시멘트 바이패스 더스트 내 염화칼륨 수득을 위한 이산화탄소 적용 연구 (Study of Using Carbon Dioxide for Obtaining Potassium Chloride from Cement By-Pass Dust)

  • 시멘트 제조 공정에서 발생하는 바이패스 더스트는 지정폐기물로서 고가의 처리비용이 요구되고 있다. 바이패스 더스트의 주성분은 KCl이며, KCl은 주로 비료에 사용되고 있다. 비료에 사용하기 위해서는 pH 수준이 중성 혹은 약산성 이어야 한다. 그러나 바이패스 더스트의 pH는 12.0~12.5 수준으로, 또다른 전처리 없이 비료로 사용될 수 없다. 본 연구에서는 시멘트 바이패스 더스트에서 KCl을 수득하는 과정 중, 이산화탄소를 사용하여 산화칼슘을 제거하고, 동시에 pH 변화량을 제어하고자 하였다. 탄산화시험기 내부 분위기를 $25^{\circ}C$-50RH%로 고정한 후, 이산화탄소 20 vol% 조건에서 0~7시간으로 유지하고, 각 시간별 산화칼슘 함량 및 pH 값을 분석하였다. 이산화탄소 유지시간 증가에 따라 산화칼슘 함량 및 pH 값은 감소하였으며, 6시간 경과 후 pH 값은 7에 근접하였다.

Bacillus sp. GS가 생산하는 Xylanase의 정제 및 특성 (Purification and Characterization of Xylanase from Bacillus sp. GS)

  • Xylanase from Bacillus sp. GS was purified through acetone precipitation, DEAE-Sephadex A-50 ion exchange chromatography and Sephadex G-100 gel filtration. The optimum reaction temperature of purified xylanase was 50t . Its optimum pH was between pH 6.0 and pH 6.5. This enzyme was stable below 5$0^{\circ}C$ for several hours and stable at between pH 5.5 and pH 8.0. The enzyme activity of xylanase was remarkably increased by Co++ and Cu++ ions. According to the study of hydrolysis mode of this enzyme, it was turned out to be ends type xylanase that can produce xylooligosaccharides, known as bifidogenic factor, from xylan.

  • Extracellular nuclease(s) in buffalo rumen fluid were purified from strained rumen fluid by a procedure involving Seitz filtration, acetone fractionation and gel filtration on Sephadex G-100. The enzyme resolved into two peaks exhibiting both DNase and RNase activities. The molecular weight of enzyme corresponding to peaks I and II were approximately 30,000 and 12,000 respectively. The properties of enzymes from the two peaks, however, were same. Optimum temperature for both DNase and RNase activities was at $50^{\circ}C$. Whereas DNase activity was stable upto $60^{\circ}C$, RNase activity was stable only up to $50^{\circ}C$. DNase activity recorded two pH optima, one at pH 5.5 and the other at pH 7.0. RNase activity recorded a broad pH optimum between pH 6.0-8.0. pH stability of the enzyme coincided with pH optima for both the activities. DNase activity was stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. RNase activity was also stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Cu^{2+}$, $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. Reducing agents stimulated both the activities.

콩(Glycine max Merr)유묘를 이용한 수질정화에 관한 연구 (Studies on the Water Purification Using Glycine max Merr Seedling)

  • The removal efficiency of nutrient was investicated by using Glycine max Meer seedling. After budding, Glycine max Merr was raised at darkness for 4 days. During cultivation, the removal efficiency of $NO_2-N+NO_3-N$ was up to 90% with initial concentration of 20-100 ppm. The removal efficiency of PO$_4$-P was up to 80% with initial concentration at 30 ppm, but it was down to 22% and 27% at 40 ppm and 50 ppm. When the removal efficiency of nutrient was compared with alternating 12 hours' light and darkness, the removal efficiency of NO$_2$-N + NO$_3$-N was up to 90% at below 60 ppm. It was not different from each other. But it was particularly low about 62% and 34% at 80 ppm and 100 ppm in alternating 12 hours' light. The removal efficiency of PO$_4$-P was low at alternating 12 hours' light between 10-50 ppm on the whole range. The neutralizing capacity of pH was shown in acidity and alkalinity except strong acidity(below pH 3). The initial pH was neutralized at 6.0-7.7 of pH after 4 days. Particularly, Glycine max Meer seedling that was difference from other water plants, was shown the neutralizing capacity in strong alkalinity.

Aureobasidium pullulans에 의한 플루란 생산에서 pH와 질소원의 영향 (Effects of pH and Nitrogen sources on the Pullulan Production by Aureobasidium pullulans)

  • 본 연구에서는 질소원의 종류와 농도 그리고 pH가 플루란 생산에 미치는 영향을 고찰하였다. A Aureobasidium pullulans에 의한 플루란 생산시 가장 좋은 질소원으로는 수율 62%를 얻은 peptone 이었고, 최적 질소원 농도는 유기질소원의 경우 탄 소원/질소의 비가 50j0.15N이었고 그 이상에서는 저해가 일어났다. 훈합질소원 I, II, III을 사용하 였을 때 각각 29.1, 27.4, 26.5g/L의 플루란 생산량 을 보였다. 그러므로, 혼합 질소원을 잘 이용하면 플 루란을 효과적으로 생산할 수 있었다. pH를 조절하 지 않을 때가 pH를 일정하게 할 때보다는 플루란 수율이 높았다. 또, 유가식배양시 탄소원과 질소원을 함께 공급하였을 때가 질소원만을 공급하였을 때 더 큰 플루란 수율을 보였다.

