본 연구는 1-세포기배의 glucose에 대한 노출이 상실배기 이후의 배발생에 미치는 영향을 검토하고자 실시되었다. hCG 주사 후 24~25시간 째에, F1hybrid(C57BL/6, ♀ $\times$CBA/N, ♂) 계통 생쥐를 도살하여 1-세포기배를 회수한 후 0.1% hyaluronidase로 처리하여 난구세포를 제거하였다. 1-세포기배는 hCG 주사 후 72시간째 다양한 농도의 glucose(5.5, 16.5, 27.5 및 38.5mM)에 1분 노출 후 glucose가 첨가되어 있거나 혹은 첨가되지 않은 CR1aa배양액에서 계속 배양함으로써 배발생을 유도하였다. 이 실험의 결과를 요약하면 다음과 같다. 1. M2 배양액에서 회수한 후 3mg/ml의 Fatty-acid free BSA가 첨가된 배양액에서 배양한 경우 27.5%의 확장배반포까지의 배발생율과 16.6%의 탈출 배반포까지의 배발생율을 나타낸 반면, TL HEPES 배양액에서 회수한 경우는 전혀 상실배기 이후의 배발생이 나타나지 않았다. 2. hCG 주사 후 72시간째에 단 1분간의 27.5mM glucose에 대한 노출만으로도 68.8% (CR1aa+BSA)와 77.1%(CR1aa+FBS)의 확장배반포까지의 발생을 유도할 수 있었다. 그러나 1분 노출과 이후 계속되는 노출간에는 배발생에 있어서 유의차는 인정되지 않았다. 3. hCG 주사 후 72시간째에 5.5, 16.5, 27.5 및 38.5mM의 glucose 첨가에 따른 확장 배반포까지의 배발생율은 45.7~61.5%로 각 처리군의 유의차는 없었으며, 따라서 고농도의 glucose 첨가에 따른 저해효과는 확인할 수 없었다.
순무 싹의 생리활성 기능과 이용 가능성에 관한 연구의 일환으로 순무 싹의 영양성분 및 생리활성 효능을 검증하여 측정한 결과는 다음과 같다. 순무 싹 분말의 일반성분은 건물(dry basis)을 기준으로 수분함량은 2.35%, 조단백질 22.51%, 조지방 21.60%, 조회분 4.35% 및 탄수화물 49.19%를 함유하였다. 구성당은 glucose가 73.39 mg/L로 가장 많았고, fructose가 12.62 mg/L 검출되어 총 2종이 검출되었다. 아미노산은 glutamic acid 함량이 242.57 mg%로 가장 많이 함유되었고, 다음으로 leucine, aspartic acid, valine 순이었다. 순무 싹의 구성 지방산 중 포화지방산 함량은 tricosanoic acid가 가장 높았으며 heneicosanoic acid, palmitic acid 순으로 나타났다. 불포화지방산은 linoleic acid가 가장 높았으며 oleic acid, linolenic acid 순으로 나타났다. 유기산은 총 2종의 유기산이 검출되었으며, tartaric acid 함량이 2,962.74 mg/L로 가장 높았고 다음으로 malic acid 순이었다. 비타민 A와 E의 함량은 각각 0.09 mg%와 3.06 mg%이었다. 무기질은 총 8종의 성분이 검출되었고, 이중 K 함량이 가장 많았으며, 다음으로 Mg, Ca, Na 순이었고 Fe, Zn, Mn, Cu의 함량은 미량이었다. 순무 싹 에탄올 추출물의 기능성을 측정한 결과 순무 싹 에탄올 추출물 100 ${\mu}g$/mL 농도에서 35.24%의 HMG-CoA reductase 저해활성을 나타내었다. 또한 항산화 활성은 시료의 농도가 1,000 ${\mu}g$/mL일 때 아질산염 소거능 및 DPPH radical 소거능이 각각 64.25% 및 69.29%로 매우 높은 항산화 활성을 나타내었다. 이상의 결과 순무 싹 분말은 필수아미노산을 비롯한 항산화 비타민과 무기질을 다량 함유하고 있으며 순무 싹 에탄올 추출물은 HMG-CoA reductase 저해활성 및 항산화 활성이 우수한 것으로 나타나 순무 싹을 이용한 기능성식품의 개발가치가 한층 더 높아질 것으로 기대되어진다.
