석문(石門)($CV_5$) 애구(艾灸)가 자성(雌性) 백서(白鼠)의 $3{\beta}-hydroxysteroid\;dehydrogenasa/{\Delta}^5-{\Delta}^4$ isomerase $(3{\beta}-HSD)$의 발현(發顯) 양상(樣相)에 미치는 영향(影響) (Effects of Moxibustion at the CV5(Serk-Moon) on the Expression of $3{\beta}-Hydroxysteroid\; dehydrogenasa/{\Delta}5-{\Delta}4$ isomerase $(3{\beta}-HSD)$ in Ovary of the Rats)

  • We demonstrated the moxibution on CV5(Serk-Moon) and CV3(Chung-Guk), as we have known contraceptive acupuncture point and pregnancy acupuncture point in oriental medcine, effected on concentration of progesteron and estrogen. As the reports, $3{\beta}-hydroxysteroid\;dehydrogenase/{\Delta}5-{\Delta}4$ isomerase $(3{\beta}-HSD)$ is an enzyme that has catalytic oxidation and isomerase reaction on steroidhormon synthase including progesteron and estrogen. So we observated the concentration and mass of $(3{\beta}-HSD)$ by immuno-chemicalstain, western blot and Enzyme-Linked Immunosorbent Assay in rat ovary. In results, we detected that the mass of $(3{\beta}-HSD)$ decreased significantly in moxibution treated group on CV5($p{\leq}0.05$) in rat ovary and proposed that the moxibution of CV5 make possible to contraception because of blocking estrogen synthesis by $(3{\beta}-HSD)$ decrease. But we didn't know moxibution effects exactly whether is biochemical reaction or thermal reaction on acupuncture point.

무지개송어 (Oncorhynchus mykiss)의 난소내 협막층(theca layers)에서의 Pregnenolone 대사: cyanoketone과 trilostane의 저해 효과 (Pregnenolone Metabolism in the Ovarian Thecal Layers of the Rainbow Trout, Oncorhynchus mykiss: in vitro Inhibitory Effects of Cyanoketone and Trilostane)

  • [ $3\beta-$ ]히드록시-$\Delta^5$-스테로이드 탈수소효소 ($3\betaβ-HSD,$ $3\beta-hydroxy-\Delta^5-steroid\;dehydrogenase$ $\Delta^5$-스테로이드$\rightarrow\Delta^4$-스테로이드로의 대사경로에 관여하는 효소)에 대하여 특이적 저해제인 cyanoketone과 trilostane의 저해 효과가 $^3H-pregnenolone$ 전구체를 이용하여 무지개송어 난소에서 분리한 여포층, 협막층(theca layers)을 대상으로 비교 관찰되었다. Pregnenolone으로부터 $17\alpha-hydroxyprogesterone$으로의 대사과정에서 주요 효소인 $(3\beta-HSD$ 활성은cyanoketone $10^{-6}$$10^{-5}\;M$, 그리고 trilostane $10^{-5}$$10^{-4}\;M$의 농도에서 억제되었으며, trilostane이 cyanoketone보다 더 효과적인 억제반응을 보이는 것으로 나타났다. Pregnenolone으로부터 $\Delta^4$-스테로이드 대사물 축적에 대한 trilostane의 저해작용은 사용한 농도 즉, $10^{-8}, 10^{-7}, 10^{-6}$ 그리고 $10^{-5}\;M$에 비례하여 나타났으나 완전한 저해효과는 보이지 않았다.

Antisteroidogenic activity of Raphanus sativus seed extract in female albino mice

  • The defatted methanol extract of Raphanus sativus Linn. (Cruciferae) seed (MERS) was evaluated for its antisteroidogenic potential in mature female Swiss albino mice. The methanol extract at the doses of 100 and 200 mg/kg body weight significantly elevated the levels of cholesterol and ascorbic acid contents which serve as a precursor for the synthesis of steroid hormones in ovaries. The extract also significantly inhibited glucose-6-phosphate dehydrogenase and ${\Delta}^5-3{\beta}$-hydroxy steroid dehydrogenase, the two key enzymes involved in ovarian steroidogenesis. Hence the extract (MERS) exhibited significant antisteroidogenic activity.

Effect of Soyabean Isoflavones Exposure on Onset of Puberty, Serum Hormone Concentration and Gene Expression in Hypothalamus, Pituitary Gland and Ovary of Female Bama Miniature Pigs

  • This study was to investigate the effect of soyabean isoflavones (SIF) on onset of puberty, serum hormone concentration, and gene expression in hypothalamus, pituitary and ovary of female Bama miniature pigs. Fifty five, 35-days old pigs were randomly assigned into 5 treatment groups consisting of 11 pigs per treatment. Results showed that dietary supplementation of varying dosage (0, 250, 500, and 1,250 mg/kg) of SIF induced puberty delay of the pigs with the age of puberty of pigs fed basal diet supplemented with 1,250 mg/kg SIF was significantly higher (p<0.05) compared to control. Supplementation of SIF or estradiol valerate (EV) reduced (p<0.05) serum gonadotrophin releasing hormone and luteinizing hormone concentration, but increased follicle-stimulating hormone concentration in pigs at 4 months of age. The expression of KiSS-1 metastasis-suppressor (KISS1), steroidogenic acute regulatory protein (StAR) and 3-beta-hydroxysteroid dehydrogenase/delta-5-delta-4 isomerase ($3{\beta}-HSD$) was reduced (p<0.01) in SIF-supplemented groups. Expression of gonadotropin-releasing hormone receptor in the pituitary of miniature pigs was reduced (p<0.05) compared to the control when exposed to 250, 1,250 mg/kg SIF and EV. Pigs on 250 mg/kg SIF and EV also showed reduced (p<0.05) expression of cytochrome P450 19A1 compared to the control. Our results indicated that dietary supplementation of SIF induced puberty delay, which may be due to down-regulation of key genes that play vital roles in the synthesis of steroid hormones.
