Cytotoxicity and Structure Activity Relationship of Dammarane-Type Triterpenoids from the Bark of Aglaia elliptica against P-388 Murine Leukemia Cells

  • Six dammarane-type triterpenoids, dammar-24-en-$3{\beta}$-ol (1), $3{\beta}$-epicabraleahydroxy lactone (2), (E)-25-hydroperoxydammar-23-en-$3{\beta}$,20-diol (3), dammar-24-en-$3{\beta}$,20-diol (4), $3{\beta}$-acetyl-20S,24S-epoxy-25-hydroxydammarane (5), and $3{\beta}$-epiocotillol (6) were isolated from the methanolic extract of the bark of Aglaia elliptica. The chemical structure were identified on the basis of spectroscopic evidence and by comparison with those spectra previously reported. Compounds 1 - 6 were isolated first time from this plant. Compounds 1 - 6, along with a known synthetic analog, cabraleone (7) were evaluated their cytotoxic activity against P-388 murine leukimia cells in vitro. Among those compounds $3{\beta}$-acetyl-20S,24S-epoxy-25-hydroxydammarane (5) showed strongest cytotoxic activity with $IC_{50}$ value of $8.02{\pm}0.06{\mu}M$.

꾸지 뽕나무로부터 분리한 flavonoid계 화합물의 암세포성장 저해 및 항산화 활성 (Tumor cell growth inhibition and antioxydative activity of flavonoids from the stem bark of Cudrania tricuspidata)

  • 꾸지 뽕나무의 줄기껍질로부터 P388 tumor cell line에 대한 세포 성장 저해활성과 흰쥐 간 microsome 분획의 과산화 지질을 사용하여 $Fe^{++}/\;ascorbate$법으로 측정한 결과 세포 성장 저해활성과 항산화활성을 갖는 5개의 flavonoid계 화합물을 분리하였다. 분리된 화합물의 구조분석을 실시한결과 각종 spectral data와 보고된 문헌에 의하여 taxifolin, orobol, eriodictyol, dihydrokaempferol, steppogenin으로 각각 동정되었다. 이 화합물들의 항산화활성은 $IC_{50}$이 각각 6, 3, 3, >50, $10\;{\mu}g/ml$이었고 taxifolin을 제외한 P388 cell line에 대한 세포 성장저해는 각각 $IC_{50}$이 0.18, 3.3, 15, $6.2\;{\mu}g/ml$이었다. 한편 Escherichia coli BE 1186, Salmonella thyphimurium SL 1102, Staphylococcus aureus IFO 12732, Staphylococcus aureus R 209, Candida albicans에 대해서는 항균활성을 나타내지 않았다.

Synthesis and Evaluation of Antitumor Activity of Novel 1,4-Naphthoquinone Derivatives (IV)

  • 1,4-Naphthoquinones are widely distributed in nature and many clinically important antitumor drugs containing a quinone moiety, such as anthracyclines, mitoxantrones and saintopin, show excellent anticancer activity. In this study, 2- or 6-substituted 5,8-dimethoxy-1,4-naphthoquinone (DMNQ) and 5,8-dihydroxy-1,4-naphthoquinone (DHNQ) derivatives were synthesized, and their cytotoxic activity against L1210 and P388 cancer cells was examined. Their antitumor activity was also assessed in mice bearing S-180 cells in the peritoneal cavity. In comparison with the DMNQ derivatives, the DHNQ derivatives exhibited more potent bioactivities than the DMNQ derivatives against both L1210 and P388 cells in vitro and S-180 cells in vivo. The $ED_{50}$ values of the DHNQ derivatives against P388 cells were in the range of 0.18-1.81 ${\mu}g/mL$ whereas those of the DMNQ derivatives were in the range of 0.26-40.41 ${\mu}g/mL$. The T/C ($\%$) values of the DHNQ derivatives, 8, 17, 18, 19, and 20, were found to be comparable to or even better than that of adriamycin. It was also observed that the 2-substituted derivatives (8, 19, 20) showed better antitumor activity than the 6-substituted derivatives (7, 17, 18) in the mice bearing S-180 cells in the peritoneal cavity.

