Ultrasonic Velocity and Absorption Measurements for Polyacrylamide and Water Solutions

  • Both ultrasonic velocity at 3 MHz and absorption coefficient for the frequency range of 0.2-2 MHz were measured in an aqueous solution of polyacrylamide for the concentration range of 0.5% to 2.5% by weight. Pulse echo overlap method was taken for measuring the ultrasonic velocity over the temperature range of 10-90℃ and the high-Q ultrasonic resonator method was used for the absorption coefficient at 30℃. The velocity exhibited a maximum value at approximately 70℃, 71℃, 72℃, 73℃ and 74℃ in 2.5%, 2.0%, 1.5%, 1.0%, and 0.5% solutions, respectively. The velocity increased with the concentration at a given temperature. The ultrasonic absorption (a/f²) at a given temperature increased linearly with the concentration for the concentration below 1.5%, but suddenly increased for the concentration above 1.5% concentration. The value of a/f² at 1MHz was entirely due to the classical Stoke's viscous effect. The ultrasonic relaxation in polyacrylamide aqueous solutions, which may be the result of structural fluctuations of polymer molecules such as the segmental motion of the polymer chains, was observed, and at 2.5%, the value of a/f² was found to suddenly increase as frequency decreased.

  • Recently various attempts have been made to apply HSPF model to calculate runoff and diffuse pollution loads of stream and reservoir watersheds. Because the role of standard flow is very important in the water quality modelling of Total Water Pollution Load Management, HSPF was used as a means of estimating standard flow. In this study, BASINS 4.0 and WinHSPF was applied to the Gomakwoncheon watershed, genetic algorithm(GA) and influence coefficient algorithm were used to calibrate the runoff parameters of the WinHSPF. The objective function is the sum of the squares of the normalized residuals of the observed and calculated flow and it is optimized using GA. Estimates of the optimum runoff parameters are made at each iteration of the influence coefficient algorithm. The calibration results showed a relatively good correspondence between the observed and the calculated values. The standard flow(Q275) of the Gomakwoncheon watershed was estimated using the ten years of weather data.

  • Recently, copy number variations (CNV) of genes or genomic segments have been intensively studied and various analysis methods have been developed. In this study, quantitative oligonucleotide ligation assay (qOLA) was applied to investigate CNV of KIT gene in the Landrace breed. A combined assay using qOLA and pyrosequencing, 6 genotype classes, I1/I1 or I3/i (IBe), I1/I2 or I3/IP, I1/I3, I1/IP or I2/i (IBe), I2/I2and I2/IP, were identified from 44 Landrace pigs. Genotype assignment using grouping features of measurements on a scatter plot showed 100% agreement with those using a statistical assignment by PROC FASTCLUS procedure implemented in the SAS package. Two versions (3100 and 3130) of ABI sequencers gave the same genotyping results, indicating there was no influence on qOLA by different versions of instrument, however, the means of standard deviation and coefficient of variation from the qOLA on a ABI 3130 (2.33 and 4.10) was lower than those from the qOLA on a ABI 3100 (2.67 and 4.81). Effect of proteinase K treatment on the PCR product followed by qOLA was very clear because noise peaks were disappeared and the observed ration fit better to the reference ratio corresponding to each genotype.

Effect of the Application of Temporal Mask Map on the Relationship between NDVI and Rice Yield (시계열 마스크 맵이 논벼 NDVI와 단수와의 관계에 미치는 영향)

  • The objectives of this study were (1) to develop a temporal mask map using MCD12Q1 data, and (2) to extract the annual variations in paddy, (3) to investigate the correlation analysis between MYD13Q1 NDVI and rice yield, and (4) to review its applicability. For these purposes, the temporal mask map was created using annual MCD12Q1 PFT data from 2002 to 2019, and compared with the fixed mask map. As a result, it found that the temporal mask map well reflected the variations of the paddy area. In addition, the correlation coefficient between NDVI and rice yield was also high significant as compared to the fixed mask map. Therefore, the temporal mask map will be useful for NDVI extraction, crop monitoring, and estimation of rice yield.

