzyme activity was 5.0 and activity was retained 78% at pH value 3.5. (2) Hesperidinase activity retained 95% of its full activity after treatment at 60$^{\circ}C$ for 30min at pH value 4.0., 70% at 70$^{\circ}C$ and 65% at 80$^{\circ}C$. Most stable pH of this enzyme was showed 5.0 after treatment for 24hr at 4$^{\circ}C$ (3) Only Magnesium ion activated enzyme reaction, while other metallic ions, Cu$\^$++/, Mn$\^$++/, Pb$\^$++/, Mo$\^$++/, Ag$\^$++/, Hg$\^$++/ inhibited. (4) Eleven fold purification with 35% recovery was obtained in the case of 60% aceton treatment and 10-fold purification with 5.6% recovery was showed with 40% aceton comparing to the crude extract Enzyme. (5) Crude enzyme precipitated with 0.4-0.6 saturated ammonium sulfate contained 13f6 of the original enzyme activity with 48-fold increase in specific activity and enzyme has been purified 25 fold with a yield 19% by 0.6-5.8 saturation. (6) Hesperidinase formation was noticeably increased by addition of small amount of orange-peel extraction on the wheat bran medium.

Production and Action of Microbial Piscicidal Substance (미생물에 의한 살어성물질의 생성 및 그 작용)

  • Piscicidal substance produced by Streptomyces sp. isolated from soil was toxic against various kinds of fish. After extraction with CH$Cl_3$ from the culture medium, the substance was purified by avicel column chromatography. In order to test toxicity, various kinds of fish were subjected to the acqueous solution of 100 us of the substance per liter of water. Generally, the substance was toxic to most fish, but Macropodus chinenes and Misgurnus mizolepis are resistant to the substance than Gobius similis and Pseudorasbora parva. The substance was stable at pH range, 3.0 to 7.0, but labile at alkaline pH, and to heat as well. Succinic dehydrogenase on most of tissue cell of Cyprinus carpio was inhibited by this substance strongly, but spinal cord was not inhibited. By addition of Cu and Pb salts to the culture medium, piscicidal substance producibility was activated.

Formation of D-Glucose Isomerase by Streptomyces sp. (Streptomyces sp.에 의한 포도당 이성화효소의 생성)

  • A source of D-xylose was required for the enhanced production of D-glucose isomerase of Streptomyces sp. strain K-17. D-glucose supported the luxuriant growth of the organism as well as D-xylose, but D-glucose isomerase activity was hardly detected in the D-glucose-grown cells. When the D-glucose-grown cells were incubated aerobically for a few hours in 0.5% xylose solution in 0.05 M phosphate buffer, pH 7.0, it was found that inductive formation of D-glucose isomerase occurred in the cells without multiplication. In the non-growth phase of cells the inductive formation of D-glucose isomerase occurred because a source of nitrogen for the synthesis of enzymes was obtained from turnover of protein accumulated in cells. D-ribose, L-arabinose, D-glucose, D-mannose, citrate, succinate and tartrate could not induce the formation of D-glucose isomerase, but D-xylose could induce. Inductinn of D-glucose isomerase was repressed by D-glucose and its catabolites : glycerol, succinate and citrate. Inductive formation of the enzymes in the non-growth phase was stimulated by $Ba^{2+}$, $Mg^{2+}$ and $Co^{2+}$, and inhibited by C $u^{2+}$, C $d^{2+}$, A $g^{+}$and H $g^{2+}$. The synthesis of enzymes in the induction system composed of 0.5% xylose solution was disrupted by actinomycin D, streptomycin, chloramphenicol, kanamycin, tetracycline, p-chloromercuribenzo ate, arsenate and 2, 4-dinitrophenol, but not disrupted by mitomycin C and penicillin G.icillin G.

Studies on the Cause and Control for Non-Sprouting in the Mulberry Field of Kyung-pook Province (경북지방의 뽕밭에 발생한 발아불량현상의 원인 및 방제에 관한 연구)

  • This study was conducted to bring light on inducing factors for non-sprouting occured in the mulberry field of Kyungpook Province in 1983. The results are as follws ; 1. In spring, winter buds were suddenly died during germination, necrosis appeared in pholem and cortex of non-sprouting stem and measles at the bottom of non-sprouting stem. 2. The depth of available of soil was more shallow in the non-sproutiong field than in healthy field. 3. There was no signifioant difference between the healthy field and non-sprouting field in siol PH, the content of organic matter, available phosphorous and exchangeble cations. 4. Available boron content in soil was significantly more in healthy field than in non-sprouting field. 5. Boron content in leaf and bark was significantly lower in the non-sproutiong tree than in the healthy tree. 6. The non-sprouting fields were completely controlled by the application of 6kg/10a borax. 7. Cold tolerance of the mulberry tree was higher in the mulberry tree with boron than in the mulberry tree without boron. 8. Mulberry tree in Kyungpook Province in 1983 may absorb water earlier compared with the average year because of high temperature at the end of March, 1983 and they had been damaged by frost injuary due to the lowest temperature (-6.4$^{\circ}C$) on the grass at the beginning of April 1983. As the above results, non-sprouting bud in the mulberry fields of Kyungpook Province in 1983 had been occured by low temperature and boron deficiency and boron deficiency of mulberry trees accelerared the frost injury.

  • Deterioration of fish muscle is known to occur more quickly in the dark fleshed fish than in the white fleshed fish, causing by their high intestinal proteolytic activity. Muscle degradation which suffer post-mortem autoproteolysis is affected by trypsin with its unique activation function towards other enzymes. To compare physicochemical and enzymatic properties for the trypsins of the dark fleshed fish, trypsins from the viscera of anchovy (Engraulis japonica), and the pyloric caeca of mackerel (Scomber japonicus), yellowfin tuna (Thunnus albacores) and albacore (Thunnus alalunga) were purified through ammonium sulfate fractionation, benzamidine-Sepharose 6B, DEAE-Sephadex A-50, and Sephadex G-75 chromatography Two trypsins from mackerel (designated mackerel trypsin A and mackerel trypsin B), and one each from anchovy, yellowfin tuna and albacore were isolated as electrophoretical homogeneity, The purities of anchovy trypsin, mackerel trypsin A and B, yellowfin tuna trypsin, and albacore trypsin increased to 78.1, 4.8, 9.3, 120, and 160-fold, respectively, compared to crude enzyme solutions. Molecular weights of the trypsins from the dark fleshed fish estimated by SDS-polyacrylamide electrophoresis were ranged from 22kDa to 26kDa. The trypsins contained higher amount of glycine, serine and aspartic acid, and less amount of tryptophan, methionine, lysine and tyrosine. Optimal conditions for amidotici reactions of the enzymes were pH 8.0 and 45$^{\circ}C$ for anchovy trypsin, pH 8.0 and 5$0^{\circ}C$ for mackerel trypsin A and B, pH 9.0 and 55$^{\circ}C$ for yellowfin tuna trypsin, and pH 9.0 and 5$0^{\circ}C$ for albacore trypsin. It was supposed that the habitat temperature of the dark fleshed fish is slightly connected with the optimal reaction temperature of the trypsins of the fish.