Cytotoxic activities of various fractions extracted from some pharmaceutical insect relatives

  • This research was performed to screen the cytotoxic activities of some pharmaceutical insect relatives. Cytotoxic activities of total extract and fractions of hexane, ethyl acetate, methanol, water and boiling water were extracted from four pharmaceutical insect relatives: the Chinese gall, the cicada slough, the hornet nest and the batryticated silkworm. These extracts were investigated against the cancer cell lines of L1210, P388 and SNU-1 in vitro tests. Results showed that, ED, , against the cancer cell lines of L1210, P388 and SNU-1 were 0.55, 0.50, and $0.83{\mu}g/ml$ in the ethyl acetate fraction from the Chinese gall; 1.07, 2.19, and $2.24{\mu}g/ml$in the ethyl acetate fraction, 1.51, 1.26, and $1.45{\mu}g/ml$ in the water fraction and 1.48, 2.29, and $1.29{\mu}g/ml$in the boiling water fraction from the cicada slough; 3.31, 2.00, and $6.61\mug/ml$ in the water fraction from the hornet nest and 13.80, 19.95, and $31.62{\mu}g/ml$in the hexane fraction and 33.88, 21.88, and $25.12{\mu}g/ml$in the ethyl acetate fraction from the batryticated silkworm, respectively. All of the fractions mentioned above showed high cytotoxic activities and could be suggested for further studies in vivo tests.

솔잎, Pinus densiflora Sieb.et Zucc., 의 항암효과(抗癌效果)에 대한 연구(硏究) (Studies on antitumor effects of pine needles, Pinus densiflora Sieb.et Zucc)

  • The pine needles, Pinus densiflow Sieb. et Zucc., which is a feed for goats showing a low incidence rate of cancer were evaluated to confirm the potent anticancer effects, with or without several conventional anticancer drugs. The pine needles collected from Mt. Buk-Han located near Seoul were extracted with 95% methanol and methand and concentrated. From the methanol extract, SOM-A, was extracted dichlormethane and SOM-B was extracted with ethyl acetate. SOM-C was extracted with distilled water. These extracts were tested for their antitumor activities in vitro and in vivo. Among them, SOM-A and SOM-C exhibited potent antitumor activities described as belows. 1. The cytotoxic effects of SOM-A and SOM-C were examined against in vitro cultured murine and humman tumor cells. SOM-A showed strong cytotoxicity against human tumor cell lines and SOM-C showed strong cytotoxicity against murine tumor cell lines tested. 2. The antitumor effects of SOM-A and SOM-C were examined against P388 and L1210 of mouse ascitic tumors. The highest mean survival time(MST) ration was 151%(P388) for SOM-C(90mg/kg). 3. To compare the antitumor effects of SOM-A, SOM-B, and SOM-C against solid tumors, S-180 and Ehrlich carcinoma were implanted subcutaneously to mice on Day O. The drugs were given intraperitoneally to mice once a day on Days 1-20, and the tumor weights were measured on Day 21. SOM-A showed inhibition of tumor growth more than 50% in the experiment on S-180 and Ehrlich, and SOM-C also markedly inhibited tumor growth. However, SOM-B had no effect. 4. SOM-C combined with ${\alpha}$-interferon and SOM-C combined with Mitomycin-C enhanced the antitumor activities against murine ascitic tumors P388 leukemia.

Taxol 유도체들의 생물학적 거동에 관한 연구 (Studies on the Biological Behaviors of Taxol Derivatives)

  • 본 연구에서는 항암제인 taxol의 $^{111}In$ 방사성표지화합물을 합성하여 암진단제로서의 이용 가능성을 보기 위한 기초 연구를 수행하였다. Taxol의 $^{111}In$ 표지화합물을 얻기 위해 taxol구조에서 C-13의 곁가지에 있는 C-2' 부분의 hydroxyl기를 DTPA anhydride 및 succinic anhydride와 반응시켜 taxol-DTPA와 2'-hemisuccinyltaxol을 합성하였다. 반응수율은 taxol-DTPA 접합체의 경우 34%이었으며, 2'-hemisuccinyltaxol은 80%이었다. MTT법을 사용하여 HT29, B16, P388, CT26 세포주에서 taxol-DTPA와 2'-hemisuccinyltaxol의 세포독성능실험에서는 taxol 보다는 못미치나 그 세포독성이 유지됨을 확인하였다. 합성된 taxol 유도체들을 리간드 교환법과 직접법을 사용하여 In-111을 표지하였다. Taxol-DTPA 접합체의 In-111 표지반응의 경우, 리간드교환법은 반응도중 침전이 생겨 반응이 어려워 직접법으로 In-111 표지화합물을 얻을 수 있었으며 그 표지수율은 100%이었다. 2'-hemisuccinyltaxol은 두 방법을 모두 시도하였으나 반응이 진행되지 않음을 확인하였다. In-111의 taxol-DTPA 접합체 및 2'-hemisuccinyltaxol에 대한 표지반응 수율은 HPLC, paper, instant thin-layer chromatography를 실시하여 결정하였다. Li-pophilicity의 실험에서는 친수성임이 확인되었으며, 세포결합능의 실험에서는 HT29, B16, P388, CT26 세포주와의 결합이 매우 낮음을 나타내었다. 혈청단백 질과의 결합능을 보기위하여 30% trichloroacetic acid 법을 수행하였으며, 약 30%정도만이 혈청단백질과 결합하여 그 값이 크지 않았다.

