A STUDY OF THE MECHANISM OF IMPROVING ACID RESISTANCE OF BOVINE TOOTH ENAMEL AFTER PULSED Nd-YAG LASER IRRADIATION (펄스형 Nd-YAG 레이저 조사에 의한 법랑질 내산성 증가 기전에 관한 연구)

  • The purpose of this study was to examine the mechanism of improving acid resistance of Nd-YAG laser irradiated tooth enamel and determine the most effective energy density for improving acid resistance. The bovine tooth enamel were lased with a pulsed Nd-YAG laser. The energy densities of exposed laser beam were varied from 10 to $70\;J/cm^2$. To investigate the degree of improving acid resistance by irradiation, all the samples were submerged to demineralize in 0.5 N $HClO_4$ solution for 1 minute. After 1 minute, 0.05 % $LaCl_3$ was added to the solution for interrupting the demineralization reaction. The amounts of dissolved calcium and phosphate in the solution were measured by using an atomic absorption spectrophotometer and the UV/VIS spectrophotometer, respectively. To examine the mechanism of improving acid resistance, X-ray diffraction analysis, infrared spectroscopy, and scanning electron microscopy were taken. The X-ray diffraction pattern of the samples were obtained in the $10^{\circ}{\sim}80^{\circ}2{\theta}$ range with $Cu-K{\alpha}$ radiation using M18HF(Mac Science Co.) with X-ray diffractometer operating at 40 KV and 300 mA. The infra-red spectra of the ground samples in 300 mg KBr pellets 10 mm diameter were obtained in the $4000cm^{-1}\;to\;400cm^{-1}$ range using JASCO 300E spectrophotometer. The scanning electron microscopy was carried out using JSM6400(JEOL Co.) with $500{\sim}2000$ times magnification. The results were as follow 1. The concentration of calcium dissolved from laser irradiated enamel with $50J/cm^2$ was significantly lesser than that of unlased control group (p<0.05) 2. From the result of the X-ray diffraction analysis, $\beta$-TCP, which increases acid solubility, was identified in lased enamel but the diffraction peaks of (002) and (004) became sharp with increasing energy density of laser irradiation. This means that the crystals in lased samples were grown through the c-axis and subsequently, the acid solubility of enamel decreased. 3. The a-axis parameter was slightly increased by laser irradiation, whereas the c-axis parameter was almost constant except for a little decrease at $50J/cm^2$. 4. In the infra-red spectra of lased enamels, phosphate bands ($600{\sim}500cm^{-1}$), B-carbonate bands (870, $1415{\sim}1455cm^{-1}$), and A-carbonate band ($1545cm^{-1}$) were observed. The amounts of phosphate bands and the B-carbonate bands were reduced, on the other hand, the amount of the A-carbonate band was increased by increase the energy density. 5. The SEM experiments reveal that the surface melting and recrystallization were appeared at $30J/cm^2$ and the cracks were observed at $70J/cm^2$. From above results, It may be suggested that the most effective energy density for improving acid resistance of tooth enamel with the irradiation of Nd-YAG laser was $50J/cm^2$. The mechanism of improving acid resistance were reduction of permeability due to surface melting and recrystallization of lased enamel and reduction of acid solubility of enamel due to decrease of carbonate content and growth of crystal.

Molecular Cloning of nifH, D from Frankia EuIK1 Strain, A Symbiont of Elaeagnus umbellata Root Nodules (보리수나무 뿌리혹 공생균주인 Frankia EuIK1의 nifH, D클로닝)

  • Genomic Southern hybridization of Frankia EuIKl strain, a nitrogen fixing symbiont of Elaeagnus umbellate root nodules, with nifH,D of K. pneumoniae as a probe, showed that 3.2 Kb and 5.5 Kb of BamHI fragments and 15 Kb PstI fragment were strongly hybridized with the probe, indicating nifH,D are located on these fragments. Using the same probe, one clone(pEuNIF) was isolated from the genomic library constructed into pWE15 cosmid vector by colony hybridization. The 3.2 Kb and 5.5 Kb BamHI fragments of this clone were hybridized with the same probe and this result corresponds to the genomic Southern hybridization data. However, using nifH of Frankia FaCl strain as a probe, only the 3.2 Kb BamHI fragment showed hybridization signal. Amino acid sequence deduced from nucleotide sequence of 3' terminus of the 3.2 Kb and 5' terminus of the 5.5 Kb fragments showed that the former was highly homologous with that of ArI3 nifD from 182nd to 240th amino acids, while the latter was from 241st to 282nd amino acids. These results show that nifH and partial nifD sequences are located on the 3.2 Kb fragment and residual sequences of nifH on the 5.5 Kb fragment which is contiguous to the 3.2 Kb fragment.

