N Mineralization and Nitrification in Forest Soils : Effect of Chemical Treatment on N Adsorption by Ion Exchange Resin (산림토양내(山林土壤內) 질소(窒素)의 양료화(養料化)와 질산화(窒酸化)에 관(關)한 연구(硏究) : ion 교환수지(交換樹指)의 처리(處理) 방법(方法)에 따른 질소(窒素)의 흡수율(吸收率) 변화(變化))

  • Soil N mineralization and nitrification can be measured conveniently using mixed bed (cation and anion) exchange resin bags. However, appropriate use of these resin bags requires pretreatment to avoid colorimetric interference and standardize N ion adsorption. Three pretreatments were evaluated : control (untreated), 2 M NaCl with a distilled water rinse, and 4 M NaCl. The 4 M NaCl treatment was effective at removing background levels of $NH_4{^+}$ and $NO_3{^-}$, but adsorbed low amounts (about 40%) of inorganic N from standard solutions. Untreated resin bags adsorbed a constant, higher amount of $NO_3{^-}$ (60%), but did not remove background levels of $NH_4{^+}$. The 2 M NaCl treatment followed by a distilled water rinse performed best : it removed background $NH_4{^+}$ and adsorbed a constant amount of both $NH_4{^+}$ (70%) and $NO_3{^-}$ (60%). Because the ion exchange resin is fairly expensive, we also tested if the resin bags could be reused. Resin bags were either loaded with $NH_4{^+}$ and $NO_3{^-}$ in the laboratory or incubated in soil in the field, desorbed with the 2 M NaCl treatment, and then loaded with standard $NH_4{^+}$ and $NO_3{^-}$ solutions. Lab loaded resin bags adsorbed about 60% of inorganic N then loaded with 2.5 or $5.0mgN\;1^{-1}$ and 70% when loaded at 10 or $20mgN\;1^{-1}$, whereas reused field incubated bags showed the opposite adsorption efficiency. These results demonstrate that resin bags can give reproducible results, but care must be taken to evaluate the effect of pretreatment and potential for reuse.

  • Experimental investigations on the effect of two kinds of additives ; aqueous NaOH solution and ammonia(NH$_3$) for removal of NOx and SO$_2$ simultaneously by corona discharge were carried out. The simulated combustion flue gas was[NO(0.02[%])-SO$_2$(0.08[%])-$CO_2$-Air-$N_2$] Volume percentage of aqueous NaOH solution used was 20[%] and $N_2$flow rate was 2.5[$\ell$/min] for bubbling aqueous NaOH solution Ammonia gas(14.81[%]) balanced by argon was diluted by air. NH$_3$ molecular ratios(MR) based on [NH$_3$] and [NO+SO$_2$] were 1, 1.5 and 2.5 The vapour of aqueous NaOH solution and NH$_3$was introduced to the main simulated combustion flue gas duct through injection systems which were located at downstream of corona discharge reactor. NOx(NO+NO$_2$) removal rate by injecting the vapour of aqueous NaOH solution was much better than that by injecting NH$_3$however SO$_2$removal rate by injecting NH$_3$was much better than that by injecting the vapour of aqueous NaOH SO$_2$removal rate slightly increased with increasing applied voltage. When the vapour of aqueous NaOH solution and NH$_3$were simultaneously injection NOx and SO$_2$ removal rate were significantly increased.

  • Glasses of Na2O-Al2O3-SiO2 system were prepared and ion-exchange characteristics change of properties and bactericidal effects by Na+↔Ag+ ion exchange were studied. Parent glasses with three compositions of varying Na2O in the 20~30 wt% were ion-exchanged in the molten salt of 2 mol% AgNO3+98 mol% NaNO3 at 320~36$0^{\circ}C$ for 15~16min. Amount of ion exchange and penetration depth of Ag+ ion increased with Na2O content in the parent glass ion exchange temperature and time. After ion exchange densities and Vickers hardness of the glasses increased and the glasses showed yellow-brown color and as amount of ion exchange increased the color turned deep because partial reduction and agglomeration of Ag+ into Ag0 results in absorption of visible light. After ion exchange chemical durability of the glass to wter was enhanced compara-bly that weight loss and change of surface of the glass were not found for the leaching test in 5$0^{\circ}C$ K.I water for 240hrs. Bactericidal effect of ion exchanged glass on Staphylococcus aureus and E. coli was determined by microorganism test and bactericidal effect increased with amount of ion exchange and incubation time.

