Purification and Characterization of Xylanase from Bacillus sp. A-6 (Bacillus sp. A-6의 Xylanase의 정제와 특성)

  • A xylanase was purified from the culture supernatant of Bacillus sp. A-6 by using ultrafiltration and ion exchange chromatography on the column of SP-Sepharose using 5 mM acetate buffer, pH 5.0. The xylanase was eluted from the column at the concentration less than 0.05 M NaCl. The eluted xylanase was shown to be a single protein band in SDS-PAGE. Zymogram analysis indicated that the protein band in SDS-PAGE had the enzyme activity to hydrolyze oat spelt xylan. The molecular weights of the xylanase were 15,000 based on SDS-PAGE and 14,100 based on gel filtration chromatography. Thin layer chromatography showed that the xylanase hydrolyzed oat spelt xylan into xylobiose and high-molecular-weight xylooligosaccharides. The relative activities of the heated xylanase decreased to 80% at $40^{\circ}C$ after 7 hr and less than 40% at $60^{\circ}C$ after 1 hr.

Concentration of Polyunsaturated Fatty Acids from Anchovy Oil by Supercritical Carbon Dioxide (초임계 이산화탄소에 의한 멸치어유의 고도불포화지방산 농축)

  • To concentrate polyunsaturated fatty acids from anchovy oil by supercritical carbon dioxide $(SC-CO_2)$, effects of different adsorbents, cosolvents and extraction pressures on the extraction yield and fatty acid composition in the extract and the residue fractions were investigated. Anchovy oil mixed with silver nitrate-coated silica gel showed higher extraction yield and concentration of EPA and DHA in the residual fraction than that mixed with only silica gel at $60^{\circ}C/345{\;}bar$. Ethyl acetate was a promising cosolvent for concentrating polyunsaturated fatty acids in the residual fraction from anchovy oil mixed with silica gel. For the extraction pressures tested, 276 bar showed a maximum value in the extraction yield and concentration of EPA and DHA in the residue. Starting with anchovy oil containing 13.3% EPA and 16.2% DHA mixed with a silver nitrate-coated silica gel, the residue fraction containing 28.2% EPA and 38.3% DHA was obtained when ethyl acetate was used as a cosolvent with $SC-CO_2$ at $60^{\circ}C/276{\;}bar$.

Synthesis and Structural Characterization of Five- and Six-Coordinate Cobalt(Ⅱ) Complexes of Tripodal Liand. Tris-(2-benzimidazolylmethyl)amine

  • The various cobalt(Ⅱ) complexes were synthesized and characterized using tris-(2-benzimidazolylmethyl)amine (ntb) as a ligand where the ntb plays as a tripodal tetradentate ligand to form complexes with a trigonal pyramidal geometry. The complexes have 5 and 6 coordinate cobalt(Ⅱ) ions depending on the additional ligand used. In each complex the additional ligand, chloride anion, or acetate anion occupies the "open" site trans to the apical tertiary nitrogen atom of ntb ligand. Complex 1, [Co(Ⅱ)(ntb)Cl]Cl has a trigonal bipyramidal geometry. This geometry was easily constructed using ntb as a tetradentate ligand and chloride as a monodentate ligand. The complex is isostructural to the corresponding manganese(Ⅱ) complex. Crystal data are as follows: [Co(Ⅱ)(ntb)Cl]Cl·MeOH, 1. triclinic space group P1; a=13.524(2) Å, b=14.037(2) Å, c=17.275(1) Å; α=78.798(9), β=84.159(8)°, γ=65.504(9)°; V=2929.6(6) Å3; Z=4; R1=0.0715, wR2=0.1461 for reflections of I > 2σ(I). Six coordinate complex 2 [Co(ntb)(OAc)](OAc) was synthesized using ntb as a tetradentate ligand and acetate as a bidentate chelating ligand.

  • Condensation of salicylaldehyde $(2-HOC_6H_4C(O)H)$ with allylamine afforded the unsaturated salicylaldimine, $2-HOC_6H_4C(H)=NCH_2CH=CH_2$. Similar reactivity was observed with substituted salicylaldehydes. Further reaction of these Schiff bases with palladium acetate or $Na_2PdCl_4$ afforded complexes of the type $PdL_2$, where L = deprotonated Schiff base. The molecular structure of the parent salicylaldimine palladium complex $[trans-(2-OC_6H_4C(H)=NCH_2CH=CH_2)_2Pd]$ (1) was characterized by an X-ray diffraction study. Crystals of 1 were monoclinic, space group $P2_1/n,\;a\;=\;14.0005(9)\;{\AA},\;b\;=7.2964(5)\;{\AA},\;c\;=\;17.5103(12)\;{\AA},\;{\beta}\;=\;100.189(1)^{\circ}$, Z = 4. Analogous chemistry with 4-vinylaniline also gave novel palladium complexes containing a pendant styryl group. Crystals of $[trans-(2-HOC_6H_4C(H)=N-4-C_6H_4CH=CH_2)_2Pd]$ (4) were monoclinic, space group $P2_1/c$, a = 13.7710(14) ${\AA}$, b = 11.0348(11) ${\AA}$, c = 7.8192(8) ${\AA}$, ${\beta}\;=\;98.817(2)^{\circ}$, Z = 2.

