Effects of Dexamethasone and DHEA on the Changes of Glutamate and Polyamine Uptake in Rat Astrocytes by Lipopolysaccharide and Antimycin A

  • Interactions among dexamethasone, dehydroepiandrosterone (DHEA), lipopolysaccharide (LPS), and antimycin A on the glutamate uptake and the polyamine uptake were investigated in primary cultures of rat cerebral cortical astrocytes to examine the effects of dexamethasone and DHEA on the regulatory role of astrocytes in conditions of increased extracellular concentrations of glutamate or polyamines. 1. $[^3H]Glutamate$ uptake: LPS and antimycin A decreased $V_{max},$ but both drugs had little effect on $K_m.$ Dexamethasone also decreased basal $V_{max}$ without any significant effect on $K_m.$ And dexamethasone further decreased the antimycin A-induced decrease of $V_{max}.$ DHEA did not affect the kinetics of basal glutamate uptake and the change by LPS or antimycin A. 2. $[^{14}C]Putrescine$ uptake: LPS increased $V_{max},$ and antimycin A decreased $V_{max}.$ They showed little effect on $K_m.$ Dexamethasone decreased $V_{max}$ of basal uptake and further decreased the antimycin A-induced decrease of $V_{max},$ and also decreased $V_{max}$ to less than control in LPS-treated astrocytes. DHEA did not affect $K_m$ and the change of $V_{max}$ by LPS or antimycin A. 3. $[^{14}C]Spermine$ uptake: Antimycin A decreased $V_{max},$ and LPS might increase $V_{max}.\;K_m$ was little affected by the drugs. Dexamethasone decreased basal $V_{max}$ and might further decrease the antimycin A-induced decrease of $V_{max}.$ And dexamethasone also decreased $V_{max}$ to less than control in LPS-treated astrocytes. DHEA might increase basal $V_{max}$ and $V_{max}$ of LPS-treated astrocytes. 4. $V_{max}$ of glutamate uptake by astrocytes was increased by putrescine (1000 ${\mu}M$ & 2000 ${\mu}M$) and spermidine (200 ${\mu}M,$ 500 ${\mu}M$ & 2000 ${\mu}M$). Spermine, 200 ${\mu}M$ (and 100 ${\mu}M$), also increased $V_{max},$ but a higher dose of 2000 ${\mu}M$ decreased $V_{max}.\;K_m$ of glutamate uptake was not significantly changed by these polyamines, except that higher doses of spermine showed tendency to decrease $K_m$ of glutamate uptake. In astrocytes, dexamethasone inhibited the glutamate uptake and the polyamine uptake in normal or hypoxic conditions, and the polyamine uptake might be stimulated by LPS and DHEA. Polyamines could aid astrocytes to uptake glutamate.

Prediction of VO2max Using Submaximal PACER in Obese Middle School Boys (최대하 PACER 검사를 통한 비만 남자 중학생의 VO2max 추정)

  • The purpose of this study was to develop the equation of $\dot{V}O_{2max}$ by $sub_{max}imal$ PACER method for obese middle school boys. For this, $_{max}$imal test using Bruce protocol in lab was performed and then PACER $_{max}imal$ test with portable $\dot{V}O_{2max}$ equipment. To decide the level of submaximal test, during PACER with portable equipment, we found the section in which target hreat rate(over 75%$HR_{max}$) and then per section(75%,80%,85%,90%,95%) metabolic responses were recorded, with which we analyzed multiple regression by stepwise method. Model 1(at 90%$HR_{max}$): $\dot{V}O_{2max}$(ml/kg/min) = 142.721-0.275(repetition)-0.48(HR)+0.177(weight)-1.536(age)[%error 3.90ml/kg/min; performance until 2 stage(13 repetition)]. Model 2(at 95%$HR_{max}$): $\dot{V}O_{2max}$(ml/kg/min) = 182.851-0.103(repetition)-0.744(HR)+0.186(weight)-0.324(age)[%error 4.51ml/kg/min; performance until 3 stage(25 repetitions)]. estimated $\dot{V}O_{2max}$ from Model 1 was different about $3.25{\pm}6.32ml/kg/min$(%error=6.84%), otherwise model 2 was $3.16{\pm}4.54ml/kg/min$(%error=5.75%). considering %HRmax, as the submaximal test model 1 might be fit more than model 2 for obese middle school boys.