Kinetics and Mechanism of the Anilinolysis of Dibutyl Chlorophosphate in Acetonitrile

  • The nucleophilic substitution reactions of dibutyl chlorophosphate (3) with substituted anilines ($XC_6H_4NH_2$) and deuterated anilines ($XC_6H_4ND_2$) are investigated kinetically in acetonitrile at $55.0^{\circ}C$. The obtained deuterium kinetic isotope effects (DKIEs; kH/kD) are secondary inverse ($k_H/k_D$ = 0.86-0.97) with the strongly basic anilines while primary normal ($k_H/k_D$ = 1.04-1.10) with the weakly basic anilines. The DKIEs, steric effects of the two ligands, activation parameters, cross-interaction constants, variation trends of the kH/kD values with X, and mechanism are discussed for the anilinolyses of the nine ($R_1O$)($R_2O$)P(=O)Cl-type chlorophosphates. A concerted mechanism is proposed with a backside nucleophilic attack transition state for the strongly basic anilines and with a frontside attack involving a hydrogen-bonded four-center-type transition state for the weakly basic anilines on the basis of the magnitudes, secondary inverse and primary normal, and variation trends of the $k_H/k_D$ values with X.

Enhanced Electrochemifluorescence and Reduction Mechanism of Acetoxy Coumarin Derivatives in Acetonitrile Solution

  • The electrochemical reduction of coumarin, 7-acetoxy-4-methyl coumarin (AMC), and 7-acetoxy-4-bromomethyl coumarin (ABMC), in 0.1 M tetraethyl ammonium perchlorate/acetonitrile solution was carried out by direct current, differential pulse polarography, cyclic voltammetry, and controlled potential coulometry. The electrochemical reduction of ABMC was proceeded through three steps of electron transfer coupled with the chemical reactions. The color of solution was changed to yellow when the carbonyl group was reduced during 2nd step (-1.8 volts) and independented with cleavage of bromo group. Highest fluorescence intensity showed when the electrochemical reduction of AMC was controlled at near the potential (-2.3 volts vs. Ag/AgCl).

  • Stainless steel has been stably used closed by passivity oxidation films($Cr_2O_3$) is made by neutral atmospheric environment. However, passivity oxidation films of the surface of stainless steel occasionally comes to be destroyed in seawater which is influenced by an environment having halogen ion like $Cl^-$, then, localization corrosion comes to occur. Stainless steel 304 for shaft system material of the small-size FRP fishing boat on seawater environments made an experiment on simulation of sacrifical anode(Al, Zn). Through these experiment and study, following results have been obtained ; According to the field inspection and corrosion simulation, the corrosion on the 2nd class stainless steel shaft(STS 304) in FRP fishing boat has been verified to occur by crevice corrosion and galvanic corrosion etc.. According to the comparison and analysis of Stainless steel 304 shaft materials after simulation leaving unprotected and applying cathodic protection, unprotected shaft specimen of stainless steel 304 was severely corroded, but, protected shaft specimen was not totally corroded. This result is assumed to be made by the facts that anodic reaction, $Fe{\rightarrow}Fe^{2+}$ + $2e^-$, has been restricted by the cathodic protection current of sacrificial anode material.

Removal of methylene blue using lemon grass ash as an adsorbent

  • Wastewater from textile industries is a major cause of water pollution in most developing countries. In order to address the issues of water pollution and high cost for treatment processes, the use of an inexpensive and environmentally benign adsorbents has been studied. The objective was to find a better alternative to the conventional methods. Lemon grass waste (ash) collected from a lemon grass stream distillation subunit in Bhutan was tested for dye removal from aqueous solutions. The study investigated the removal of methylene blue using the following operational parameters: initial concentration (100-600 mg/L), contact time, adsorbent dose (0.1-0.55 gm/100 mL), and pH (3-10). It was found that the percentage removal of dye increased with a decrease of the initial concentration and increased contact time and dose of adsorbent. The basic pH solution of dye showed better adsorption capacity as compared to the acidic dye solution. Langmuir and Freundlich adsorption isotherms were fitted to the data well. Data fitted better to Lagergren pseudo 2nd order kinetics than a 1st order kinetic model. Surface morphology was also examined via scanning electron microscopy. An elemental analysis was also carried out and the chemical composition and functional groups were analyzed using energy dispersive X-ray spectroscopy and Fourier transform infrared spectroscopy techniques, respectively. The obtained results indicate that lemon grass ash could be employed as a low cost alternative to commercial activated carbon in wastewater treatment for the removal of dyes.

  • Recently, rapid palatal expansion technique is widely used for the correction of the skeletal imbalance in Cl III malocclusion patients. There were many studies about the cephalometric changes to rapid palatal expansion but quantitative analysis were small. The purpose of this study was to analysis the stresses and displacement of the maxilla in human dry skull to rapid palatal expansion. The results were as follows: 1. The anterior portion of palate show more lateral and inferior displacement than the posterior portion. But the posterior portion show more anterior displacement. 2. In transpalatal suture area, the medial portion show more anterior and inferior displacement than the lateral portion. But the lateral portion show more lateral displacement than the medial portion. 3. In mid-sagittal plane, the lower portion (palatal area) of maxilla show more anterior, lateral, inferior displacement than the upper portion (frontamaxillary stuture area). 4. In zygomatic arch, the adjacent area to maxilla show tonsil. stresses and the adjacent area to frontal bone show compressive stresses. 5. The sequence of stress bearing area to R.P.E. is upper retromolar area, upper 1st molar, 1st premolar, 2nd premolar, anterior segment of teeth.

