Annual Variations in Community Structure of Mesozooplankton by Short-term Sampling in Jangmok Harbor of Jinhae Bay (진해만의 장목항에서 단주기 샘플링에 의한 중형동물플랑크톤 군집의 연변동)

  • The annual variation of mesozooplankton community in the Jangmok harbor of Jinhae Bay was studied in relation to environmental variables. Sampling was carried out weekly from January to December 2009. During the study periods, mesozooplankton community consisted of 44 taxa and the annual mean abundance was 8308 inds. $m^{-3}$. The maximum abundance was observed to be 50043 inds. $m^{-3}$ in August and the minimum in April with 1013 inds. $m^{-3}$. Of these, Penilia avirostris, cirripedia larvae, Evadne tergestina, Acartia omorii, Oikopleura s, Paracalanus parvus s. l., Eurytemora pacifica, Podon s, Oithona s, and Acartia steueri were observed as dominant species in Jangmok bay and they also contributed to 79% of total mesozooplankton. According to non-metric multidimensional scaling (nMDS) and cluster analysis based on the mesozooplankton community data from each season, the community was divided into three groups. The first group included appearence species in winter and spring season, which is mainly dominated the copepod such as A. omorii and E. pacifica. The second and third group was composed with observed species in summer and autumn, respectively. Based on the SIMPER (similarity percentages), P. avirostris in summer and cirripedia larvae in autumn were significantly dominated. Our results indicate that although the mesozooplankton abundances in Jangmok harbor fluctuated abruptly, its annual variation was strongly influenced by water temperature.

Rational Introduction of Disulfide Bond to Enhance Optimal Temperature of Lipomyces starkeyi $\alpha$-Dextranase Expressed in Pichia pastoris

  • $\alpha$-Dextranase, which can hydrolyze dextran, is largely used in the sugar industry. However, a thermostable $\alpha$-dextranase is needed to alleviate the viscosity of syrups and clean blocked machines. Thus, to improve the optimal temperature of Lipomyces starkeyi $\alpha$-dextranase expressed by Pichia pastoris, the rational introduction of a de novo designed disulfide bond was investigated. Based on the known structure of Penicillium minioluteum dextranase, L. starkeyi $\alpha$-dextranase was constructed using homology modeling. Four amino acids residues were then selected for site-directed mutagenesis to cysteine. When compared with the wild-type dextranase, the mutant DexM2 (D279C/S289C) showed a more than $13^{\circ}C$ improvement on its optimal temperature. DexM2 and DexM12 (T245C/N248C, D279C/S289C) also showed a better thermal stability than the wild-type dextranase. After the introduction of two disulfide bonds, the specific activity of DexM12 was evaluated and found to be two times higher than that of the wild-type. Moreover, DexM12 also showed the highest $V_{max}$.

ACE-Inhibitory Properties of Proteolytic Hydrolysates from Giant Jellyfish Nemopilema nomurai

  • This study aimed to determine the degree of hydrolysis and angiotensin-I-converting enzyme (ACE)-inhibitory activity of Giant Jellyfish Nemopilema nomurai (jellyfish) hydrolysates. The degree of hydrolysis using six proteolytic enzymes (Alcalase, Flavozyme, Neutrase, papain, Protamex, and trypsin) ranged from 13.1-36.8% and the inhibitory activities from 20.46-79.58%. Using papain hydrolysate, we newly isolated and characterized ACE-inhibitory peptides with a molecular weight of 3,000-5,000 Da that originated from jellyfish collagen. The purified peptide (FII-b) was predicted to be produced from an alpha-2 fragment of the type IV collagen of jellyfish. The N-terminal sequence of FII-b was Asp-Pro-Gly-Leu-Glu-Gly-Ala-His-Gly- and showed 87% identity to the collagen type IV alpha-2 fragment of Rattus norvegicus and a predicted protein from Nematostella vectensis, indicating that the ACE-inhibitory peptide originated from the collagen hydrolysate and had an $IC_{50}$ value of 3.8 ${\mu}g$/mL. The primary structure of the fragment is now being studied; this peptide represents an interesting new type of ACE inhibitor and will provide knowledge of the potential applications of jellyfish components as therapies for hypertension.