An Exploratory Study of Electrochemical Skin Conductance for the Deficiency Pattern Identification in Diabetic Patients (당뇨병 환자의 허증별 전기전도도 특성에 대한 탐색적 관찰 연구)

  • Objectives The objective of this study is to examine the interpretability of the questionnaire-based pattern identification in terms of biosignals. For this purpose, we investigate the relationship between electrochemical skin conductance (ESC) and Qi-Blood-Yin-Yang Deficiency Questionnaire (QBYY-Q) in diabetic patients. Methods A total of 40 patients with diabetes mellitus answered the QBYY-Q and their ESC were measured by SUDOSCAN device (a diabetes screening device, France). To analyze the relationship between QBYY-Q and ESC, ANOVA analysis and Scheffe test were performed and Pearson correlation coefficients were obtained. Results Of the 40 diabetic patients, 23 (57.5%) were males and 17 (42.5%) were females. According to the QBYY-Q, 9 patients were classified into Qi deficiency pattern (QD), 9 patients were Blood deficiency pattern (BD), 10 patients were Yin deficiency pattern (YiD) and 12 patients were Yang deficiency pattern (YaD). Demographic information (age, body mass index, duration of illness, etc.), signs of vitality (blood pressure, body temperature, etc.), fasting plasma glucose and glycated hemoglobin were not significantly different in each deficiency pattern. The ESC of the right leg was significantly lower in the BD group compared to the YiD group (p<0.022). Pearson's correlation coefficient was negatively correlated with the BD questionnaire score (r=-0.343, p <0.05). Finally, ESC showed a positive correlation with hemoglobin and erythrocyte levels in all limbs (r=0.483, p<0.01). Conclusions We showed that ESC could be used to classify the Deficiency pattern identifications in diabetic patients. Especially, the ESC was significantly lower in the BD group and was negatively correlated with the BD scores. It implies the potential utility of the ESC to understand the BD in terms of modern biosignals.

  • In the classification of satellite images, the representative of training of classes is very important factor that affects the classification accuracy. Hence, in order to improve the classification accuracy, it is required to optimize pre-classification stage which determines classification parameters rather than to develop classifiers alone. In this study, the normality of training are calculated at the preclassification stage using SPOT XS and LANDSAT TM. A correlation coefficient of multivariate Q-Q plot with 5% significance level and a variance of initial training are considered as an object function of genetic algorithm in the training normalization process. As a result of normalization of training using the genetic algorithm, it was proved that, for the study area, the mean and variance of each class shifted to the population, and the result showed the possibility of prediction of the distribution of each class.

Derivation of Roughness Coefficient Relationships Using Field Data in Vegetated Rivers (식생하천의 현장자료를 이용한 조도계수 관계식 유도)

  • Field measurements of resistance to flow are analyzed for 739 rivers vegetated with grass (281 channels), shrubs (150 channels) and trees (308 channels). The measured distribution of Manning roughness coefficients ranges from 0.015~0.250 for grass, 0.016~0.250 for shrubs, 0.018~0.310 for trees. Significant trends are obtained between Darcy-Weisbach (or Manning roughness coefficients) and flow discharge, friction slope, and relative submergence. The regression equations for Darcy-Weisbach and Manning roughness coefficients in vegetated rivers are: $f_{veg}=0.436Q^{-0.363}$, $f_{veg}=3.305S_f^{0.508}$, and $n_{veg}=0.061Q^{-0.124}$, $n_{veg}=0.144S_f^{0.199}$, $V=5.3(h/d_{50})^{1/8.3}{\sqrt{ghS_f}}$, $\sqrt{8/f}(=V/u*)=5.75log(5h/d_{50})$, respectively. These semi-empirical relationships should be useful for hydraulic engineering practice.

  • We investigated statistical evaluation parameters for 3D meteorological and air quality models and selected several quantitative indicator references, and summarized the reference values of the statistical parameters for domestic air quality modeling researcher. The finally selected 9 statistical parameters are MB (Mean Bias), ME (Mean Error), MNB (Mean Normalized Bias Error), MNE (Mean Absolute Gross Error), RMSE (Root Mean Square Error), IOA (Index of Agreement), R (Correlation Coefficient), FE (Fractional Error), FB (Fractional Bias), and the associated reference values are summarized. The results showed that MB and ME have been widely used in evaluating the meteorological model output, and NMB and NME are most frequently used for air quality model results. In addition, discussed are the presentation diagrams such as Soccer Plot, Taylor diagram, and Q-Q (Quantile-Quantile) diagram. The current results from our study is expected to be effectively used as the statistical evaluation parameters suitable for situation in Korea considering various characteristics such as including the mountainous surface areas.