Accuracy of 14 intraoral scanners for the All-on-4 treatment concept: a comparative in vitro study

  • PURPOSE. This in vitro study aimed to evaluate the accuracy of 14 different intraoral scanners for the All-on-4 treatment concept. MATERIALS AND METHODS. Four implants were placed in regions 13, 16, 23, and 26 of an edentulous maxillary model that was poured with scannable Type 4 gypsum to imitate the All-on-4 concept. The cast was scanned 10 times for each of 14 intraoral scanners (Primescan, iTero 2, iTero 5D, Virtuo Vivo, Trios 3, Trios 4, CS3600, CS3700, Emerald, Emerald S, Medit i500, BenQ BIS-I, Heron IOS, and Aadva IOS 100P) after the polyether ether ketone scanbody was placed. For the control group, the gypsum model was scanned 10 times with an industrial scanner. The first of the 10 virtual models obtained from the industrial model was chosen as the reference model. For trueness, the data of the 14 dental scanners were superimposed with the reference model; for precision, the data of all 14 scanners were superimposed within the groups. Statistical analyses were performed using the Kolmogorov-Smirnov, Shapiro-Wilks, and Dunn's tests. RESULTS. Primescan showed the highest trueness and precision values (P < .005), followed by the iTero 5D scanner (P < .005). CONCLUSION. Some of these digital scanners can be used to make impressions within the All-on-4 concept. However, the possibility of data loss due to artifacts, reflections, and the inability to combine the data should be considered.

간호대학생의 학업 및 임상실습 스트레스와 전공만족도와의 관계 (Relationship between Academic and Clinical Practice Stress and Major Satisfaction in Nursing Students)

  • 본 연구는 간호대학생의 학업 및 임상실습 스트레스와 전공 만족도의 관계를 확인학기 위하여 시도되었다. 자료는 140명의 B대학교의 간호대학생으로부터 온라인 설문 조사를 통해 수집되었다. 수집된 자료는 SPSS 18.0을 사용하여 분석되었다. 일반적인 특성에 따른 학업 및 임상실습 스트레스, 전공 만족도와의 차이를 분석한 결과, 학업 스트레스는 종교와 차이가 있었으며(F=14.348, p<.001), 전공 만족도는 성적에 따라 차이가 있는 것으로 나타났다(F=4.582, p=0.004). 학업 스트레스, 임상실습 스트레스 및 전공 만족도의 상관 관계를 분석한 결과, 학업 스트레스는 임상실습 스트레스와 양의 상관관계가 있고(r=.388, p<.01), 임상실습 스트레스는 전공 만족도와 양의 상관 관계가 있는 것으로 나타났다 (r=.336, p<.01). 따라서 간호학 전공의 만족도를 높이기 위해서는 학업 스트레스를 낮추는 프로그램 개발이 필요하다고 판단된다.

중소 제조업체의 친환경적 경영활동이 직무향상에 미치는 영향 (Effect of Environmentally Friendly Management Activities of SMEs on Job Improvement)

  • 본 연구는 중소 제조업체의 친환경적 경영활동이 직무향상에 미치는 실증 연구로서 친환경 경영의 현황을 파악하고자 중소기업 제조업체를 대상으로 설문지 총 128개를 분석하였다. 분석결과를 요약하면 다음과 같다. 첫째. 중소기업의 친환경적 경영활동 중 사회적 활동은 직무향상에 통계적으로 유의한 영향을 미치지 못한 것으로 분석되었고, 둘째. 환경적 활동은 업무생산성에 𝛽= .394(p<.001)로 유의미한 영향을 미치는 것으로 나타났고, 직무만족에서는 𝛽=.216, p<.05로 낮은 인과관계를 보이며, 셋째. 경제적 경영활동은 직무만족에서 𝛽= .388(p<.001)로 유의한 영향을 미친 것으로 나타났다. 또한, 업무생 산성에는 β= .267(p<.01)로 낮은 인과관계를 확인하였다. 따라서 직무향상에 미치는 영향 중 업무생산성은 환경적 활동이, 직무만족은 경제적 경영활동이 직무향상에 영향을 미치는 것을 알 수 있었다. 본 연구는 중소기업의 친환경적 경영활동을 위한 환경조성과 경쟁력 있는 전략적 방향 제시 및 연구의 기초자료 제공에 의미가 있다.