Anticancer activity and potential mechanisms of 1C, a ginseng saponin derivative, on prostate cancer cells

  • Background: AD-2 (20(R)-dammarane-3b, 12b, 20, 25-tetrol; 25-OH-PPD) is a ginsenoside and isolated from Panax ginseng, showing anticancer activity against extensive human cancer cell lines. In this study, effects and mechanisms of 1C ((20R)-3b-O-(L-alanyl)-dammarane-12b, 20, 25-triol), a modified version of AD-2, were evaluated for its development as a novel anticancer drug. Methods: MTT assay was performed to evaluate cell cytotoxic activity. Cell cycle and levels of reactive oxygen species (ROS) were determined using flow cytometry analysis. Western blotting was employed to analyze signaling pathways. Results: 1C concentration-dependently reduces prostate cancer cell viability without affecting normal human gastric epithelial cell line-1 viability. In LNCaP prostate cancer cells, 1C triggered apoptosis via Bcl-2 family-mediated mitochondria pathway, downregulated expression of mouse double minute 2, upregulated expression of p53 and stimulated ROS production. ROS scavenger, N-acetylcysteine, can attenuate 1C-induced apoptosis. 1C also inhibited the proliferation of LNCaP cells through inhibition on $Wnt/{\beta}-catenin$ signaling pathway. Conclusion: 1C shows obvious anticancer activity based on inducing cell apoptosis by Bcl-2 family-mediated mitochondria pathway and ROS production, inhibiting $Wnt/{\beta}-catenin$ signaling pathway. These findings demonstrate that 1C may provide leads as a potential agent for cancer therapy.

Evolution of Magnetic Property in Ultra Thin NiFe Films (나노두께 퍼말로이에서의 계면효과에 의한 자기적 물성 변화)

  • We prepared ultra thin film structure of Si(100)/ $SiO_2$(200 nm)/Ta(5 nm)/Ni$_{80}$Fe$_{20/(l~15 nm)}$Ta(5 nm) using an inductively coupled plasma(ICP) helicon sputter. Magnetic properties and cross-sectional microstructures were investigated with a superconduction quantum interference device(SQUID) and a transmission electron microscope(TEM), respectively. We report that NiFe films of sub-3 nm thickness show the B$_{bulk}$ = 0 and B$_{surf}$=-3 ${\times}$ 10$^{-7}$(J/$m^2$). Moreover, Curie temperature may be lowered by decreasing thickness. Coercivity become larger as temperature decreased with 0.5 nm - thick Ta/NiFe interface intermixing. Our result implies that effective magnetic properties of magnetoelastic anisotropy, saturation magnetization, and coercivity may change abruptly in nano-thick films. Thus we should consider those abrupt changes in designing nano-devices such as MRAM applications.

Statistical Analysis for Turbulence Properties of the Orion Molecular Cloud

  • Turbulence plays an important role in molecular clouds. However, the properties of turbulence are poorly understood. In order to study the influence of turbulence in molecular clouds, we need to sample the turbulent properties in the full range of scales down to sonic scale. We mapped the $20^{\prime}{\times}60^{\prime}$ area covering the Orion Molecular Cloud (OMC) 1-4 region in HCN 1-0 and HCO+ 1-0 with Taeduk Radio Astronomy Observatory (TRAO) 14-m telescope as part of the TRAO key science program, "Mapping turbulent properties of star-forming molecular clouds down to the sonic scale (PI: Jeong-Eun Lee)". In addition, we combine our TRAO data with other molecular line maps ($^{13}CO$ 1-0, $C^{18}O$ 1-0, CS 1-0, $N_2H^+$ 1-0) obtained with the Nobeyama Radio Observatory (NRO) 45-m telescope. To analyze these data, we apply statistical methods, the principal component analysis (PCA) and spectral correlation function (SCF), which are known to be useful to study underlying turbulent properties and to quantitatively characterize cloud structure. We will present the preliminary results of observations and analyses.