Kinetics and Mechanism of the Anilinolysis of Dipropyl Chlorophosphate in Acetonitrile

  • The kinetic studies on the reactions of dipropyl chlorophosphate (3O) with substituted anilines ($XC_6H_4NH_2$) and deuterated anilines ($XC_6H_4ND_2$) have been carried out in acetonitrile at $55.0^{\circ}C$. The obtained deuterium kinetic isotope effects (DKIEs; $k_H/k_D$) are primary normal ($k_H/k_D$ = 1.09-1.01) with the strongly basic anilines while secondary inverse ($k_H/k_D$ = 0.74-0.82) with the weakly basic anilines. The steric effects of the two ligands on the rates are extensively discussed for the anilinolyses of the ($R_1O$)($R_2O$)P(=O or S)Cl-type chlorophosphates and chlorothiophosphates. A concerted mechanism is proposed with a frontside nucleophilic attack involving a hydrogen-bonded four-center-type transition state for the strongly basic anilines and with a backside attack transition state for the weakly basic anilines on the basis of the DKIEs, primary normal and secondary inverse with the strongly and weakly basic anilines, respectively.

Behavior of Radioactive Metal Surrogates Under Various Waste Combustion Conditions

  • A laboratory investigation of the behavior of radioactive metals under the various waste combustion atmospheres was conducted to predict the parameters that influence their partitioning behavior during waste incineration. Neodymium, samarium, cerium, gadolinium, cesium and cobalt were used as non-radioactive surrogate metals that are representative of uranium, plutonium, americium, curium, radioactive cesium, and radioactive cobalt, respectively. Except for cesium, all of the investigated surrogate metal compounds converted into each of their stable oxides at medium temperatures from 400 to 90$0^{\circ}C$, under oxygen- deficient and oxygen-sufficient atmospheres (0.001-atm and 0.21-atm $O_2$). At high temperatures above 1,40$0^{\circ}C$, cerium, neodymium and samarium in the form of their oxides started to vaporize but the vaporization rates were very slow up to 150$0^{\circ}C$ . Inorganic chlorine (NaCl) as well as organic chlorine (PVC) did not impact the volatility of investigated Nd$_2$O$_3$, CoO and Cs$_2$O. The results of laboratory investigations suggested that the combustion chamber operating parameters affecting the entrainment of particulate and filtration equipment operating parameters affecting particle collection efficiency be the governing parameters of alpha radionuclides partitioning during waste incineration.

THE HISTOPATHOLOGICAL STUDY ON THE RESPONSE OF THE REMAINING PULP TISSUES TO THE ADHESIVE RESIN AFTER PULPOTOMY (치수절단 후 접착레진 도포에 대한 잔존치수조직 반응에 관한 연구)

  • This study was performed to observe the histopathological response to the bonding resin directly applied on the remaining pulp tissues. 40 teeth from 3 adult dogs were pulpotomized with a sterile round bur and sharp excarvater. In the control group, $Ca(OH)_2$ powder was applied on the pulp tissue and the cavities were sealed with IRM cement. In the experimental group 1, Superbond C&B was applied on the remaining pulp and the cavities conditioned with 10-3 solution were filled with the mixture of the MMA liquid, PMMA powder and Catalyst. Multi-purpose adhesive was used on the remaining pulp tissue in the experimental group 2 and Z-100 was filled in the cavities. In the experimental group 3, Clearfil photobond applied and directly photo-cured on the pulp tissue, then the cavities were treated with CA agent (10% citric acid and 20% $CaCl_2$ aqueous solution) for 20 seconds, washed and applied with Clearfil photobond then filled with Protect liner. The experimental animals were sacrified at the 1st, 2nd, and 4th week. The specimens were routinely processed and stained with H-E for light microscopic observation. The results were as followed : 1. In the experimental group 1, the number and characteristics of the dentin bridge formation case was similar to those in the control group and less cases were observed in the experimental group 2 and 3 than experimental group 3. The inflammatory response in experimental group 1 was less than that in the control group at 1st week but there had been little difference at between 2nd and 4th week. 2. The number of the dentin bridge in experimental group 2 was less than that in control group and experimental group 1. The inflammatory response of the experimental group 1 was similar to that of experimental group 1 but less than that of the control group. A number of bleeding and vascular congestion were observed. The least inflammatory response was seen in the experimental group 2 among all groups. 3. In the experimental group 3, one case of the dentin bridge formation was observed and that was the same as that in the experimental group 2 but smaller than that of the control and experimental group 1. The inflammatory response of the experimental group 3 was least at the 1st week and most at the 4th week in the all group.