  • 본 논문에서는 AC-AC 직접 변환 방식인 매트릭스 컨버터의 진압 변조 방법 중 하나인 $V_{max}-V_{mid}$ PWM 변조 방법을 간략화한 Simple $V_{max}-V_{mid}$ 전압 변조 방법에 대해 소개한다. $V_{max}-V_{mid}$ 전압 변조 방법은 실행 전에 입력전압 위상각$(\theta_E)$을 판별하는 PLL(Phase Looked Loop)과정을 거친 후, 얻은 위상 정보를 사용하석 게이트 신호를 만들어 낸다. 하지만 PLL의 사용이 연산 시간의 딜레이를 발생시키고, 처리과정을 복잡하게 만드는 단점으로, PLL과정을 생략하여 입력 전압의 크기 정보만으로 섹터를 판별하고 게이트 신호를 발생시켜 스위치를 조작하는 변조 방범을 사용하였다. 이로 인해 연산시간의 단축과 처리 과정의 축소 등의 이점을 취하고, 이를 시뮬레이션으로 검증한다.

  • Es konnte nachgewiesen werden, daß die erwartete lineare Funktion der $V_{max}$- und $V_{HWB}$-Großen in Abhangigkeit vom Saulenquerschnitt vorhanden ist. Allerdings ließ sich diese Abhangigkeit noch bei Saulen mit 3 mm Durchmesser bestatigen, was bei 20∼50 mesh Kornern wesentlich unter der Grenze der ublicherweise angenommen Randgangigkeit liegt. Der Einfluß der Stromugsgeschwindigkeit macht sich bei den $V_{max}$-Werten ales kleines negatives und bei den V_{HWB}-Werten als starkeres positives Glied bemerkbar. Die damit nachgewiesene Bandeverbreiterung weist auf den Vorteil moglichst geringer Stromungsgeschwindigkeiten bei schwierigen Trennproblemen hin. Die Auftragungsmengen beeinflußten die $V_{max}$- and $V_{HWB}$-Werte durch additive Glieder. Die Unterschiede in den $V_{max}$}-Werte wurden bei den verwente Nukliden (Co und Mn) bei hoheren applizierten Mengen so gering, daß kein Trenneffekt mehr zustande kam. Durch Trennung von Co($K_D$ = 68.2) und Mn($K_D$ = 71.8) konnten die erhaltenen Formulierungen uberpruft und verhaltnismaßig gut bestatigt werden.

Comparative Analysis of Offset Voltage PWM and $V_{max}-V_{mid}$ PWM Method for 3 Phase Matrix Converter (3상 매트릭스 컨버터에 사용되는 옵셋전압 PWM 방법과 $V_{max}-V_{mid}$ PWM 방법의 비교분석)

  • In this paper, comparative analysis of offset voltage PWM method and $V_{max}-V_{mid}$ PWM method for three-phase matrix converter is addressed by using a simple analytical and graphical method. Offset voltage PWM method calculates PWM patterns in terms of offset voltage and variable slope of carrier, and it simplifies matrix converter modulation algorithm significantly. $V_{max}-V_{mid}$ PWM method generates patterns by using two phases and maintaining a remaining phase to base phase, and it is implemented in the industrial products. The most important performance criterion of modulation method is a magnitude of current ripples and it is analytically modelled. The graphical illustration of theses complex multivariable functions make per-carrier cycle and per fundamental cycle behavior of two PWM methods understood. Two modulation methods are analysed with the analytical formulas and graphics, and the analysis shows offset voltage PWM method is superior to $V_{max}-V_{mid}$ PWM method with respect to input current ripples and output voltage ripples.

