가토 신피질 절편에서 PAH$(\rho-aminohippuric\;acid)$ 이동에 미치는 음이온의 영향 (Effects of Anions on PAH Transport in Rabbit Kidney Cortical Slices)

  • The effects of anions on net accumulation of $(\rho-aminohippuric\;acid)$(PAH) were studied in rabbit kidney cortical slices. Experiments were carried while varying the major anionic composition of the incubation medium(replacement of $Cl^-$ by isethionate and $SCN^-$). The total replacement of $Cl^-$ with isethionate, $SO_4\;^{2-}$ and $SCN^-$ in the incubation medium decreased the 60-min slice-to-medium concentration(S/M) ratio of PAH to 60%, 40% and 50% of control value, respectively. The degree of inhibition in PAH accumulation by the replacement of isethionate and $SCN^-$ was increased with increasing of both preincubation and incubation time. The influence of isethionate and $SCN^-$ on PAH uptake was fully reversible. Both isethionate and $SCN^-$ increased the apparent Km value significantly with no change on the apparent Vmax value, suggesting a competitive inhibition on PAH uptake. And the inhibitory effect of $SCN^-$ on PAH uptake decreased with increase of pH in the incubation medium while that of isethionate increased with increase of pH. Intracellular water content, intracellular electrolyte concentration and oxygen consumption were not influenced by the replacement of $Cl^-$ with isethionate or $SCN^-$ in the incubation medium. These results suggest that both $isethionate^-$ and $SCN^-$ inhibit the PAH uptake by binding to some site necessary for normal PAH transport without affecting the cellular viability.

한국재래산양 시각교차위핵에 관한 형태학적 연구 (Morphological study on suprachiasmatic nucleus of the Korean native goat)

  • The present study was undertaken to investigate the morphological characteristics of the suprachiasmatic nucleus (SCN) of the Korean native goat by cresyl violet stain. The SCN was located ventrolateral to the third ventricle and dorsal to the optic chiasm. This nucleus was showed carrot form in longitudinal section. Its size was 1.8 mm in length, 0.9 mm in maximum height and 1.6 mm in maximum width. In coronal sections, the SCN shaped very thin plate in rostral part, but it was changed to sweet-potato form in middle part, and ornamental jade form in caudal part. The size of SCN was larger in caudal part than in rostral part. The size of the neuron of SCN was about $10{\mu}m$ in diameter with round or oval shape. The boundary of SCN was not firmly defined in caudal part because the neurons were dispersed into the hypothalamic region. It is concluded that the SCN of the Korean native goat has a morphological characteristics.

  • 콩 품종 Essex와 PI 437654간 교잡 후 F$_2$유래 F$_3$ 계통들을 재료로 하여 작성된 RAPD 유전자지도상에 cyst 선충 race 5에 대한 저항성 QTLs 분석을 실시한 바 결과를 요약하면 다음과 같다. 1. 회귀분석 결과 26개의 marker들(22 RAPD, 4 RFLP)에서 cyst 선충 race 5 저항성 반응에 대한 유의성이 인정되었다. 2. MAPMAKER /QTL 분석 결과2개의 저항성 QTL들이 탐색되었는데, 이 QTL들은 2개의 linkage groups(LGC-20와 Group 2)에 위치하였다. 3. 탐색된 2개의 QTL들 중 1개는 우성유전, ?고 나머지 하나는 열성유전양상을 나타내었다. 4. 콩 cyst 선충 race 5의 저항성에 대한 유의성이 인정되는 5개의 marker들간 상호작용을 알아보기 위한 다중회귀분석 결과 총 26개의 조합들 중 4개의 marker들(E02$^3$, G$10^1$, W03, pK418C)로 구성된 조합에서 가장 높은 표리적 변이의 값(35.2%)을 나타내었다.

질산술코나졸겔의 제조 및 약물방출에 관한 연구 (Study on Preparation and Drug Release of Sulconazole Nitrate Gels)

  • Sulconazole nitrate(SCN), an imidazole derivative which has been effective in the treatment of dermatophytosis, tinea versicolor and candidiasis, was formulated as a gel containing drug, poloxamer 407, ethanol and propylene glycol. The resulting SCN gels were evaluated with respect to their viscosity, drug release rate, skin permeation rate. The apparent viscosity of SCN gel increased in proportion to poloxamer 407, drug and propylene glycol concentration. In case ethanol was added, the apparent viscosity decreased. The drug release rate of SCN gel increased in proportion to temperature and ethanol concentration. But the drug release rate decreased as the concentration of poloxamer 407 increased. The increase of drug concentration induced nonlinear increase of drug release rate. When propylene glycol was added at the level of 10%, the drug release rate increased but from 15% it decreased. The skin permeation rate decreased in high concentration of poloxamer 407. The skin permeation rate of SCN gel containing 15% ethanol increased about twice than that of gel without ethanol. The increase of drug concentration induced nonlinear increase of skin permeation rate. When propylene glycol was added at the level of 10%, the skin permeation rate increased but from 15% it decreased.

