A STUDY ON SPECIFIC ACTIVITIES OF ENZYMES IN 7, 12-DIMETHYLBENZ(A)ANTHRACENE(DMBA)-INDUCED RAT SUBMAXILLARY GLAND CARCINOGENESIS (백서의 7, 12-Dimethylbenz(a)anthracene 유도 악하선 종양발암과정에서의 효소 특이활성도에 대한 연구)

  • In recent years, tissue antigens and enzymes that will serve as phenotypic markers for malignant cells are becoming increasingly important as diagnostic aids. This study was undertaken to investigate the specific activities of these enzymes in DMBA-induced rat submaxillary gland carcinogenesis. One hundred and twenty Sprague-Dawley rats about 100 gms of body weight were used. In experimental group, DMBA pellet (5mg) was implanted into right submaxillary gland and sham operation was performed into left gland to serve as control. The animals were sacrificed every three weeks up to 15 weeks. Submaxillary glands were excised on both sides and enzyme assays for ${\gamma}-glutamyl$ transpeptidase (GGT), 5'-Nucleotidase, Ornithine decarboxylase(ODC) and Acetyl-Co A carboxylase were carried out biochemically. The obtained results were as follows ; 1. In control group, there was no significant weight change of submaxillary gland, while experimental group, weight was increased remarkably about 7-fold at 15th week since DMBA implantation. 2. In control group, there was no change in specific activities of enzymes during the experimental period. 3. GGT activity was rapidly increased reaching a peak of 1.766${\pm}$0.082units/mg of DNA, 8-fold greater than that of onset. 4. 5'-Nucleotidase activity was increased reaching a peak of $362.1{\pm}53.2{\mu}moles/mg$ of DNA at 9th week. 5. ODC activity was rapidly increased, reaching a peak of 26.2${\pm}$4.8nmoles/mg of DNA at 9th week and quickly returned to that of control at 15th week. 6. Acetyl-Co A carboxylase activity was rapidly increased earlier than other enzymes, reaching a peak of 0.178${\pm}$0.013units/mg of DNA at 6th week and quickly declined to the control level at 15th week.

Growth Performance of Nursery Pigs Fed 30% Distillers Dried Grain with Solubles (DDGS) and the Effects of Pelleting on Performance and Nutrient Digestibility

  • Four experiments with 2,020 nursery pigs (Triumph-4${\times}$PIC Camborough 22) were conducted at a commercial research site to evaluate the effects of including 30% distillers dried grain with solubles (DDGS) in late nursery diets on pig growth performance and to compare the effects of pelleted and meal diets containing 30% DDGS on growth performance and nutrient digestibility. In Exp. 1, 312 pigs (10.54${\pm}$0.16 kg) were allotted to two diets: corn-soybean meal based control diet and a corn-soybean meal diet containing 30% DDGS. In Exp. 2, 337 pigs (16.70${\pm}$0.14 kg) were used to validate Exp. 1 with similar diets. In Exp. 3, 665 pigs (11.77${\pm}$0.12 kg) were allotted to diets containing 30% DDGS, but in different forms: meal or pellet. Fecal samples were collected during the final day of the trials to measure nutrient and energy apparent total tract digestibility (ATTD). In Exp. 4, 706 pigs (18.40${\pm}$0.18 kg) were allotted to the same diets as Exp. 3. In Exp. 1 and 2, there was no (p>0.05) effect of DDGS on ADG, ADFI or G:F. In Exp. 3, pelleting improved (p<0.01) ADG (578 vs. 541 g/d) and G:F (714 vs. 674 g/kg), with no difference (p>0.05) in ADFI. The ATTD of most nutrients and energy were improved by pelleting (p<0.05). In Exp. 4, ADG (717 vs. 675 g/d) and G:F (680 vs. 648 g/kg) were improved by pelleting (p<0.01), with no difference (p>0.05) in ADFI. In conclusion, 10-23 kg pigs in late nursery phase can be fed up to 30% DDGS without having a detrimental effect on pig performance, and pelleting the diet will increase nutrient and energy availability.

A Study on the Design Considerations of Vol-Oxidizer for High-Capacity Uranium Dioxide Pellets (대용량 우라늄디옥사이드 펠릿 산화를 위한 공기산화로의 설계 고려사항에 대한 연구)

  • This study deals with the design and implementation results for a high-capacity vol-oxidizer that can convert Uranium Dioxide pellets to $U_3O_8$ powder for up to several tens of kg HM/batch. We developed two versions of the $1^{st}$ vol-oxidizer and the $2^{nd}$ vol-oxidizer. Through an experiment with the $1^{st}$ vol-oxidizer, we deduced some problems concerning the design considerations such as the recovery rate of $U_3O_8$, the oxidation time of the Uranium Dioxide pellets, the exothermic reaction, and the powder dispersion. From the analyses of the drawbacks of the $1^{st}$ vol-oxidizer, we devised some novel items such as a folding type mesh, vibrators, and mixing blades. Also, we used the Stokes and Density ratio Eq. to determine the most reasonable flux for preventing a powder dispersion. Compared with the results of the $1^{st}$ vol-oxidizer, we showed that both the permeability of the $U_3O_8$ powders and the oxidation rate of the Uranium Dioxide pellets of the $2^{nd}$ vol-oxidizer were remarkably increased, and the temperature of the reactor was controlled well in spite of an exothermic reaction. Also, the powder was not entirely dispersed through the outlet of the voloxidizer. The experimental results of this work can help in the design of a novel and efficient vol-oxidizer with a higher capacity.