Maximum Oxygen Consumption Determined by the Bruce and Inclined Treadmill Protocols

  • The aim of the present study was to derive regression equations for $\dot{V}o_{2max}\;vs.\;\dot{V}o_{2peak},\;and\;\dot{V}o_2\;vs.$ heart rate, exercise time, and other variables from maximal exercise tests on a treadmill using the Bruce and inclined protocols. Twelve male and 10 female Korean college students aged between 19 and 23 years voIunteered for this study. After the resting measurements, the subjects performed a maximal exercise on a treadmill according to the Bruce protocol. When the resting conditions were restored, the subjects performed another maximal exercise according to an inclined protocol where the speed was fixed at 8.05 $km{\cdot}h^{-1}$ and the grade was incremented starting from 09t by 2.5% for every 2 min. Peak $\dot{V}o_2$ observed during the Bruce exercise $(\dot{V}o_{2peak})$ was $37.7{\pm}2.4\;and\;31.7{\pm}1.8\;ml\;kg^{-1}\;min^{-1}$ in the male and female groups, respectively. Peak $\dot{V}o_2$ observed during the inclined exercise was higher than $\dot{V}o_{2peak}$ during the Bruce exercise. Maximum $\dot{V}o_2$ value observed during the tyro exercises $(\dot{V}o_{2max})$ was $43.0{\pm}2.8\;and\;36.2{\pm}1.4\;ml\;kg^{-1}\;min^{-1}$ in the male and female groups, respectively. Thus, $\dot{V}o_{2peak}$ by the Bruce protocol was about 12% (male) or 13% (female) lower than $\dot{V}o_{2max}$, and a linear relationship was found between $\dot{V}o_{2peak}$ and $\dot{V}o_{2max}$. The peak values of % $\dot{V}o_{max}$ with the Bruce protocol were $89.2{\pm}3.3\;and\;87.5{\pm}3.6%$ and those with the inclined protocol $97.7{\pm}1.8\;and\;96.9{\pm}2.0%$ in the male and female groups, respectively. In the female group, $%\dot{V}o_{2max}$ at a given workload was higher than in the male group, while $\dot{V}o_{2}$ per kg body weight was the same. Maximum HR observed during the two exercises was $204{\pm}2\;and\;195{\pm}3\;beat\;min^{-1}$ in the male and female groups, respectively. Linear relationships were found, excluding the last points, between 1) $\dot{V}o_{2}$ and exercise time, 2) $\dot{V}o_{2}$ and $%\dot{V}o_{2max}\;and\;%HR_{max}$.

Changes in the Spinal Motor Neuron Excitability Depending on Postural Changes in Post Stoke Hemiplegics (뇌졸중 후 편마비 환자의 체위변화에 따른 척수운동신경원 흥분성 변화)

  • Purpose: The purpose of this study was to measure changes in the H-reflex and V wave under loading conditions (e.g. prone and standing position) and to investigate whether postural change would affect the H-reflex and V wave in post stroke hemiplegic patients. Methods: Thirty persons with hemiplegia resulting from stroke (20 males, 10 females) participated in this study. Electromyography (EMG) was used to electrically stimulate and record the soleus H-reflexes and V waves under various loading conditions. The normality of the distribution of each variable (H latency, $H_{max}/M_{max}$ ratio, $V_{max}/M_{max}$ ratio) was tested using the Kolmogorov-Smirnov test. The means of normally distributed continuous data were assessed by independent t-test (${\alpha}$=0.05). Results: There were statistically significant differences in $H_{max}/M_{max}$ ratio (p<0.01), $V_{max}/M_{max}$ ratio (p<0.01), H latency (p<0.01) among the prone and standing position. Conclusion: We found that the H-reflex and V wave in standing position was more active to weight bearing load than prone position.