  • 식이섬유와 무기질의 장내 흡수의 관계를 알아보기 위해 율무겨에서 추출한 식이섬유의 Ca 및 Fe 결합력을 장의 pH 조건과 유사한 범위에서 측정하였다. 율무겨에서 추출한 식이섬유의 내인성 Ca및 Fe함량을 원소 흡광 분석기로 측정한 결과, 총 식이섬유에는 산성세제 저항섬유보다 자체내 Ca함량을 300배 이상이나, Fe함량은 4배 이상 함유되어 있었다. 식이섬유와 칼슘과의 결합 정도를 pH 5.0-7.0 범위에서 측정한 결과 TDF는 pH가 증가함에 따라 50.77%에서 86.64%로 증가했고, ADF는 40-50% 정도를 나타내어, TDF가 pH의 변화에 민감하게 반응하여 칼슘과 결합한다는 것을 알 수 있었다. 또한 식이섬유와 철분과의 결합 정도는 pH 5.0∼7.0 사이에서 TDF는 95∼97%, ADF는 85∼95%를 나타내, 철분은 pH변화에 영향을 받음이 없이 섬유소에 높은 비율로 결합하고 있음을 알 수 있었다.

니켈 함유폐수의 독성평가 및 처리방안에 대한 연구 (Research of the Toxic Estimation and Treatment Method of Wastewater Including Nickel)

  • 본 연구는 산업단지 폐수종말처리 시설내로 유입되는 니켈폐수의 독성평가 및 이의 효과적 처리방안을 도출하기 위하여 수행되었다. 처리장내로 유입되는 폐수를 24시간 동안 1시간 간격으로 시료를 채취하여 니켈 농도를 분석한 결과 0.33 mg/L~116.0 mg/L의 농도범위로 조사되었으며, 평균 24.0 mg/L로 조사되었다. 본 대상폐수의 농축질화균을 이용한 니켈의 독성도 조사 결과 Nitrosomonas와 Nitrobacter에 대한 $IC_{50}$값이 각각 5.5 및 4.9 mg/L인 것으로 나타났으며, 이는 본 유입폐수를 5배 희석하여 주입하여도 질산화 미생물의 50%를 사멸시킬 수 있는 농도인 것으로 나타났다. 또한 Nickel hydroxide형성에 의한 니켈 제거효율 실험결과 응집 pH에 따라 잔류 니켈 농도는 pH 11에서 1.7 mg/L, pH 12에서 0.6 mg/L로 나타났으며, 이는 상기 니켈 함유 폐수의 $NH_4$-N 및 $NO_3$-N에 대한 $IC_{50}$ 독성농도 조사 결과와 비교시 69.1~89.1% 및 65.3~87.8%의 독성도가 제거될 것으로 판단된다. 따라서 pH 10~11 이상으로 니켈 함유폐수를 응집처리 후 생물학적 처리를 수행한 결과 83.8%~99.4%의 범위로 평균 97.6%의 질산화 반응효율을 얻을 수 있었다.

비체중(比體重)의 보험의학적고찰(保險醫學的考察) (A Study on the Ratio of Weight-far-Height in Insurance Medicine)