This study was performed to analyze the distribution of air current speed, $CO_2$ concentration, and photosynthetic photon flux (PPF) in a closed transplants production system (CTPS) for producing quality transplants. And the effect of PPF on the growth of potato (Solanum tuberosum L. cv. Dejima) plug seedlings was analyzed. Uniformity of the air current speed in CTPS was improved by installing perforated floors in duct for air circulating and by adjusting of air flow rate of the fan connected to air conditioning unit used in this study, Measured $CO_2$ concentrations were measured $409{\pm}13$, $950{\pm}25$, and $1,550{\pm}35\;{\mu}mol{\cdot}mol^{-1}$ for setting values of 400, 950, and $1,550\;{\mu}mol{\cdot}mol^{-1}$, respectively. Uniformity of PPF by adding each one the single fluorescent lamp of 20 W at both ends of the single fluorescent lamps of 40 W was highly improved. While the average PPF measured under the twin fluorescent lamps of 55 W installed at regular intervals of 10 cm was decreased by increasing the vertical distance from the lighting sources, the ratio of average PPF measured at both ends to PPF measured in the center was 74-79%. Five levels ($100{\pm}9$, $150{\pm}14$, $200{\pm}17$, $250{\pm}24$ and $300{\pm}31{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) of PPF were provided to investigate the effect of PPF on plant height, fresh weight and dry weight of potato plug seedlings produced in CTPS. Plant height was decreased by increasing PPF. Maximum fresh weight and dry weight were shown under PPF of $250{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. Thus PPF of $250\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ was enough to produce quality potato transplants under air temperature, photoperiod, and relative humidity of $20^{\circ}C$, 16/8 h, and 70%, respectively. It was concluded that quality indices such as plant height, fresh weight and dry weight could be improved by illuminating of adequate PPF from artificial lighting sources.
To investigate an efficacy of mixed extract with Ginseng radix, Puerariae lobata, Puerariae radix, Rubi pructus, Gomi pructus, Hoelen, Dried orange peel and Parvum comus cervi etc., on the hangover elimination, 12 hr-fasted male Sprague-Dawley rats weighing 150-200 g were given mixed extract (5 mL/kg, p.o.) and administered ethanol at a dose of 3 g/kg bw (25% in distilled water) orally 30 min postdosing. Blood was collected from caudal artery at 0.25, 0.5, 1, 2, 4, 6, 8, 10, and 12hr and then the animals were sacrificed at 24hr after the ethanol treatment. In these experiments, liver function indices, such as alanine aminotransferase and alkaline phosphatase activities, showed unaltered results in all treated groups compared with the normal group. The pharmacokinetics of ethanol after oral administration of mixed extract were also evaluated. From 0 min to 12hr, the administration of mixed extract showed 14% reduction of the area under the serum concentrations-versus-time curves (AUC) compared with the control group. The activities of alcohol dehydrogenase and aldehyde dehydrogenase measured at 24hr postdosing were also not altered by the administration of mixed extract compared with the control group. These studies demonstrate that oral administration of mixed extract, prepared by traditional prescription, decreases the ethanol concentration in serum and reduces AUC, suggesting that the mixed extract is effective for elimination of ethanol-induced hangover.
