Dual positional substrate specificity of rice allene oxide synthase-1: insight into mechanism of inhibition by type II ligand imidazole

  • Phylogenetic and amino acid sequence analysis indicated that rice allene oxide synthase-1 (OsAOS1) is CYP74, and is clearly distinct from CYP74B, C and D subfamilies. Regio- and stereo-chemical analysis revealed the dual substrate specificity of OsAOS1 for (cis,trans)-configurational isomers of 13(S)- and 9(S)-hydroperoxyoctadecadienoic acid. GC-MS analysis showed that OsAOS1 converts 13(S)- and 9(S)-hydroperoxyoctadecadi(tri)enoic acid into their corresponding allene oxide. UV-Visible spectral analysis of native OsAOS1 revealed a Soret maximum at 393 nm, which shifted to 424 nm with several clean isobestic points upon binding of OsAOS1 to imidazole. The spectral shift induced by imidazole correlated with inhibition of OsAOS1 activity, implying that imidazole may coordinate to ferric heme iron, triggering a heme-iron transition from high spin state to low spin state. The implications and significance of a putative type II ligand-induced spin state transition in OsAOS1 are discussed.

  • A Process of 3-D Particle image velocimetry, called here, as '3-D volume PIV' was developed for the full-field measurement of 3-D complex flows. The present method includes the coordinate transformation from image to camera, calibration of camera by a calibrator based on the collinear equation, stereo matching of particles by the approximation of the epipolar lines, accurate calculation of 3-D particle positions, identification of velocity vectors by 3-D cross-correlation equation, removal of error vectors by a statistical method followed by a continuity equation criterior, and finally 3-D animation as the post processing. In principle, as two frame images only are necessary for the single instantaneous analysis 3-D flow field, more effective vectors are obtainable contrary to the previous multi-frame vector algorithm. An Experimental system was also used for the application of the proposed method. Three analog CCD camera and a Halogen lamp illumination were adopted to capture the wake flow behind a bluff obstacle. Among 200 effective particle s in two consecutive frames, 170 vectors were obtained averagely in the present study.

  • A value net is a business design that uses digital supply chain concepts to achieve both superior customer satisfaction and company profitability. In order to implement the value net model, information processing and distribution needs to occur in real time. Software agent technology is becoming popular due to the inherent characteristics of autonomy, distributedness and modularity. In this paper, we adopt agent technology to handle all real time decision process, making the value net model a complex multi-agent network of decision makers. For the agents to properly coordinate their respective activities we develop MAVN model, a Web-based multi-agent language grounded in the XML and Java.

  • The objective of the present study is to investigate experimentally the mean flow characteristics of the three-dimensional turbulent boundary layer over a rotating disk with an impinging jet at the center of the disk, which may be regarded as one of the simplest models for the flow in turbomachinery. A relatively strong radial outflow (crossflow) generated from the impinging jet is added to the radial outflow (crossflow) induced by the centrifugal force in order to create the three-dimensional boundary layer. A new calibration technique has been introduced to determine the velocity direction and magnitude using an I-wire probe, where the uncertainties are ${\pm}1.5^{\circ}$ and ${\pm}0.35\;m/s$, respectively, in the laminar boundary layer region, compared with the known exact solutions. The flow in the tangential direction is of similar type to that associated with a favorable pressure gradient, considering that no wake region appears in wall coordinate velocity profiles and the Clauser shape factor is between 4.0 and 5.3. The flow angle is significantly changed by the crossflow generated by the impinging jet.

  • It is very important to understand characteristics of solution due to the variation of computational grid sizes, especially when turbulence model not incorporating wall-function is used. The present paper performs numerical investigation on the grid dependency of numerical solution for three dimensional turbulent flow field around a ship. In the present study a finite volume method with a modified sub-grid scale turbulence model and a numerically constructed non-orthogonal curvilinear coordinate system capable of conforming complex ship geometries are used. Numerical studies are then performed for a mathematical Wigley hull and the Series 60, $C_B=0.8$ hull forms. The results for various grid sizes are compared with each other and with measured data to show grid dependencies of numerical solutions.