Biodegradation of Azo and Reactive Dyes with Pseudomonas strains (Pseudomonas 속의 균주를 이용한 Azo계와 Reactive계의 Dye의 생분해)

  • This study describes biodegradation of dyes which are used in textile industries. Dyes released into the environment from industrial waste water are considered to be a serious pollution problem because of the wide spread into environment with a variety of colors. The microorganisms used in this experiment were Pseudomonas species, which had been screened from aeration tank of waste water treatment. It was found that optimum concentrations for culture media were 14g of glucose, 6g of peptone, 160 mg of Na2HPO4, 200mg of KCl, 140mg of MgSO4,.7H2O,1.0g of KH2PO4, 400mg of NaCl, 200mg of CaCl2 and dye 10ppm per litre of distilled water. The high efficiency of dye degradation was obtained at pH 7-8 and $30-35^{\circ}C$. Strains screened are excellent for removal of azo and reactive dyes, which are relatively stable and difficult to degrade. Dyes of 10ppm such as mono-azo (Lot No. 180), di-azo (Lot No. 138) and reactive red(Lot No. 2) were mostly decolorlzed within 2 days and di-azo (Lot No. 151) and reactive red(Lot No. 34, No. 00166) were decolorized within 5 days in the controlled fermenter. In the case of reactive dyes, oxygen supplies showed lower biodegradability compared to anaerobic culture.

Effects of low NaNO2 and NaCl concentrations on Listeria monocytogenes growth in emulsion-type sausage

  • Objective: The objective of this study was to evaluate the effect of combinations of $NaNO_2$ and NaCl concentrations on Listeria monocytogenes (L. monocytogenes) growth in emulsion-type sausage. Methods: Emulsion-type sausages formulated with different combinations of $NaNO_2$ (0 and 10 ppm) and NaCl (1.00%, 1.25%, and 1.50%) were inoculated with a five-strain L. monocytogenes mixture, and stored at $4^{\circ}C$, $10^{\circ}C$, and $15^{\circ}C$, under aerobic or vacuum conditions. L. monocytogenes cell counts were measured at appropriate intervals, and kinetic parameters such as growth rate and lag phase duration (LPD) were calculated using the modified Gompertz model. Results: Growth rates increased (0.004 to 0.079 Log colony-forming unit [CFU]/g/h) as storage temperature increased, but LPD decreased (445.11 to 8.35 h) as storage temperature and NaCl concentration increased. The effect of combinations of NaCl and low-$NaNO_2$ on L. monocytogenes growth was not observed at $4^{\circ}C$ and $10^{\circ}C$, but it was observed at $15^{\circ}C$, regardless of atmospheric conditions. Conclusion: These results indicate that low concentrations of $NaNO_2$ and NaCl in emulsion-type sausage may not be sufficient to prevent L. monocytogenes growth, regardless of whether they are vacuum-packaged and stored at low temperatures. Therefore, additional techniques are necessary for L. monocytogenes control in the product.

  • The corrosion behavior of a rebar in concrete according to the amount of NaCl and $LiNO_2$ was observed by using Electrochemical Impedance Spectroscopy. The corrosion was accelerated in a short time by using dry/wet cycles method, which is one of the corrosion acceleration methods, and though the value of measured impedance, equivalent circuit can be introduced. It was confirmed that the passive film of a embedded rebar in concrete with NaCl was broken quickly, and when $0.6M\;LiNO_2$ was added, the velocity of ongoing corrosion was declined considerably compared to the amount of NaCl. However, when $1.2M\;LiNO_2$ was added, it was confirmed that the passive film was not broken and its performance remained, compared to the amount of NaCl.

  • Glass-ceramics composed of $5Na_2O-36CaO-10TiO_2-xP_2O_5$ could be obtained with $P_2O_5$ content as following procedure: 1) leaching out $Ca_3(PO_4)_2$ crystals between $Ca_3(PO_4)_2$ and $NaTi_2(PO_4)_3$ selectively in 1 N HCl solution for 2 days, and 2) exchanging $Na^+$ ion to $Ag^+$ ion in Ag($NO_3$) solution for 1 day. Scanning electron microscope (SEM) and X-rat diffractometer (XRD) were measured to verify the proper synthesis of glass-ceramics. And anti-bacteria test was accomplished using Staphylococus aureus bacteria. In the results of anti-bacteria test, the bacteria were died perfectly after 3 hours.