본 연구는 산업단지의 화학제품제조업 및 석유화학제품제조업에서 발생하는 유기성슬러지에 대하여 삼성분, 원소조성, 발열량, 열중량 분석 및 연소시험을 실시하였다. 삼성분 분석 결과 "화합물 및 화학제품 제조업"의 평균값은 수분함량 75.11, 가연분 17.42, 불연분 7.45%로 나타났으며, "코크스, 석유 정제품 및 핵연료 제조업"의 경우 평균값은 수분함량 77.54, 가연분 18.25, 불연분 4.22%로 나타났다. 원소분석 결과 평균값은 "화합물 및 화학제품 제조업"의 경우 C 33.06, H 4.34, O 24.81, N 5.18, S 0.72wt%로 나타났으며, "코크스, 석유 정제품 및 핵연료 제조업"의 경우 C 36.58, H 4.74, O 26.79, N 5.09, S 0.49wt%로 나타났다. 열중량분석 결과, B사에서 배출되는 슬러지는 $700^{\circ}C$ 이상에서 그리고 F와 N사에서 배출되는 슬러지는 $600^{\circ}C$ 정도의 온도에서도 연소가 가능할 것으로 판단된다. 연소테스트 결과, 산화반응과 동시에 열분해 반응으로 고농도의 일산화탄소가 배출되므로 이로 인한 로의 국부과열이나 파손현상에 대한 기초자료를 얻는데 이용될 수 있을 것으로 판단된다.
BACKGROUND/OBJECTIVES: Gastrodia elata Blume (GEB), a traditional herbal medicine, has been used to treat a wide range of neurological disorders (e.g., paralysis and stroke) and skin problems (e.g., atopic dermatitis and eczema) in oriental medicine. This study was designed to investigate whether GEB extract inhibits melanogenesis activity in murine B16F10 melanoma. MATERIALS/METHOD: Murine B16F10 cells were treated with 0-5 mg/mL of GEB extract or $400{\mu}g/mL$ arbutin (a positive control) for 72 h after treatment with/without 200 nM alpha-melanocyte stimulating hormone (${\alpha}$-MSH) for 24 h. Melanin concentration, tyrosinase activity, mRNA levels, and protein expression of microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein (Trp)1, and Trp2 were analyzed in ${\alpha}$-MSH-untreated and ${\alpha}$-MSH-treated B16F10 cells. RESULTS: Treatment with 200 nM ${\alpha}$-MSH induced almost 2-fold melanin synthesis and tyrosinase activity along with increased mRNA levels and protein expression of MITF, tyrosinase, Trp1 and Trp2. Irrespective of ${\alpha}$-MSH stimulation, GEB extract at doses of 0.5-5 mg/mL inhibited all these markers for skin whitening in a dose-dependent manner. While lower doses (0.5-1 mg/mL) of GEB extract generally had a tendency to decrease melanogenesis, tyrosinase activity, and mRNA levels and protein expression of MITF, tyrosinase, Trp1, and Trp2, higher doses (2-5 mg/mL) significantly inhibited all these markers in ${\alpha}$-MSH-treated B16F10 cells in a dose-dependent manner. These inhibitory effects of the GEB extract at higher concentrations were similar to those of $400{\mu}g/mL$ arbutin, a well-known depigmenting agent. CONCLUSIONS: These results suggest that GEB displays dose-dependent inhibition of melanin synthesis through the suppression of tyrosinase activity as well as molecular levels of MITF, tyrosinase, Trp1, and Trp2 in murine B16F10 melanoma. Therefore, GEB may be an effective and natural skin-whitening agent for application in the cosmetic industry.
Paracoccus sp.의 astaxanthin 생산성 증가를 위해 반응표면 분석법을 사용하여 최적의 배지조성을 설계하였다. Paracoccus의 성장 배지인 Marine Broth와 modified Blaszczyk 배지를 사용하여 astaxanthin 생산성을 측정한 결과 각각 0.39 mg/L, 0.40 mg/L의 astaxanthin 농도를 보였다. Modified Blaszczyk 배지의 조성을 각각 변수로 두어 가장 많은 영향을 주는 성분을 알아본 결과 $MgSO_4$와 yeast extract로 확인되었다. 중심합성계획법에 따라 $MgSO_4$ ($0.397{\sim}4.621$ g/L), yeast extract ($2.879{\sim}7.121$ g/L)를 달리하였을 때, astaxanthin 생상성에 대한 회귀식의 $R^2$은 0.894로 나타났고, 이에 따른 최대 생산량에 0.925 mg/L로 예상되었으며 이때의 $MgSO_4$와 yeast extract의 농도는 각각 2.83과 7.02 g/L로 나타났다. 이에 대한 확인실험 결과 2.83 $MgSO_4$ g/L, 7.02 yeast extract g/L에서 1.021 mg/L의 astaxanthin이 생산되었으며, 배지의 최적화에 따라 250% 이상의 생산성 증가가 확인되어졌다.