Rheological Behavior of Lyotropilc Solutions of Cellulose in the $NH_3/NH_4SCN$ Solvent System

  • In the past, facile dissolution of cellulose has been hampered by the lack of suitable nondegrading solvents. Recently, this problem has been solved in our laboratory by the discovery of an inexpensive, convenient solvent system, that is the mixture of $NH_3\;and\;NH_4SCN$, for cellulose. Also, the $cellulose/NH_3/NH_4SCN$ solution system has been found to form the anisotropic, i.e., liquid crystalline phase. It is believed that both the cholesterio and the nematic phase occur. This finding has prompted extensive on-going researoh on the formation of the liquid crystalline phase from an inexpensive natural source such as cellulose since the nematic phase is envisioned as an excellent precursor sources for products with desirable properties, for example, high modulus and high strength. This interest naturally leads to a desire to understand the theological properties of the nematic phase so that the transformation of the nematic phase to the solid state with desirable properties can be efficiently accomplished, ;From this point of view, the theological behavior of the $cellulose/NH3_/NH_4SCN$ system has been studied as a function of shear rate and shear stress over a wide range of solvent compositions, cellulose concentration, centrifugation and urea contents, Results indicate that the viscosity decreases with increasing shear rate. A marked shear thinning behavior and a quasi-Newtonian behavior were observed in the low shear rate region and in the high shear rate region, respectively for all solvent compositions. The $cellulose/NH_3/NH_4SCN$ solution system only exhibited the viscosity increase with increasing cellulose concentration and failed to show the viscosity drop generally observed at the point of incipience of liquid crystal formation, This may be due to the gel-like nature of the solution by the association of the rodlike molecules into bundles which may serve as crosslinking points giving the cellulose solution a network structure. Also, simply hydrogen bonding may be so restrictive of molecular mobility that a viscosity drop is blocked. In addition to the above results, yield stress and thixotropy were also observed in the $cellulose/NH_3/NB_4SCN$ solution system which are characteristics of liquid crystal and gel, The results of the effect of centrifugation on viscosity show that viscosity decreases by the application of centrifugation. This may be explained by the change of the piled polydomain structure to the dispersed polydomain structure due to the pressure gradient generated during centrifugation.ation.

  • 친핵치환반응 ([Pd(ONN)Cl] + Y$^-\;{\rightleftharpoons}$ [Pd(ONN)Y] + Cl$^-$ ; Y = SCN$^-$, CN$^-$, N$_3^-$, imidazole, pyridine)속도를 25 ~ 45$^{\circ}C$ 범위에서 분광광도법으로 측정하였다. 친핵체의 반응성상수, n$_{Pd}^{\circ}$를 계산했으며 그 값은 CN$^-$ > SCN$^-$ > N$_3^-$ > Imidazole > Pyridine의 순서였다. 활성화엔탈피(${\Delta}H^{\neq}$)는 작은 양(+)의 값을, 활성화엔트로피(${\Delta}S^{\neq}$)는 큰 음(-)의 값을 가졌다. 이 결과로 보아 이 착물의 친핵 치환반응은 회합(A) 메카니즘으로 진행됨을 알 수 있었다.

In Situ Scanning Tunneling Microscope of Cyanide and Thiocyanate Adsorption on Pt(111)

  • Cyclic voltammetry and in situ STM were employed to examine the interfacial structures of a Pt(111) electrode in 0.1 mM KCN (pH9.5) and 0.1 mM KSCN (pH7) solutions. In situ STM atomic resolution revealed well ordered (2${\surd}$3${\times}$2${\surd}$3)$R30^{\circ}$-6CN and ($2{\times}2$)-2SCN structures within the double layer charging region. Six CN adsorbates formed a hollow hexagon, which embraced a coadsorbed $K^+$ cation. In contrast, the coadsorbed $K^+$ cations on the SCN covered Pt(111) were poorly ordered, despite adsorbed SCN formed a long range ordered ($2{\times}2$)-2SCN adlattice. In situ STM revealed the pronounced influence of potential in controlling the structures of compact layers at the proximity of a Pt electrode. Cathodic polarization facilitated the replacement of the coadsorbed cations by protons.