An Experimental Study on Drilling Conditions for the Instrumentation of Nuclear Fuel (핵연료 계장을 위한 천공조건에 대한 실험적 연구)

  • To develop a new nuclear fuel, it needs to make a test fuel rod and carry out burn-up test in the test loop of a research reactor to check the irradiation characteristics of the nuclear fuel. At that time, several sensors such as thermocouples, LVDTs and SPNDs are needed to be attached in and out of the fuel rod and connect them with instrumentation cables. Then, the instrumentation cables deliver the signals measured by the sensors to the measuring device located outside of the reactor pool. In particular, to install a thermocouple in a fuel rod, it needs to drill off holes on the alumina blocks and sintered $UO_2$ pellets. However, because the hardness of a sintered $UO_2$ pellet is 700 Hv (or HRC 61) and that of an alumina block is 1480 Hv, a special drilling machine which adapts a diamond coated drill bit had developed. In this study, several case experiments have been carried out to find an optimal drilling condition of the drilling machine. And, using the optimal drilling condition, minimum numbers of the holes that a drill bit can drill off are verified.

Developmemt of Rice Husk Pellets as Bio-filter Media of Bio Scrubber Odor Removal System (왕겨펠렛 생물담체 개발 및 이를 이용한 bio scrubber형 악취제거 시스템 성능평가)

  • The rice husk contains nutrients which can be easily utilized by microorganisms, and also has a water retaining ability, which played a crucial part in enabling it to become a biofilter media. In this study, we evaluated the applicability of rice husk pellet bio-scrubber as a microbiological carrier. The pelletization experiment of rice husk as a biological media was performed using PVA and EVA binder. Also, the feasibility tests of rice husk as a biological media for odor removal were carried out in order to know whether rice-husk contains useful components as a media for microbiological growth or not. Lastly, a combined test for odor gas absorption and biological oxidation was conducted using a lab scale bio-filter set-up packed with rice-husk pellets as wet-scrubber. The major components of the rice husk were carbon, hydrogen, nitrogen, and oxygen, while carbon acted as the main ingredient which comprised up to 23.00%. The C : N : P ratio was calculated as 45 : 1 : 2. Oxygen uptake rate, yield and decay rate of the rice husk eluent was calculated to be $0.0049mgO_2/L/sec$, 0.24 mgSS/mgCOD and 0.004 respectively. The most stable form of rice husk pellets was produced when the weight of the rice husk, EVAc, PVAc, and distilled water was 10 : 2 : 0.2 : 10. The prepared rice husk pellets had an apparent density of 368 g/L and a porosity of 59.00% upon filling. Dry rice husks showed high adsorption capacity for ammonia gas but low adsorption capacity for hydrogen sulfide. The bio-filter odor removal column filled with rice husk pellets showed more than 99.50% removal efficiency for NH3 and H2S gas. Through the analysis of circulation water, the prime removal mechanism is assumed to be the dissolution by water, microbial nitrification, and sulfation. Finally, it was confirmed that the microorganisms could survive well on the rice husk pellets, which provided them a stable supply of nutrients for their activity in this long-term experiment. This adequate supply of nutrients from the rice husk enabled high removal efficiency by the microorganisms.

  • Improvements in understanding the effects of dietary fermentable carbohydrates and their interaction with supplemental feed enzymes and the feed manufacturing process may lead to reductions in volatile organic compound (VOC) emissions from poultry manure. Starch digestibility has been improved by replacing ground wheat or barley with whole wheat or barley, but there was no consistent effect of cereal species or feed form on the pH value of the gizzard contents. Pelleting results in improvements in feed conversion from 0 to 12%. Starch digestibility has been reported to account for up to 35 % of the improvement in available metabolic energy as a result of xylase supplementation. Factors which affect starch utilization and non-starch polysaccharide (NSP) absorption include the presence of anti-nutrient facto. (ANF) in grains, the nature of grain starch, NSP and the digestive capacity of animals. Improvements in feed production technology have been made in enzyme stabilization, allowing some dry enzyme products to be pelleted after conditioning at up to $87.69^{\circ}C$ and liquid enzymes to be stored in the feed mill for up to low months prior to use. The soluble NSP, arabinokylans and beta-glucans are partially degraded into smaller fragments by enzymes. With fragmentation, the water holding capacity is decreased, which leads to a reduction in digesta moisture, wet feces, and dirty eggs from hens fed diets containing viscosity-inducing ingredients.