It is well-known method to use Ethylene Oxide (ETO) as a fumigant to preserve cultural heritage from the attack of some bacteria and insects. In this article, we investigated co]or change of some cultural material upon treating ETO. The cultural material was selected from Korean paper (hanji), ancient books, blue prints, pigments (DA, DB, DC, DD, DE and DF). hemp cloths, and colored pictures (DG, DH, Dl, DJ, DK). The $\Delta$E value in chromaticity coordinates for each item was obtained upon treatment with ETO at concentration of 200, 250, 380, 500, 710 g/m$^3$ respectively. The $\Delta$E value was derived from the equation of L*at with the data checked 24 hr, 48 hr, and 72 hrs respectively. The Korean papers(hanji) showed slightly change in color by $\Delta$E 0.8. In the case of ancient books, it showed $\Delta$E 2.91 which was easily distinguishable with the naked eyes. For the almost 73% of blue prints, the value of $\Delta$E was as much as more than 4.0 that it is recommended not to use. In the case of pigments, the value of $\Delta$E was ranged from 0.15 up to 4.0 so that it should be very careful before use. The hemp cloth dyed with natural indigo showed less than 1.00 in $\Delta$E, while various colored hemp cloth showed wide range of $\Delta$E from 1.00 to 4.00. Finally, from the fact that the color change was as small as less than 1.00 for the colored pictures, it is thought that treatment with ETO is one of method to be used to this purpose.
The effects of extracts from Lentinula edodes (L. edodes) and Aquilariae agallocha (A. agallocha) on the DNA damage response in ultraviolet A (UVA)-exposed HaCaT cells and on the allergic contact dermatitis caused by 2,4-dinitro-chlorobezene (DNCB) were investigated. When UVA-exposed cells were incubated for 24 hours in medium containing L. edodes or A. agallocha extract, the level of 8-OHdG and CPD decreased in a concentration-dependent manner. The combined treatment with both extracts potentiated the decrease in UVA-induced 8-OHdG and CPD levels as compared with those following treatment with a single extract. In addition, the two extracts showed preventive effects against the UVA-induced reduction in collagen levels. Furthermore, the blood levels of IgE, IL-6, and histamine decreased more significantly upon combined treatment with L. edodes and A. agallocha extracts as compared with those following treatment with single extracts in DNCB-induced allergic contact dermatitis in the ICR mouse. The results of the present study suggest that the components with in the extracts of L. edodes and A. agallocha can help to prevent of UVA-induced genomic instability via a decrease in DNA damage, and to decrease the DNCB-induced allergic dermatitis via modulation of relevant proteins including IgE and IL-6. Further study is needed to clarify the purified components related to the preventative effects of the two extracts against UVA- or DNCB-induced genomic damage.
The effects of BHSH on L-DOPA-induced increase in dopamine content in PC12 cells were investigated. L-DOPA treatment at $20\;or\;50\;{\mu}M$ increased dopamine content after both 24 and 48 h of incubation in PC12 cells. However, the co-treatments of BHSH $(10-50\;{\mu}M)$ with L-DOPA $(20\;or\;50\;{\mu}M)$ significantly inhibited the increase of dopamine content induced by L-DOPA. BHSH treatment at $10-50\;{\mu}M$ significantly inhibited basal aromatic L-amino acid decarboxylase (AADC) activity in a concentration-dependent manner at 15 min, and then AADC activity was rapidly recovered to the control level at about 2 h. These results indicate that the inhibition of AADC activity by BHSH was, in part, contributed to the early-stage decrease of dopamine content induced by LDOPA in PC12 cells. Taken together, it is proposed that the short-term inhibition of dopamine biosynthesis by BHSH was mediated by the regulation of tyrosine hydroxylace (TH).

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