Preparation and Crystal Structures of Silver(I), Mercury(II), and Lead(II) Complexes of Oxathia-Tribenzo-Macrocycles

  • An investigation of the coordination behavior of sulfur-containing mixed-donor tribenzo-macrocycles $L^1-L^3$ ($L^1$: 20-membered $O_3S_2$, $L^2$: 20-membered $O_2S_3$, and $L^3$: 23-membered $O_4S_2$) with $d^{10}$-metal ($Ag^+$, $Hg^{2+}$, and $Pb^{2+}$) salts is reported. The X-ray structures of five complexes (1-5) with different structural types and stoichiometries, including mono- to dinuclear species have been determined. Reactions of $L^2$ and $L^3$ with the silver(I) salts ($PF_6{^-}$ and $SCN^-$) afforded two dinuclear 2:2 (metal-to-ligand) complexes with different arrangements: a sandwich-type cyclic dinuclear complex $[Ag_2(L^2)_2](PF_6)_2{\cdot}3CH_2Cl_2$ (1) and a linear dinuclear complex $[Ag_2(L^3)_2(SCN)_2]$ (2), in which two monosilver(I) complex units are linked by an Ag-Ag contact. Reactions of $L^1$ and $L^2$ with mercury(II) salts ($SCN^-$ and $Cl^-$) gave a mononuclear 1:1 complexes $[Hg(L^1)(SCN)_2]$ (3) and $[Hg(L^2)Cl_2]$ (4) with anion coordination in both cases. $L^2$ reacts with lead(II) perchlorate to yield a mononuclear sandwich-type complex $[Pb(L^2)_2(ClO_4)_2]$ (5), giving an overall metal coordination geometry of eight with a square antiprism arrangement. From these results, the effects of the donor variation and the anioncoordination ability on the resulting topologies of the soft metal complexes are discussed.

아끼바레 및 밀양 23호 쌀전분의 이화학적 성질비교 (Comparison of Physicochemical Properties of Akibare and Milyang 23 Rice Starch)

  • 아끼바레 및 밀양 23호 쌀 전분의 이화학적 성질, 전분 gel의 노화 및 음이온에 의한 호화양상을 조사하였다. 두 전분입자의 성상은 다각형이며 입자의 크기는 $3{\sim}6{\mu}m$ 정도였으며, 아끼바레가 밀양 23호 보다 다소 작았다. 두 전분의 X-ray 회절도는 A형으로 peak 형태와 강도에서 약간의 차이를 보였다. 아끼바레 및 밀양 23호 쌀 전분의 아밀로스함량은 각각 18.5 및 19.5%이었고 물 결합능력은 106 및 100%이었다. 팽화력은 $50^{\circ}C$까지는 큰 변화가 없었으나 이후에 급격히 증가하였고 밀양 23호 쌀 전분의 팽화력이 더 컸다. 전분 현탁액(0.1%)의 광투과도는 두 전분 모두 $60^{\circ}C$ 이후에서 급격히 증가하였으며 밀앙 23호 쌀 전분이 더 큰 광투과도를 보였다. 음이온에 의한 아밀로 그람 pattern은 NaCl과 NaF는 동일하였고 NaSCN과 NaI는 유사였다. 호화온도 및 최고점도시의 온도는 SCN-${SO_4}^=$의 순으로 증가하였고 SCN- 및 I-이온은 최고점도를 증가시켰으나 NaCl은 감소시켰다. ${SO_4}^=$ 이온을 첨가한 경우 점도는 계속 증가하였고 최고점도에 도달하지 않았다. 아끼바레 및 밀양 23호 전분 gel을 $21^{\circ}C$에서 저장한 경우 시간 상수가 각각 6.72 및 6.42일이었다.

Pivalic Acid-Ethanol 및 Succinonitrile-Salol 계에서의 수지상정 선단의 형상 (Dendrite Tip Shapes of Pivalic Acid-Ethanol and Succinonitrile-Salol Systems)

  • The shape of a dendrite tip has long been approximated by a paraboloid of revolution, but many attempts have been made as well to more accurately match the dendrite tip profile using other mathematical functions: power function, 4th order polynomial, and hyperbolic function. In the present work, dendrite tip shapes were matched by parabolic function. The differences between the dendrite tip shapes of pivalic acid(PVA)-ethanol(Eth) and succinonitrile(SCN)-salol systems, characterized by anisotropic and isotropic solid-liquid interfacial properties, respectively, were quantitatively treated using shape parameters. The PVA-Eth system showed a slightly higher Z/R value than the SCN-salol system, their Z/R values lying in the range 2-4. (Z is the distance from the tip beyond which the parabolic fit starts to deviate from the profile, and R the tip radius.) ${\lambda}_P$ is the distance from the tip beyond which side branching starts to appear, and is larger in the PVA-Eth system than the SCNsalol system. ${\lambda}_P$ is different for both sides of the 2-dimensional dendrite profile. The difference of ${\lambda}_P$ between both sides of the dendrite is larger for PVA-Eth system than for SCN-salol, implying that the dendrite of PVA-Eth is less symmetric than that of SCN-salol.