  • A statistical analysis on the ratio of weight-for-height in insurance medicine was carried out in a total of 12,690 insured persons who were medically examined at the Honam medical department, Dong Bang life insurance company, ltd. from June, 1979 to February, 1984. The results were as follows: 1. The mean value of the ratio of weight-for-height(W/H ratio) was $102.3{\pm}11.3%$ in all males, and $105.2{\pm}103.6%$ in all females. The difference of the values between males and females showed statistical significance(P<0.001). The mean value of W/H ratio in all the insureds was $103.8{\pm}12.9%$. In each age group, the mean value of female was higher than that of male without exception(P<0.001). The mean value of both sexes in each age group showed gradual increase with age except for the age group over 50(P<0.001). 2. The incidences of the average weighted, overweighted and underweighted lives were respectively 91.6% in all males, 84.2% in all females(P<0.001); 7.9% in all males, 15.4% in all females(P<0.001); 0.5% in all males, 0.4% in all females(P>0.05). The incidences of average weighted, overweighted and underweighted lives in all the insureds were respectively 87.6%, 12.0% and 0.5%. The incidence of the average weighted lives in male in each age group was always higher than that in female with the exception of the age group below 19(P<0.001), whereas the incidence of the overweighted lives in male in each age group was always lower than that in female(P<0.05 in the age group below 19, P<0.001 in the other age groups). The incidence of average weighted lives of both sexes in each age group showed gradual decrease with age from the second to fifth decade(P<0.001), whereas the incidence of the overweighted lives of both sexes in each age group showed gradual increase with age from the second to fifth decade(P<0.001). 3. The relative frequency distribution polygons of W/H ratio of both sexes drawn in a pair on one coordinate plane revealed lying in juxtaposition each other horizontally and showed the shifting of female's polygon to male's one toward the direction of greater value of W/H ratio at a short distance which increased gradually with age. 4. Correlations between both sexes and among age groups relating to W/H ratios of the insureds seem to be a physiological phenomenon of body build and should be considered on the rating of the build. Author prepared a rating table of build based on the correlations above menthioned. Some gradients by sex and age in the rating table of build are organized; in female group, the ratings of 0, 30-50, 50-100 and 100-D are to calculate by the build with W/H ratio of 80-125%, $126{\sim}145%,\;146{\sim}165%$ and over 166% respectively in the age group of 29 or under, by the build with W/H ratio of $80{\sim}130%,\;131{\sim}150%,\;151{\sim}170%$ and over 171% respectively in the age group of $30{\sim}59$, and by the build with W/H ratio of $80{\sim}135%,\;136{\sim}155%,\;156{\sim}175%$ and over 176% respectively in the age group of 60 or over. In male group, the ratings of 0, $30{\sim}50$, 50-100, and 100-D are to calculate by the build with W/H ratio of $80{\sim}120%,\;121{\sim}140%,\;141{\sim}160%$ and over 161% respectively in all ages. Of under weighted lives, in both sexes, the ratings of 30-50 and 50-D are to calculate by the build with W/H ratio of $76{\sim}79%$ and under 75% respectively. The results of the ratings according to a rating table by author were as follows: The incidence of the ratings of 0, 30-50, 50-100 and 100-D showed no difference between male and female. The incidence of the ratings of 0(indicating the average weighted lives) showed gradual decrease with age and the incidence of the ratings of 30-50 and 50-100(indicating the overweighted lives) showed gradual increase with age. These results are thought to be considerably reasonable because the incidence of the builds in each age group is supposed to have no difference by sex and the incidence of the overweighted lives would increase with age.

pH 조건에 따른 기공성 실리콘의 나노구조 및 광학적 특성의 변화 (Variation of the Nanostructural and Optical Features of Porous Silicon with pH Conditions)

  • The effect of chemical treatments of porous silicon in organic solvents on its nanostructural and optical features was investigated. When the porous Si was dipped in the organic solvent with various PH values, the morphological, chemical, and structural properties of the porous silicon was sensitively affected by the chemical conditions of the solvents. The size of silicon nanocrystallites in the porous silicon decreased from 5.4 to 3.1 nm with increasing pH values from 1 to 14. After the samples were dipped in the organic solvents, the Si-O-H bonding intensity was increased while that of Si-H bonding decreased. Photoluminescence peaks shifted to a shorter wavelength region in the range of 583 to 735 nm as the pH value increased. PL intensity was affected by the size as well as the volume fraction of the nanocrystalline silicon in the porous silicon.

난황 유래 인지철로 합성한 리포좀의 물리적 특성과 안정성 (Physical Property and Stability of Liposome Prepared from Egg Yolk Phospholipids at Various Storage Conditions)

  • 난황 유래의 인지질을 주원료로 리포좀을 제조하여 서로 다른 저장조건에서의 리포좀의 물리적인 안정성을 연구하였다. 리포좀 용액의 pH를 달리하여 저장한 후 리포좀 입자크기와 리포좀 용액의 제타전위와 형광도를 측정한 결과, 리포좀은 pH 1-2에서 입자크기의 증가를 나타내었고 pH 4 이하에서 제타전위의 절대값이 감소하였으며 리포좀의 내부에 포집한 수용성물질의 손실량 지표인 형광도의 측정치 또한 pH 2-4에서 급격하게 증가한 것으로 보아 산성의 저장조건에서는 리포좀이 불안정함을 추론할 수 있다. 저장 온도별 리포좀의 안정성 평가에서는 $4^{\circ}C$에서 저장하였을 때 10일째부터 입자의 크기 변화가 있었고 20, $35^{\circ}C$에서 저장할 경우, 저장기간 40일째 이후부터 입자의 크기변화가 있었다. 제타전위는 3가지 온도조건에서 동일하게 저장 말기에 측정값의 절대값이 미세하게 감소하는 경향을 보였다. 또한, $50^{\circ}C$에서 저장된 리포좀의 형광도 측정실험에서 포집된 SRB의 누출량이 급격히 증가하는 양상을 나타내었다. 따라서 pH 6 이하의 산성의 조건에서 리포좀은 불안정하며 $50^{\circ}C$와 같이 높은 온도에서 인지질로 구성된 리포좀의 안정성을 유지하기 어렵다는 것을 알 수 있었다. 본 연구의 결과는 난황 유래 인지질로 구성된 리포좀을 이용한 진단용 시약 개발 및 포집 특성 연구에 유용한 기초 자료로 활용될 수 있을 것이다.