Determination of 5-HT2C agonist KKHQ80114 and KKHQ80109 in Rat Plasma and Urine by Gas-Chromatography/Mass Spectrometry

  • 5-HT$_{2C}$ receptors have been considered as therapeutic targets for the treatment of various central nervous system disorders such as depression, anxiety, epilepsy, schizophrenia and sleep disorders. We chemically synthesized KKHQ80114 (K14) and KKHQ80109 (K09), selective 5-HT$_{2C}$ agonists, with the purpose of developing therapeutic agents for the treatment of obesity. The objective of this work is to investigate analytical methods of these compounds in the plasma and urine of rats by gas chromatography/mass spectrometry. In this experiment, K14 was determined in plasma and urine by using K09 as internal standard. Calibration curves give a good linearity in plasma (r$^2$=0.9993) and urine (r$^2$=0.9988). Among hexane, ethyl acetate and diethyl ether, the highest peak was observed in diethyl ether. However, ethyl acetate was used since more interfering peaks were observed with diethyl ether. Inter-day precision and accuracy were determined in the ranges of 50-500 ng/mL for plasma and 10-500 ng/ml for urine. Quantitation limits were 50 ng/mL plasma and 25 ng/ mL urine. These data may be applicable for further studies of these compounds including absorption and metabolism due to no pharmacokinetic or analytical data available.

Volatile Flavor Components in Pumpking Vinegar

  • Pumpkin venegar was produced using autoclaved pumpkin juice by fermenting with cetobacter starter and ethanol at the ratio of 4 % and 10% to the volume of pumpkin juice, respectivley. Fermentation was carried out at 2$0^{\circ}C$ for 14 days followed by aging at 1$0^{\circ}C$ for 14 days. Flavor components of pumpkin vinegar was determined using GC/MS. Identified components, were 2 aldehydes (4.74%), 5 acohols (30.06%), 4 ketones (8.99%), 4 acids (16.39%), 5 alkanes (11.10%), 11 miscellaneous compounds (27.01%) and 9 unknown compounds (1.71%). Pumpkin vinegar showed very similar flavor characteristics to those of conventional wien vinegar and sherry wine vinegar in particular , acetioin, methyl acetate, and butanoic acid were typical volatile components of these three kinds of vinegar. Pumpkin vinegar showed possiblity to compete with European wine vinegar.

Quantitative Analysis of Phenolic Compounds in Different parts of Panax ginseng C.A. Meyer and Its Inhibitory Effect on Melanin Biosynthesis (인삼의 부위별 페놀성 성분 함량 및 멜라닌 생성억제효과)

  • Comparison of four phenolic compounds among three different parts of Panax ginseng C.A. Meyer was performed in this study. Contents of esculetin, p-coumaric acd, quercetin, maltol in the ethyl acetate fraction were determinated by HPLC analysis. The main root has a highest concentration of p-coumaric acid and the leave has a highest concentration of quercetin and esculetin. This ethyl acetate fraction of the leaves exhibited higher depigmenting activity and lower cell toxicity than the other parts of ginseng. In addition, it showed that quercetin and esculetin among phenolic compounds highly inhibited melanin biosynthesis in ginseng. It implies that leaves of ginseng may be used as skin whitening herb. Quercetin and esculetin proved to be active compounds in the leaves of ginseng.

  • The oral administration of deer horn extract was proved to cause a considerable amelioration to the impairment of hepatic oxidative phosphorylation and to the decrease of ATP content in liver tissue induced by cholesterol ingestion. Deer horn extract was shown to accelerate considerably incorporation of acetate-1-C$^{14}$ into cholesterol in liver tissue and it tends to restore a decrease of GOT activity of liver tissue, caused by cholesterol administration. Histological examination showed that deer horn extract had a remarkable preventive effect against fatty infiltration of organs such as liver, heart, spleen, and adrenal gland of cholesterol given rabbits.

And the chemical structure of its permethyl ether was suggested as 6-[2(3,4-dimethoxyphenyl)ethenyl]-5-(3,4-dimethoxyphenyl)2,8,10-trimethoxybenzo[a] xanthen-9-one by spectroscopic and chemical degradative evidences.

Influence of Medium Composition on the Production of $\gamma$-Linolenic Acid by Mucor sp. KCTC 8405P (Mucor sp. KCTC 8405P의 배지조성이 감마 리놀렌산의 생산에 미치는 영향)

  • As a way to determine the optimal culture conditions for the production of ${\gamma}$-linolenic acid by Mucor sp. KCTC 8405P, the influence of different carbon and nitrogen sources, initial pH, and C/N ratio of medium was investigated. Glucose was found to be the best carbon source in terms of lipid content and ${\gamma}$-linolenic acid yield. Ammonium sulfate and organic nitrogen sources such as urea and peptone resulted in relatively increased lipid and ${\gamma}$-linolenic acid production. The highest accumulation of lipid was obtained at a C/N ratio of 56.6 using glucose and (NH$_4$)$_2$SO$_4$ as carbon and nitrogen source, respectively. It was found that the lipid content increased significantly with increasing initial pH of medium up to pH 9.0. The influence of mixed carbon source on the ${\gamma}$-linolenic acid yield was also investigated. High accumulation of lipids, 315 mg/100 ml medium, and 13-14% of ${\gamma}$-linolenic acid content in the cellular lipid were obtained in a shaking culture containing 3% of glucose and 2% sodium acetate as carbon source and 0.1% of (NH$_4$)$_2$SO$_4$ as nitrogen source at pH 8.0.