Effect of NaCl Treatment on Growth and Physiologycal Activity of Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara (NaCl 처리가 눈개승마[Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara]의 생육과 생리활성에 미치는 영향)

  • This study was conducted to investigate the growth and physiologycal activitis of Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara under the NaCl treatment conditions (0, 100, 200, 300 mM). After 30 days treatment, the growth and physiologycal activitis were investigated. In the growth of plants, the plant height, leaf width, leaf length and ion level were reduced at NaCl treatments of more than 100 mM. The total polyphenol content was decreased by NaCl in a concentration-dependent manner compared to the control group. The contents of total flavonoids did not show any difference at the concentration of 200 mM and 300 mM. However, the content of total flavonoid decreased compared with that of control. In antioxidant activity, the DPPH and ABTS radical scavenging activity and reducing power activity were decreased by NaCl concentration compared to the control. When changes in the content of NO production was monitored by ELISA, production inhibitory effect was 94.5%, 70%, 63%, 56.9% in NaCl concentration of 0, 100, 200, 300 mM, respectively. The growth, ion level, antioxidant and anti-inflammation activity of Aruncus dioicus var. kamtschaticus was reduced at NaCl treatments of more than 100 mM.

The Effects of Salt and NaNO2 on Fatty Acid Composition, Free Amino Acids, Microbial Counts and Sensory Characteristics of Dry-cured Ham Processed under Korean Environment (소금과 아질산염 처리수준이 자연숙성 건염햄의 지방산 조성, 유리아미노산, 미생물수 및 관능적 특성에 미치는 영향)

  • The study analyzed the effects of salt concentration [high salt (HS) and low salt (LS)] and sodium nitrite ($NaNO_2$), which are typically utilized in Korean processing facilities, on fatty acid composition, free amino acids, microbial counts and sensory characteristics of processed dry-cured ham. Four different treatments were considered: three hams (11.30 kg) salted with 92 g/kg salt (w/w) (HS), three hams (10.65 kg) treated with HS and 100 ppm $NaNO_2$ (HS+$NaNO_2$), three hams (11.42 kg) salted with 62 g/kg salt (w/w) (LS), and three hams (10.62 kg) treated with LS and 100 ppm $NaNO_2$ (LS+$NaNO_2$). Fatty acid composition analysis revealed significantly (p<0.05) higher saturated fatty acid and lower (p<0.05) unsaturated fatty acid in the HS+$NaNO_2$ group compared with the other groups. Glutamate, alanine and lysine free amino acids were higher than the other free amino acids. The processing conditions did not significantly affect the free amino acids of biceps femoris muscles, except for the proline content (p>0.05). In sensory evaluation, the fermentation aroma of the LS group was higher than that of the HS group. The aerobic counts consistently ranged from from $2.3{\times}10^2$ to $1.11{\times}10^4$ CFU/g. Escherichia coli including strain O157:H7, Staphylococcus aureus, and Salmonella spp. were not detected.

Optimization of Red Pigmentation and Effect of the Metabolites Produced by Monascus Strains on Microbial Inhibition and Colorization in Processed Ham (Monascus 균주의 적색색소 생산 특성과 육제품에서의 항균 및 착색 효과)

  • In this study, we tested possibility of replacing nitrite salts, which were always added during the meat product processing, with the metabolites produced by antimicrobial and red pigment producing Monascus strains. We have already shown that Monascus No. 116 strain has the highest antimicrobial activity among the strains isolated from Ang-Khak. Monascus isolate No. 229 was chosen due to its outstanding red pigment producing ability. The red pigment production by No. 229 was highest in the medium containing 8% sucrose, 2% yeast extract, 0.1% K2HPO4, 0.5% MgSO4. Optimum pH and temperature for the red pigment production were pH 6.2 and 3$0^{\circ}C$, was found in spot or Rf value 0.54 on TLC plate using ethyl acetate-acetone-water (4:4:1, v/v/v) as development solvent system. Isolate No. 116 and No. 229 were cultured in a optimal condition for the antimicrobial activity and red pigmentation. The culture concentrates were applied in situ to the production of instantly processed ham. Mixed application of 89 ppm Na-nitrite and 300 ppm of culture broth concentrate of Monascus isolate No. 116 and 500 ppm of red color produced by Monascus isolate No. 229 showed similar results with the single application of 94 ppm Na-nitrite. These results confirmed that the antimicrobial activity and red pigment of Monascus strains might be valuable to replace Na-nitrite salt in meat processing.