A Study on the Reduction of Iron Oxide from Slag in the EAF Process (전기로 공정에서 슬래그 중 산화철의 환원 회수에 관한 연구)

  • EAF processed slag which contains about 20 ~ 35 weight percent FetO is poured to slag pot and cooled. If we recover Fe from molten slag by the reduction, we will improve steel yield rate and reduce slag quantity poured from the furnace. Usually, carbon is used as a reductant and slag foaming agent in the EAF process. In this experiment, after melt the metal in induction furnace and then add slag with carbon and Al dross powder as a reductant, we investigated the reduction of FetO from slag and change of Phophorus content. As the result, when we use Al dross as a reductant, recovery rate is two times more than carbon. Phosphorus pick up is less than 50ppm with reduction of EAF slag.

Effect of light, ultrasonication and liquid smoke on germination of proso millet (Panicum miliaceum L.) seeds

  • High quality seed of proso millet that has high germination percentage, germination speed, and uniformity demanded to increases rates of mechanization in cereal crop cultivation. In order to improve germination characteristics, proso millet seeds were treated with red light, ultrasonication and liquid smoke (LS) solution that generated from hickory wood. All treatments were performed in seed priming solution with 100mM $NH_4NO_3$ at $15^{\circ}C$ for 24hrs under aeration condition. Seeds were exposed under light intensity of 2000 lux for 15m, 30m, 60m, and 120m in priming solution. Ultrasonic treatment was performed at 60%, 80%, and 100% intensity of 21.6 KHz for 5m, 10m, and 20m in priming solution. For LS treatment seed were soaked in 0%, 0.5%, 1.0%, 5.0% and 10.0% of diluted solution with $dH_2O$ or 100mM $NH_4NO_3$ solution. The effect of seed treatment was evaluated with germination percentage (GP), mean germination time (MGT), germination index (GI), germination rate (GR), Germination uniformity (GU) and heath seed percentage (HS). Our results demonstrate that red light (15min) or ultrasonication (21.6kHz, 5min) treatment improved MGT, GI, GR, and GU comparing to untreated control. Importantly, we show that LS treatments have significant effect on the health seedling and germination characteristics. Proso millet seeds that treated with 5% LS solution for 24hrs improves HS up to 97% that similar results obtained in 100mM $NH_4NO_3$ priming for 24hrs. The combined treatment with LS solution and 100mM $NH_4NO_3$ priming were not effective in all treatments. Our results demonstrate that treating seeds with LS or 100mM $NH_4NO_3$ priming or ultrasonication improves germination characteristics. The methods described here will help advance research using this species by increasing the germination performance at which successive seed pellet process.

  • The effect of desorption pressure on $CO_2/N_2$ breakthrough behaviors for 4 different adsorbents was studied at a fixed bed. Zeolite 3A, 4A, 5A, and 13X pellets were used as adsorbents. Cyclic operations were executed with varying desorption pressure from vacuum (0 bar) to 3 bar while other conditions such as adsorption step pressure (3 bar), temperature (293 K), composition ($CO_2:N_2=10:90$vol%) and flow rate (400 ccm) were fixed at constant values. Each adsorption and desorption step was set as 80 min, which totaled up to 160 min per a cycle. 5 cycles with adsorption and desorption steps were run overall. After the experiment, breakthrough time, saturation time, and adsorption amount were measured and compared in order to find an optimum adsorbent and a proper operating condition for a post combustion $CO_2$ capture process.

Induced Breeding and Indoor Culture of the Catfish, Silurus asotus (Teleostomi : Siluridae) (메기(Silurus asotus)의 산란 유도 및 실험실 사육에 관한 연구)

  • Induced breeding and indoor rearing of the catfish, Silurus asotus were performed. Ovulation was induced by injection of 5 mg dried carp pituitary per kg body weight. Fertilization rates were $81.5\~98.0{\%}$ and hatching success was $67.0\~82.0{\%}$. There were no significant differences of daily growth rate (DGR) and food conversion ratio (FCR) between the groups fed extruded eel pellet and extruded carp pellet. No significant differences in DGR and FCR were also found between $3{\%}$ and $4{\%}$ daily feeding rates. DGR and FCR of small fry were slightly affected by dissolved oxygen (DO) level in aquaria. When the DO level maintained at $4.5\;mg/{\ell}$, the best result was obtained and DGR and FCR were $5.7{\%}$ and 0.65, respectively. At the higher level of DO, $5.5\;mg/{\ell}$, DGR and FCR were $5.52{\%}$ and 0.77, respectively, whereas at the lower level of DO, $3.5\;mg/{\ell}$, the DGR was decreased to $3.45{\%}$ and FCR was increased up to 1.01.