ON SPATIAL DISTRIBUTION OF SHORT GAMMA-RAY BURSTS FROM EXTRAGALACTIC MAGNETAR FLARES

  • Recently, one interesting possibility is proposed that a magnetar can be a progenitor of short and hard gamma-ray bursts (GRBs). If this is true, one may expect that the short and hard GRBs, at least some of GRBs in this class, are distributed in the Euclidean space and that the angular position of these GRBs is correlated with galaxy clusters. Even though it is reported that the correlation is statistically marginal, the observed value of < $V/V_{max}$ > deviates from the Euclidean value. The latter fact is often used as evidence against a local extragalactic origin for short GRB class. We demonstrate that GRB sample of which the value of < $V/V_{max}$ > deviates from the Euclidean value can be spatially confined within the low value of z. We select very short bursts (TgO < 0.3 sec) from the BATSE 4B catalog. The value of < $V/V_{max}$ > of the short bursts is 0.4459. Considering a conic-beam and a cylindrical beam for the luminosity function, we deduce the corresponding spatial distribution of the GRB sources. We also calculate the fraction of bursts whose redshifts are larger than a certain redshift z', i.e. f>z'. We find that GRBs may be distributed near to us, despite the non-Euclidean value of < $V/V_{max}$ >. A broad and uniform beam pattern seems compatible with the magnetar model in that the magnetar model requires a small $z_{max}$.

Respiratory Gas Exchange and Ventilatory Functions at Maximal Exercise (최대운동시의 호흡성 가스교환 및 환기기능)

  • Background: Although graded exercise stress tests are widely used for the evaluation of cardiorespiratory performance, normal standards on respiratory gas exchange and ventilatory functions at maximal exercise in Koreans have not been well established. The purpose of this study is to provide reference values on these by sex and age, along with derivation of some of their prediction equations. Method: Symptom-limited maximal exercise test was carried out by Bruce protocol in 1,000 healthy adults consisting of 603 males and 397 females, aged 20~66 years. Among them VC, $FEV_1$ and MVV were also determined in 885 cases. All the subjects were members of a health center, excluding athletes. During the exercise, subjects were allowed to hold on to front hand rail of the treadmill for safety purpose. Results: The $VO_2\;max/m^2$, $VCO_2\;max/m^2$ and $V_E\;max/m^2$ were greater in males than in females and decreased with age. The RR max in men and women was similar but decreased slightly with age. The $V_T$ max was markedly greater in men but showed no significant changes with age in either gender. The mean of $V_T$ max/VC, $V_E$ max/MVV and BR revealed that there were considerable ventilatory reserves at maximal exercise even in older females. The regression equations of the cardinal parameters obtained using exercise time(ET, min), age(A, yr), height(Ht, cm), weight(W, kg), sex(S, 0=male; 1=female), VC(L), $FEV_1$(L) and $V_E$ max(L) as variables are as follows: $VO_2\;max/m^2$(L/min)=1.449+0.073 ET-0.007A+0.010W-0.006Ht-0.209S, $VCO_2\;max/m^2$(L/min)=1.672+0.063ET-0.008A+0.010W-0.005Ht-0.319S, VE max/$m^2$(L/min)=58.161+1.503ET-0.315A-9.871S or VE max/$m^2$(L/min)=47.873+6.548 $FEV_1$-5.715 S, and VT max(L)=1.497+0.223VC-0.493S. Conclusion: Respiratory gas exchange and ventilatory variables at maximal exercise were studied in 1,000 non-athletes by Bruce protocol. During exercise, the subjects were allowed to hold on to hand rail of the treadmill for safety purpose. We feel that our results would provide ideal target values for patients and healthy individuals to be achieved, since our study subjects were members of a health center whose physical fitness levels were presumably higher than ordinary population.