Directed Evolution of Beta-galactosidase from Escherichia coli into Beta-glucuronidase

  • In vitro directed evolution through DNA shuffling is a powerful molecular tool for creation of new biological phenotypes. E. coli $\beta$-galactosidase and $\beta$-glucuronidase are widely used, and their biological function, catalytic mechanism, and molecular structures are well characterized. We applied an in vitro directed evolution strategy through DNA shuffling and obtained five mutants named YG6764, YG6768, YG6769, YG6770 and YG6771 after two rounds of DNA shuffling and screening, which exhibited more $\beta$-glucuronidase activity than wild-type $\beta$-galactosidase. These variants had mutations at fourteen nucleic acid sites, resulting in changes in ten amino acids: S193N, T266A, Q267R, V411A, D448G, G466A, L527I, M543I, Q626R and Q951R. We expressed and purified those mutant proteins. Compared to the wild-type protein, five mutant proteins exhibited high $\beta$-glucuronidase activity. The comparison of molecular models of the mutated and wildtype enzymes revealed the relationship between protein function and structural modification.

Analysis of Treatment Efficiency according to Open-water in Constructed Wetland (인공습지 내 개방수역 조성에 따른 처리효율분석)

  • The field scale experiment which is constructed with four sets (0.88 ha for each set) of wetland (0.8 ha) and pond (0.08 ha) systems was performed to examine the effect of plant coverage on the constructed wetland performance and to recommend the optimum development and management of macrophyte communities. After six growing seasons of wetlands, plant coverage was about 100%. And the concentration of DO showed low value (1.0~5.4 mg/L). This is caused by a blighted plant consumed dissolved oxygen with decay in water column. As the result, water column went to be anaerobic conditions and T-N removal rate are 58~67%. Dead vegetation increased nitrogen removal during winter because it is a source of organic carbon which is an essential parameter in denitrification. However, wetland released phosphorus caused by a blighted plant and accumulation, the removal rate of phosphorus might be decreased. To rise of DO concentration, the three open-waters were constructed in cell 3 and 4. Cell 3 has two open-waters (width 10 m, depth 1.8 m) and cell 4 has one open-water (width 20 m, depth 1.8 m). As the result, DO concentration and treatment efficiency of nutrient and BOD were improved. In case that constructed wetland is operated for a long time, physical circulation structure such as open water help continuous circulation of aerobic and anaerobic conditions. Through the constructed open-water, treatment efficiency of phosphorus and nitrogen in wetland could be improved effectively.

  • This paper introduces the high-resolution channel estimation technique which are used to estimate the first arrival multipath delay component. The proposed technique yields the precise estimate of the first time arrival which is directly related to the performance of TDOA-based position location. The proposed technique utilizes the transformed auto-correlation function of received common pilot signal in frequency domain, its samples compose the hermitian Toeplitz matrix at sequel. Then the time delay components could be estimated with precision by the analysis of eigen-structure of corresponding matrix. In this paper, obeying the modified CODIT model, the performance of the PR-IPDL(Pseudo Random-Idle Period Downlink) and TA-IPDL(Time Aligned-Idle Period Downlink considered as 3GPP position location technique will be exploited systematically through the computer simulations with applying the proposed technique.

  • Spot welding is the primary method of joining sheet metals in the automotive industry. As automobiles are subjected to fatigue loading, some spot welds may fracture before the whole system has failed. This local fracture of spot welds may lead to change in the dynamic response and consequently affect fatigue behavior of an automobile. Therefore, this change in dynamic response should be taken into consideration to assess the fatigue life of structures subjected to spectrum loading, such as automobiles. In this study, vibration fatigue analysis was performed by taking into consideration the change in the dynamic response due to accumulated damage at spot-welded parts. Fatigue tests were carried out on tensile-shear spot-welded specimens under constant amplitude loading condition. And the fatigue life of spot welds under spectrum loading was predicted using vibration fatigue analysis method based on finite element analysis.