Influence of Nd2O3 Addition to 0.3CaTiO3-0.7(Li1/2Nb1/2)TiO3 on their Microwave Dielectric Properties (Nd2O3 첨가가 0.3CaTiO3-0.7(Li1/2Nb1/2)TiO3 세라믹스의 마이크로파 유전특성에 미치는 영향)

  • The effects of $Nd_2O_3$ addition to $Q{\cdot}f_{0}(GHz)$ ceramics with ${\varepsilon}_r$ of 126, $Q{\cdot}f_{0}(GHz)$ of 2240 and of $68\;ppm/^{\circ}C$ on their microwave properties were investigated. For the addition of 5 wt% $Nd_2O_3$, the dielectric constant (${\varepsilon}_r$) showed maximum value of 131, then decreased with the further addition of $Nd_2O_3$. $Q{\cdot}f_{0}(GHz)$ value was still increased to 3533 with 9 wt% $Nd_2O_3$ addition, it is influenced by densification of grain boundary. With more addition of $Nd_2O_3$ up to 18 wt%, the abnormal grain growth have influence on the decreasing of $Q{\cdot}f_{0}(GHz)$ value. But with the further addition of $Nd_2O_3$ over 25 wt%, the $Q{\cdot}f_{0}(GHz)$ value was again increased by the effect of the second phase ($Nd_2Ti_2O_7$) forming. The temperature coefficient of resonant frequency (${\tau}_f$) was decreased from $+\;68\;ppm/^{\circ}C$ with the addition of $Nd_2O_3$, reached $0\;ppm/^{\circ}C$ at 12 wt% addition, and became negative with the further addition of $Nd_2O_3$. The optimum microwave dielectric properties were obtained for $0.3CaTiO_3-0.7(Li_{1/2}Nd_{1/2})TiO_3$ with 9 wt% $Nd_2O_3$ sintered at $1425^{\circ}C$ for 3 hrs. The dielectric constant (${\varepsilon}_r$), the $Q{\cdot}f_{0}(GHz)$ value, and the temperature coefficient of resonant frequency (${\tau}_f$) were 108, 3533, and $+\;6\;ppm/^{\circ}C$, respectively.

Validation of Stem-loop RT-qPCR Method on the Pharmacokinetic Analysis of siRNA Therapeutics (Stem-loop RT-qPCR 분석법을 이용한 siRNA 치료제의 생체시료 분석법 검증 및 약물 동태학적 분석)

  • The first small interfering RNA (siRNA) therapeutics have recently been approved by the Food and Drug Administration in the U.S., and the demand for a new RNA therapeutics bioanalysis method-which is essential for pharmacokinetics, including the absorption, distribution, metabolism, and excretion of siRNA therapeutics-is rapidly increasing. The stem-loop real-time qPCR (RT-qPCR) assay is a useful molecular technique for the identification and quantification of small RNA (e.g., micro RNA and siRNA) and can be applied for the bioanalysis of siRNA therapeutics. When the anti-HPV E6/E7 siRNA therapeutic was used in preclinical trials, the established stem-loop RT-qPCR assay was validated. The limit of detection was sensitive up to 10 fM and the lower limit of quantification up to 100 fM. In fact, the reliability of the established test method was further validated in three intra assays. Here, the correlation coefficient of $R^2$>0.99, the slope of -3.10 ~ -3.40, and the recovery rate within ${\pm}20%$ of the siRNA standard curve confirm its excellent robustness. Finally, the circulation profiles of siRNAs were demonstrated in rat serum, and the pharmacokinetic properties of the anti-HPV E6/E7 siRNA therapeutic were characterized using a stem-loop RT-qPCR assay. Therefore, the stemloop RT-qPCR assay enables accurate, precise, and sensitive siRNA duplex quantification and is suitable for the quantification of small RNA therapeutics using small volumes of biological samples.