  • The recognition of the esthetics as well as their function of the teeth in the dental field is growing. Evaluating the esthetics of a tooth concerns shape, texture, and color. And among these, color has been the most difficult element to make natural expression. Color is a very subjective element affected by many different factor like light source, object, observer. Numerous efforts to quantifying color has been made. Currently the use of colorimeter and spectrophotometercolor that the color measuring device has been suggested. In this study, the normal range of the tooth color in canines, lateral incisors and central incisors for Koreans was evaluated with a modified spectrophotometer for intraoral use. The color differences in terms of teeth positions, age and area within the tooth was evaluated and comparatively analyzed. Results were as follow ; 1. In all age groups, the central incisors are most bright and least yellowish opposit to the canines are most dark and yellowish. The central incisors and lateral incisors showed the similar color patterns. 2. In the comparison between gender, it showed that in the 10' s group male have more yellowish, the 20's female have lighter and more redness, the 30's indicated more redness in female and in the 40's the lightness is more in female. 3. With aging, there was no significant change in $L^*$ and $a^*$ coordinates but a shift was in $b^*$ to yellowish in the 30' sand 40's in central incisors. Lateral incisors and canines showed a lighter and more yellowish tooth color with an irregular change of color coordinate in $a^*$. 4. In all teeth the middle and cervical portion showed close correlation with over 0.7(0.05>p) but color ($a^*$, $b^*$) showed higher correlation than lightness($L^*$). 5. It has been shown that through the regression test the cervical color could be calculated by the middle color value.

  • Wireless LAN (WLAN) has greatly benefited from the introduction of various technologies, such as MAC protocol and scheduling algorithm. The majority of these technologies focus on fairness or service differentiation. However, it is difficult to use these technologies to provide many benefits to WLAN simultaneously because the current WLAN system only focuses on the provision of a single aspect of QoS. Unfortunately, multimedia applications require both service differentiation and fairness. Therefore, this paper combines Distributed Fair Scheduling (DFS) and Enhanced Distributed Coordinate Function (EDCF), to provide both fairness and service differentiation simultaneously. Furthermore, we show numerical analysis using Markov process. The simulation results demonstrate that F-EDCF outperforms the EDCF, in terms of throughput, fairness, and delay viewpoints.

  • A 30kVA superconducting generator (S.G.) has been designed, developed and successfully tested in KERI before the end of last year. The design is based on 2-dimensional electromagnetic field analysis of cylindrical coordinate. The field winding of rotor has been wound with superconducting wire of Nb-Ti alloy and tested after assembled with other rotor components. The stator has air-gap type armature windings which allow higher terminal voltage and more sinusoidal voltage waveform than conventional iron cored machines. Steady-state open & short-circuit test and lamp load test have been conducted also. The results of tests are given in this paper and compared with design quantities. Moreover the cooling scheme and characteristics of test system is included.

  • In this paper, a new onto-digital stereo object-tracking system using hierarchical digital algorithms and optical BPEJTC is proposed. This proposed system can adaptively track a moving target by controlling the convergence of stereo camera. firstly, the target is detected through the background matching of the sequential input images by using optical BPEJTC and then the target area is segmented by using the target projection mask which is composed by hierarchical digital processing of image subtraction, logical operation and morphological filtering. Secondly, the location's coordinate of the moving target object for each of the sequential input frames can be extracted through carrying out optical BPEJTC between the reference image of the target region mask and the stereo input image. Finally, the convergence and pan/tilt of stereo camera can be sequentially controlled by using these target coordinate values and the target can be kept in tracking. Also, a possibility of real-time implementation of the adaptive stereo object tracking system is suggested through optically implementing the proposed target extraction and convergence control algorithms.

Leksell Frame-Based Stereotactic Biopsy for Infratentorial Tumor : Practical Tips and Considerations

  • The Leksell frame-based transcerebellar approach was proposed with the arc support frame attached upside down to the Z coordinate. This study presented practical tips and considerations for obtaining adequate tissue samples for deep-seated cerebellar lesions or lower brainstem lesions specifically those accessible via the cerebellar peduncle. For practical insights, the Leksell coordinate frame G was fixed to prevent the anterior screw implantation within the temporalis muscle, to avoid interference with the magnetic resonance (MR)-adapter, and taking into account the magnetic field of MR in close proximity to the tentorium. After mounting of indicator box, the MR imaging evaluation should cover both the indicator box and the infratentorial region that deviated from it. The coordinates [X, Y, Za, Arc0, Ringa0] obtained from Leksell SurgiPlan® software (Elekta, Stockholm, Sweden) with arc 00 located on the patient's right side were converted to [X, Y, Zb=360-Za, Arc0, Ringb0=Ringa0-1800]. The operation was performed in the prone position under general anesthesia in four patients with deep cerebellar (n=3) and brainstem (n=1) tumors. The biopsy results showed two cases of diffuse large B-cell lymphoma, one metastatic braintumor and one glioblastoma. One patient required frame repositioning as a complication. Drawing upon the methodology outlined in existing literature, we anticipate that imparting supplementary expertise could render the stereotactic biopsy of infratentorial tumors more consistent and manageable for the practitioner, thereby facilitating adequate tissue samples and minimizing patient complications.
